1Effect on platelet aggregation activity: extracts from 31 Traditional Chinese Medicines with the property of activating blood and resolving stasis显示文摘OBJECTIVE: To evaluate the anti-platelet aggregation effects of extracts from 31 Traditional Chinese Medicines(TCM) with the property of activating blood and resolving stasis in terms of TCM theory.METHODS: The 31 TCMs extracts were prepared using water, 90% ethanol and ethyl acetate., and the effects on anti-platelet aggregation were tested on a platelet aggregation analyzer in vitro with adenosine 5'-diphosphate, bovine thrombin and arachi-donic acid(AA) as aggregation inducers, respectively. Aspirin was the positive control.RESULTS: Lots of the tested TCMs had inhibitory effects with concentration-dependent manner on platelet aggregations induced by various agonists.Especially, some of the TCMs such as Chuanxiong(Rhizoma Chuanxiong), Yanhusuo(Rhizoma Corydalis Yanhusuo) and Danshen(Radix Salviae Miltiorrhizae) showed good anti-platelet aggregation effect similar or higher than that in positive control group.CONCLUSION: The study provided scientific references that several TCMs such as Chuanxiong(Rhizoma Chuanxiong), Yanhusuo(Rhizoma Corydalis Yanhusuo) and Danshen(Radix Salviae Miltiorrhizae),possess the property of anti-platelet aggregation.Chen Cen Wang Fengqin Xiao Wen Xia Zhining Hu Guang Wan Jianbo Yang Fengqing 2017Journal of Traditional Chinese Medicine2017,37,1:7
2A high-quality genome provides insights into the new taxonomic status and genomic characteristics of Cladopus chinensis(Podostemaceae)显示文摘The Podostemaceae are ecologically and morphologically unusual aquatic angiosperms that survive only in rivers with pristine hydrology and high water quality and are at a relatively high risk of extinction.The taxonomic status of Podostemaceae has always been controversial.Here,we report the first high-quality genome assembly for Cladopus chinensis of Podostemaceae,obtained by incorporating Hi-C,Illumina and PacBio sequencing.We generated an 827.92 Mb genome with a contig N50 of 1.42 Mb and 27,370 annotated protein-coding genes.The assembled genome size was close to the estimated size,and 659.42 Mb of the assembly was assigned to 29 superscaffolds(scaffold N5021.22 Mb).A total of 59.20%repetitive sequences were identified,among which long terminal repeats(LTRs)were the most abundant class(28.97%of the genome).Genome evolution analysis suggested that the divergence time of Cladopus chinensis(106 Mya)was earlier than that of Malpighiales(82 Mya)and that this taxon diverged into an independent branch of Podestemales.A recent whole-genome duplication(WGD)event occurred 4.43 million years ago.Comparative genomic analysis revealed that the expansion and contraction of oxidative phosphorylation,photosynthesis and isoflavonoid metabolism genes in Cladopus chinensis are probably related to the genomic characteristics of this growing submerged species.Transcriptome analysis revealed that upregulated genes in the shoot group compared to the root group were enriched in the NAC gene family and transcription factors associated with shoot development and defense responses,including WUSCHEL(WUS),ASYMMETRIC LEAVES(ASL),SHOOT MERISTEMLESS(STM),NAC2,NAC8,NAC29,NAC47,NAC73,NAC83 and NAC102.These findings provide new insights into the genomic diversity of unusual aquatic angiosperms and serve as a valuable reference for the taxonomic status and unusual shoot apical meristem of Podostemaceae.Ting Xue Xuehai Zheng Duo Chen Limin Liang Nan Chen Zhen Huang Wenfang Fan Jiannan Chen Wan Cen Shuai Chen Jinmao Zhu Binghua Chen Xingtan Zhang Youqiang Chen 2020Horticulture Research2020,7,1:1
3mTOR regulates cocaine-induced behavioural sensitization through the SynDIG1–GluA2 interaction in the nucleus accumbens显示文摘Behavioral sensitization is a progressive increase in locomotor or stereotypic behaviours in response to drugs. It is believed to contribute to the reinforcing properties of drugs and to play an important role in relapse after cessation of drug abuse. However, the mechanism underlying this behaviour remains poorly understood. In this study, we showed that mTOR signaling was activated during the expression of behavioral sensitization to cocaine and that intraperitoneal or intra-nucleus accumbens (NAc) treatment with rapamycin, a specific mTOR inhibitor, attenuated cocaine-induced behavioural sensitization. Cocaine significantly modified brain lipid profiles in the NAc of cocaine-sensitized mice and markedly elevated the levels of phosphatidylinositol-4-monophosphates (PIPs), including PIP, PIP2, and PIP3. The behavioural effect of cocaine was attenuated by intra-NAc administration of LY294002, an AKT-specific inhibitor, suggesting that PIPs may contribute to mTOR activation in response to cocaine. An RNA-sequencing analysis of the downstream effectors of mTOR signalling revealed that cocaine significantly decreased the expression of SynDIG1, a known substrate of mTOR signalling, and decreased the surface expression of GluA2. In contrast, AAV-mediated SynDIG1 overexpression in NAc attenuated intracellular GluA2 internalization by promoting the SynDIG1–GluA2 interaction, thus maintaining GluA2 surface expression and repressing cocaine-induced behaviours. 5Biosynthesis of rumbrins and inspiration for discovery of HIV inhibitors显示文摘Investigation on how nature produces natural compounds with chemical and biological diversity at the genetic level offers inspiration for the discovery of new natural products and even their biological targets.The polyketide rumbrin(1)is a lipid peroxide production and calcium accumulation inhibitor,which contains a chlorinated pyrrole moiety that is a rare chemical feature in fungal natural products.Here,we identify the biosynthetic gene cluster(BGC)rum of 1 and its isomer 12E-rumbrin(2)from Auxarthron umbrinum DSM3193,and elucidate their biosynthetic pathway based on heterolo-gous expression,chemical complementation,and isotopic labeling.We show that rumbrins are assembled by a highly reducing polyketide synthase(HRPKS)that uniquely incorporates a proline-derived pyrrolyl-CoA starer unit,and followed by methylation and chlorination.Sequent precursor directed biosynthesis was able to yield a group of rumbrin analogues.Remarkably,inspired by the presence of a human immunodeficiency virus(HIV)-Nef associated gene in the rum cluster,we predicted and pharmacologically demonstrated rumbrins as potent inhibitors of HIV at the nanomolar level.This work enriches the recognition of unconventional starter units of fungal PKSs and provides a new strategy for genome mining-guided drug discovery.Beifen Zhong Jun Wan Changhui Shang Jiajia Wen Yujia Wang Jian Bai Shan Cen Youcai Hu 2022Acta Pharmaceutica Sinica B2022,12,11:1
6Study of the decorporation efficacy and toxicity of tetradentate 3-hydroxy- 2-pyridinone ligands at the cellular level显示文摘Objective:A series of experiments were carried out to study the comparative decorporation efficacy and toxicity of three tetradentate hydroxypyridinone ligands(3LI-1-Cm-3,2-HOPO,5LIO-1-Cm-3,2-HOPO,and 7LIO-1-Cm-3,2-HOPO)with the variation of the linker length and the substituents.Methods:Three ligands were obtained from a 4-step synthesis via the amidation of HOPO unit and different backbones.Potentiometric titrations were carried out to evaluate the formation constants of their corresponding uranyl complexes.CCK-8 and colony formation assays were used to compare the cytotoxicity of the three ligands and uranyl using renal proximal tubular epithelial(NRK-52E)cells.In vitro uranium removal assays were performed to assess decorporation efficiencies of those ligands by co-incubating uranium and chelators with NRK-52E cells for 48 h.Results:Among the three ligands,5LIO-1-Cm-3,2-HOPO exhibits the highest uranyl affinity(logβ110?18.6(7))in comparison to the other two ligands(logβ110?14.9(2)for 3LI-1-Cm-3,2-HOPO and logβ110?16.7(1)for 7LIO-1-Cm-3,2-HOPO).The results of CCK-8 tests and colony formation assays further elucidate that 5LIO-1-Cm-3,2-HOPO and 7LIO-1-Cm-3,2-HOPO both show a similar level of cytotoxicity compared with ZnNa3-DTPA at relatively low ligand concentrations.In contrast,in the 3LI-1-Cm-3,2-HOPO treated group,the cell viability decreased markedly with the increase of the ligand concentrations,and the colony formation capability of NRK-52E cells was inhibited.Furthermore,the comparative decorporation efficacy of the three ligands was obtained from in vitro uranium removal assays,suggesting that these ligands could significantly inhibit the cellular uptake and prompt the cellular release of uranium from NRK-52E cells by the level of 81.9%,91.8%,and 87.1%,respectively.Conclusion:Our current results demonstrate that the linker/backbone species would significantly affect the decorporation efficacy and toxicity of the tetradentate 3-hydroxy-2-pyridinone ligands,which is informative for the future design and modification of novel decorporation ligands.Xiaomei Wang Cen Shi Manni Gao Ying Xu Yang Jiao Jianmei Wan Jianping Cao Zhifang Chai Juan Diwu 2020Radiation Medicine and Protection2020,1,4:1
7Electrochemical analysis of microRNAs with hybridization chain reaction-based triple signal amplification显示文摘Selective and sensitive detection of trace microRNA is important for early diagnosis of diseases due to its expression level related to diseases.Herein,a triple signal amplification strategy is developed for trace microRNA-21 (miRNA-21) detection by combining with target-triggered cyclic strand displacement reaction (TCSDR),hybridization chain reaction (HCR) and enzyme catalytic amplification.Four DNA hairpins(H1,H2,H3,H4) are employed to form an ultralong double-strand DNA (dsDNA) structure,which is initiated by target miRNA-21.As H3 and H4 are labeled with horseradish peroxidase (HRP),numerous HRPs are loaded on the long dsDNA,producing significantly enhanced electrocatalytic signals in the hydrogen peroxide (H_(2)O_(2)) and 3,3,5,5-tetramethylbenzidine (TMB) reaction strategy.Compared with single signal amplification,the triple signal amplification strategy shows higher electrochemical response,wider dynamic range and lower detection limit for miRNA-21 detection with excellent selectivity,reproducibility and stability.Taking advantage of the triple signal amplification strategy,the proposed electrochemical biosensor can detect miRNA-21 in 10 He La cell lysates,suggesting that it is a promising method for fruitful assay in clinical diagnosis.Jianfeng Ma Lingbo Gong Yingying Cen Lin Feng Yan Su Xingfen Liu Jie Chao Ying Wan Shao Su Lianhui Wang 2023Chinese Chemical Letters2023,34,7:0
8Research on Various Factors Affecting the Dedifferentiation of Different Floral Organs of Jinxiu Wild Chive显示文摘[Objective]This study aimed to investigate various factors affecting the dedifferentiation of different floral organs of Jinxiu wild chive.[Method]Floral organs of Jinxiu wild chive were cultured in vitro,to analyze the effects of basic medium,exogenous hormone type and combination,and blossoming status of buds on callus induction.[Result]MS medium was the most suitable medium for callus induction from buds.NAA was conducive to improving callus formation rate compared with other auxins.Non-blossomed buds exhibited the highest rate of callus induction.Under the same culture conditions,the rate of callus induction varied extremely significantly among various floral organs;to be specific,the rate of callus induction from floral disc reached the highest,followed successively by bud and pedicel,but no callus was formed in floral axis group.[Conclusion]This study laid the foundation for further establishment of tissue culture and rapid propagation system of Jinxiu wild chive and other wild chives and provided technical support for the preservation,development and utilization of wild chive germplasm resources.Xi WANG Lifeng LONG Pengxiao WEI Xiufen CEN Zhenglin WAN Minghua LONG 2015Agricultural Biotechnology2015,4,2:0
9Simvastatin Blocks Reinstatement of Cocaine-induced Conditioned Place Preference in Male Mice with Brain Lipidome Remodeling显示文摘Drug-associated reward memories are conducive to intense craving and often trigger relapse.Simvastatin has been shown to regulate lipids that are involved in memory formation but its influence on other cognitive processes is elusive.Here,we used a mass spectrometry-based lipidomic method to evaluate the impact of simvastatin on the mouse brain in a cocaine-induced reinstatement paradigm.We found that simvastatin blocked the reinstatement of cocaine-induced conditioned place preference(CPP)without affecting CPP acquisition.Specifically,only simvastatin administered during extinction prevented cocaine-primed reinstatement.Global lipidome analysis showed that the nucleus accumbens was the region with the greatest degree of change caused by simvastatin.The metabolism of fatty-acids,phospholipids,and triacylglycerol was profoundly affected.Simvastatin reversed most of the effects on phospholipids induced by cocaine.The correlation matrix showed that cocaine and simvastatin significantly reshaped the lipid metabolic pathways in specific brain regions.Furthermore,simvastatin almost reversed all changes in the fatty acyl profile and unsaturation caused by cocaine.In summary,pre-extinction treatment with simvastatin facilitates cocaine extinction and prevents cocaine relapse with brain lipidome remodeling.Wei Xu Yuman He Jiamei Zhang Hongchun Li Xuemei Wan Menglu Li Yonghai Wang Rui Xu Haoluo Zhang Yanping Dai Haxiaoyu Liu Linhong Jiang Ying Zhao Xiaobo Cen 2021Neuroscience Bulletin2021,37,12:0
10Defining stratigraphic oil and gas plays by modifying structural plays:A case study from the Xihu Sag,east China Sea Shelf Basin显示文摘The study focuses on an area of relatively mature oil and gas exploration located in the western slope of the Xihu Sag,East China Sea Shelf Basin.Three stratigraphic oil and gas plays are introduced to replenish the shrinking prospect inventory in the area.The reservoirs of these plays are traditional pay intervals of the Eocene Pinghu Formation and Baoshi Formation composed of tide-dominated delta front,tidal flat and delta front sand bar facies.By investigating conventional seismic data,seismic inversion volumes,and well log data,multiple tidal-influenced delta front channels,beach sand packages have been mapped out in different reservoir sand intervals of the Pinghu Formation and Baoshi Formation.We recognize four types of stratigraphic terminations:channel flank pinch-outs,axial pinch-outs of sand-fills,channel mouth sand bar pinch-outs,and onlap pinch-outs for the Baoshi Formation.Both the depositional facies and relatively low sand-shale ratio of the reservoir formations favor the occurrence of stratigraphic terminations.The closure mechanism of all these plays are similar to the traditional structure plays in the area:fault blocks or fault-bounded rollover anticlines,except that one or more closing faults are replaced by these stratigraphic terminations.Several small-sized closures should exist but the relatively large ones are centered around a major intra-slope rise,raising the exploration potential in this maturely explored area.The new exploration drilling campaign has so far tested the play of delta front channel facies,with three out of four wells commercially successful.These play concepts open up a novel approach to identify new traps in the study area and many other rift basins with similar settings in eastern China.Feng Ding Chunyu Xie Xinghai Zhou Cen Jiang Kun Li Lifeng Wan Pei Zhang Huawei Niu 2021Energy Geoscience2021,2,1:0
11Mycobacteriaceae Phenome Atlas(MPA):A Standardized Atlas for the Mycobacteriaceae Phenome Based on Heterogeneous Sources显示文摘The bacterial family Mycobacteriaceae includes pathogenic and nonpathogenic bacteria,and systematic research on their genome and phenome can give comprehensive perspectives for exploring their disease mechanism.In this study,the pheno-types of Mycobacteriaceae were inferred from available phenomic data,and 82 microbial phenotypic traits were recruited as data elements of the microbial phenome.This Mycobacteriaceae phenome contains five categories and 20 subcategories of polyphasic phenotypes,and three categories and eight subcategories of functional phenotypes,all of which are complemen-tary to the existing data standards of microbial phenotypes.The phenomic data of Mycobacteriaceae strains were compiled by literature mining,third-party database integration,and bioinformatics annotation.The phenotypes were searchable and comparable from the website of the Mycobacteriaceae Phenome Atlas(MPA,http://gffzz9fa96046bf0d4f3esovcwuonu6vuo6c0v.ffgz.tsg.suse.edu.cn no.org/mpa/).A topological data analysis of MPA revealed the co-evolution between Mycobacterium tuberculosis and virulence factors,and uncovered potential pathogenicity-associated phenotypes.Two hundred and sixty potential pathogen-enriched pathways were found by Fisher's exact test.The application of MPA may provide novel insights into the pathogenicity mechanism and antimicrobial targets of Mycobacteriaceae.Wan Liu Hui Cen Zhile Wu Haokui Zhou Shuo Chen Xilan Yang Guoping Zhao Guoqing Zhang 2023Phenomics2023,3,5:0
12Stability evaluation of the PROSPECT model for leaf chlorophyll content retrieval显示文摘The radiative transfer model,PROSPECT,has been widely applied for retrieving leaf biochemical traits.However,little work has been conducted to evaluate the stability of the PROSPECT model with consideration of multiple factors(i.e.,spectral resolution,signal-to-noise ratio,plant growth stages,and treatments).This study aims to investigate the stability of the PROSPECT model for retrieving leaf chlorophyll(Chl)content(Cab).Leaf hemispherical reflectance and transmittance of oilseed rape were acquired at different spectral resolutions,noise levels,growth stages,and nitrogen treatments.The Chl content was also measured destructively by using a microplate spectrophotometer.The performance of the PROSPECT model was compared with a commonly used random forest(RF)model.The results showed that the prediction accuracy of PROSPECT and RF models for Cab did not produce significant differences under varied spectral resolutions ranging from 1 to 20 nm.The ranges of the relative root mean square errors(rRMSE)of the PROSPECT and RF models were 12%-13%and 11.70%-12.86%,respectively.However,the performance of both models for leaf Chl retrieval was strongly influenced by the noise level with the rRMSE of 13-15.37%and 12.04%-15.80%for PROSPECT and RF,respectively.For different growth stages,the PROSPECT model had similar prediction accuracies(rRMSE=9.26%-12.41%)to the RF model(rRMSE=9.17%-12.70%).Furthermore,the superiority of the PROSPECT model(rRMSE=10.10%-12.82%)over the RF model(rRMSE=11.81%-15.47%)was prominently observed when tested with plants growth at different nitrogen treatment levels.The results demonstrated that the PROSPECT model has a more stable performance than the RF model for all datasets in this study.Li Zhai Liang Wan Dawei Sun Alwaseela Abdalla Yueming Zhu Xiaoran Li Yong He Haiyan Cen 2021International Journal of Agricultural and Biological Engineering2021,14,5:0
