2Capecitabine maintenance therapy for XT chemotherapy-sensitive patients with metastatic triple-negative breast cancer显示文摘Objective: To investigate the efficacy and safety of capecitabine maintenance therapy(MT) after initial capecitabine plus docetaxel(XT) chemotherapy in patients with metastatic triple-negative breast cancer(m TNBC).Methods: Fifty-five m TNBC patients treated with XT chemotherapy between May 2007 and June 2013 were retrospectively analyzed. When initial disease control was achieved by the combination chemotherapy, capecitabine was continued for 32 patients(MT), while 23 patients remained without any treatment(nonMT). We compared progression-free survival(PFS) and safety of both groups.Results: The median PFS of 55 patients was 8.1 months, overall median PFS time of 32 patients in the capecitabine MT group and 23 in the non-MT group was 10.1 vs. 6.7 months(P=0.032), respectively. When compared PFS time of maintenance treatment, single-agent capecitabine prolonged PFS by 7.1 months, for non-MT patients, the PFS without any treatment was 3.1 months, and this between-group difference was statistically significant(P=0.003). Adverse events, including of hematologic toxicity, gastrointestinal toxicities, hand-foot syndrome and abnormal liver function were not significantly different between two groups.Conclusions: After initial disease control was achieved with the XT combination chemotherapy, capecitabine MT can significantly prolong PFS time with a favorable safety profile in m TNBC patients.Xu Liang Lijun Di Guohong Song Ying Yan Chaoying Wang Hanfang Jiang Huiping Li 2014Chinese Journal of Cancer Research2014,26,5:8
3Blunted nitric oxide regulation in Tibetans under high-altitude hypoxia显示文摘Nitric oxide(NO) is an important molecule for vasomotor tone, and elevated NO signaling was previously hypothesized as a unique and adaptive physiological change in highland Tibetans. However, there has been lack of NO data from Tibetans living at low altitude and lowlander immigrants living at high altitude, which is crucial to test this hypothesis. Here, through cross-altitude(1990–5018 m) and cross-population(Tibetans and Han Chinese) analyses of serum NO metabolites(NOx) of 2086 individuals, we demonstrate that although Tibetans have a higher serum NOx level compared to lowlanders, Han Chinese immigrants living at high altitude show an even higher level than Tibetans. Consequently, our data contradict the previous proposal of increased NO signaling as the unique adaptive strategy in Tibetans.Instead, Tibetans have a relatively lower circulating NOx level at high altitude. This observation is further supported by data from the hypoxic experiments using human umbilical vein endothelial cells and gene knockout mice. No difference is detected between Tibetans and Han Chinese for endothelial nitric oxide synthase(e NOS), the key enzyme for circulating NO synthesis, suggesting that e NOS itself is unlikely to be the cause. We show that other NO synthesis-related genes(e.g. GCH1) carry Tibetan-enriched mutations significantly associated with the level of circulating NOx in Tibetans. 4Transcriptome analysis reveals key differentially expressed genes involved in wheat grain development显示文摘Wheat seed development is an important physiological process of seed maturation and directly affects wheat yield and quality. In this study, we performed dynamic transcriptome microarray analysis of an elite Chinese bread wheat cultivar(Jimai 20) during grain development using the Gene Chip Wheat Genome Array. Grain morphology and scanning electron microscope observations showed that the period of 11–15 days post-anthesis(DPA) was a key stage for the synthesis and accumulation of seed starch. Genome-wide transcriptional profiling and significance analysis of microarrays revealed that the period from 11 to 15 DPA was more important than the 15–20 DPA stage for the synthesis and accumulation of nutritive reserves.Series test of cluster analysis of differential genes revealed five statistically significant gene expression profiles. Gene ontology annotation and enrichment analysis gave further information about differentially expressed genes, and Map Man analysis revealed expression changes within functional groups during seed development. Metabolic pathway network analysis showed that major and minor metabolic pathways regulate one another to ensure regular seed development and nutritive reserve accumulation. We performed gene co-expression network analysis to identify genes that play vital roles in seed development and identified several key genes involved in important metabolic pathways. The transcriptional expression of eight key genes involved in starch and protein synthesis and stress defense was further validated by q RT-PCR. 5A semiempirical algorithm for SAR wave height retrieval and its validation using Envisat ASAR wave mode data显示文摘Many synthetic aperture radar(SAR) wave height retrieval algorithms have been developed.However,the wave height retrievals from most existing methods either depend on other input as the first guess or are restricted to the long wave regime.A semiempirical algorithm is presented,which has the objective to estimate the wave height from SAR imagery without any prior knowledge.The proposed novel algorithm was developed based on the theoretical SAR ocean wave imaging mechanism and the empirical relation between two types of wave period.The dependency of the proposed model on radar incident and wave direction was analyzed.For Envisat advanced synthetic aperture radar(ASAR) wave mode data,the model can be reduced to the simple form with two input parameters,i.e.,the cutoff wavelength and peak wavelength of ocean wave,which can be retrieved from SAR imagery without any prior knowledge of wind or wave.Using Envisat ASAR wave mode data and the collocated buoy measurements from NDBC,the semiempirical algorithm is validated and compared with the Envisat ASAR level 2 products.The root-mean-square-error(RMSE) and scatter index(SI) in respect to the in situ measurements are 0.52 m and 19% respectively.Validation results indicate that,for Envisat ASAR wave mode data,the proposed method works well.WANG He ZHU Jianhua YANG Jingsong SHI Chaoying 2012Acta Oceanologica Sinica2012,31,3:6
6Evaluation of a laying-hen tracking algorithm based on a hybrid support vector machine显示文摘Background: Behavior is an important indicator reflecting the welfare of animals. Manual analysis of video is the most commonly used method to study animal behavior. However, this approach is tedious and depends on a subjective judgment of the analysts. There is an urgent need for automatic identification of individual animals and automatic tracking is a fundamental part of the solution to this problem.Results: In this study, an algorithm based on a Hybrid Support Vector Machine(HSVM) was developed for the automated tracking of individual laying hens in a layer group. More than 500 h of video was conducted with laying hens raised under a floor system by using an experimental platform. The experimental results demonstrated that the HSVM tracker outperformed the Frag(fragment-based tracking method), the TLD(Tracking-Learning-Detection),the PLS(object tracking via partial least squares analysis), the Mean Shift Algorithm, and the Particle Filter Algorithm based on their overlap rate and the average overlap rate.Conclusions: The experimental results indicate that the HSVM tracker achieved better robustness and state-of-theart performance in its ability to track individual laying hens than the other algorithms tested. 7De novo assembly of a Tibetan genome and identification of novel structural variants associated with high-altitude adaptation显示文摘Structural variants(SVs) may play important roles in human adaptation to extreme environments such as high altitude but have been under-investigated. Here, combining long-read sequencing with multiple scaffolding techniques, we assembled a high-quality Tibetan genome(ZF1), with a contig N50 length of 24.57 mega-base pairs(Mb) and a scaffold N50 length of 58.80 Mb. The ZF1 assembly filled 80 remaining N-gaps(0.25 Mb in total length) in the reference human genome(GRCh38). Markedly, we detected17 900 SVs, among which the ZF1-specific SVs are enriched in GTPase activity that is required for activation of the hypoxic pathway. Further population analysis uncovered a 163-bp intronic deletion in the MKL1 gene showing large divergence between highland Tibetans and lowland Han Chinese. This deletion is significantly associated with lower systolic pulmonary arterial pressure, one of the key adaptive physiological traits in Tibetans. Moreover, with the use of the high-quality de novo assembly, we observed a much higher rate of genome-wide archaic hominid(Altai Neanderthal and Denisovan) shared non-reference sequences in ZF1(1.32%–1.53%) compared to other East Asian genomes(0.70%–0.98%),reflecting a unique genomic composition of Tibetans. One such archaic hominid shared sequence-a662-bp intronic insertion in the SCUBE2 gene-is enriched and associated with better lung function(the FEV1/FVC ratio) in Tibetans. 8Spatial heterogeneity of a microbial community in Kongsfjorden,Svalbard during late summer 2006 and its relationship to biotic and abiotic factors显示文摘The 16S and 18S ribosomal ribonucleic acid genes of microbial organisms collected from the contrasting environments (temperature,salinity,silicate,phosphate and nitrate,p <0.05) of the inner and outer basins of Kongsfjorden (Spitsbergen,Arctic) were studied using polymerase chain reaction-denaturing gradient gel electrophoresis(DGGE) fingerprinting.Comparison of the microbial fingerprints and the physicochemical parameters revealed that molecular methodology exhibited a greater sensitivity.Sequences obtained from bacterial DGGE were affiliated with four main phylogenetic groups of bacteria:Proteobacteria(Alpha,Beta and Gamma),Bacteroidetes,Verrucomicrobia and Cyanobacteria.The relationships between the genotype distribution of these microbes and associated biotic/abiotic factors,revealed by canonical correspondence analysis,showed that Station 1 at 30 m (outer fjord) was grouped separately from the other sites.This difference could be a consequence of the thermocline and base of the euphotic layer at this depth where the Atlantic and Arctic-type waters overlapped.GUO ChaoYing HE JianFeng ZHANG Fang CAI MingHong WANG GuiZhong 2011Chinese Journal of Polar Science2011,22,1:3
9Cloud-based data management system for automatic real-time data acquisition from large-scale laying-hen farms显示文摘Management of poultry farms in China mostly relies on manual labor.Since such a large amount of valuable data for the production process either are saved incomplete or saved only as paper documents,making it very difficult for data retrieve,processing and analysis.An integrated cloud-based data management system(CDMS)was proposed in this study,in which the asynchronous data transmission,distributed file system,and wireless network technology were used for information collection,management and sharing in large-scale egg production.The cloud-based platform can provide information technology infrastructures for different farms.The CDMS can also allocate the computing resources and storage space based on demand.A real-time data acquisition software was developed,which allowed farm management staff to submit reports through website or smartphone,enabled digitization of production data.The use of asynchronous transfer in the system can avoid potential data loss during the transmission between farms and the remote cloud data center.All the valid historical data of poultry farms can be stored to the remote cloud data center,and then eliminates the need for large server clusters on the farms.Users with proper identification can access the online data portal of the system through a browser or an APP from anywhere worldwide.Chen Hongqian Hongwei Xin Teng Guanghui Meng Chaoying Du Xiaodong Mao Taotao Wang Cheng 2016International Journal of Agricultural and Biological Engineering2016,9,4:3
10Study on the Supply Efficiency of Rural Public Service in China Based on Three-stage DEA Model显示文摘Improving the supply efficiency of rural public service is an important way to solve the severe shortage of rural public service. In this article,we use three-stage DEA model to carry out empirical research of the supply efficiency of rural public service in 31 provinces and regions of China. The results show that if without control over exogenous environment variables and random brunt,the classic DEA method will overestimate the rural public service efficiency; after controlling the impact of the external environmental factors,the mean of supply efficiency of rural public service in 31 provinces and regions of China is 0. 697; the improved rural per capita income,population density,population size and the educational level of residents,is a significantly favorable factor for enhancing the supply efficiency of rural public service,while the increase in the proportion of fiscal spending on rural public service to GDP plays no significant role in improving the rural public service efficiency; according to their efficiency type,the provinces and regions should adopt some measures,such as improving the management level or expanding the supply scale,to improve the supply efficiency.Zongbing DENG Chaoying WU Junliang Zhang Ju WANG 2014Asian Agricultural Research2014,6,1:3

14Acupoint electrogymnastics therapy in stroke hemiplegia显示文摘BACKGROUND: Electrogymnastics can offer a range of proprioceptive, motor, and cutaneous sensation impulses to the central nerve system. The center receives perception of the paralyzed muscle through the aid of these impulses. During this process, functional reorganization of connecting network between segment and intersegment takes place. OBJECTIVE: To observe the therapeutic efficacy of acupoint electrogymnastics and traditional electroacupuncture on stroke hemiplegia. DESIGN, TIME AND SETTING: A multicenter, randomized, controlled, blinded, clinical study was performed at the College of Acu-moxibustion and Massage in Shanghai University of Traditional Chinese Medicine from May 2004 to September 2006. PARTICIPANTS: A total of 153 patients suffering from stroke hemiplegia, comprising 83 males and 70 females, aged 63-70 years, were admitted to outpatient and inpatient at LongHua Hospital Affiliated Shanghai University of Traditional Chinese medicine, Putuo District Traditional Chinese Medicine Hospital and Changqiao Street Community Health Service Center of Shanghai. METHODS: The patients were randomly divided into treatment (n = 77) and control (n = 76) groups. They were treated with acupoint electrogymnastics and traditional electroacupuncture, respectively. In the treatment group, two pairs of positive and negative JD-2008 type electrodes from a hemiplegia treatment apparatus were directly pasted on the Shousanli (LI 10) and Waiguan (SJ 5) acupoints of the upper limb, as well as the Zusanli (S 36) and Yanglingquan (GB 34) acupoints of the lower limb, respectively. In the control group, needles were consecutively inserted into the above acupoints. Using the method of lifting-inserting and twisting-rotating, the needle was manipulated with small amplitude of 5-7 mm and a fast frequency of 80-120 times/min when the needle was inserted to a suitable depth. When the sensation of needling was attained, the two pairs of positive and negative electrodes of type G6805-II electro-acupuncture apparatus were connected to the Shousanli (LI 10) and Waiguan (SJ 5) acupoints of the upper limb, as well as the Zusanli (S 36) and Yanglingquan(GB 34) acupoints of the lower limb, respectively. MAIN OUTCOME MEASURES: The Fugl-Meyer evaluation method was used to assess upper limb movement, lower limb movement, sensory stimulation of all limbs, balance, and joint range of motion. RESULTS: Upper-lower limb motor function, limb sensory function, balance, and joint range of motion were significantly ameliorated following treatment, compared with pre-treatment (P < 0.01). There was no significant difference in various indexes between the two groups after treatment (P > 0.05). 20A loss-of-function mutant allele of a glycosyl hydrolase gene has been co-opted for seed weight control during soybean domestication显示文摘The resultant DNA from loss-of-function mutation can be recruited in biological evolution and development.Here,we present such a rare and potential case of“to gain by loss”as a neomorphic mutation during soybean domestication for increasing seed weight.Using a population derived from a chromosome segment substitution line of Glycine max(SN14)and Glycine soja(ZYD06),a quantitative trait locus(QTL)of 100-seed weight(q HSW)was mapped on chromosome 11,corresponding to a truncatedβ-1,3-glucosidase(βGlu)gene.The novel gene hsw results from a 14-bp deletion,causing a frameshift mutation and a premature stop codon in theβGlu.In contrast to HSW,the hsw completely lostβGlu activity and function but acquired a novel function to promote cell expansion,thus increasing seed weight.Overexpressing hsw instead of HSW produced large soybean seeds,and surprisingly,truncating hsw via gene editing further increased the seed size.We further found that the core 21-aa peptide of hsw and its variants acted as a promoter of seed size.Transcriptomic variation in these transgenic soybean lines substantiated the integration hsw into cell and seed size control.Moreover,the hsw allele underwent selection and expansion during soybean domestication and improvement.Our work cloned a likely domesticated QTL controlling soybean seed weight,revealed a novel genetic variation and mechanism in soybean domestication,and provided new insight into crop domestication and breeding,and plant evolution.Siming Wei Bin Yong Hongwei Jiang Zhenghong An Yan Wang Bingbing Li Ce Yang Weiwei Zhu Qingshan Chen Chaoying He 2023Journal of Integrative Plant Biology2023,65,11:1
