| 1 | Broaching Performance of Superalloy GH4169 Based on FEM显示文摘The nickel-based superalloy GH4169 is an important material for high temperature applications in the aerospace industry. However, due to its poor machinability, GH4169 is hard to be cut and generates saw-tooth chips during high speed machining, which could significantly affect the dynamic cutting force, cutting temperature fluctuation, tool life, and the surface integrity of the parts. In this paper, the saw-tooth chip formation mechanism of superalloy GH4169 was investigated by the elasto-viscoplastic finite element method (FEM). Using the finite element software of ABAQUS/Explicit, the deformation of the part during high speed machining was simulated. The effective plastic strain, the temperature field, the stress distribution, and the cutting force were analyzed to determine the influence of the cutting parameters on the saw-tooth chip formation. | 3 | Sarm1-mediated neurodegeneration within the enteric nervous system protects against local inflammation of the colon显示文摘Axonal degeneration is one of the key features of neu-rodegenerative disorders.In the canonical view,axonal degeneration destructs neural connections and promotes detrimental disease defects.Here,we assessed the enteric nervous system(ENS)of the mouse,nonhuman primate,and human by advanced 3D imaging.We observed the profound neurodegeneration of catecholaminergic axons in human colons with ulcerative colitis,and similarly,in mouse colons during acute dextran sulfate sodium-induced colitis.However,we unexpectedly revealed that blockage of such axonal degeneration by the Sarml deletion in mice exacerbated the colitis condition.In contrast,pharmacologic ablation or chemogenetic inhibition of catecholaminergic axons suppressed the colon inflammation.We further showed that the catecholaminergic neurotransmitter norepinephrine exerted a pro-inflammatory function by enhancing the expression of IL-17 cytokines.Together,this study demonstrated that Sarm1-mediated neurodegeneration within the ENS mitigated local inflammation of the colon,uncovering a previously-unrecognized beneficial role of axonal degeneration in this disease context. | Yue Sun Qi Wang Yi Wang Wenran Ren Ying Cao Jiali Li Xin Zhou Wei Fu Jing Yang | 2021 | Protein & Cell2021,12,8: | 5 |
| 4 | Curcumin inhibits the replication of enterovirus 71 in vitro显示文摘Human enterovirus 71(EV71)is the main causative pathogen of hand,foot,and mouth disease(HFMD)in children.The epidemic of HFMD has been a public health problem in Asia-Pacific region for decades,and no vaccine and effective antiviral medicine are available.Curcumin has been used as a traditional medicine for centuries to treat a diversity of disorders including viral infections.In this study,we demonstrated that curcumin showed potent antiviral effect again EV71.In Vero cells infected with EV71,the addition of curcumin significantly suppressed the synthesis of viral RNA,the expression of viral protein,and the overall production of viral progeny.Similar with the previous reports,curcumin reduced the production of ROS induced by viral infection.However,the antioxidant property of curcumin did not contribute to its antiviral activity,since N-acetyl-L-cysteine,the potent antioxidant failed to suppress viral replication.This study also showed that extracellular signal-regulated kinase(ERK)was activated by either viral infection or curcumin treatment,but the activated ERK did not interfere with the antiviral effect of curcumin,indicating ERK is not involved in the antiviral mechanism of curcumin.Unlike the previous reports that curcumin inhibited protein degradation through ubiquitin–proteasome system(UPS),we found that curcumin had no impact on UPS in control cells.However,curcumin did reduce the activity of proteasomes which was increased by viral infection.In addition,the accumulation of the short-lived proteins,p53 and p21,was increased by the treatment of curcumin in EV71-infected cells.We further probed the antiviral mechanism of curcumin by examining the expression of GBF1 and PI4KB,both of which are required for the formation of viral replication complex.We found that curcumin significantly reduced the level of both proteins.Moreover,the decreased expression of either GBF1 or PI4KB by the application of siRNAs was sufficient to suppress viral replication.We also demonstrated that curcumin showed anti-apoptotic activity at the early stage of viral infection.The results of this study provide solid evidence that curcumin has potent anti-EV71 activity.Whether or not the down-regulated GBF1 and PI4KB by curcumin contribute to its antiviral effect needs further studies. | Ying Qin Lexun Lin Yang Chen Shuo Wu Xiaoning Si Heng Wu Xia Zhai Yan Wang Lei Tong Bo Pan Xiaoyan Zhong Tianying Wang Wenran Zhao Zhaohua Zhong | 2014 | Acta Pharmaceutica Sinica B2014,4,4: | 5 |
| 5 | Stress Granule Formation is One of the Early Antiviral Mechanisms for Host Cells Against Coxsackievirus B Infection显示文摘Stress granules(SGs) are intracellular granules formed when cellular translation is blocked and have been reported to be involved in a variety of viral infections. Our previous studies revealed that SGs are involved in the coxsackievirus B(CVB)infection process, but the role of SGs in CVB infection has not been fully explored. In this study, we found that CVB type 3(CVB3) could induce SG formation in the early phase of infection. Results showed that levels of CVB3 RNA and protein were significantly inhibited during the early stage of CVB3 infection by the elevated formation of SGs, while viral RNA and protein synthesis were significantly promoted when SG formation was blocked. | 7 | Expression Profile and Function Analysis of Long Non-coding RNAs in the Infection of Coxsackievirus B3显示文摘The roles of IncRNAs in the infection of enteroviruses have been barely demonstrated.In this study,we used coxsackievirus B3(CVB3),a typical enterovirus,as a model to investigate the expression profiles and functional roles of IncRNAs in enterovirus infection.We profiled IncRNAs and mRNA expression in CVB3-infected HeLa cells by IncRNA-mRNA integrated microarrays.As a result,700 differentially expressed IncRNAs(431 up-regulated and 269 down-regulated)and 665 differentially expressed mRNAs(299 up-regulated and 366 down-regulated)were identified in CVB3 infection.Then we performed IncRNA-mRNA integrated pathway analysis to identify potential functional impacts of the differentially expressed mRNAs,in which IncRNA-mRNA correlation network was built.According to IncRNA-mRNA correlation,we found that XLOC-001188,an IncRNA down-regulated in CVB3 infection,was negatively correlated with NFAT5 mRNA,an anti-CVB3 gene reported previously.This interaction was supported by qPCR detection following siRNA-mediated knockdown of XLOC-001188,which showed an increase of NFAT5 mRNA and a reduction of CVB3 genomic RNA.In addition,we observed that four most significantly altered IncRNAs,SNHG11,RP11-145F16.2,RP11-1023L17.1 and RP11-1021N1.2 share several common correlated genes critical for CVB3 infection,such as BRE and IRF2BP1.In all,our studies reveal the alteration of IncRNA expression in CVB3 infection and its potential influence on CVB3 replication,providing useful information for future studies of enterovirus infection. | Lei Tong Ye Qiu Hui Wang Yunyue Qu Yuanbo Zhao Lexun Lin Yan Wang Weizhen Xu Wenran Zhao Hongyan He Guangze Zhao Mary HZhang Decheng Yang Xingyi Ge Zhaohua Zhong | 2019 | Virologica Sinica2019,34,6: | 2 |

| 11 | Proteomic profiling of cotton fiber developmental transition from cell elongation to secondary wall deposition显示文摘Cotton fiber developmental transition from elongation to secondary cell wall biosynthesis is a critical growth shifting phase that affects fiber final length,strength,and other properties.Morphological dynamic analysis indicated an asynchronous fiber developmental pattern between two most important commercial cotton species,Gossypium hirsutum(Gh)and G.barbadense(Gb).Using isobaric tags for relative and absolute quantitation techniques,we examined the temporal changes of protein expression at three representative development periods(15-19,19-23,and 23-27 dpa)in both species.Strikingly,a large proportion of differentially expressed proteins(DEPs)were identified at 19-23 dpa in Gh and at 23-27 dpa in Gb,corresponding to their fiber developmental transition timing.To better understand fiber transitional development,we comparatively analyzed those DEPs in 19-23 dpa of Gh vs.in 23-27 dpa of Gb,and noted that these cotton species indeed share fundamentally similar fiber developmental features under the biological processes.We also showed that there are limited overlaps in both specific upregulated and downregulated proteins between the two species,suggesting species-specific protein regulations in the development process.Proteomic profiling results revealed dynamic changes of several key proteins and biological processes that are potentially correlated with fiber developmental transition.During the transition,upregulated proteins are mainly involved in carbohydrate/energy metabolism,oxidation-reduction,cytoskeleton,protein turnover,Ca2+signaling,etc.,whereas important downregulated proteins are mostly involved in phenylpropanoid and flavonoid secondary metabolism pathways.The gene expressions of several changed proteins in this key stage were also examined by quantitative reverse transcription polymerase chain reaction.Overall,the present study provides accurate pictures of the regulatory networks of functional proteins during the fiber developmental transition,therefore highlighting candidate genes/proteins and related pathways for the cotton fiber improvement. | Xiaoyun Zhou Wenran Hu Bo Li Yang Yang Yong Zhang Kieran Thow Ling Fan Yanying Qu | 2019 | Acta Biochimica et Biophysica Sinica2019,51,11: | 1 |
