2Chromosome-scale genome assembly of sweet cherry(Prunus avium L.)cv.Tieton obtained using long-read and Hi-C sequencing显示文摘Sweet cherry(Prunus avium)is an economically significant fruit species in the genus Prunus.However,in contrast to other important fruit trees in this genus,only one draft genome assembly is available for sweet cherry,which was assembled using only Illumina short-read sequences.The incompleteness and low quality of the current sweet cherry draft genome limit its use in genetic and genomic studies.A high-quality chromosome-scale sweet cherry reference genome assembly is therefore needed.A total of 65.05 Gb of Oxford Nanopore long reads and 46.24 Gb of Illumina short reads were generated,representing~190x and 136x coverage,respectively,of the sweet cherry genome.The final de novo assembly resulted in a phased haplotype assembly of 344.29 Mb with a contig N50 of 3.25 Mb.Hi-C scaffolding of the genome resulted in eight pseudochromosomes containing 99.59%of the bases in the assembled genome.Genome annotation revealed that more than half of the genome(59.40%)was composed of repetitive sequences,and 40,338 protein-coding genes were predicted,75.40%of which were functionally annotated.With the chromosomescale assembly,we revealed that gene duplication events contributed to the expansion of gene families for salicylic acid/jasmonic acid carboxyl methyltransferase and ankyrin repeat-containing proteins in the genome of sweet cherry.Four auxin-responsive genes(two GH3s and two SAURs)were induced in the late stage of fruit development,indicating that auxin is crucial for the sweet cherry ripening process.In addition,772 resistance genes were identified and functionally predicted in the sweet cherry genome.The high-quality genome assembly of sweet cherry obtained in this study will provide valuable genomic resources for sweet cherry improvement and molecular breeding.Jiawei Wang Weizhen Liu Dongzi Zhu Po Hong Shizhong Zhang Shijun Xiao Yue Tan Xin Chen Li Xu Xiaojuan Zong Lisi Zhang Hairong Wei Xiaohui Yuan Qingzhong Liu 2020Horticulture Research2020,7,1:4
3ITO@TiO_(2)nanoarray:An efficient and robust nitrite reduction reaction electrocatalyst toward NH_(3)production under ambient conditions显示文摘Ambient electrochemical nitrite(NO_(2)^(-))reduction is viewed as an effective and sustainable approach for simul-taneously removing NO_(2)^(-)and producing ammonia(NH_(3)).However,the complex multi-electron transfer steps involved in the NO_(2)^(-)reduction reaction(NO_(2)^(-)RR)lead to sluggish kinetics and low product selectivity toward NH_(3),underscoring the need for NH_(3)synthesis electrocatalysts with high activity and durability.Herein,we report amorphous indium-tin oxide sputtered on a TiO_(2)nanobelt array on a Ti plate(ITO@TiO_(2)/TP)as a 3D NH_(3)-producing catalyst for the NO_(2)^(-).In 0.5 M LiClO_(4)with 0.1 M NO_(2)^(-),it shows greatly boosted NO_(2)^(-)RR activity toward NH_(3)production,with excellent selectivity,achieving a large NH_(3)yield of 411.3μmol h^(-1)cm^(-2)and a high Faradaic efficiency of 82.6%.It also shows high durability for continuous electrolysis.A Zn-NO_(2)^(-)battery with ITO@TiO_(2)/TP cathode offers an NH_(3)yield of 23.1μmol h^(-1)cm^(-2)and a peak power density of 1.22 m.Shaoxiong Li Jie Liang Peipei Wei Qian Liu Lisi Xie Yonglan Luo Xuping Sun 2022eScience2022,2,4:3

7Research on the Equivalence Between Double Differential- mode Current Injection and Radiation Test Method显示文摘There are the application scope limits for single differential-mode current injection test method, so in order to carry out injection susceptibility test for two-pieces equipment interconnected with both ends of a cable simultaneously, a double differential-mode current in- jection test method (DDMCI) is proposed. The method adopted the equivalence source wave theorem and Baum-Liu-Tesche(BLT) equation as its theory foundation. The equivalent corresponding relation between injection voltage and radiation electric field intensity is derived, and the phase relation between the two injection voltage sources is confirmed. The results indicate that the amplitude and phase of the equivalent injection voltage source is closely related to the S parameter of directional coupling device, the transmission line length, and the source vector in BLT equation, but has nothing to do with the reflection coefficient between the two equipment pieces. 8Assembly dynamics of East Asian subtropical evergreen broadleaved forests: New insights from the dominant Fagaceae trees显示文摘The dominant species of a biome can be regarded as its genuine indicator.Evergreen broadleaved forests(EBLFs) in subtropical East Asia harbor high levels of species biodiversity and endemism and are vital to regional carbon storage and cycling.However,the historical assembly of this unique biome is still controversial.Fagaceae is the most essential family in East Asian subtropical EBLFs and its dominant species are vital for the existence of this biome.Here,we used the dominant Fagaceae species to shed light on the dynamic process of East Asian subtropical EBLFs overtime.Our results indicate high precipitation in summer and low temperature in winter are the most influential climatic factors for the distribution of East Asian subtropical EBLFs.Modern East Asian subtropical EBLFs did not begin to appear until 23 Ma,subsequently experienced a longlasting development in the Miocene and markedly deteriorated at about 4 Ma,driven jointly by orogenesis and paleoclimate.We also document that there is a lag time between when one clade invaded the region and when its members become dominant species within the region.This study may improve our ability to predict and mitigate the threats to biodiversity of East Asian subtropical EBLFs and points to a new path for future studies involving multidisciplinary methods to explore the assembly of regional biomes.Lisi Hai Xiao-Qian Li Jing-Bo Zhang Xiao-Guo Xiang Rui-Qi Li Florian Jabbour Rosa del COrtiz An-Ming Lu Zhi-Duan Chen Wei Wang 2022Journal of Integrative Plant Biology2022,64,11:1

13The development of an iridium(Ⅲ) complex functionalized G-quadruplex probe for the stability of G-quadruplex and lifetime image in cytoplasm显示文摘G-quadruplex(G4) is widely known as a non-classical secondary structure of nucleic acid. With the indepth study of G4, it is an urgent need for a phosphorescent probe with a high G4 binding ability to evaluate the level of G4 in the cytoplasm. Thus, this study designed and synthesized Ir-PDP where an Ir(Ⅲ)complex was used as a phosphorescent emitter. Meanwhile, two installed PDPs(pyridostatin derivatives)were used to improve the combination ability with G4 and reduced the cytotoxicity of the Ir(Ⅲ) complex.Compared with other nucleic acid secondary structures, Ir-PDP produced a higher phosphorescence lifetime after interacting with G4. Ir-PDP was distributed in the cytoplasm of living cells, and two-photon phosphorescence lifetime imaging can detect the binding events of the probe in the cytoplasm. The addition of G4 binder PDS significantly regulated cytoplasmic phosphorescence lifetime.
