| 1 | Effects of phosphorus and nitrogen limitation on PHA production in activated sludge显示文摘The effects of phosphorus and nitrogen limitation on polyhydroxyalkanoate(PHA) production and accumulation by activated sludge biomass with acetate as a carbon source were investigated.Pre-selected influent carbon-phosphorus(C:P,W/W) of 100,160,250,500 and 750,and carbon-nitrogen(C:N,W/W) of 20,60,100,125 and 180 were applied in the phosphorus limitation experiments and the nitrogen limitation experiments,respectively.The maximum PHA accumulation up to 59%of the cell dry weight with a PHA productivity of 1.61 mg PHA/mg COD consumed was observed at the C:N 125 in the nitrogen limitation experiment.This value was much higher than that obtained in previous studies with a normal substrate feeding.The study showed that activated sludge biomass would produce more polyhydroxybutyrate than polyhydroxyvalerate under the stress of nutrient limitation,especially under phosphorus limitation conditions.The experimental result also indicated that both phosphorus and nitrogen limitation may cause sludge bulking. | Qinxue Wen,Zhiqiang Chen,Ting Tian,Wei Chen State Key Laboratory of Urban Water Resource and Environment,Harbin Institute of Technology,Harbin 150090,China | 2010 | Journal of Environmental Sciences2010,22,10: | 9 |
| 2 | The involvement of wheat U-box E3 ubiquitin ligase TaPUB1 in salt stress tolerance显示文摘U-box E3 ubiquitin ligases play important roles in the ubiquitin/26 S proteasome machinery and in abiotic stress responses. Ta PUB1-overexpressing wheat(Triticum aestivum L.) were generated to evaluate its function in salt tolerance. These plants had more salt stress tolerance during seedling and flowering stages, whereas the Ta PUB1-RNA interference(RNAi)-mediated knock-down transgenic wheat showed more salt stress sensitivity than the wild type(WT). Ta PUB1 overexpression upregulated the expression of genes related to ion channels and increased the net root Na+efflux, but decreased the net K+efflux and H+influx, thereby maintaining a low cytosolic Na+/K+ratio, compared with the WT. However,RNAi-mediated knock-down plants showed the opposite response to salt stress. Ta PUB1 could induce the expression of some genes that improved the antioxidant capacity of plants under salt stress. Ta PUB1 also interacted with Ta MP(Triticum aestivum α-mannosidase protein), a regulator playing an important role in salt response in yeast and in plants. Thus, low cytosolic Na+/K+ratios and better antioxidant enzyme activities could be maintained in wheat with overexpression of Ta PUB1 under salt stress. | 5 | DC-SIGN promotes Japanese encephalitis virus transmission from dendritic cells to T cells via virological synapses显示文摘Skin-resident dendritic cells(DCs) likely encounter incoming viruses in the first place, and their migration to lymph nodes following virus capture may promote viral replication. However, the molecular mechanisms underlying these processes remain unclear. In the present study, we found that compared to cell-free viruses, DC-bound viruses showed enhanced capture of JEV by T cells.Additionally, JEV infection was increased by co-culturing DCs and T cells. Blocking the C-type lectin receptor DC-specific intercellular adhesion molecule-3-grabbing non-integrin(DC-SIGN) with neutralizing antibodies or antagonists blocked JEV transmission to T cells. Live-cell imaging revealed that DCs captured and transferred JEV viral particles to T cells via virological synapses formed at DC-T cell junctions. | 6 | Binding of HIV-1 virions to α_4β_7 expressing cells and impact of antagonizing α_4β_7 on HIV-1 infection of primary CD4^+ T cells显示文摘HIV-1 envelope glycoprotein is reported to interact with α4β7, an integrin mediating the homing of lymphocytes to gut-associated lymphoid tissue, but the significance of α4β7 in HIV-1 infection remains controversial. Here, using HIV-1 strain Ba L, the gp120 of which was previously shown to be capable of interacting with α4β7, we demonstrated that α4β7 can mediate the binding of whole HIV-1 virions to α4β7-expressing transfectants. We further constructed a cell line stably expressing α4β7 and confirmed the α4β7-mediated HIV-1 binding. In primary lymphocytes with activated α4β7 expression, we also observed significant virus binding which can be inhibited by an anti-α4β7 antibody. Moreover, we investigated the impact of antagonizing α4β7 on HIV-1 infection of primary CD4+ T cells. In α4β7-activated CD4+ T cells, both anti-α4β7 antibodies and introduction of shorthairpin RNAs specifically targeting α4β7 resulted in a decreased HIV-1 infection. Our findings indicate that α4β7 may serve as an attachment factor at least for some HIV-1 strains. | 8 | Phenological metrics-based crop classification using HJ-1 CCD images and Landsat 8 imagery显示文摘Crop type data are an important piece of information for many applications in agriculture.Extracting crop type using remote sensing is not easy because multiple crops are usually planted into small parcels with limited availability of satellite images due to weather conditions.In this research,we aim at producing crop maps for areas with abundant rainfall and small-sized parcels by making full use of Landsat 8 and HJ-1 charge-coupled device(CCD)data.We masked out non-vegetation areas by using Landsat 8 images and then extracted a crop map from a longterm time-series of HJ-1 CCD satellite images acquired at 30-m spatial resolution and two-day temporal resolution.To increase accuracy,four key phenological metrics of crops were extracted from time-series Normalized Difference Vegetation Index curves plotted from the HJ-1 CCD images.These phenological metrics were used to further identify each of the crop types with less,but easier to access,ancillary field survey data.We used crop area data from the Jingzhou statistical yearbook and 5.8-m spatial resolution ZY-3 satellite images to perform an accuracy assessment.The results show that our classification accuracy was 92%when compared with the highly accurate but limited ZY-3 images and matched up to 80%to the statistical crop areas. | Xiaochun Zhang Qinxue Xiong Liping Di Junmei Tang Jin Yang Huayi Wu Yan Qin Rongrui Su Wei Zhou | 2018 | International Journal of Digital Earth2018,11,12: | 0 |
