| 1 | Spatio-temporal Pattern and Spatial Heterogeneity of Ecotones Based oh Land Use Types of Southeastern Da Hinggan Mountains in China显示文摘Ecotones have received great attention due to its critical function in energy flux, species harbor, global carbon sequestration, and land-atmosphere interaction. This study investigated land use pattern and spatial heterogeneity of the ecotones among agricultural land, forest land, and grassland of the southeastern Da Hinggan Mountains in the northeastern China. The change of these delineated ecotones under different slopes and aridity conditions was analyzed by two landscape indices, edge density(ED) and core area percentage of landscape(CPL), to explore the inter-linkage between spatial structure of ecotones and socioeconomic development and land management. Specifically, the ecotones such as agriculture-forest(AF) ecotone, forest-grassland(FG) ecotone, and agriculture-forestgrassland(AFG) ecotone moved from the arid southeast to the humid northwest. The flat area with small slope is more edge-fragmented than the steep area since the ED decreases as the slope increases. | 2 | Comparison and Analysis of Agricultural and Forest Land Changes in Typical Agricultural Regions of Northern Mid-latitudes显示文摘The northeastern China, the United States, and the western Europe are important agricultural regions both on the global and regional scales. The western Europe has a longer history of agricultural land development than the eastern United States. These two regions have changed from the deforestation and reclamation phase in the past to the current land abandonment and reforestation phase. Compared with the two regions, large-scale land exploitation has only been practiced in the northeastern China during the last century. After a short high-intensity deforestation and reclamation period, agricultural and forest lands are basically in a dynamic steady state. By comparing domestic and international agro-forestry development and considering the ecological environment and socio-economic benefits that can be derived from agro-forestry, this paper suggests that large area of reforestation would be inevitable in future though persistent and large agricultural demand in coming decades even more. And local reforestation at slope farmland with ecological vulnerability should be imperative at present to avoid severer damage. | 3 | A MODIS Time Series Data Based Algorithm for Mapping Forest Fire Burned Area显示文摘Burned area mapping is an essential step in the forest fire research to investigate the relationship between forest fire and climate change and the effect of forest fire on carbon budgets. This study proposed an algorithm to map forest fire burned area using the Moderate-Resolution Imaging Spectroradiameter (MODIS) time series data in Heilongjiang Province, China. The algorithm is divided into two steps: Firstly, the′core′pixels were extracted to represent the most possible burned pixels based on the comparison of the temporal change of Global Environmental Monitoring Index (GEMI), Burned Area Index (BAI) and MODIS active fire products between pre- and post-fires. Secondly, a 15-km distance was set to extract the entire burned areas near the′core′pixels as more relaxed conditions were used to identify the fire pixels for reducing the omission error as much as possible. The algorithm comprehensively considered the thermal characteristics and the spectral change between pre-and post-fires, which are represented by the MODIS fire products and the spectral index, respectively. Tahe, Mohe and Huma counties of Heilongjiang Province, China were chosen as the study area for burned area mapping and a time series of burned maps were produced from 2000 to 2011. | 4 | Effects of electro-acupuncture on brain tissue norepinephrine contents in a morphine withdrawal anxiety mouse model显示文摘BACKGROUND: Following morphine withdrawal, anxiety is associated with abnormal norepinephrine content change. However, increasing blood lactic acid content can induce anxiety or panic in patients with anxiety disorder or endogenous opioid peptide functional disorder. OBJECTIVE: This study was designed to observe the effects of electro-acupuncture, at the 'Sanyinjiao' point (SP 6), on brain tissue norepinephrine and blood lactic acid content in anxiety-model mice after morphine withdrawal. DESIGN: A randomized controlled animal experiment. SETTING: This study was performed in the Laboratory of Acupuncture, Electro-acupuncture & Tuina College, Chengdu University of Traditional Chinese Medicine, from June to September 2001. MATERIALS: A total of 50 healthy Kunming male mice were provided by the Laboratory Animal Center of Chengdu University of Traditional Chinese Medicine. The protocol was performed in accordance with ethical guidelines stated in the Guide for the use and care of laboratory animals, approved by the Committee on the Care and Use of Laboratory Animals of the Institute of Laboratory Animal Resources Commission on Life Sciences, National Research Council, China (1985). Experimental reagents and equipment used were as follows: morphine hydrochloride (Lot No. 930503, Shenyang No.1 Pharmaceutical Factory, China), norepinephrine (Sigma Chemical Company, USA), fluorospectrophotometer (RF-510, Shimadzu Corporation, Japan), Han electro-acupuncture apparatus (WQ 1002, No. zun (91)-227270-588, Beijing Anlong Photoelectricity-Technique Company, China), and T-maze (self-made). METHODS: A total of 50 mice were randomly divided into 5 groups, with 10 mice in each group: blank control, T-maze, model, model + electro-acupuncture, and electro-acupuncture groups. Establishment of anxiety model after morphine withdrawal: the mouse hot plate assay was used to detect the activity of morphine. The median effective dose of morphine, 2.95 mg/kg, was defined as the base. Mice were subcutaneously administered morphine, 3 times a day, for 4 days successively (initially 2.95 mg/kg, then increased day by day, as described below). Interventions: In the model + electro-acupuncture group, after model induction, mice were subjected electro-acupuncture at bilateral 'Sanyinjiao' (SP6) points using a Han electro-acupuncture apparatus with sparse-dense waves and frequency of 2-100 Hz, once a day, for 6 days. In the model group, after anxiety-model induction, mice were subjected to fixation as same as model + electro-acupuncture group within 6 days of model induction. In the electro-acupuncture group, the anxiety model was not induced and mice were subjected to fixation, electro-acupuncture and the T-maze test. In the T-maze group, the anxiety model was not induced and mice were subjected to fixation. The T-maze test was performed in the 4 groups after experiment. In the blank control group, the anxiety model was not induced and mice were subjected to fixation only. MAIN OUTCOME MEASURES: Brain tissue norepinephrine content of morphine-withdrawal anxiety mice was detected by fluorospectrophotometry after 6 days of electro-acupuncture. Blood lactic acid content was detected by visible spectrophotometry. RESULTS: A total of 50 mice were included in the final analysis. Brain norepinephrine content was significantly greater in the model group compared to the T-maze, blank control, electro-acupuncture and model + electro-acupuncture groups, (P < 0.05-0.01). Brain norepinephrine content was similar between the model + electro-acupuncture and the blank control groups (P > 0.05). There was no significant difference in blood lactic acid content among the groups (P > 0.05). CONCLUSION: Electro-acupuncture lowers brain norepinephrine content but does not influence peripheral blood lactic acid content in morphine-withdrawn, anxiety-modeled mice. | 7 | Identification of a key G-protein coupled receptor in mediating appressorium formation and fungal virulence against insects显示文摘Fungal G-protein coupled receptors(GPCRs)play essential roles in sensing environmental cues including host signals.The study of GPCR in mediating fungus-insect interactions is still limited.Here we report the evolution of GPCR genes encoded in the entomopathogenic Metarhizium species and found the expansion of Pth11-like GPCRs in the generalist species with a wide host range.By deletion of ten candidate genes MrGpr1–MrGpr10 selected from the six obtained subfamilies in the generalist M.robertsii,we found that each of them played a varied level of roles in mediating appressorium formation.In particular,deletion of MrGpr8 resulted in the failure of appressorium formation on different substrates and the loss of virulence during topical infection of insects but not during injection assays when compared with the wild-type(WT)strain.Further analysis revealed that disruption of MrGpr8 substantially impaired the nucleus translocation of the mitogen-activated protein kinase(MAPK)Mero-Fus3 but not the MAPK Mero-Slt2 during appressorium formation.We also found that the defect ofΔMrGpr8 could not be rescued with the addition of cyclic AMP for appressorium formation.Relative to the WT,differential expression of the selected genes have also been detected inΔMrGpr8.The results of this study may benefit the understanding of fungus-interactions mediated by GPCRs. | Junmei Shang Yanfang Shang Guirong Tang Chengshu Wang | 2021 | Science China(Life Sciences)2021,64,3: | 1 |

| 10 | Construction of a Multipurpose M13KE Phage Display System显示文摘In this study,a multipurpose M13 KE phage display vector was constructed from wild-type M13 KE phage for long peptide or protein display libraries without helper phage to expand the scope of targeted high-throughput screening. Based on the relationship between the structure and function of minor coat protein of wild-type M13KE( wt-p III),a truncated gene III( tg III) encoding minor coat protein from M13 KE phage was cloned. A fusion gene fragment harboring a lac / tac promoter,signal peptide and C-terminal region sequence of g III was assembled with SOEing-PCR( splice-overlapping-extension polymerase chain reaction)method and inserted into M13 KE vector. SDS-PAGE and Western blot analysis with anti-M13 p III monoclonal antibody were employed to detect the expression of recombinant protein. c-Myc and HA tag sequences were fused into the recombinant protein. The results showed that tg III was inserted into an unessential region of M13 KE. According to the results of SDS-PAGE and Western blot with anti-M13 p III antibody,p III was expressed by wt-g III and tg III. g III harboring two tags expressed both c-Myc and HA peptides using SDS-PAGE and Western blot with the corresponding monoclonal antibodies. In this study,a multipurpose M13 KE phage display system was successfully constructed,which could express both short and long peptide libraries without helper phage. | 11 | Phenological metrics-based crop classification using HJ-1 CCD images and Landsat 8 imagery显示文摘Crop type data are an important piece of information for many applications in agriculture.Extracting crop type using remote sensing is not easy because multiple crops are usually planted into small parcels with limited availability of satellite images due to weather conditions.In this research,we aim at producing crop maps for areas with abundant rainfall and small-sized parcels by making full use of Landsat 8 and HJ-1 charge-coupled device(CCD)data.We masked out non-vegetation areas by using Landsat 8 images and then extracted a crop map from a longterm time-series of HJ-1 CCD satellite images acquired at 30-m spatial resolution and two-day temporal resolution.To increase accuracy,four key phenological metrics of crops were extracted from time-series Normalized Difference Vegetation Index curves plotted from the HJ-1 CCD images.These phenological metrics were used to further identify each of the crop types with less,but easier to access,ancillary field survey data.We used crop area data from the Jingzhou statistical yearbook and 5.8-m spatial resolution ZY-3 satellite images to perform an accuracy assessment.The results show that our classification accuracy was 92%when compared with the highly accurate but limited ZY-3 images and matched up to 80%to the statistical crop areas. | Xiaochun Zhang Qinxue Xiong Liping Di Junmei Tang Jin Yang Huayi Wu Yan Qin Rongrui Su Wei Zhou | 2018 | International Journal of Digital Earth2018,11,12: | 0 |
