1Olmutinib(HM61713) reversed multidrug resistance by inhibiting the activity of ATPbinding cassette subfamily G member 2 in vitro and in vivo显示文摘Overexpressing of ATP-binding cassette(ABC) transporters is the essential cause of multidrug resistance(MDR), which is a significant hurdle to the success of chemotherapy in many cancers.Therefore, inhibiting the activity of ABC transporters may be a logical approach to circumvent MDR.Olmutinib is an epidermal growth factor receptor(EGFR) tyrosine kinase inhibitor(TKI), which has been approved in South Korea for advanced EGFR T790 M-positive non-small cell lung cancer(NSCLC). Here,we found that olmutinib significantly increased the sensitivity of chemotherapy drug in ABCG2-overexpressing cells. Furthermore, olmutinib could also increase the retention of doxorubicin(DOX) and rhodamine 123(Rho 123) in ABC transporter subfamily G member 2(ABCG2)-overexpressing cells. In addition, olmutinib was found to stimulate ATPase activity and inhibit photolabeling of ABCG2 with [^(125) I]-iodoarylazidoprazosin(IAAP). However, olmutinib neither altered ABCG2 expression at protein and m RNA levels nor blocked EGFR, Her-2 downstream signaling of AKT and ERK. Importantly,olmutinib enhanced the efficacy of topotecan on the inhibition of S1-MI-80 cell xenograft growth. All the results suggest that olmutinib reverses ABCG2-mediated MDR by binding to ATP bind site of ABCG2 and increasing intracellular chemotherapeutic drug accumulation. 2Identification of microRNA-like RNAs in Ophiocordyceps sinensis显示文摘Ophiocordyceps sinensis is well known as a traditional Chinese medicine and has widely been used for over 2,000 years to stimulate immune system, decrease blood pressure and to inhibit tumor growth. While miRNAs are increasingly recognized for their roles in post-transcriptional regulation of gene expression in animals and plants, miRNAs in fungi were less studied until the discovery of microRNA-like RNA(milRNA). High-throughput sequencing and bioinformatics approaches were used to identify conserved and novel milRNAs in O. sinensis. 40 conserved milRNAs were identified, while 23 pre-miRNA candidates encoding 31 novel milRNAs were predicted. Furthermore, the potential target genes of milRNAs in human were predicted and gene ontology analysis was applied to these genes. Enrichment analysis of GO-represented biological process showed that target genes of both conserved and novel milRNAs are involved in development, metabolic and immune processes, indicating the potential roles of milRNAs of O. sinensis in pharmacological effects as health food and traditional Chinese medicine. 3Exploitation of methane in the hydrate by use of carbon dioxide in the presence of sodium chloride显示文摘The replacement process of CH4 from CH4 hydrate formed in NaCl solution by using pressurized CO2 was investigated with a self-designed device at temperatures of 271.05,273.15 and 275.05 K and a constant pressure of 3.30 MPa.The mass fraction of the NaCl solution was either 0.5 wt% or 1.0 wt%.The effects of temperature and concentration of NaCl solution on the replacement process were investigated.Experimental results showed that high temperature was favorable to the replacement reaction but high NaCl concentration had a negative effect on the replacement process.Based on the experimental data,kinetic models of CH4 hydrate decomposition and CO2 hydrate formation in NaCl solution were established.The calculated activation energies suggested that both CH4 hydrate decomposition and CO2 hydrate formation are dominated by diffusion in the hydrate phase.Zunzhao Li Xuqiang Guo Lanying Yang Xiaona Ma 2009Petroleum Science2009,6,4:5

5Comparative analysis by simulating and testing pole impact for hot stamping front crossbeam of automobile显示文摘Hot stamping 22MnB5 steel plate with ultra-high strength has been widely used for body structural members in consideration of automobile safety and lightweight.This paper presents a verification program of simulating and testing pole impact in order to verify if the front crossbeam reinforcement assembly can meet the design requirements,reduce the overall vehicle simulation and test cost and shorten the operation period.In the same condition,the simulation proved conforming to the design requirements;however,the bumper cracked at the impacting point in the course of pole impact test.The analysis of the crack by optical microscope,Vickers and scanning electron microscope indicates that mixture of ferrite,bainite and martensite was produced in the weld heat-affected zone of the 340/590DP tow hook holder and the 22MnB5 front crossbeam;therefore,their hardness and mechanical property were reduced obviously,so that they fractured when impacted.No welding process with continuous weld but spot welding or other bonding method may be employed for the reason that the capacity of the 22MnB5 steel plate in the weld heat-affected zone was reduced obviously.Ma Fangwu Yao Zaiqi Liu Qiang Wang Xiaona Zhao Fuquan Li Fangfang Lin Jianping 2012Engineering Sciences2012,10,6:2
6Spatial Pattern and Influencing Factor of County-level Industrial Development in Liaoning Province of China显示文摘County-level industrial development and structure upgrade is one of the most important issues of revitaliz- ing old industrial base of China. After the cluster analysis on GDP per capita and GDP per area of each county in Liaoning Province, this paper finds the similarity of population size, land use intensity, and economic development of each county. Location quotient reflects the specialization intensity of industries in each county, and it also reflects the spatial differences of county-level industrial development. Economic development level is higher in the southeast than in the northwest of Liaoning, and the industry driving effect on county-level economy is apparent. The main influenc- ing factors include location, industrial foundation and economic system reform, capital input level, knowledge and technology dissemination, conditions of domestic and overseas markets, population and labor force transfer. Industri- alization is an important approach to urbanization for the counties in Liaoning Province. 9Hupo powder promotes autophagy of menstrual blood-derived stem cells from patients with endometriosis显示文摘Objective:To explore the effect of Hupo powder(HP)on autophagy in menstrual blood-derived stem cells(MenSCs)with endometriosis(EMT).Methods:EMT MenSCs(E-MenSCs)and healthy MenSCs(H-MenSCs)were isolated from the menstrual blood of patients with EMT and healthy female participants,respectively.We identified their stem cells’characteristics via adipogenic and osteogenic differentiation.Twelve male SpragueeDawley rats received 0.9% NaCl and HP-dispensing granules by gastric irrigation to prepare blank serum and medicated serum,respectively.We used serum concentrations of 5%,10%,and 20%,each at administered times of 12,24,and 48 h to select the best condition.These cells were divided into three groups:blank serum of the control group,blank serum of the model group,and medicated serum of the HP group.H-MenSCs were used in the control group,while E-MenSCs were used in the model and HP groups.We analyzed cell viability using a cell counting kit-8 assay,observed cell morphology,evaluated the amounts of auto-phagosomes and autolysosomes by transmission electron microscopy,and detected the protein expression of autophagy markers(LC3-II and Beclin1)by Western blot.Results:E-MenSCs and H-MenSCs became long fusiform with a diffuse radial pattern,forming lipid droplets and calcium nodules after adipogenic and osteogenic differentiation.We then used the best conditiond 20% serum and 48 hdfor the subsequent experiments.In contrast to the model group,the HP group exhibited lower cell viability(=0.007),larger amounts of autophagosomes and autolysosomes(P<0.001 and P=0.001,respectively),and higher expression of LC3-II and Beclin1(P=0.021 and P=0.019,respectively).Conclusion:Hupo powder can promote autophagy in E-MenSCs,which might be one of the mechanisms underlying its therapeutic effects.Yuejian Zhang Changxiang Li Conglu Sui Xiuping Zhang Ya'nan Guo Tiantian He Taoxiu Lin Xiaona Ma 2023Journal of Traditional Chinese Medical Sciences2023,10,2:1
