1The FTO/miR-181b-3p/ARL5B signaling pathway regulates cell migration and invasion in breast cancer显示文摘Background:N6-methyladenosine(m6A)RNA modification has been demonstrated to be a significant regulatory process in the progression of various tumors,including breast cancer.Fat mass and obesity-associated(FTO)enzyme,initially known as the obesity-related protein,is the first identified m6A demethylase.However,the relationship between FTO and breast cancer remains controversial.In this study,we aimed to elucidate the role and clinical significance of FTO in breast cancer and to explore the underlying mechanism.Methods:We first investigated the expression of FTO in breast cancer cell lines and tissues by quantitative reverse transcription-PCR(qRT-PCR),Western blotting,and immunohistochemistry.Wound healing assay and Transwell assay were performed to determine the migration and invasion abilities of SKBR3 and MDAMB453 cells with either knockdown or overexpression of FTO.RNA sequencing(RNA-seq)was conducted to decipher the downstream targets of FTO.qRT-PCR,luciferase reporter assay,and Western blotting were employed to confirm the existence of the FTO/miR-181b-3p/ARL5B axis.The biological function of ADP ribosylation factor like GTPase 5B(ARL5B)in breast cancer cells was evaluated by wound healing assay and Transwell invasion assay.Results:High FTO expression was observed in human epidermal growth factor receptor 2(HER2)-positive breast cancer,predicting advanced progression(tumor size[P<0.001],nuclear grade[P=0.001],peritumoral lymphovascular invasion[P<0.001),lymph node metastasis[P=0.002],and TNM stage[P=0.001])and poor prognosis.Moreover,FTO promoted cell invasion and migration in vitro.Mechanistically,RNA-seq and further confirmation studies suggested that FTO up-regulated ARL5B by inhibiting miR-181b-3p.We further verified that ARL5B also displayed carcinogenic activity in breast cancer cells.Conclusion:Our work demonstrated the carcinogenic activity of FTO in promoting the invasion and migration of breast cancer cells via the FTO/miR-181b-3p/ARL5B signaling pathway.Yuanyuan Xu Shuang Ye Nan Zhang Shuhui Zheng Huatao Liu Kewen Zhou Ling Wang Yue Cao Peng Sun Tinghuai Wang 2020Cancer Communications2020,40,10:24
2Fucoxanthin Isolated from Undaria pinnatifida Can Interact with Escherichia coli and lactobacilli in the Intestine and Inhibit the Growth of Pathogenic Bacteria显示文摘Fucoxanthin is a xanthophyll-type carotenoid that provides many benefits to human health. However, the mechanism by which fucoxanthin interacts with microbes and inhibits pathogenic bacteria is unknown. In this study, we investigated the effects of fucoxanthin isolated from the edible seaweed Undaria pinnatifida on pathogenic bacteria Escherichia coli and lactobacilli both in vitro and in vivo. Fucoxanthin strongly inhibited the growth of Gram-positive pathogenic bacteria but was less effective against Gram-negative bacteria. Fucoxanthin extracted from the crude mixture had a recovery rate of 93.38% and a purity of 82.70%, which were higher than those of fucoxanthin extracted using a previous method. Fucoxanthin also promoted the growth of intestinal mi- crobes in mice. Fucoxanthinol, a metabolite of fucoxanthin, was generated in the culture media. Fucoxanthin can be deacetylated into fucoxanthinol not only by conventional digestive enzymes in the digestive tract, but also by E. coli and lactobacilli in the intestine. These results indicate that fucoxanthin interacts with and influences E. coli and lactobacilli in the intestine. 5Effects of Organic Enrichment on Sandy Beach Meiofauna:A Laboratory Microcosm Experiment显示文摘Meiofauna samples from intertidal sediments of Qingdao No.2 Bathing Beach, China, were collected for field study, and subjected to organic enrichment in a laboratory microcosm experiment for 21 d. There were three different treatments including non-organic addition as the control, low-organic enrichment (2 g DW green algae per 150 mL) and high-organic enrichment (10 g DW green algae per 150 mL). After 21 d, the meiofauna richness decreased in both organic enrichment treatments. Among the three treatments, total meiofauna abundance was significantly different, and the control groups had higher abundance than the other two treatment groups. However, the responses of the meiofauna abundance in the two organic enrichment treatments were non-significantly different. 6Bioresponsive nanotherapy for preventing dental caries by inhibiting multispecies cariogenic biofilms显示文摘Early childhood caries(ECC)is a public healthcare concern that greatly reduces the quality of life of young children.As a leading factor of ECC,cariogenic biofilms are composed of acidogenic/aciduric pathogens and extracellular polysaccharides(EPSs),creating an acidic and protected microenvironment.Antimicrobial photodynamic therapy(aPDT)is a noninvasive,painless,and efficient therapeutic approach that is suitable for treating ECC.However,due to the hyperfine structure of cariogenic biofilms,most photosensitizers(PSs)could not access and penetrate deeply in biofilms,which dramatically hamper their efficiency in the clinic.Herein,bioresponsive nanoparticle loaded with chlorin e6(MPP-Ce6)is developed,which largely increases the penetration depth(by over 75%)and retention(by over 100%)of PS in the biofilm compared with free Ce6.Furthermore,MPP-Ce6-mediated aPDT not only kills the bacteria in preformed biofilms but also inhibits multispecies biofilm formation.A rampant caries model is established to mimic ECC in vivo,where the population of cariogenic bacteria is decreased to 10%after MPP-Ce6-mediated aPDT.Importantly,the number and severity of carious lesions are efficiently reduced via Keyes’scoring and micro-CT analysis.This simple but effective strategy can serve as a promising approach for daily oral hygiene in preventing ECC.Danfeng Liu Xianbin Ma Yaoting Ji Rourong Chen Shuhui Zhou Hantao Yao Zichen Zhang Mengjie Ye Zhigang Xu Minquan Du 2022Bioactive Materials2022,7,8:2

17Discriminating hydrocarbon generation potential of coaly source rocks and their contribution:a case study from the Upper Paleozoic of Bohai Bay Basin,China显示文摘Although various coaly source rocks widely developed in the Carboniferous-Permian(C-P)of the Bohai Bay Basin,their geochemical characteristics and hydrocarbon generation potential are poorly understood.This study aims to discriminate the contribution of hydrocarbon generation from different C-P coaly source rocks and clarify the differences within generated oils using organic geochemistry,organic petrology,and thermal simulation experiments.The coaly source rocks containt coal clarain and durain,carbonaceous shale,and shale deposited in deltaic and lagoonal environment.The results indicated that clarain,durain,and carbonaceous shale exhibited higher hydrogen index and liquid-gas hydrocarbon yields than lagoonal and deltaic shales,which was mainly associated with the concentrations of sporinite,cutinite,and hydrogen-rich collodetrinite.Aliphatic hydro-carbons originated firom coal and carbonaceous shale presented lower Ts/(Ts+Tm),Ga/17α(H)21β(H)-C_(30)hopane,18α(H)-oleanane/17α(H)21β(H)-C_(30)hopane ratios,and higher 17β3(H)21α(H)-C_(30)Morane/17α(H)21β(H)-C_(30)hopane than deltaic lagoonal shales.Parameters of aromatic hydrocarbons generated from five lithologies of coaly source rocks trended as clear group distribution,e.g.,clarain and durain showing lower MNR,DBT/Fluorene(F)ratios and higher DBF/F ratio than coaly shales.The distinct descending trend of hydrocarbon potential is obtained from clarain,durain,carbonaceous shale to lagoonal and deltaic shales,implying dominated the petroleum and natural gas supplement from coal and carbonaceous shale.The difference between aliphatic and aromatic hydrocarbons provides a significant contribution to analyze the generic relationship between coaly source rock and lacustrine shale.Our results illustrate the importance of coaly source rocks for the in-depth oil-gas exploration of the Bohai Bay Basin and understanding hydrocarbon generation potential of source rocks in coal bearing strata.Jinjun XU Da LOU Qiang JIN Lixin FU Fuqi CHENG Shuhui ZHOU Xiuhong WANG Chao LIANG Fulai LI 2021Frontiers of Earth Science2021,15,4:1
