2Blunted nitric oxide regulation in Tibetans under high-altitude hypoxia显示文摘Nitric oxide(NO) is an important molecule for vasomotor tone, and elevated NO signaling was previously hypothesized as a unique and adaptive physiological change in highland Tibetans. However, there has been lack of NO data from Tibetans living at low altitude and lowlander immigrants living at high altitude, which is crucial to test this hypothesis. Here, through cross-altitude(1990–5018 m) and cross-population(Tibetans and Han Chinese) analyses of serum NO metabolites(NOx) of 2086 individuals, we demonstrate that although Tibetans have a higher serum NOx level compared to lowlanders, Han Chinese immigrants living at high altitude show an even higher level than Tibetans. Consequently, our data contradict the previous proposal of increased NO signaling as the unique adaptive strategy in Tibetans.Instead, Tibetans have a relatively lower circulating NOx level at high altitude. This observation is further supported by data from the hypoxic experiments using human umbilical vein endothelial cells and gene knockout mice. No difference is detected between Tibetans and Han Chinese for endothelial nitric oxide synthase(e NOS), the key enzyme for circulating NO synthesis, suggesting that e NOS itself is unlikely to be the cause. We show that other NO synthesis-related genes(e.g. GCH1) carry Tibetan-enriched mutations significantly associated with the level of circulating NOx in Tibetans. 3De novo assembly of a Tibetan genome and identification of novel structural variants associated with high-altitude adaptation显示文摘Structural variants(SVs) may play important roles in human adaptation to extreme environments such as high altitude but have been under-investigated. Here, combining long-read sequencing with multiple scaffolding techniques, we assembled a high-quality Tibetan genome(ZF1), with a contig N50 length of 24.57 mega-base pairs(Mb) and a scaffold N50 length of 58.80 Mb. The ZF1 assembly filled 80 remaining N-gaps(0.25 Mb in total length) in the reference human genome(GRCh38). Markedly, we detected17 900 SVs, among which the ZF1-specific SVs are enriched in GTPase activity that is required for activation of the hypoxic pathway. Further population analysis uncovered a 163-bp intronic deletion in the MKL1 gene showing large divergence between highland Tibetans and lowland Han Chinese. This deletion is significantly associated with lower systolic pulmonary arterial pressure, one of the key adaptive physiological traits in Tibetans. Moreover, with the use of the high-quality de novo assembly, we observed a much higher rate of genome-wide archaic hominid(Altai Neanderthal and Denisovan) shared non-reference sequences in ZF1(1.32%–1.53%) compared to other East Asian genomes(0.70%–0.98%),reflecting a unique genomic composition of Tibetans. One such archaic hominid shared sequence-a662-bp intronic insertion in the SCUBE2 gene-is enriched and associated with better lung function(the FEV1/FVC ratio) in Tibetans. 4Adsorption of phenol from aqueous solution by a hierarchical micro-nano porous carbon material显示文摘A hierarchical micro-nano porous carbon material (MNC) was prepared using expanded graphite (EG), sucrose, and phosphoric acid as raw materials, followed by sucrose-phosphoric acid solution impregnation, solidification, carbonization and activation. Nitrogen adsorption and mercury porosimetry show that mixed nanopores and micropores coexist in MNC with a high specific surface area of 1978 m2·g-1 and a total pore volume of 0.99 cm3·g-1. In addition, the MNC is found to consist of EG and activated carbon with the latter deposited on the interior and the exterior surfaces of the EG pores. The thickness of the activated carbon layer is calculated to be about one hundred nanometers and is further confirmed by scanning electron microscope (SEM) and transmission election microscope (TEM). A maximum static phenol adsorption of 241.2 mg·g-1 was obtained by using MNC, slightly higher than that of 220.4 mg·g-1 by using commercial activated carbon (CAC). The phenol adsorption kinetics were investigated and the data fitted well to a pseudo-second-order model. Also, an intra-particle diffusion mechanism was proposed. Furthermore, it is found that the dynamic adsorption capacity of MNC is nearly three times that of CAC. 5Large-scale Ni-MOF derived Ni_(3)S_(2) nanocrystals embedded in N-doped porous carbon nanoparticles for high-rate Na+ storage显示文摘Metal organic framework(MOF) has been confirmed as the promising precursor to develop the conve rsion-typed anode mate rials of sodium-ion batteries(SIBs) because of the tunable structure design and simple functional modification.Here,we prepare the ultrasmall Ni_(3)S_(2) nanocrystals embedded into N-doped porous carbon nanoparticles using the scalable Ni-MOF as precursor(denoted as Ni_(3)S_(2)@NPC).The ultrasmall size of Ni_(3)S_(2) can work for accelerated electro n/ion transfer to facilitate the electrochemical reaction kinetics.Moreover,the robust conductivity network originated from N-doped porous carbon nanoparticles can not only improve the electron conductivity,but also enhance the electrode integrity and stability of the electrode/electrolyte interface.In addition,the N heteroatoms provide extra Na storage sites.Accordingly,the electrode delivers the obviously competitive capacities and high-power output with respect to the currently reported Ni_(3)S_(2)/C composites.This study provides a scalable and universal strategy to develop the advanced transition metal sulfides for practically feasible SIBs.Miao Yang Qiuli Ning Chaoying Fan Xinglong Wu 2021Chinese Chemical Letters2021,32,2:3
6Study on the Supply Efficiency of Rural Public Service in China Based on Three-stage DEA Model显示文摘Improving the supply efficiency of rural public service is an important way to solve the severe shortage of rural public service. In this article,we use three-stage DEA model to carry out empirical research of the supply efficiency of rural public service in 31 provinces and regions of China. The results show that if without control over exogenous environment variables and random brunt,the classic DEA method will overestimate the rural public service efficiency; after controlling the impact of the external environmental factors,the mean of supply efficiency of rural public service in 31 provinces and regions of China is 0. 697; the improved rural per capita income,population density,population size and the educational level of residents,is a significantly favorable factor for enhancing the supply efficiency of rural public service,while the increase in the proportion of fiscal spending on rural public service to GDP plays no significant role in improving the rural public service efficiency; according to their efficiency type,the provinces and regions should adopt some measures,such as improving the management level or expanding the supply scale,to improve the supply efficiency.Zongbing DENG Chaoying WU Junliang Zhang Ju WANG 2014Asian Agricultural Research2014,6,1:3

13Transcriptomic responses in resistant and susceptible maize infected with Fusarium graminearum显示文摘Gibberella ear rot(GER)caused by Fusarium graminearum(teleomorph Gibberella zeae)is a common maize disease that not only severely reduces grain yield but also contaminates maize grain with mycotoxins.We investigated the molecular mechanism underlying the host defense responses against pathogen infection using comparative transcriptomic analysis.We injected F.graminearum spore suspensions into plants of resistant(IBM-81)and a susceptible(IBM-85)maize inbred line after pollination and performed RNA-seq 48,72,and 96 h after inoculation.Respectively 487 and 410 differentially expressed genes(DEGs)were induced in the resistant and susceptible lines across three time points,indicating that a stronger defense response was activated in the resistant than in the susceptible line.Among them,198 genes commonly induced in the two lines were subjected to pathway analysis,revealing that most of the DEGs were closely associated with defense and a wide range of metabolic activities.DEGs associated with pathogenesis-related protein 1(PR1)and regulation of salicylic acid were significantly enriched during F.graminearum infection,suggesting that these DEGs play dominant roles in maize resistance to GER.Our results provide a resource for future gene discovery and facilitate elucidation of the complex defense mechanisms involved in resistance to GER.Guangsheng Yuan Xiujing He Hui Li Kui Xiang Li Liu Chaoying Zou Haijian Lin Jiali Wu Zhiming Zhang Guangtang Pan 2020The Crop Journal2020,8,1:1

19Expression of inducible nitric oxide synthase in trophoblasts and deciduas in early medical abortion显示文摘The purpose of this study was to investigate the expression of inducible nitric oxide synthase(iNOS)in trophoblasts and deciduas in early medical abortion,and study the relationship of medical abortion through mifepristone and iNOS in early pregnancy.Expression of iNOS in trophoblasts and deciduas was detected by both in situ hybridization and immunohistochemical assay in 40 patients(experimental group);the positive expression of iNOS was represented by number density(N/S)and positive unit(Pu)using computer color magic image analysis system(CMIAS).All results were compared with those obtained from vacuum aspiration.In the experi-mental group,N/S and Pu in trophoblasts were 0.120�0.010 and 15.3�2.6,respectively,while in the control group,they were 0.021�0.003 and 3.1�0.5,respectively,and there were significant differences between the two groups.By immunohistochemical assay,N/S and Pu were 0.090�0.010,10.24�1.55 vs 0.016�0.002,1.26�0.33 in the trophoblasts of the two groups;there were also significant differences between the two groups.There were lower iNOS expression in deciduas by in situ hybridization and immunohistochem-ical assay,and the difference between the two groups was not significant.It was concluded that mifepristone induced medical abortion through the expression of iNOS in trophoblasts but not in deciduas.Geqing XIA Chaoying WU 2009Frontiers of Medicine2009,3,2:0
