1Feasibility and safety of autologous bone marrow mononuclear cell transplantation in patients with advanced chronic liver disease显示文摘AIM: To evaluate the safety and feasibility of bone marrow cell (BMC) transplantation in patients with chronic liver disease on the waiting list for liver transplantation. METHODS: Ten patients (eight males) with chronic liver disease were enrolled to receive infusion of autologous bone marrow-derived cells. Seven patients were classified as Child-Pugh B and three as Child-Pugh C. Baseline assessment included complete clinical and laboratory evaluation and abdominal MRI. Approximately 50 ml of bone marrow aspirate was prepared by centrifugation in a ficoll-hypaque gradient. At least of 100 millions of mononuclear-enriched BMCs were infused into the hepatic artery using the routine technique for arterial chemoembolization for liver tumors. Patients were followed up for adverse events up to 4 mo. RESULTS: The median age of the patients was 52 years (range 24-70 years). All patients were discharged 48 h after BMC infusion. Two patients complained ofmild pain at the bone marrow needle puncture site. No other complications or specific side effects related to the procedure were observed. Bilirubin levels were lower at 1 (2.19 ± 0.9) and 4 mo (2.10 ± 1.0) after cell transplantation that baseline levels (2.78 ± 1.2). Albumin levels 4 mo after BMC infusion (3.73 ± 0.5) were higher than baseline levels (3.47 ± 0.5). International normalized ratio (INR) decreased from 1.48 (SD = 0.23) to 1.43 (SD = 0.23) one month after cell transplantation. CONCLUSION: BMC infusion into hepatic artery of patients with advanced chronic liver disease is safe and feasible. In addition, a decrease in mean serum bilirubin and INR levels and an increase in albumin levels are observed. 9The Interrelation of Urban Health Indicators and COVID-19:A Study of Passo Fundo/RS显示文摘The COVID-19(Coronavirus Disease 19)pandemic has demonstrated that cities are at the center of major contemporary events.The epidemiological crisis has highlighted the importance of the urban environment,challenging public managers on managing cities.An initiative that aims to assist in this management process is the concept of Smart Cities,which uses ICT(Information and Communication Technology)as tools for transforming urban dynamics,and through urban indicators,measures information about cities.Thus,the research aimed to analyze the health indicators of Passo Fundo/RS,seeking to analyze the interrelationship of these indicators with the epidemiological data from COVID-19.In the methodology,multi-method procedures were applied,using the indicators of the Connected Smart Cities Ranking as reference,as well as a regional selection of medium-sized cities in the southern region of Brazil.The results show that the health indices of Passo Fundo are,for the most part,lower than those of the analyzed cities,with the indicator related to Population Coverage of the Family Health team as the main weakness.However,it also presents satisfactory indices as is the case of the indicator of Beds/1,000 inhabitants.Regarding the epidemiological picture of COVID-19,Passo Fundo had a high lethality rate when compared to the other analyzed cities.Letícia Müller Thaísa Leal da Silva Wilson Levy Braga da Silva Neto 2021Journal of Civil Engineering and Architecture2021,15,10:0

12In vitro antileishmanial effects of Physalis angulata root extract on Leishmania infantum显示文摘Objective: In the present study, we evaluated the effects of the aqueous extract of Physalis angulata root(AEPa) on Leishmania infantum proliferation, morphology, and the driving mechanism in leishmanicidal activity and modulatory action on macrophages.Methods: L. infantum promastigotes were treated with 50 and 100 μg/mL AEPa for 72 h and then antipromastigote assay was performed by counts in a Newbauer chamber, morphological changes were analyzed by transmission electron microscopy and the mechanism of the leishmanicidal activity was detected. In addition, macrophages were infected with L. infantum and were used to evaluate anti-amastigote activity of AEPa and effects of AEPa on cytokine secretion after 72-hour treatment.Results: Treatment with AEPa reduced the numbers of L. infantum promastigotes(50% inhibitory concentration(IC_(50))= 65.9 μg/mL; selectivity index(SI) = 22.1) and amastigotes(IC_(50)=37.9 μg/mL; SI = 38.5)compared with the untreated control. Amphotericin B reduced 100% of the promastigote numbers after72 h of treatment(IC_(50) = 0.2 μg/mL). AEPa induced several morphological changes and increased the production of reactive oxygen species and apoptotic death in promastigotes after treating for 72 h.AEPa(100 μg/mL) promoted tumor necrosis factor-α secretion in macrophages infected with L. infantum after 72 h of treatment, but did not induce an increase in this cytokine in noninfected macrophages.
