| 2 | Abnormal expression of VEGF and its gene transcription status as diagnostic indicators in patients with non-small cell lung cancer显示文摘Objective Angiogenesis is known to be essential for the survival,growth,invasion,and metastasis of lung cancer cells. Vascular endothelial growth factor(VEGF) is an important factor regulating angiogenesis of non-small cell lung cancer(NSCLC); however,its pathologic features and significance are unclear. In this study,the tissue VEGF expression levels and its gene transcriptional status,as well as circulating VEGF levels,were investigated in patients with lung disease. Methods VEGF protein and m RNA expression levels in 38 lung tissue samples were analyzed by immunohistochemistry and reverse transcription-polymerase chain reaction(RT-PCR),respectively. Circulating VEGF levels were detected quantitatively by an enzyme linked immuno-sorbent assay. Results The level of VEGF expression was significantly higher in lung cancer tissue than in the corresponding paracancerous or non-cancerous tissues. The average level of VEGF-positive staining was 76% in tissue samples from NSCLC patients; the levels were 89% in tissue samples from stage III patients and 92% in stage IV patients. High VEGF expression was also evident in cases with lymph node metastasis(84%),distant metastasis(90%),and lower differentiation degree(89%). VEGF m RNA in cancerous tissues was represented predominantly by the VEGF121 and VEGF165 isoforms. Circulating VEGF levels were significantly higher in NSCLC patients [(840 ± 324) pg/m L] than in patients with benign lung diseases [(308 ± 96) pg/m L] or in healthy individuals serving as controls [(252 ± 108) pg/m L]. | 4 | Enhanced dynamic deformability and strengthening effect via twinning and microbanding in high density NiCoFeCrMoW high-entropy alloys显示文摘High density alloys with enhanced deformability and strength are urgently required in energy,military and nuclear industries,etc.In this work,we present a new kind of NiCoFeCrMoW high entropy alloys(HEAs)which possess higher densities and sound velocities than copper.We systematically investigate the phase structure,quasi-static tensile,dynamic compression and related deformation mechanism of these HEAs.It is shown that single FCC or FCC+μdual phases were formed in the HEAs depending on Mo and W content and annealing temperature.Excellent quasi-static tensile and dynamic compression properties have been achieved for these HEAs,e.g.Ni_(30)Co_(30)Fe_(21)Cr_(10)W_(9)HEA annealed at 1573 K exhibited a yield and ultimate tensile strength and elongation of~364 MPa,~866 MPa and~32%,respectively,in quasi-static test;a yield strength of~710 MPa and no fracture under the dynamic strain rate of 4100 s^(-1).Superior strain rate sensitivity(SRS)of yield strength than that of previously reported FCC HEAs have been evidenced.The dynamic stress-strain constitutive relation can be described by the modified Johnson-Cook model.As for the dynamic deformation mechanism,it is envisaged that the regulation of stacking fault energy and Peierls barrier in current HEAs resulted in occurrences of abundant nanoscale deformation twins and microbands during high strain rate compression.The synergistic microbanding and twinning effectively contributes to the enhanced dynamic deformability and strengthening effect.Besides,the interactions of dislocations with precipitates,stacking faults(SFs)with twins,and between SFs also contribute to extraordinary work-hardening capacity. | Xuli Liu Yidong Wu Yansong Wang Jinbin Chen Rui Bai Lei Gao Zhe Xu William Yi Wang Chengwen Tan Xidong Hui | 2022 | Journal of Materials Science & Technology2022,,32: | 1 |

| 6 | Soil C, N and P stocks and stoichiometry under different vegetation on the surface of the Leshan Giant Buddha显示文摘The accumulation of soil organic matter and nutrients is an important pathway in effectivelyunderstanding the mechanisms of plant settlement and rock weathering, while the characteristics ofsoil organic carbon (C), nitrogen (N) and phosphorus (P) under different vegetation remain unclear.In this study, the stocks and stoichiometry of soil organic C, N and P were determined in differentpositions and types of vegetation on the surface of the Leshan Giant Buddha. We found that the totalstocks of soil organic C, N and P were 1689.77, 134.6 and 29.48 kg, respectively, for the Buddha.The stocks of soil organic C, N and P under vascular plants were higher than those under othervegetation, with highest values observed under herb. Higher stocks per unit area (m2) of soil organicC, N and P were found on the left and right arms, shoulders, and two platforms. | 8 | Identification of potential anti-pneumonia pharmacological components of Glycyrrhizae Radix et Rhizoma after the treatment with Gan An He Ji oral liquid显示文摘Glycyrrhizae Radix et Rhizoma,a traditional Chinese medicine also known as Gan Cao(GC),is frequently included in clinical prescriptions for the treatment of pneumonia.However,the pharmacological components of GC for pneumonia treatment are rarely explored.Gan An He Ji oral liquid(GAHJ)has a simple composition and contains GC liquid extracts and paregoric,and has been used clinically for many years.Therefore,GAHJ was selected as a compound preparation for the study of GC in the treatment of pneumonia.We conducted an in vivo study of patients with pneumonia undergoing GAHJ treatments for three days.Using the intelligent mass spectrometry data-processing technologies to analyze the metabolism of GC in vivo,we obtained 168 related components of GC in humans,consisting of 24 prototype components and 144 metabolites,with 135 compounds screened in plasma and 82 in urine.After analysis of the metabolic transformation relationship and relative exposure,six components(liquiritin,liquiritigenin,glycyrrhizin,glycyrrhetinic acid,daidzin,and formononetin)were selected as potential effective components.The experimental results based on two animal pneumonia models and the inflammatory cell model showed that the mixture of these six components was effective in the treatment of pneumonia and lung injury and could effectively downregulate the level of inducible nitric oxide synthase(iNOS).Interestingly,glycyrrhetinic acid exhibited the strongest inhibition on iNOS and the highest exposure in vivo.The following molecular dynamic simulations indicated a strong bond between glycyrrhetinic acid and iNOS.Thus,the current study provides a pharmaceutical basis for GC and reveals the possible corresponding mechanisms in pneumonia treatment. | Xiaojuan Jiang Yihua Lin Yunlong Wu Caixia Yuan Xuli Lang Jiayun Chen Chunyan Zhu Xinyi Yang Yu Huang Hao Wang Caisheng Wu | 2022 | Journal of Pharmaceutical Analysis2022,12,6: | 0 |
| 9 | Analysis of Biological Activity and Immunogenicity of Extracellular Products from Streptococcus iniae显示文摘[Objectives] This study aimed to analyze the biological activity and immunogenicity of extracellular products from Streptococcus iniae. [Methods] S.iniae was incubated with brain heart infusion agar medium( BHIA + 4% calf serum) for 60 h. The bacterial liquid was rinsed with PBS,centrifuged,and filtered through microporous filtering film to collect extracellular products( ECPs). [Results]Extracellular proteinase( ECPase) of S. iniae exhibited amylase,protease,lecitinase,gelatinase,lipase activities and hemolytic activity but had no urease activity. EDTA,DTT and PMSF could reduce ECPase activity to 72. 4%,77. 6%and 72. 4%,respectively. Cu^(2+),Ca^(2+),K+and Mg^(2+)exhibited an inhibitory effect on ECPase activity,whereas Fe^(3+),Co^(2+)and Mn^(2+)could activate ECPase activity. ECPs had good heat stability and exhibited relatively high activities under alkaline conditions. The optimal temperature for ECPs was 55 ℃. Two-dimensional electrophoresis was performed to analyze the main protein of ECPs. The results indicated that there are 12 main bands of ECPs,and the molecular weights mainly ranged between 28-68 kD a. About 120 protein spots were detected,and the molecular weights mainly ranged between 26-95 kD a. The mouse anti-S. iniae was used for Western-blot analysis of ECPs,and the results showed that there were four proteins,with molecular weights of 26,37,95,and 97 kD a,respectively. The pathogenicity assay indicated that ECPs of S. iniae were highly pathogenic to tilapia. The mortality rate of tilapia was enhanced as the concentration of ECPs increased. | 10 | Optimized One-Step Preparation of a Bioactive Natural Product,Guaiazulene-2,9-dione显示文摘We previously isolated a natural product, namely guaiazulene-2,9-dione showing strong antibacterial activity against Vibrio anguillarum, from a gorgonian Muriceides collaris collected in South China Sea. In this experiment, guaiazulene-2,9-dione was quantitatively synthesized with an optimized one-step bromine oxidation method using guaiazulene as the raw material. The key reaction condition including reaction time and temperature, drop rate of bromine, concentration of aqueous THF solution, respective molar ratio of guaiazulene to bromine and acetic acid, and concentration of guaiazulene in aqueous THF solution, were investigated individually at five levels each for optimization. Combined with the verification test to show the absolute yield of each optimization step, the final optimal condition was determined as: when a solution of 0.025 mmol m L-1 guaiazulene in 80% aqueous THF was treated with four volumes of bromine at a drop rate of 0.1 m L min-1 and four volumes of acetic acid at-5℃ for three hours, the yield of guaiazulene-2,9-dione was 23.72%.
