1Prospect of the QTL-qSB-9^(Tq) utilized in molecular breeding program of japonica rice against sheath blight显示文摘The major QTL-qSB-9Tq conferring partial resistance to rice (Oryza sativa L.) sheath blight (Rhizoctonia solani Kühn) has been verified on chromosome 9 of the indica rice cultivar, Teqing. In this study, the prospect of this QTL utilized in molecular breeding program of japonica rice for sheath blight resistance was investigated. Most of the japonica rice cultivars showed lower level of sheath blight resistance than the indica rice cultivars. At the corresponding site of qSB-9Tq, nine typical japonica rice cultivars from different ecological regions or countries proved to possess the susceptible allele(s). Introgression of qSB-9Tq into these cultivars enhanced their resistance level by decreasing sheath blight score of 1.0 (0.5–1.3), which indicated that qSB-9Tq had a large potential in strengthening the resistance of japonica rice to sheath blight. 23D-QSAR study on atmospheric half-lives of POPs using CoMFA and CoMSIA显示文摘3D-QSAR studies of persistent organic pollutants(POPs)screening for atmosphere persistence were performed by comparative molecular field analysis(CoMFA)and comparative molecular similarity index analysis(CoMSIA)methods.The mean and maximum half-life estimations for degradation in air of 49 UNEP POPs and possible POPs were modeled.Both groups’data have been modeled to obtain an average estimate and a predictive value for ranking and screening purposes.CoMFA and CoMSIA models have given cross-validation regression coeffcient(q2)values of more than 0.69 and correlation coeffcient(r2)value of more than 0.84,which validated for their prediction,could be applied to predict unavailable data.Yuyin LV Chunsheng YIN Hongyan LIU Zhongsheng YI Yang WANG 2008Journal of Environmental Sciences2008,20,12:7

4Validation and invalidation of SARS-CoV-2 main protease inhibitors using the Flip-GFP and Protease-Glo luciferase assays显示文摘SARS-CoV-2 main protease(M^(pro))is one of the most extensively exploited drug targets for COVID-19.Structurally disparate compounds have been reported as M^(pro) inhibitors,raising the question of their target specificity.To elucidate the target specificity and the cellular target engagement of the claimed M^(pro) inhibitors,we systematically characterize their mechanism of action using the cell-free FRET assay,the thermal shift-binding assay,the cell lysate Protease-Glo luciferase assay,and the cell-based FlipGFP assay.Collectively,our results have shown that majority of the M^(pro) inhibitors identified from drug repurposing including ebselen,carmofur,disulfiram,and shikonin are promiscuous cysteine inhibitors that are not specific to M^(pro),while chloroquine,oxytetracycline,montelukast,candesartan,and dipyridamole do not inhibit M^(pro) in any of the assays tested.Overall,our study highlights the need of stringent hit validation at the early stage of drug discovery.Chunlong Ma Haozhou Tan Juliana Choza Yuyin Wang Jun Wang 2022Acta Pharmaceutica Sinica B2022,12,4:3
5Cardiomyocytes induced from hiPSCs by well-defined compounds have therapeutic potential in heart failure by secreting PDGF-BB显示文摘Recent studies have suggested that transplant of hiPS-CMs is a promising approach for treating heart failure.However,the optimally clinical benefits have been hampered by the immature nature of the hiPS-CMs,and the hiPS-CMs-secreted proteins contributing to the repair of cardiomyocytes remain largely unidentified.Here,we established a saponin+compound optimally induced system to generate hiPS-CMs with stable functional attributes in vitro and transplanted in heart failure mice.Our study showed enhanced therapeutic effects of optimally induced hiPS-CMs by attenuating cardiac remodeling and dysfunction,these beneficial effects were concomitant with reduced cardiomyocytes death and increased angiogenesis.Moreover,the optimally induced hiPS-CMs could gathering to the injured heart and secret an abundant PDGF-BB.The reparative effect of the optimally induced hiPS-CMs in the hypoxia-injured HCMs was mimicked by PDGF-BB but inhibited by PDGF-BB neutralizing antibody,which was accompanied by the changed expression of p-PI3K and p-Akt proteins.It is highly possible that the PI3K/Akt pathway is regulated by the PDGF-BB secreted from the compound induced hiPS-CMs to achieve a longer lasting myocardial repair effect compared with the standard induced hiPS-CMs.Taken together,our data strongly implicate that the compound induced hiPS-CMs promote the recovery of injured hearts via paracrine action.In this process,the paracrine factor PDGF-BB derived from the compound induced hiPS-CMs reduces isoproterenol-induced adverse cardiac remodeling,which is associated with improved cardiac function,and these effects are mediated by the PI3K/Akt pathway,suggesting that the optimally induced hiPS-CMs may serve as a new promising cell therapy for clinical applications.Hongmei Li Fenfang Wu Guangrui Huang Di Wu Ting Wang Xiashuang Wang Kai Wang Yuyin Feng Anlong Xu 2022Signal Transduction and Targeted Therapy2022,7,8:2

10The Phytophthora effector Avh241 interacts with host NDR1-like proteins to manipulate plant immunity显示文摘Plant pathogens rely on effector proteins to suppress host innate immune responses and facilitate colonization.Although the Phytophthora sojae RxLR effector Avh241 promotes Phytophthora infection,the molecular basis of Avh241 virulence remains poorly understood.Here we identified non-race specific disease resistance 1(NDR1)-like proteins,the critical components in plant effector-triggered immunity(ETI)responses,as host targets of Avh241.Avh241 interacts with NDR1 in the plasma membrane and suppresses NDR1-participated ETI responses.Silencing of GmNDR1s increases the susceptibility of soybean to P.sojae infection,and overexpression of GmNDR1s reduces infection,which supports its positive role in plant immunity against P.sojae.Furthermore,we demonstrate that GmNDR1 interacts with itself,and Avh241 probably disrupts the self-association of GmNDR1.These data highlight an effective counter-defense mechanism by which a Phytophthora effector suppresses plant immune responses,likely by disturbing the function of NDR1 during infection.Bo Yang Sen Yang Baodian Guo Yuyin Wang Wenyue Zheng Mengjun Tian Kaixin Dai Zehan Liu Haonan Wang Zhenchuan Ma Yan Wang Wenwu Ye Suomeng Dong Yuanchao Wang 2021Journal of Integrative Plant Biology2021,63,7:1
