1Pharmacokinetics and enterohepatic circulation of jervine,an antitumor steroidal alkaloid from Veratrum nigrum in rats显示文摘Jervine,a novel steroidal alkaloid from Veratrum nigrum L.,exhibits both antitumor effect and potential toxicity.The aim of study was to characterize the pharmacokinetic behaviors and enterohepatic circulation of jervine in rats.A rapid and simple ultra-high performance liquid chromatography-tandem mass spectrometric method was developed and validated for quantification of jervine and alpinetin (internal standard) in rat plasma.After extraction from rat plasma by a simple protein-precipitation method,the analyte was separated on a C18 column (2.1 mm×50 mm,1.7μm) using water with 0.1%formic acid and acetonitrile as the mobile phase delivered at a flow rate of 0.4mL/min.Jervine and alpinetin were determined in the positive mode with multiple reaction monitoring (MRM) of the ion transitions at m/z426.3→108.8 and m/z 271.0 166.9,respectively.Molecular docking method was used to investigate the binding of jervine to p-glycoprotein and dehydroepiandrosterone sulfotransferase.The method was well validated within acceptance limits including specificity,matrix effect,recovery,precision,accuracy,and stability,and was successfully applied to the pharmacokinetic study of jervine after oral and intravenous administration to rats.Jervine presented a small volume of distribution,fast absorption,high oral bioavailability,and enterohepatic circulation.The enterohepatic circulation was first observed in veratrum alkaloids,and was further investigated by molecular docking studies,which was related to the binding of jervine to p-glycoprotein and dehydroepiandrosterone sulfotransferase.The pharmacokinetic properties and enterohepatic circulation of jervine in rats provided a significant basis for the drug-drug interaction and toxicity study in the future.Bingjing Zheng Caihong Wang Wenwen Song Xiaoxia Ye Zheng Xiang 2019Journal of Pharmaceutical Analysis2019,9,5:4
2Graph-ranking collective Chinese entity linking algorithm显示文摘Entity linking(EL)systems aim to link entity mentions in the document to their corresponding entity records in a reference knowledge base.Existing EL approaches usually ignore the semantic correlation between the mentions in the text,and are limited to the scale of the local knowledge base.In this paper,we propose a novel graphranking collective Chinese entity linking(GRCCEL)algorithm,which can take advantage of both the structured relationship between entities in the local knowledge base and the additional background information offered by external knowledge sources.By improved weighted word2vec textual similarity and improved PageRank algorithm,more semantic information and structural information can be captured in the document.With an incremental evidence mining process,more powerful discrimination capability for similar entities can be obtained.We evaluate the performance of our algorithm on some open domain corpus.Experimental results show the effectiveness of our method in Chinese entity linking task and demonstrate the superiority of our method over state-of-the-art methods.Tao XIE Bin WU Bingjing JIA Bai WANG 2020Frontiers of Computer Science2020,14,2:3
3Image-based numerical study of three-dimensional meso-structure effects on damage and failure of heterogeneous coal-rock under dynamic impact loads显示文摘This paper proposes a numerical three-dimensional(3D)mesoscopic approach based on the discrete element method combined with X-ray computed tomography(XCT)images to characterize the dynamic impact behavior of heterogeneous coal-rock(HCR).The dynamic impact loading in three directions was modelled to investigate the effects of the 3D meso-structure on the failure patterns and fracture mechanism,with different impact velocities.The XCT image-based discrete element model of HCR was calibrated through appropriate standard uniaxial compression tests.Numerical simulations were carried out to investigate how the breakage behaviors are affected by different loading directions with different impact velocities.The loading direction,input energy,and spatial distribution of the mineral phase had a remarkable influence on the failure patterns and load-carrying capacities.The shape of the gangue phase and the approximate location of the gangue interfaces are key parameters to consider when investigating the failure patterns and fracture mechanism of heterogeneous rock materials.The damage and fracture tended to propagate from the surfaces to the HCR interior.The gangue phase area contacting the loading wall,growth direction of the strong gangue interfaces,and loading directions greatly influenced the failure patterns of the heterogeneous rock materials.Kehong Zheng Bingjing Qiu Zhenyu Wang Xuefeng Li Jianping Li Kuidong Gao 2020Particuology2020,18,4:1
4RNA-binding protein ZCCHC4 promotes human cancer chemoresistance by disrupting DNA-damage-induced apoptosis显示文摘RNA-binding proteins(RBPs)play important roles in cancer development and treatment.However,the tumor-promoting RBPs and their partners,which may potentially serve as the cancer therapeutic targets,need to be further identified.Here,we report that zinc finger CCHC domain-containing protein 4(ZCCHC4)is of aberrantly high expression in multiple human cancer tissues and is associated with poor prognosis and chemoresistance in patients of hepatocellular carcinoma(HCC),pancreatic cancer and colon cancer.ZCCHC4 promotes chemoresistance of HCC cells to DNA-damage agent(DDA)both in vitro and in vivo.HCC cell deficiency of ZCCHC4 reduces tumor growth in vivo and intratumoral interference of ZCCHC4 expression obviously enhances the DDAinduced antitumor effect.Mechanistically,ZCCHC4 inhibits DNA-damage-induced apoptosis in HCC cells by interacting with a new long noncoding RNA(lncRNA)AL133467.2 to hamper its pro-apoptotic function.Also,ZCCHC4 blocks the interaction between AL133467.2 andγH2AX upon DDA treatment to inhibit apoptotic signaling and promote chemoresistance to DDAs.Knockout of ZCCHC4 promotes AL133467.2 andγH2AX interaction for enhancing chemosensitivity in HCC cells.Together,our study identifies ZCCHC4 as a new predictor of cancer poor prognosis and a potential target for improving chemotherapy effects,providing mechanistic insights to the roles of RBPs and their partners in cancer progression and chemoresistance.Ha Zhu Kun Chen Yali Chen Juan Liu Xiaomin Zhang Yumei Zhou Qiuyan Liu Bingjing Wang Taoyong Chen Xuetao Cao 2022Signal Transduction and Targeted Therapy2022,7,8:1

6The Impact of Climatic and Environmental Factors on n-Alkanes Indices in Southwestern Tibetan Plateau显示文摘n-Alkanes are widely used in paleoenvironmental reconstructions.However,our understanding of changes in the distribution of n-alkanes with climatic and environmental factors remains unclear in arid/semi-arid regions.We sampled 26 surface sediments from three climatic zones across the southwestern Tibetan Plateau to evaluate the sensitivity of chain length distributions of n-alkanes to climatic and environmental parameters.Our observations demonstrate that average chain length(ACL),proportion of aquatic macrophyte(Paq),carbon preference index(CPI)and ratio of the contents of nC_(27)and nC_(31)(nC_(27)/nC_(31))are all sensitive to hydroclimatic conditions.In contrast to commonly-adopted assumptions,the correlations between these indices and hydrological parameters are not always good,which indicates that the interpretation of n-alkane indices is special on the southwestern Tibetan Plateau.These might be related to the vegetation characteristics and seasonality of biological activity,and need to be considered in paleoclimatic reconstruction.The impact of seasonal precipitation on n-alkanes indices was also evaluated.LING Yuan ZHENG Mianping WANG Shuxian SUN Qing XIE Bingjing ZHANG Chengjun 2021Acta Geologica Sinica(English Edition)2021,95,2:1

12Preparation and Application of Biomass-based Sprayable Hydrogels显示文摘Hydrogels have three-dimensional network structures that have been widely applied owing to their high water content,excellent biocompatibility,and physicochemical properties.Compared with conventional hydrogels,sprayable hydrogels exhibit excellent temporal and spatial controllability.Biomass materials offer easy accessibility,biocompatibility,biodegradability,and other physicochemical properties that are extensively used in the formation of sprayable hydrogels.In situ formed biomass-based sprayable hydrogels are realized by chemical or physical crosslinking.Rapid spray filming,in situ drug delivery,high permeability,and flexible portability enable biomass-based sprayable hydrogels to show great potential for topical drug delivery,wound healing,and other applications.This review describes in detail the status of research on the preparation and application of biomassbased sprayable hydrogels and suggests prospects for their future development.Yujiao Tan Bingjing Cai Xiaoyun Li Xiaoying Wang 2023Paper And Biomaterials2023,8,2:0
