| 1 | ALDH2 contributes to melatonin-induced protection against APP/PS1 mutation-prompted cardiac anomalies through cGAS-STING-TBK1-mediated regulation of mitophagy显示文摘Ample clinical evidence suggests a high incidence of cardiovascular events in Alzheimer’s disease(AD),although neither precise etiology nor effective treatment is available.This study was designed to evaluate cardiac function in AD patients and APP/PS1 mutant mice,along with circulating levels of melatonin,mitochondrial aldehyde dehydrogenase(ALDH2)and autophagy.AD patients and APP/PS1 mice displayed cognitive and myocardial deficits,low levels of circulating melatonin,ALDH2 activity,and autophagy,ultrastructural,geometric(cardiac atrophy and interstitial fibrosis)and functional(reduced fractional shortening and cardiomyocyte contraction)anomalies,mitochondrial injury,cytosolic mtDNA buildup,apoptosis,and suppressed autophagy and mitophagy.APP/PS1 mutation downregulated cyclic GMP-AMP synthase(cGAS)and stimulator of interferon genes(STING)levels and TBK1 phosphorylation,while promoting Aβaccumulation.Treatment with melatonin overtly ameliorated unfavorable APP/PS1-induced changes in cardiac geometry and function,apoptosis,mitochondrial integrity,cytosolic mtDNA accumulation(using both immunocytochemistry and qPCR),mitophagy,and cGAS-STING-TBK1 signaling,although these benefits were absent in APP/PS1/ALDH2−/−mice.In vitro evidence indicated that melatonin attenuated APP/PS1-induced suppression of mitophagy and cardiomyocyte function,and the effect was negated by the nonselective melatonin receptor blocker luzindole,inhibitors or RNA interference of cGAS,STING,TBK1,and autophagy.Our data collectively established a correlation among cardiac dysfunction,low levels of melatonin,ALDH2 activity,and autophagy in AD patients,with compelling support in APP/PS1 mice,in which melatonin rescued myopathic changes by promoting cGAS-STING-TBK1 signaling and mitophagy via an ALDH2-dependent mechanism. | Shuyi Wang Lin Wang Xing Qin Subat Turdi Dongdong Sun Bruce Culver Russel JReiter Xiaoming Wang Hao Zhou Jun Ren | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 9 |
| 2 | Synthesis of ordered mesoporous carbonaceous materials and their highly efficient capture of uranium from solutions显示文摘An extremely effortless method was applied for successful synthesis of mesoporous carbonaceous materials(MCMs) using well-ordered mesoporous silica as template. Various characterizations(scanning electron microscopy(SEM), transmission electron microscopy(TEM), X-ray diffraction(XRD), Fourier transform infrared spectroscopy(FT-IR), Raman, X-ray photoelectron spectroscopy(XPS), Brunner-Emmet-Teller(BET) and Zeta potential) confirmed that MCMs had large surface area, uniform pore size distribution, and abundant oxygen-containing functional groups. The batch techniques were employed to study U(VI) adsorption on MCMs under a wide range of experiment conditions. The adsorption kinetics of U(VI) onto MCMs were well-fitted by pseudo-second-order kinetic model, indicating a chemisorption process. The excellent adsorption capacity of MCMs calculated from the Langmuir model was 293.95 mg g^(-1) at pH 4.0. The FT-IR and XPS analyses further evidenced that the binding of U(VI) onto MCMs was ascribed to the plentiful adsorption sites(–OH and –COOH groups) in the internal mesoporous structure, which could efficiently trap guest U(VI) ions. | 5 | Fouling community characteristics in subtropical coastal waters of the southwestern East China Sea显示文摘A fouling study was conducted in coastal waters southwest of the East China Sea between December 2013 and November 2014. A total of 84 species of fouling organisms belonging to 69 genera, 49 families, and 10 phyla were recorded over the entire year. The community composition was dominated by coastal warm-water species belonging to typical subtropical inner bay communities. The prosperous stage of settlement lasted from April to September, and the adhesion strength of the fouling organisms was the highest in summer. Sessile suspension feeders constituted the main core of settlement for the fouling community. Amphibalanus reticulatus was the most dominant and representative species of fouling organism, and other dominant species included Caprella equilibra, Ectopleura crocea, Anthopleura nigrescens, Stylochus ijimai, Spirobranchus kraussii, Crassostrea angulata, Perna viridis, Jassa falcata, Stenothoe valida, Sphaerozius nitidus, and Biflustra grandicella. The individuals in the fouling community showed a mutual dependence or constraint relationship due to competition for settlement space and food, and they exhibited a particular spatiotemporal distribution in accordance with adaptation to environmental factors. Temperature was the most important environmental factor determining the geographic distribution of fouling organisms. The temperature characteristics of species essentially reflect the differences in the fouling community composition in various climate zones. The species number, settlement stage, and settlement rate of fouling organisms are closely related to water temperature. | 9 | An ecological survey of the abundance and diversity of benthic macrofauna in Indonesian multispecific seagrass beds显示文摘Seagrasses are one of the most productive ecosystems in coastal areas and support a wide variety of associated fauna. The tropical Indo-Pacific region is considered to have the highest diversity of seagrass plant species and the largest distribution areas of seagrass, yet the seagrass macrofauna in this region are poorly understood. To help fill this gap in our knowledge, an ecological survey was conducted to describe the abundance and diversity of benthic macrofauna from tropical seagrass beds and to determine between-station variations within a transect and between-site variations in macrofaunal abundance, taxa richness and community structure. Benthic macrofaunal samples associated with seagrass beds were collected with a core sampler on the east coast of North Sulawesi in May 2014 and on the west coast in October 2015. A total of 149 species from 14 higher taxa was collected. The most species-rich groups were polychaetes(56 species, 26% of total individual numbers), decapods(20 species, 9% of total numbers) and amphipods(18 species, 35% of total numbers). Between-station variations within a transect displayed different patterns between the east coast and the west coast. On the east coast, there were marked variations in abundance between stations within a transect for the macrofauna and amphipod assemblages. Both taxa richness and abundance varied with station for the macrofauna and polychaete assemblages on the west coast, resulting from the heterogeneity of the substrate along a transect. One-way ANOSIM together with MDS ordination indicated that macrofaunal community structure in seagrasses differed significantly between the east coast and the west coast, corresponding with the division of seagrasses into two broad categories of habitats, i.e.,mangrove-seagrass-reef continuum and seagrass-reef continuum. Compared with other studies in tropical areas,the abundance and diversity of benthic macrofauna in the present study were moderate. | 12 | E17110 promotes reverse cholesterol transport with liver X receptor β agonist activity in vitro显示文摘Liver X receptor(LXR) plays an important role in reverse cholesterol transport(RCT), and activation of LXR could reduce atherosclerosis. In the present study we used a cell-based screening method to identify new potential LXRβ agonists. A novel benzofuran-2-carboxylate derivative was identified with LXRβ agonist activity: E17110 showed a significant activation effect on LXRβ with an EC50 value of 0.72 μmol/L. E17110 also increased the expression of ATP-binding cassette transporter A1(ABCA1) and G1(ABCG1) in RAW264.7 macrophages. Moreover, E17110 significantly reduced cellular lipid accumulation and promoted cholesterol efflux in RAW264.7 macrophages. Interestingly, we found that the key amino acids in the LXRβ ligand-binding domain had distinct interactions with E17110 as compared to TO901317. | 14 | Identification of anti-Gram-negative bacteria agents targeting the interaction between ribosomal proteins L12 and L10显示文摘Gram-negative bacteria have become the main pathogens and cause serious clinical problems with increased morbidity and mortality. However, the slow discovery of new antimicrobial agents is unable to meet the need for the treatment of bacterial infections caused by drug-resistant strains. The interaction of L12 and L10 is essential for ribosomal function and protein synthesis. In this study, a yeast two-hybrid system was established to successfully detect the interaction between L12 and L10 proteins from gram-negative bacteria Escherichia coli, which allows us to screen compounds that specifically disrupt this interaction. With this system, we identified two compounds IMB-84 and IMB-87 that block L12-L10 interaction and show bactericidal activity against E. coli. We used glutathione-S-transferase(GST) pull-down and surface plasmon resonance(SPR) assays to demonstrate that these compounds disrupt L12-L10 interaction in vitro and the target of compounds was further confirmed by the overexpression of target proteins. Moreover, protein synthesis and elongation factor G-dependent GTPase activities are inhibited by two compounds. OBJECTIVE:To investigate whether effects of Ginsenoside Rg1 against Aβare associated with activity of nuclear factor-kappa B(NF-κB). DESIGN,TIME AND SETTING:The randomized,controlled,cell biological experiment was performed at the DME Center,Institute of Clinical Pharmacology,Guangzhou University of Chinese Medicine,China from July 2005 to May 2006. MATERIALS:Beta-amyloid fragment 25-35(Aβ_(25-35)) was supplied by the Neural Biochemical Laboratory,Xuanwu Hospital,Capital Medical University,China.Ginsenoside Rg1 was obtained from National Institute for the Control of Pharmaceutical and Biological Products,China.Rabbit anti-rat NF-κB p65 antibody was purchased from Santa Cruz Biotechnology,USA. METHODS:Hippocampal neurons and cortical astrocytes of neonatal Sprague Dawley rats were harvested and treated with various concentrations(0,5,10,20,and 40μmol/L) of Aβfor 6,12,and 24 hours to establish cellular models of Alzheimer's disease.Cellular models were pretreated with various concentrations of Ginsenoside Rg1(1,2,4,8,and 16μmol/L).According to cell morphology and activity,the following conditions were selected:40μmol/L Aβfor 24 hours,as well as 2,4,and 8μmol/L Ginsenoside Rg1.NF-κB activity was observed using immunofluorescence and cytochemical staining. MAIN OUTCOME MEASURES:Morphology and viability of hippocampal neurons and cortical astrocytes,and activities of NF-κB were measured. RESULTS:Hippocampal neuron activity was significantly greater in the normal and 2 and 4μmol/L Ginsenoside Rg1 groups compared with the model group(P<0.05).Astrocyte activity was significantly greater in the normal,1,2,4,8,and 16μmol/L Ginsenoside Rg1 groups compared with the model group(P<0.05).NF-κB activity of hippocampal neurons was significantly greater in the normal,2,4,and 8μmol/L Ginsenoside Rg1 groups compared with the model group(P<0.01). NF-κB activity of astrocytes was significantly less in the normal,2,4,and 8μmol/L Ginsenoside Rg1 groups compared with the model group(P<0.01 or P<0.05).No significant difference in NF-κB activity was determined between the 2μmol/L Ginsenoside Rg1 and normal groups(P>0.05). | 17 | Radial acceleration estimation of multiple high maneuvering targets显示文摘The acceleration of a high maneuvering target in signal processing is helpful to enhance the performance of the tracker and facilitate the classification of targets. At present, most of the research on acceleration estimation is carried out in cases of a single target with time-frequency analysis methods such as fractional Fourier transform(FRFT), Hough-ambiguity transform(HAT), and Wigner-Ville distribution(WVD), which need to satisfy enough time duration and sampling theorem. Only one reference proposed a method of acceleration estimation for multiple targets based on modified polynomial phase transform(MPPT) in the linear frequency modulation(LFM) continuous-wave(CW) radar. The method of acceleration estimation for multiple targets in the pulse Doppler(PD) radar has not been reported so far. Compressive sensing(CS) has the advantage of sampling at a low rate and short duration without sacrificing estimation performance. Therefore, this paper proposes a new method of acceleration estimation for multiple maneuvering targets with the unknown number based on CS with pulse Doppler signals. Simulation results validate the effectiveness of the proposed method under several conditions with different duration, measurement numbers, signal to noise ratios(SNR), and regularization parameters, respectively. | 19 | The novel ER stress inducer Sec C triggers apoptosis by sulfating ER cysteine residues and degrading YAP via ER stress in pancreatic cancer cells显示文摘Pancreatic adenocarcinoma(PAAD) is one of the most lethal malignancies. Although gemcitabine(GEM) is a standard treatment for PAAD, resistance limits its application and therapy. Secoemestrin C(Sec C) is a natural compound from the endophytic fungus Emericella, and its anticancer activity has not been investigated since it was isolated. Our research is the first to indicate that Sec C is a broad-spectrum anticancer agent and could exhibit potently similar anticancer activity both in GEM-resistant and GEM-sensitive PAAD cells. Interestingly, Sec C exerted a rapid growth-inhibiting effect(80% death at 6 h), which might be beneficial for patients who need rapid tumor shrinkage before surgery. Liquid chromatography/mass spectrometry and N-acetyl-L-cysteine(NAC) reverse assays show that Sec C sulfates cysteines to disrupt disulfide-bonds formation in endoplasmic reticulum(ER) proteins to cause protein misfolding, leading to ER stress and disorder of lipid biosynthesis. Microarray data and subsequent assays show that ER stress-mediated ER-associated degradation(ERAD) ubiquitinates anddownregulates YAP to enhance ER stress via destruction complex(YAP-Axin-GSK-βTr CP), which also elucidates a unique degrading style for YAP.
