1醒脑静注射液对急性脑梗死血瘀证患者外周血单个核细胞T淋巴细胞的影响显示文摘目的:探讨醒脑静注射液对急性脑梗死(ACI)血瘀证患者外周血单个核细胞(PBMC)T淋巴细胞的影响。方法:70例患者随机分为治疗组以及对照组各35例。对照组用常规治疗,治疗组在常规治疗基础上加醒脑静注射液。疗程均为14 d,观察两组患者治疗前后中医疗效和NIHSS评分,检测治疗前及治疗后Th1、Th2百分率和Th1/Th2比值。结果:治疗组和对照组治疗前Th1、Th2百分率、Th1/Th2和NIHSS评分比较差异无统计学意义(P>0.05);治疗3、7、14 d后两组患者NIHSS评分与治疗前比较,均有统计学差异(P<0.05)。治疗组患者Th1、Th1/Th2、NIHSS评分在治疗后3、14 d均明显低于对照组(P<0.05),Th2以及中医疗效总有效率明显高于对照组(P<0.05或0.01)。结论:醒脑静注射液明显调控急性脑梗死血瘀证患者Th1/Th2平衡,对神经功能缺损具有较好的改善作用。卢志刚 刘芸 卢青 WANG Chenming 2016中华中医药学刊2016,34,12:4
2A multi-axis robot-based bioprinting system supporting natural cell function preservation and cardiac tissue fabrication显示文摘Despite the recent advances in artificial tissue and organ engineering,how to generate large size viable and functional complex organs still remains as a grand challenge for regenerative medicine.Three-dimensional bioprinting has demonstrated its advantages as one of the major methods in fabricating simple tissues,yet it still faces difficulties to generate vasculatures and preserve cell functions in complex organ production.Here,we overcome the limitations of conventional bioprinting systems by converting a six degree-of-freedom robotic arm into a bioprinter,therefore enables cell printing on 3D complex-shaped vascular scaffolds from all directions.We also developed an oil bath-based cell printing method to better preserve cell natural functions after printing.Together with a self-designed bioreactor and a repeated print-and-culture strategy,our bioprinting system is capable to generate vascularized,contractible,and long-term survived cardiac tissues.Such bioprinting strategy mimics the in vivo organ development process and presents a promising solution for in vitro fabrication of complex organs.Zeyu Zhang Chenming Wu Chengkai Dai Qingqing Shi Guoxin Fang Dongfang Xie Xiangjie Zhao Yong-Jin Liu Charlie CLWang Xiu-Jie Wang 2022Bioactive Materials2022,7,12:4
3Application of resist-profile-aware source optimization in 28 nm full chip optical proximity correction显示文摘As technology node shrinks, aggressive design rules for contact and other back end of line(BEOL)layers continue to drive the need for more effective full chip patterning optimization. Resist top loss is one of the major challenges for 28 nm and below technology nodes, which can lead to post-etch hotspots that are difficult to predict and eventually degrade the process window significantly. 9Relevant Region sampling strategy with adaptive heuristic for asymptotically optimal path planning显示文摘Sampling-based planning algorithm is a powerful tool for solving planning problems in highdimensional state spaces.In this article,we present a novel approach to sampling in the most promising regions,which significantly reduces planning time-consumption.The RRT#algorithm defines the Relevant Region based on the cost-to-come provided by the optimal forward-searching tree.However,it uses the cumulative cost of a direct connection between the current state and the goal state as the cost-to-go.To improve the path planning efficiency,we propose a batch sampling method that samples in a refined Relevant Region with a direct sampling strategy,which is defined according to the optimal cost-to-come and the adaptive cost-to-go,taking advantage of various sources of heuristic information.The proposed sampling approach allows the algorithm to build the search tree in the direction of the most promising area,resulting in a superior initial solution quality and reducing the overall computation time compared to related work.To validate the effectiveness of our method,we conducted several simulations in both SE(2)and SE(3)state spaces.And the simulation results demonstrate the superiorities of proposed algorithm.Chenming Li Fei Meng Han Ma Jiankun Wang Max Q-H.Meng 2023Biomimetic Intelligence & Robotics2023,3,3:1

12Josephin domain containing 2(JOSD2)promotes lung cancer by inhibiting LKB1(Liver kinase B1)activity显示文摘Non-small cell lung cancer(NSCLC)ranks as one of the leading causes of cancer-related deaths worldwide.Despite the prominence and effectiveness of kinase-target therapies in NSCLC treatment,these drugs are suitable for and beneficial to a mere~30%of NSCLC patients.Consequently,the need for novel strategies addressing NSCLC remains pressing.Deubiquitinases(DUBs),a group of diverse enzymes with well-defined catalytic sites that are frequently overactivated in cancers and associated with tumorigenesis and regarded as promising therapeutic targets.Nevertheless,the mechanisms by which DUBs promote NSCLC remain poorly understood.Through a global analysis of the 97 DUBs’contribution to NSCLC survival possibilities using The Cancer Genome Atlas(TCGA)database,we found that high expression of Josephin Domain-containing protein 2(JOSD2)predicted the poor prognosis of patients.Depletion of JOSD2 significantly impeded NSCLC growth in both cell/patient-derived xenografts in vivo.Mechanically,we found that JOSD2 restricts the kinase activity of LKB1,an important tumor suppressor generally inactivated in NSCLC,by removing K6-linked polyubiquitination,an action vital for maintaining the integrity of the LKB1-STRAD-MO25 complex.Notably,we identified the first small-molecule inhibitor of JOSD2,and observed that its pharmacological inhibition significantly arrested NSCLC proliferation in vitro/in vivo.Our findings highlight the vital role of JOSD2 in hindering LKB1 activity,underscoring the therapeutic potential of targeting JOSD2 in NSCLC,especially in those with inactivated LKB1,and presenting its inhibitors as a promising strategy for NSCLC treatment.Tao Yuan Chenming Zeng Jiawei Liu Chenxi Zhao Fujing Ge Yuekang Li Meijia Qian Jiamin Du Weihua Wang Yonghao Li Yue Liu Xiaoyang Dai Jianya Zhou Xueqin Chen Shenglin Ma Hong Zhu Qiaojun He Bo Yang 2024Signal Transduction and Targeted Therapy2024,9,2:0
13Biallelic variants in RBM42 cause a multisystem disorder with neurological,facial,cardiac,and musculoskeletal involvement显示文摘Here,we report a previously unrecognized syndromic neurodevelopmental disorder associated with biallelic loss-of-function variants in the RBM42 gene.The patient is a 2-year-old female with severe central nervous system(CNs)abnormalities,hypotonia,hearing loss,congenital heart defects,and dysmorphic facial features.Familial whole-exome sequencing(WEs)reveals that the patient has two compound heterozygous variants,c.304C>T(p.R102*)and c.1312G>A(p.A438T),in the RBM42 gene which encodes an integral component of splicing complex in the RNA-binding motif protein family.The p.A438T variant is in the RRM domain which impairs RBM42 pro-tein stability in vivo.Additionally,p.A438T disrupts the interaction of RBM42 with hnRNP K,which is the causa-tive gene for Au-Kline syndrome with overlapping disease characteristics seen in the index patient.The human R102*or A438T mutant protein failed to fully rescue the growth defects of RBM42 ortholog knockout△FgRbp1 in Fusarium while it was rescued by the wild-type(WT)human RBM42.A mouse model carying Rbm42 compound heterozygous variants,c.280C>T(p.Q94*)and c.1306_1308delinsACA(p.A436T),demonstrated gross fetal develop-mental defects and most of the double mutant animals died by E13.5.RNA-seq data confirmed that Rbm42 was involved in neurological and myocardial functions with an essential role in alternative splicing(As).Overall,we present clinical,genetic,and functional data to demonstrate that defects in RBM42 constitute the underlying etiology of a new neurodevelopmental disease which links the dysregulation of global AS to abnormal embryonic development.Yiyao Chen Bingxin Yang Xiaoyu Merlin Zhang Songchang Chen Minhui Wang Liya Hu Nina Pan Shuyuan Li Weihui Shi Zhenhua Yang Li Wang Yajing Tan Jian Wang Yanlin Wang Qinghe Xing Zhonghua Ma Jinsong Li He-Feng Huang Jinglan Zhang Chenming Xu 2024Protein & Cell2024,15,1:0
14Insulin induces PKC-dependent proliferation of mesenteric vascular smooth muscle cells from hypertensive patients显示文摘Background and objectives Proliferation of human vascular smooth muscle cells (VSMCs) induced by hyperinsulinemia is a very common clinical pathology. Extensive research has focused on PKC (Protein kinase C)-MAPK (mitogen-activated protein kinase)intracellular signal transduction and the phenotypic modulation accompanied by reorganization of intracellular F-actins in VSMCs.Methods DNA synthesis, signaling of ERK1/2 MAPKs, and changes in α-smooth muscle (SM) actin and F-actin were studied in hypertensive and normotensive human arterial VSMCs exposed to insulin and PMA with and without the PKC inhibitor, GF109203X.Results Differences among cell types in MAPK signaling, α-SM actin, and F-actin isoforms in VSMCs harvested from the arteries of patients with essential hypertension (EH) and normotension (NT) were identified in response to insulin treatment. Proliferation and activation of MAPK were more pronounced in EH VSMCs than in NEH VSMCs. Insulin exposure decreased expression of α-SM actin and was accompanied by rearrangement of intracellular F-actins in VSMCs, especially in the EH group. These effects were reversed by treatment with the PKC inhibitor.
