| 1 | Experimental investigations on validity of moving chemical reaction boundary equation for the strong acid——weak base system with an excess of potassium chloride显示文摘A moving chemical reaction boundary (MCRB) is formed by the weak alkali NH3*H2O and strong acid HCl with an excess of potassium chloride as background electrolyte. The experimental results are compared with the predictions in accordance with the moving chemical reaction boundary equation (MCRBE). If the MCRB is cathodic, the experimental results are in agreement with the predictions by the MCRBE. However, if the MCRB is anodic, the experimental results significantly deviate from the predictions of the MCRBE. A corrected MCRB equation is proposed for such conditions. The findings are evidently different from those in our early reports(J. Chromatogr. A, 891 (2000): 337~347; 907 (2001): 347~352; 922 (2001) 283~393). | 2 | Identification of Styryl Sulfonyl Fluoride as a Near-Perfect Michael Acceptor for Diversified Protein Bioconjugations显示文摘Cysteine(Cys)-specific bioconjugation has widespread applications in the synthesis of protein conjugates,particularly for the functionalization of antibodies.Here,we report the discovery of transstyryl sulfonyl fluoride(SSF)as a near-perfect Michael acceptor for Cys-specific protein bioconjugation.Compared to maleimides,which are predominantly used,SSF exhibited better chemoselectivity,selfstability,and conjugate stability while maintaining comparable reactivity.Using SSF-derived probes,proteins can be readily modified on the Cys residue(s)to install functionalities,for example,fluorescent dyes,toxins,and oligonucleotides,without influencing the activity.Further applications of SSF-derived serum-stable antibody-drug conjugates and PD-L1 nanobody-oligo conjugates demonstrate the great translational value of SSF-based bioconjugation in drug development and single-cell sequencing. | Qingsong Wu Qi Xue Ji Li Qinheng Zheng Xinlu Zhao Wannan Li Shiming Sun Wanxing Sha Yang Yang Yi Yang Jie P.Li | 2023 | CCS Chemistry2023，5，10： | 0 |
