| 1 | Therapeutic efficacy of anti-CD19 CAR-T cells in a mouse model of systemic lupus erythematosus显示文摘Dysregulated B-cell activation plays pivotal roles in systemic lupus erythematosus(SLE),which makes B-cell depletion a potential strategy for SLE treatment.The clinical success of anti-CD19 CAR-T cells in treating B-cell malignancies has attracted the attention of researchers.In this study,we aimed to investigate the feasibility of applying anti-CD19 CAR-T cell therapy to SLE treatment in a mouse disease model.We constructed murine anti-CD19 CARs with either CD28 or 4-1BB as the intracellular costimulatory motif and evaluated the therapeutic function of the corresponding CAR-T cells by infusing them into MRL-lpr mice.Furthermore,anti-CD19 CAR-T cells were transferred to MRL-lpr mice before the onset of disease to determine their role in SLE prevention.According to our observations,compared with antibody treatment,the adoptive transfer of our anti-CD19 CAR-T cells showed a more sustained B-cell-depletion effect in MRL-lpr mice.The transfer of syngeneic anti-CD19 CAR-T cells not only prevented disease pathogenesis before the onset of disease symptoms but also displayed therapeutic benefits at a later stage after disease progression.We also tried to optimize the treatment strategy and found that compared with CAR-T cells with the CD28 costimulatory motif,CAR-T cells with the 4-1BB costimulatory motif showed better therapeutic efficiency without cell enrichment.Taken together,these results show that anti-CD19 CAR-T cell therapy was effective in the prevention and treatment of a murine model of SLE,indicating its potential for clinical use in patients. | Xuexiao Jin Qin Xu Chengfei Pu Kaixiang Zhu Cheng Lu Yu Jiang Lei Xiao Yongmei Han Linrong Lu | 2021 | Cellular & Molecular Immunology2021,18,8: | 12 |

| 3 | Study on mechanical properties of soil-rock mixture of various compactness subjected to freeze-thaw cycles显示文摘The soil-rock mixture,a collection of soil particles and rock blocks,is inherently heterogeneous and anisotropic due to significant particle size and material strength differences.This study conducts triaxial tests on soil-rock mixture samples of various compactness subjected to varying freeze-thaw cycles.A mesoscopic simulation is carried out by particle flow code(PFC)to analyze the effects of freeze-thaw cycles on the mechanical properties of soil and rock particles.The results show that the mechanical properties of the soil-rock mixture under freeze-thaw cycles are greatly affected by the initial compaction.In general,when the degree of compaction is higher,the influence of freeze-thaw cycles on the soil-rock mixture is greater.The stress-strain curves of the samples with different compactness demonstrate strain-softening behavior.The freeze-thaw cycles greatly influence the failure strength of the samples with a higher degree of compaction but have little impact on the samples with a lower degree of compaction.On the microscopic level,during freeze-thaw cycles,the pore volume in the highly compacted sample is too small to accommodate the volume expansion from ice crystal formation,causing significant strength loss among the soil and rock particles and deterioration of the macroscopic properties of the soil-rock mixture. | Zhong Zhou HaoHui Ding WenYuan Gao LinRong Xu | 2021 | Research in Cold and Arid Regions2021,13,5: | 2 |

| 7 | Debris Flow Susceptibility Evaluation in Meizoseismal Region:A Case Study in Jiuzhaigou,China显示文摘Jiuzhaigou is situated on a mountain-canyon region and is famous for frequent tectonic activities.An abundance of loose co-seismic landslides and collapses were produced on gullies after the Jiuzhaigou Earthquake on August 8,2017,which was served as material source for debris flow in later years.Debris flow appears frequently which are seriously endangering the safety of people's lives and properties.Even the earliest debris flow appeared in areas where no case ever reported before.The debris flow susceptibility evaluation(DFSE)is used for predicting the areas prone to debris flow,which is urgently required to avoid hazards and help to guide the strategy of preventive measures.Therefore,this work employs debris flow in Jiuzhaigou to reveal the characteristics of disaster-pregnant environment and to explore the application of machine learning in DFSE.Some new viewpoints are suggested:(i)Material density factor of debris flow is first adopted in this work,and it is proved to be a critical factor for triggering debris flows by sensitivity analysis method.(ii)Deep neural network and convolutional neural network(CNN)achieve relatively good area under the curve(AUC)values and are 0.021-0.024 higher than traditional machine learning methods.(iii)Watershed units combined with CNNbased model can achieve more accurate,reliable and practical susceptibility map.This work provides an idea for prevention of debris flow in mountainous lands. | Yongwei Li Linrong Xu Yonghui Shang Shuyang Chen | 2024 | Journal of Earth Science2024,35,1: | 0 |
| 8 | Thymic-specific regulation of TCR signaling by Tespa1显示文摘Double-positive(DP)thymocytes undergo positive selection to become mature single-positive CD4+and CD8+T cells in response to T cell receptor(TCR)signaling.Unlike mature T cells,DP cells must respond to low-affinity self-peptide-MHC ligands before full upregulation of their surface TCR expression can occur.Thus,DP thymocytes must be more sensitive to ligands than mature T cells.A number of molecules have been found that are able to enhance the strength of the TCR signal to facilitate positive selection.However,almost all of these molecules are also active in mature T cells.Themis(thymocyte expressed molecule involved in selection)and Tespa1(thymocyte expressed positive selection associated 1)are two recently discovered molecules essential for optimal TCR signaling and thymocyte development.A deficiency in both molecules leads to defects in positive selection.Here,we compared the relative contributions of Themis and Tespa1 to positive selection in thymocytes.We show that Tespa1 deficiency led to more limited and specific gene expression profile changes in cells undergoing positive selection.In mixed bone marrow transfer experiments,Tespa1^(−/−)cells showed more severe defects in thymocyte development than Themis^(−/−)cells.However,Tespa1^(−/−)cells showed a substantial degree of homeostatic expansion and became predominant in the peripheral lymphoid organs,suggesting that Tespa1 is a thymic-specific TCR signaling regulator.This hypothesis is further supported by our observations in Tespa1 conditional knockout mice,as Tespa1 deletion in peripheral T cells did not affect TCR signaling or cell proliferation.The different regulatory effects of Tespa1 and Themis are in accordance with their nonredundant roles in thymocyte selection,during which Tespa1 and Themis double knockouts showed additive defects. | Jun Lyu Pengfei Wang Ting Xu Yichao Shen Zejin Cui Mingzhu Zheng Guo Fu Linrong Lu | 2019 | Cellular & Molecular Immunology2019,16,12: | 0 |
