2In tegrative iTRAQ-based proteomic and transcriptomic analysis reveals the accumulation patter ns of key metabolites associated with oil quality during seed ripening of Camellia oleifera显示文摘Camellia oleifera(C.oleifera)is one of the four major woody oil-bearing crops in the world and has relatively high ecological,economic,and medicinal value.Its seeds undergo a series of complex physiological and biochemical changes during ripening,which is mainly manifested as the accumulation and transformation of certain metabolites closely related to oil quality,especially flavonoids and fatty acids.To obtain new insights into the underlying molecular mechanisms,a parallel analysis of the transcriptome and proteome profiles of C.oleifera seeds at different maturity levels was conducted using RNA sequencing(RNA-seq)and isobaric tags for relative and absolute quantification(iTRAQ)complemented with gas chromatography-mass spectrometry(GC-MS)data.A total of 16,530 transcripts and 1228 proteins were recognized with significant differential abundances in pairwise comparisons of samples at various developmental stages.Among these,317 were coexpressed with a poor correlation,and most were involved in metabolic processes,including fatty acid metabolism,a-linolenic acid metabolism,and glutathione metabolism.In addition,the content of total flavonoids decreased gradually with seed maturity,and the levels of fatty acids generally peaked at the fat accumulation stage;these results basically agreed with the regulation patterns of genes or proteins in the corresponding pathways.The expression levels of proteins annotated as upstream candidates of phenylalanine ammonia-lyase(PAL)and chalcone synthase(CHS)as well as their cognate transcripts were positively correlated with the variation in the flavonoid content,while shikimate O-hydroxycinnamoyltransferase(HCT)-(encoding genes had the opposite pattern.The increase in the abundance of proteins and mRNAs corresponding to alcohol dehydrogenase(ADH)was associated with a reduction in linoleic acid synthesis.Using weighted gene coexpression network analysis(WGCNA),we further identi fied six unique modules related to flavonoid,oil,and fatty acid anabolism that contained hub genes or proteins similar to transcription factors(TFs),such as MADS intervening keratin-like and C-terminal(MIKC_MAD s),type-B authentic response regulator(ARR-B),and basic helix-loop-helix(bH〔H).Finally,based on the known metabolic pathways and WGCNA combined with the correlation analysis,fi ve coexpressed transcripts and proteins composed of cinnamyl-alcohol dehyd rogenases(CADs),caffeicacid 3-O-methyltransferase(COMT),fl avonol synthase(FLS),and 4-coumarate:CoA ligaas(4CL)were screened out.With this exploratory multiomics dataset,our results presented a dynamic picture regarding the maturation process of C.oleifera seeds on Hainan Island,not only revealing the temporal specific expression of key candidate genes and proteins but also providing a scientific basis for the genetic improvement of this tree species.Zhouchen Ye Jing Yu Wuping Yan Junfeng Zhang Dongmei Yang Guanglong Yao Zijin Liu Yougen Wu Xilin Hou 2021Horticulture Research2021,8,1:3
3Maximal methane potential of different animal manures collected in northwest region of China显示文摘Maximum methane potential(B0)is an important parameter used in assessing suitability of a substrate for biogas production.This study examined maximum methane potential of different manures generated from three major Chinese livestock,namely chicken,hog and cattle,and evaluated the important factors that affect the maximum methane potential of a substrate.The livestock manures collected from the local farms were incubated under a thermophilic anaerobic condition(55℃).The results showed that the maximum methane potential(B0)of cattle,hog and chicken manures were 292.0 mL/g VS,272.0 mL/g VS and 266.4 mL/g VS,respectively.The B0 value decreases with increasing contents of crude protein and crude fat,while increases with increasing the contents of carbohydrates and crude fiber in manures.The content of NH4+-N in chicken manure was significantly higher during the digestion period,reached as high as 1962.5 mg/L by the end of incubation period.Heavy metals of Cu and Zn in the manure also affect the B0.Empirical relationships that describe the B0 decrease in response to increase of Zn and Cu contents in manure were developed and used as a simple tool to assess the effects of these metals on the B0.It was concluded that the protein,Cu and Zn contents of manure are most important chemical compositions that negatively affect maximum methane potential.Based on the three experimental manures,the maximum methane potential was limited by either ammonium content or Cu and Zn content in the manure.For a commercial biogas production facility using these manures as main feedstock,one should consider to add co-substrate or co-substrates to reduce concentration of these chemicals to maximize biogas production.Chen Fen Yu Gao Li Wei Liu Fenwu Zhang Wuping Bu Yushan Li Xiaomei 2017International Journal of Agricultural and Biological Engineering2017,10,1:2

6Microbial community characterization,activity analysis and purifying efficiency in a biofilter process显示文摘The growth and metabolism of microbial communities on biologically activated carbon(BAC) play a crucial role in the purification of drinking water.To gain insight into the growth and metabolic characteristics of microbial communities and the efficiency of drinking water treatment in a BAC filter,we analyzed the heterotrophic plate count(HPC),phospholipid,dehydrogenase,metabolic function and water quality parameters during start-up and steady-state periods.In the start-up process of the filter with natural biofilm colonization,the variation in heterotrophic plate count levels was S-curved.The total phospholipid level was very low during the first 5 days and reached a maximum value after 40 days in the filter.The activity of dehydrogenase gradually increased during the first 30 days and then reached a plateau.The functional diversity of the microbial community in the filter increased,and then reached a relatively stable level by day 40.After an initial decrease,which was followed by an increase,the removal rate of NH4+-N and COD Mn became stable and was 80% and 28%,respectively,by day 40.The consumption rate of dissolved oxygen reached a steady level after 29 days,and remained at 18%.At the steady operation state,the levels of HPC,phospholipid,dehydrogenase activity and carbon source utilization had no significant differences after 6 months compared to levels measured on day 40.The filter was shown to be effective in removing NH4+-N,NO2--N,COD Mn,UV 254,biodegradable dissolved organic carbon and trace organic pollutants from the influent.Our results suggest that understanding changes in the growth and metabolism of microorganisms in BAC filter could help to improve the efficiency of biological treatment of drinking water.Hong Xiang Xiwu Lu Lihong Yin Fei Yang Guangcan Zhu Wuping Liu 2013Journal of Environmental Sciences2013,25,4:1
