| 1 | The Performance of Whole Genome Amplification Methods and Next-Generation Sequencing for Pre-Implantation Genetic Diagnosis of Chromosomal Abnormalities显示文摘Reliable and accurate pre-implantation genetic diagnosis(PGD) of patient's embryos by next-generation sequencing(NGS) is dependent on efficient whole genome amplification(WGA) of a representative biopsy sample. However, the performance of the current state of the art WGA methods has not been evaluated for sequencing. Using low template DNA(15 pg) and single cells, we showed that the two PCR-based WGA systems Sure Plex and MALBAC are superior to the REPLI-g WGA multiple displacement amplification(MDA) system in terms of consistent and reproducible genome coverage and sequence bias across the 24 chromosomes, allowing better normalization of test to reference sequencing data. When copy number variation sequencing(CNV-Seq) was applied to single cell WGA products derived by either Sure Plex or MALBAC amplification, we showed that known disease CNVs in the range of 3e15 Mb could be reliably and accurately detected at the correct genomic positions. | 2 | Fouling corrosion in aluminum heat exchangers显示文摘Fouling deposits on aluminum heat exchanger reduce the heat transfer efficiency and cause corrosion to the apparatus.This study focuses on the corrosive behavior of aluminum coupons covered with a layer of artificial fouling in a humid atmosphere by their weight loss,Tafel plots,electrochemical impedance spectroscopy(EIS),and scanning electron microscope(SEM)observations.The results reveal that chloride is one of the major elements found in the fouling which damages the passive film and initiates corrosion.The galvanic corrosion between the metal and the adjacent carbon particles accelerates the corrosive process.Furthermore,the black carbon favors the moisture uptake,and gives the dissolved oxygen greater chance to migrate through the fouling layer and form a continuous diffusive path.The corrosion rate decreasing over time is conformed to electrochemistry measurements and can be verified by Faraday’s law.The EIS results indicate that the mechanism of corrosion can be interpreted by the pitting corrosion evolution mechanism,and that pitting was observed on the coupons by SEM after corrosive exposure. | Su Jingxin Ma Minyu Wang Tianjing Guo Xiaomei Hou Liguo Wang Zhiping | 2015 | Chinese Journal of Aeronautics2015,28,3: | 5 |
| 3 | Collaborative target tracking in WSNs using the combination of maximum likelihood estimation and Kalman filtering显示文摘Target tracking using wireless sensor networks requires efficient collaboration among sensors to tradeoff between energy consumption and tracking accuracy. This paper presents a collaborative target tracking approach in wireless sensor networks using the combination of maximum likelihood estimation and the Kalman filter. The cluster leader converts the received nonlinear distance measurements into linear observation model and approximates the covariance of the converted measurement noise using maximum likelihood estimation, then applies Kalman filter to recursively update the target state estimate using the converted measurements. Finally, a measure based on the Fisher information matrix of maximum likelihood estimation is used by the leader to select the most informative sensors as a new tracking cluster for further tracking. Given the unstable agent dynamics and a known communication delay, two consensus protocols are designed to guarantee consensus over undirected network. By jointly researching the effects of agent dynamics and network topology, allowable delay bounds depending on the maxima of concave functions are easy to calculate. Especially, the maximum delay bound is derived when the network topology is completely connected. | 7 | The spatial-temporal differentiation of green land use in Beijing-Tianjin-Hebei urban agglomeration显示文摘As the space carrier of the construction of ecological civilization, land's green and efficient utilization is an important guarantee for realizing national sustainable development. Based on traditional land evaluation, this paper scientifically defines the green land use, puts land as one of the production factors, and brings energy consumption, environmental pollution, etc. into the input-output system to measure the green land utilization efficiency of the urban construction land of Beijing-TianjinHebei Urban Agglomeration from 2006 to 2016. The study shows that the overall efficiency variation of the urban agglomeration is related with the land and environment policies. Efficiency of 2016 is higher than that of 2006, and energy and environment are the principal factors affecting the green land use. The efficiency of each city is positively correlated with its economic development, negatively correlated with the construction land expansion. Efficiency gaps in different cities are expanding.There is positive correlation with overall weak space between cities, and the partial spatial agglomeration phenomenon appears. | 16 | Transcriptome analysis to identify candidate genes related to chlorogenic acid biosynthesis during development of Korla fragrant pear in Xinjiang显示文摘Korla fragrant pear(KFP)with special fragrance is a unique cultivar in Xinjiang,China.In order to explore the biosynthesis molecular mechanism of chlorogenic acid(CGA)in KFP,the samples at different development periods were collected for transcriptome analysis.High performance liquid chromatography analysis showed that CGA contents of KFP at 88,118 and 163 days after full bloom were(20.96±1.84),(12.01±0.91)and(7.16±0.41)mg/100 g,respectively,and decreased with the fruit development.Pears from these typical 3 periods were selected for de novo transcriptome assemble and 68059 unigenes were assembled from 444037960 clean reads.One‘phenylpropanoid biosynthesis’pathway including 57 unigenes,11 PALs,1 PTAL,64CLs,9 C4Hs,25 HCTs and 5 C3’Hs related to CGA biosynthesis was determined.It was found that the expression levels of 11 differentially expressed genes including 1 PAL,2 C4Hs,34CLs and 5 HCTs were consistent with the change of CGA content.Quantitative polymerase chain reaction analysis further showed that 8 unigenes involved in CGA biosynthesis were consistent with the RNA-seq data.These findings will provide a comprehensive understanding and valuable information on the genetic engineering and molecular breeding in KFP. | Hao Wen Wenqiang Wang Xi Jiang Minyu Wu Hongjin Bai Cuiyun Wu Lirong Shen | 2022 | Food Science and Human Wellness2022,11,4: | 1 |

| 20 | Transcriptome and weighted gene co-expression network analysis of jujube(Ziziphus jujuba Mill.)fruit reveal putative genes involved in proanthocyanin biosynthesis and regulation显示文摘Proanthocyanidin(PA)is an important bioactive compound with multiple physiological benefits in jujube(Ziziphus jujube Mill.).However,the molecular mechanisms underlying PA biosynthesis in jujube fruit have not been investigated.Here,the profiling of PA,(+)-catechin and(–)-epicatechin and transcriptome sequencing of three jujube cultivars from Xinjiang Uyghur Autonomous Region of China at five developmental stages were analyzed.The levels of total PAs and catechin exhibited a decreased trend over jujube ripening,and epicatechin content of two jujube cultivars increased first and then declined.Transcriptome analysis revealed that the differentially expressed genes(DEGs)were mainly enriched in ribosome,glycolysis/gluconeogenesis,fructose and mannose metabolism.17 DEGs encoding PAL,CHS,CHI,CHS,F3'H,LAR,ANR,C4Hs,4CLs,FLSs,DFRs and UFGTs involved in PA biosynthesis were relatively abundant.The highly transcribed LAR gene may greatly contribute to epicatechin accumulation.A weighted gene co-expression network analysis(WGCNA)was performed,and a network module including 1620 genes highly correlated with content of Pas and catechin was established.We identified 58 genes including 9 structural genes and 49 regulatory genes related to PA biosynthesis and regulation in the WGCNA module.Sixteen genes encoding 9 families of transcriptional factors(i.e.,MYB,bHLH,ERF,bZIP,NAC,SBP,MIKC,HB,WRKY)were considered as hub genes.The results of qRT-PCR analysis validating 10 genes were well consistent with the transcriptome data.These findings provide valuable knowledge to facilitate its genetic studies and molecular breeding. | Wenqiang Wang Yunfeng Pu Hao Wen Dengyang Lu Min Yan Minzhe Liu Minyu Wu Hongjin Bai Lirong Shen Cuiyun Wu | 2023 | Food Science and Human Wellness2023,12,5: | 1 |
