| 2 | Association of a SLC30A8 Genetic Variant with Monotherapy of Repaglinide and Rosiglitazone Effect in Newly Diagnosed Type 2 Diabetes Patients in China显示文摘Objective To investigate a potential relationship between Solute carrier family 30 (zinc transporter) member 8 (SLC30A8) rs13266634 variant and efficacy of rosiglitazone or repaglinide in treating newly diagnosed Chinese type 2 diabetes patients.Methods A total of 209 diabetic patients without any antihyperglycemic history were recruited and treated with repaglinide or rosiglitazone randomly for 48 weeks (104 and 105 patients,respectively).Anthropometric measurements and clinical laboratory tests were carried out before and after the treatment.An non-synonymous variant rs13266634 was genotyped by matrix-assisted laser desorption ionization-time of flight mass spectroscopy.Results Ninety-one patients in repaglinide group and ninety-three patients in rosiglitazone group completed the study.Δ value of homeostasis model assessment of beta cell function (HOMA-B)and Δ value of fasting proinsulin levels were statistically significant between three genotype groups (P=0.0149 and 0.0246,respectively) after rosiglitazone treatment.However,no genotype association was observed in the repaglinide or rosiglitazone group with other parameters.Conclusion The SLC30A8 variant was associated with the efficacy of insulin sensitizer monotherapy on insulin secretion in patients with newly diagnosed type 2 diabetes mellitus in Shanghai,China. | JIANG Feng LI Qing HU Cheng ZHANG Rong WANG Cong Rong YU Wei Hui LU Jing Yi TANG Shan Shan BAO Yu Qian XIANG Kun San JIA Wei Ping | 2012 | Biomedical and Environmental Sciences2012,25,1: | 8 |

Ltd., Kyoto, Japan), Eyesi direct ophthalmoscope simulator(VRmagic, Gmb H, Mannheim,Germany), Eyesi indirect ophthalmoscope simulator(VRmagic, Gmb H, Mannheim, Germany) and Eyesi cataract simulators(VRmagic, Gmb H, Mannheim,Germany). These simulators are thought to be able to reduce the initial learning curve for the ophthalmology training but further research will need to be conducted to assess the effectiveness of the simulation-assisted ophthalmology training. | 7 | Effects of palbociclib on oral squamous cell carcinoma and the role of PIK3CA in conferring resistance显示文摘Objective: Lack of effective therapies remains a problem in the treatment of oral squamous cell carcinoma(OSCC), especially in patients with advanced tumors. OSCC development is driven by multiple aberrancies within the cell cycle pathway, including amplification of cyclin D1 and loss of p16. Hence, cell cycle inhibitors of the CDK4/6-cyclin D axis are appealing targets for OSCC treatment. Here, we determined the potency of palbociclib and identified genetic features that are associated with the response of palbociclib in OSCC.Methods: The effect of palbociclib was evaluated in a panel of well-characterized OSCC cell lines by cell proliferation assays and further confirmed by in vivo evaluation in xenograft models. PIK3CA-mutant isogenic cell lines were used to investigate the effect of PIK3CA mutation towards palbociclib response.Results: We demonstrated that 80% of OSCC cell lines are sensitive to palbociclib at sub-micromolar concentrations.Consistently, palbociclib was effective in controlling tumor growth in mice. We identified that palbociclib-resistant cells harbored mutations in PIK3CA. Using isogenic cell lines, we showed that PIK3CA mutant cells are less responsive to palbociclib as compared to wild-type cells with concurrent upregulation of CDK2 and cyclin E1 protein levels. | 9 | A review of the thermal stability of metastable austenite in steels:Martensite formation显示文摘Metastable austenite plays a critical role in achieving improved combinations of high strength and high ductility/toughness in the design of advanced high-strength steels(AHSS). The thermal stability of metastable austenite determines the transformation characteristics of AHSS and thus primarily determines the microstructure evolution during complex processes, e.g., the quenching and partitioning process, to achieve the desirable microstructure. This study provides a review of the thermal stability of austenite and its influence on martensitic transformation from both experimental and theoretical modeling perspectives. From the experimental perspective, factors affecting the thermal stability are analyzed,the relative sensitivities are compared, and their corresponding mechanisms are discussed. From the theoretical modeling perspective, the most representative kinetic models that describe athermal and isothermal martensitic transformation are reviewed. The advantages, shortcomings, and applicability of each model are discussed. | 15 | Kinetic Characteristics of Neutralizing Antibody Responses Vary among Patients with COVID-19显示文摘Objective The coronavirus disease 2019(COVID-19) pandemic continues to present a major challenge to public health. Vaccine development requires an understanding of the kinetics of neutralizing antibody(NAb) responses to severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).Methods In total, 605 serum samples from 125 COVID-19 patients(from January 1 to March 14, 2020)varying in age, sex, severity of symptoms, and presence of underlying diseases were collected, and antibody titers were measured using a micro-neutralization assay with wild-type SARS-CoV-2.Results NAbs were detectable approximately 10 days post-onset(dpo) of symptoms and peaked at approximately 20 dpo. The NAb levels were slightly higher in young males and severe cases, while no significant difference was observed for the other classifications. In follow-up cases, the NAb titer had increased or stabilized in 18 cases, whereas it had decreased in 26 cases, and in one case NAbs were undetectable at the end of our observation. Although a decreasing trend in NAb titer was observed in many cases, the NAb level was generally still protective.Conclusion We demonstrated that NAb levels vary among all categories of COVID-19 patients.
