| 1 | Size and elemental composition of dry-deposited particles during a severe dust storm at a coastal site of Eastern China显示文摘Dry-deposited particles were collected during the passage of an extremely strong dust storm in March, 2010 at a coastal site in Qingdao(36.15°N, 120.49°E), a city located in Eastern China. The size, morphology, and elemental composition of the particles were quantified with a scanning electron microscope equipped with an energy dispersive X-ray instrument(SEM–EDX). The particles appeared in various shapes, and their size mainly varied from 0.4to 10 μm, with the mean diameters of 0.5, 1.5, and 1.0 μm before, during, and after the dust storm, respectively. The critical size of the mineral particles settling on the surface in the current case was about 0.3–0.4 μm before the dust storm and about 0.5–0.7 μm during the dust storm. Particles that appeared in high concentration but were smaller than the critical size deposited onto the surface at a small number flux. The elements Al, Si and Mg were frequently detected in all samples, indicating the dominance of mineral particles. The frequency of Al in particles collected before the dust storm was significantly lower than for those collected during and after the dust storm. The frequencies of Cl and Fe did not show obvious changes, while those of S, K and Ca decreased after the dust arrival. | 2 | Phenotypic variation in thirteen native provenances of Lespedeza bicolor Turcz显示文摘Lespedeza bicolor Turcz is a native shrub in the temperate region of China. Thirteen provenances of L. bicolor were inves-tigated in our study. Morphological diversity among and within provenances were analyzed based on 19 phenotypic traits, including seven vegetative growth characteristics, nine floral traits and three pod characteristics. The coefficient of variation (CV) ranged from 6.4% to 65.4% and the phenotypic differentiation coefficient from 43.3% to 97.3%. Analyses of variance showed that there were extremely significant variations among and within provenances in pod length. Differences in plant height, inter-nodal length, number of branches, leaf width, leaf length and leaf area were not remarkable among provenance, but distinct within provenances. Other traits showed evidence of contrasts. Variation among provenances was the main part of phenotypic variation. Correlation analysis indicated that there were highly significant positive correlations between plant height and other vegetative growth characteristics. Floral traits were not associated with vegetative growth characteristics. | 5 | Isolation and sequence analysis of a Dof protein gene(PtDof1) in Populus显示文摘Both cDNA and DNA clones of PtDof1(GenBank Accession No. FJ402844 and FJ402845) were isolated from plants grown in tissue culture of Populus tomentosa. The DNA sequence is 1597 bp including two exons and one intron. The cDNA is 969 bp in length with a 765 bp open reading frame which is capable of encoding 255 amino acids. The deduced amino acids sequence of the PtDof1 protein shares 65%,56% and 55% identity with Vitis vinifera(CAO48618) ,Nicotiana tabacum(CAA08755) and Glycine max(ABI16022) Dof protein by blast analysis in GenBank. Phylogenic analysis suggests PtDof1 gene could belong to the Dof gene family. | 6 | Identification of HMGCR as the anticancer target of physapubenolide against melanoma cells by in silico target prediction显示文摘Physapubenolide(PB),a withanolide-type compound extracted from the traditional herb Physalis minima L.,has been demonstrated to exert remarkable cytotoxicity against cancer cells;however,its molecular mechanisms are still unclear.In this study,we demonstrated that PB inhibited cell proliferation and migration in melanoma cells by inducing cell apoptosis.The anticancer activity of PB was further verified in a melanoma xenograft model.To explore the mechanism underlying the anticancer effects of PB,we carried out an in silico target prediction study,which combined three approaches(chemical similarity searching,quantitative structure-activity relationship(QSAR),and molecular docking)to identify the targets of PB,and found that PB likely targets 3-hydroxy-methylglutaryl CoA reductase(HMGCR),the rate-limiting enzyme of the mevalonate pathway,which promotes cancer cell proliferation,migration,and metastasis.We further demonstrated that PB interacted with HMGCR,decreased its protein expression and inhibited the HMGCR/YAP pathway in melanoma cells.In addition,we found that PB could restore vemurafenib sensitivity in vemurafenib-resistant A-375 cells,which was correlated with the downregulation of HMGCR.In conclusion,we demonstrate that PB elicits anticancer action and enhances sensitivity to vemurafenib by targeting HMGCR. | Hai-yan Wang Pian Yu Xi-sha Chen Hui Wei Shi-jie Cao Meng Zhang Yi Zhang Yong-guang Tao Dong-sheng Cao Feng Qiu Yan Cheng | 2022 | Acta Pharmacologica Sinica2022,43,6: | 0 |
