1Salinity Acclimation Induces Reduced Energy Metabolism,Osmotic Pressure Regulation and Transcriptional Reprogramming in Hypotrichida Ciliate Gastrostyla setifera显示文摘Coastal and estuarine protists are frequently exposed to salinity undulation.While the tolerance and stress responses of microalgae to salinity have been extensively studied,there have been scarce studies on the physiological response of heterotrophic protists to salinity stressing.In this study,we investigated the physiological response of the heterotrophic ciliate Gastrostyla setifera to a salinity of 3,via a transcriptomic approach.The first transcriptome of genus Gastrostyla was obtained utilizing a group of manually isolated ciliate individuals(cells)and RNA-seq technique.The completeness of the transcriptome was verified.Differentially expressed gene(DEG)analysis was performed among the transcriptomes of G.setifera acclimated in saline water(salinity 3)and those cultured in fresh water.The results demonstrated a significant alternation in gene transcription,in which the ciliate exhibits a transcripttomic acclimation in responding salinity stressing.The up-regulated DEGs were enriched in the pathways of cytoskeleton proteins,membrane trafficking,protein kinases and protein phosphatases.These may represent enhanced functions of ion transport,stress response and cell protections.Pathways involved in energy metabolism and biosynthesis were markedly down-regulated,reflecting decreased cell activity.Particularly,we detected significantly down-regulated genes involved in several pathways of amino acid catabolism,which may lead to accumulation of amino acids in the ciliate cell.Amino acid could act as compatible solutes in the cytoplasm to maintain the osmotic balance in saline water.Overall,this work is an initial exploration to the molecular basis of the heterotrophic protist responding to salinity stressing.The result sheds light on the mechanisms of enhancement of cell protection,reduction of cell activity,and osmotic pressure regulation in ciliates acclimated to salinity.JI Xin BI Luping ZOU Songbao LI Wenlu JI Daode ZHANG Qianqian 2024Journal of Ocean University of China2024,23,2:0
2Exploration and Validation of the Potential Downstream Genes Underlying ipa1-2D Locus for Rice Panicle Branching显示文摘In recent years,some super hybrid rice varieties were bred with strong culms and large panicles,which are mainly contributed by the ipa1-2D locus.A gain-of-function allele of OsSPL14 is the ipa1-2D and it can greatly increase the panicle primary branch number.However,the key downstream genes mediating this trait variation are not fully explored.In this study,we developed high-quality near-isogenic lines(NILs)with a difference of only 30 kb chromosomal segment covering the ipa1-2D locus.Using the NILs,we explored the impact of ipa1-2D on five sequential stages of early inflorescence development,and found that the locus can greatly enhance the initiation of primary branch meristems.A transcriptomic analysis was performed to unveil the downstream molecular network of ipa1-2D,and 87 genes were found differentially expressed,many of which are involved in metabolism and catalysis processes.In addition,transgenic lines of overexpression and RNA interference were generated to shape different levels of OsSPL14.They were also used to validate the expression variation explored by transcriptome.Based on the gene annotation,twelve potential downstream targets of ipa1-2D were selected,and their expression variation was confirmed by qRT-PCR analysis both in NILs and transgenic lines.This research expands the molecular network underlying ipa1-2D and provides novel gene information which might be involved in the control of panicle branching.We discussed the potential function of identified genes and highlighted their values for future function exploration and breeding application.Lin Zhang Dong Xie Zhong Bian Yiting Zou Han Zhou Wenlu Cai Hadi Yeilaghi Xiaolei Fan Changquan Zhang Qiaoquan Liu 2021Phyton-International Journal of Experimental Botany2021,90,3:0
3The presence of foci of Rickettsia conorii infection in China显示文摘China is not considered as an endemic area of Rickettsia conorii,so there is no routine clinical way to diagnose this infection.This study aims to determine whether 2 febrile patients who had a tick bite in East China were indeed infected with R.conorii.The citrate synthase gene(gltA)was amplified with universal rickettsial primers by real-time fluorescent PCR from the patients’blood samples.Nested PCR was used to amplify the outer membrane protein A gene(ompA)for positive specimens.PCR products were further identified and analyzed through nucleic acid sequencing.Positive amplification of the gltA and ompA genes was found in both patients.The nucleotide sequences(303 bp)of the ompA gene of the 2 patients had high homology(99%)with the R.conorii Indian tick typhus strain in GenBank.A more than 4-fold increase in IgG against R.conorii provided supportive evidence of SFG Rickettsia infection.And the rapid recovery after doxycycline treatment also supported a rickettsial cause for the disease.Physicians in East China should be aware of human infections with R.conorii.PCR-based diagnostic methods offer a rapid and precise way to diagnose rickettsiosis,improving patient identification and management.Nannan Xu Hui Liu Chunmei Qu Sai Wen Wenlu Zou Caiyun Chang Gang Wang 2023Infectious Medicine2023,2,4:0
