1CircRNA-SORE mediates sorafenib resistance in hepatocellular carcinoma by stabilizing YBX1显示文摘Sorafenib is the first-line chemotherapeutic therapy for advanced hepatocellular carcinoma(HCC).However,sorafenib resistance significantly limits its therapeutic efficacy,and the mechanisms underlying resistance have not been fully clarified.Here we report that a circular RNA,circRNA-SORE(a circular RNA upregulated in sorafenib-resistant HCC cells),plays a significant role in sorafenib resistance in HCC.We found that circRNA-SORE is upregulated in sorafenib-resistant HCC cells and depletion of circRNA-SORE substantially increases the cell-killing ability of sorafenib.Further studies revealed that circRNA-SORE binds the master oncogenic protein YBX1 in the cytoplasm,which prevents YBX1 nuclear interaction with the E3 ubiquitin ligase PRP19 and thus blocks PRP19-mediated YBX1 degradation.Moreover,our in vitro and in vivo results suggest that circRNA-SORE is transported by exosomes to spread sorafenib resistance among HCC cells.Using different HCC mouse models,we demonstrated that silencing circRNA-SORE by injection of siRNA could substantially overcome sorafenib resistance.Our study provides a proof-of-concept demonstration for a potential strategy to overcome sorafenib resistance in HCC patients by targeting circRNA-SORE or YBX1.Junjie Xu Lin Ji Yuelong Liang Zhe Wan Wei Zheng Xiaomin Song Kirill Gorshkov Qiming Sun Hui Lin Xueyong Zheng Jiang Chen Ren-an Jin Xiao Liang Xiujun Cai 2020Signal Transduction and Targeted Therapy2020,5,1:5
2The diversity of denitrifying bacteria in the alpine meadow soil of Sanjiangyuan natural reserve in Tibet Plateau显示文摘This is the first time to describe the diversity of denitrifying bacteria in Sanjiangyuan natural reserve in Tibet Plateau by investigating the molecular diversity and phylogenetic analysis of nirK and nosZ genes using PCR-RFLP and sequencing. Four soil samples were collected from alpine meadow communities from over an altitude of 4600 m which had different physicochemical properties by principal component analysis (PCA). For the genes fragment of nirK and nosZ, the diverse PCR products were characterized by cloning, restriction fragment length polymorphism (RFLP) analysis and sequenced. A total of 253 nirK clones and 283 nosZ clones were received in four samples, and 78 operational taxonomic units (OTUs) of nirK and 120 OTUs of nosZ by the restriction enzymes MspI and RsaI digested. The analysis of environmental factors showed that altitude and C/N ratio in soil may be the key factors to the denitrifying bacteria community. 36 nirK clones and 17 nosZ clones were sequenced, and their levels of nucleotide identity were from 69% to 98% and 57% to 97%, respectively. The phylogenetic tree was constructed using the Clustal W and Mega softwares, and all the sequenced clones could be subdivided into 4 groups. Some of clone sequences were related to the nirK and nosZ genes belonging to three phylogenetic subdivisions (α-, β- and γ subclasses of the Proteobacteria), while most of the clones were closely related to the genes of the uncultured bacteria. 3Mechanical activation of spike fosters SARS-CoV-2 viral infection显示文摘The outbreak of SARS-CoV-2(SARS2)has caused a global COVID-19 pandemic.The spike protein of SARS2(SARS2-S)recognizes host receptors,including ACE2,to initiate viral entry in a complex biomechanical environment.Here,we reveal that tensile force,generated by bending of the host cell membrane,strengthens spike recognition of ACE2 and accelerates the detachment of spike’s S1 subunit from the S2 subunit to rapidly prime the viral fusion machinery.Mechanistically,such mechano-activation is fulfilled by force-induced opening and rotation of spike’s receptor-binding domain to prolong the bond lifetime of spike/ACE2 binding,up to 4 times longer than that of SARS-S binding with ACE2 under 10 pN force application,and subsequently by force-accelerated S1/S2 detachment which is up to~103 times faster than that in the no-force condition.Interestingly,the SARS2-S D614G mutant,a more infectious variant,shows 3-time stronger force-dependent ACE2 binding and 35-time faster force-induced S1/S2 detachment.We also reveal that an anti-S1/S2 non-RBD-blocking antibody that was derived from convalescent COVID-19 patients with potent neutralizing capability can reduce S1/S2 detachment by 3×106 times under force.Our study sheds light on the mechano-chemistry of spike activation and on developing a non-RBD-blocking but S1/S2-locking therapeutic strategy to prevent SARS2 invasion.Wei Hu Yong Zhang Panyu Fei Tongtong Zhang Danmei Yao Yufei Gao Jia Liu Hui Chen Qiao Lu Tenny Mudianto Xinrui Zhang Chuxuan Xiao Yang Ye Qiming Sun Jing Zhang Qi Xie Pei-Hui Wang Jun Wang Zhenhai Li Jizhong Lou Wei Chen 2021Cell Research2021,31,10:2

63D Printing of Nacre-Inspired Structures with Exceptional Mechanical and Flame-Retardant Properties显示文摘Flame-retardant and thermal management structures have attracted great attention duc to the requirement of high-temperature exposure in industrial,aerospace,and thermal power fields,but the development of protoctive fire-retardant structures with complex shapes to fit arbitrary surfaces is still challenging.Herein,we reported a rotation-blade casting-assisted 3D printing process to fabricate nacre-inspired structures with exceptional mechanical and flame-retardant properties,and the relatedi fundamental mechanisms are studied.3-(Trimethoxysilyl)propyl methacrylate(TMSPMA)modified boron nitride nanoplatelets(BNs)were aligned by rotation-blade casting during the 3D printing process to build the'hrick and mortar'architecture.The 3D printed structures are more lightweight,while having higher fracture toughness than the natural nacre,which is attributed to the crack deflection,aligned BN(a-BNs)bridging,and pull-outs reinforced structures by the covalent bonding between TMSPMA grafted a-BNs and polymer matrix.Thermal conductivity is enhanced by 25.5 times compared with pure polymer and 5.8 times of anisotropy due to the interconnection of a-BNs.3D printed heat-exchange structures with vertically aligned BNs in complex shapes were demonstrated for efficient thermal control of high-power light-emitting diocles.3D printed helmet and armor with a-BNs show exceptional mechanical and fire-retardant properties,demonstrating integrated mechanical and thermal protection.Yang Yang Ziyu Wang Qingqing He Xiangjia Li Gengxi Lu Laiming Jjiang Yushun Zeng Brandon Bethers Jie Jin Shuang Lin Siqi Xiao Yizhen Zhu Xianke Wu Wenwu Xu Qiming Wang Yong Chen 2022Research2022,,2:1
7Long-Term Trend Analysis of Major Human Helminth Infections — Guangdong Province, China, 1988–2021显示文摘Introduction:Although helminth infections threaten millions of people worldwide,the spatiotemporal characteristics remain unclear across China.This study systematically describes the spatiotemporal changes of major human helminth infections and their epidemiological characteristics from 1988 to 2021 in Guangdong Province,China.Methods:The survey data in Guangdong Province were primarily obtained from 3 national surveys implemented during 1988–1992,2001–2004,and 2014–2016,respectively,and from the China Information System for Disease Control and Prevention during 2019–2021.A modified Kato-Katz technique was used to detect parasite eggs in collected fecal samples.Results:The overall standardized infection rates(SIRs)of any soil-transmitted helminths(STH)and Clonorchis sinensis decreased from 65.27%during 1988–1992 to 4.23%during 2019–2021.In particular,the SIRs of STH had even more of a decrease,from 64.41%during 1988–1992 to 0.31%during 2019–2021.The SIRs of Clonorchis sinensis in the 4 surveys were 2.40%,12.17%,5.20%,and 3.93%,respectively.This study observed different permutations of gender,age,occupation,and education level on the SIRs of helminths.Conclusions:The infection rate of STH has substantially decreased.However,the infection rate of Clonorchis sinensis has had fewer changes,and it has become the dominant helminth.Yueyi Fang Jiong Wang Guanhao He Qiming Zhang Jianpeng Xiao Jianxiong Hu Zuhua Rong Lihua Yin Fangfang Zeng Pan Yang Xiaomei Dong Dan Liu Xiaofeng Liang Zhuohui Deng Tao Liu Wenjun Ma 2022China CDC weekly2022,4,41:1
8Susceptibility gene for stroke or cerebral infarction in the Han population in Hunan Province of China显示文摘The scavenger receptor class B type I gene can protect against atherosclerosis; a mononucleotide polymorphism is associated with differences in blood lipid metabolism, postprandial serum lipid levels, insulin resistance, coronary artery disease and familial hyperlipidemia. In this study, the scavenger receptor class B type I gene exon 1 G4A gene polymorphism in atherosclerotic cerebral infarction patients, cerebral hemorrhage patients and normal controls was detected using the polymerase chain reaction-restriction fragment length polymorphism method. The results showed that the GA + AA genotype frequency of scavenger receptor class B type I gene G4A in atherosclerotic cerebral infarction patients was similar to that in cerebral hemorrhage patients and normal controls; however, the A allele frequency was significantly lower than that in normal controls. The serum level of high-density lipoprotein cholesterol in patients with the scavenger receptor class B type I gene G4A GA + AA genotype was significantly higher, while the serum level of low-density lipoprotein cholesterol was significantly lower than that in patients with the GG genotype, in both the atherosclerotic cerebral infarction and cerebral hemorrhage groups. The serum level of high-density lipoprotein cholesterol in patients with the scavenger receptor class B type I gene G4A GA + AA genotype was significantly higher, while the serum levels of low-density lipoprotein cholesterol and total cholesterol were significantly lower than those in normal controls with the GG genotype. Our experimental results suggest that the G4A polymorphism of the scavenger receptor class B type I gene is a possible predisposing risk factor for atherosclerotic cerebral infarction, and that it has no association with cerebral hemorrhage in the Han population in Hunan province of China. 10Evaluation of cable tension sensors of FAST reflector from the perspective of EMI显示文摘The active reflector of FAST(five-hundred-meter aperture spherical radio telescope) is supported by a ring beam and a cable-net structure,in which nodes are actively controlled to form series of real-time paraboloids.To ensure the security and stability of the supporting structure,tension must be monitored for some typical cables.Considering the stringent requirements in accuracy and longterm stability,magnetic flux sensor,vibrating wire strain gauge and fiber bragg grating strain gauge are screened for the cable tension monitoring of the supporting cable-net.Specifically,receivers of radio telescopes have strict restriction on electro magnetic interference(EMI) or radio frequency interference(RFI).These three types of sensors are evaluated from the view of EMI/RFI.Firstly,these fundamentals are theoretically analyzed.Secondly,typical sensor signals are collected in the time and analyzed in the frequency domain,which shows the characteristic in the frequency domain.Finally,typical sensors are tested in an anechoic chamber to get the EMI levels.Theoretical analysis shows that Fiber Bragg Grating strain gauge itself will not lead to EMI/RFI.According to GJB151 A,frequency domain analysis and test results show that for the vibrating wire strain gauge and magnetic flux sensor themselves,testable EMI/RFI levels are typically below the background noise of the anechoic chamber.FAST finally choses these three sensors as the monitoring sensors of its cable tension.The proposed study is also a reference to the monitoring equipment selection of other radio telescopes and large structures.朱明 Wang Qiming Dennis Egan Wu Mingchang Sun Xiao 2016High Technology Letters2016,22,2:0
11MAML^(2):meta reinforcement learning via meta-learning for task categories显示文摘Meta-learning has been widely applied to solving few-shot reinforcement learning problems,where we hope to obtain an agent that can learn quickly in a new task.However,these algorithms often ignore some isolated tasks in pursuit of the average performance,which may result in negative adaptation in these isolated tasks,and they usually need sufficient learning in a stationary task distribution.In this paper,our algorithm presents a hierarchical framework of double meta-learning,and the whole framework includes classification,meta-learning,and re-adaptation.Firstly,in the classification process,we classify tasks into several task subsets,considered as some categories of tasks,by learned parameters of each task,which can separate out some isolated tasks thereafter.Secondly,in the meta-learning process,we learn category parameters in all subsets via meta-learning.Simultaneously,based on the gradient of each category parameter in each subset,we use meta-learning again to learn a new metaparameter related to the whole task set,which can be used as an initial parameter for the new task.Finally,in the re-adaption process,we adapt the parameter of the new task with two steps,by the meta-parameter and the appropriate category parameter successively.Experimentally,we demonstrate our algorithm prevents the agent from negative adaptation without losing the average performance for the whole task set.Additionally,our algorithm presents a more rapid adaptation process within readaptation.Moreover,we show the good performance of our algorithm with fewer samples as the agent is exposed to an online meta-learning setting.Qiming FU Zhechao WANG Nengwei FANG Bin XING Xiao ZHANG Jianping CHEN 2023Frontiers of Computer Science2023,17,4:0

17Nanotechnology-based therapeutics: new hope for ischemic cerebral stroke intervention显示文摘Ischemic cerebral stroke is a leading cause of death and disability globally.At present,thrombolytics,such as recombinant tissue-type plasminogen activator,are the only effective treatment for acute stroke.However,usage of thrombolytics has a strict therapeutic window and cannot be applied to a number of patients.Despite the promising effects of some neuroprotectants in preclinical studies,they failed in clinical trials as a result of poor pharmacokinetic properties,particularly with regard to solubility and permeability across the blood-brain barrier(BBB).Approaches for delivering these drugs by nanotechnologies may overcome these pharmacokinetic deficits and enhance their neuroprotective effects.However,issues such as potential side effects and biosafety properties currently limit clinical application of these approaches.In this article,we reviewed recent progress of nanotechnology-based interventions for stroke treatment,and in particular,summarized novel materials applied to synthesize nanocarriers,encapsulation with neuroprotectants,and factors impacting nanodrug bioactivities to provide a theoretical basis for the development of anti-stroke drugs.Sihan Wang Qiming Gao Fuyao Xiao Yuming Zhao 2020Journal of Translational Neuroscience2020,5,1:0
