| 1 | The δ^(18)O variation of a stalagmite from Qixing Cave,Guizhou Province and indi-cated climate change during the Holocene显示文摘A stalagmite from Qixing Cave, Guizhou Province was dated with the TIMS U-series method and its oxygen isotope composition was analyzed. On the basis of the ages and the variations of the δ18O of the stalagmite, the climate change of the last 7.7 ka has been reconstructed in this area: 7.7-5.8 ka, summer monsoon maximum period; 5.8-3.8 ka summer monsoon weakening period; 3.8-0.15 ka, weakened summer monsoon and high amplitude climate fluctuations period. We put forward that the increasing trend of δ18O of the stalagmite reflected not only the weakening of the summer monsoon, which was caused by the decreasing of solar radiation in the Northern Hemisphere, but also the possibly decreasing contribution of the southwest monsoon to the rainfall of this area. These results are consistent with the output of the numerical climate-model modeling. | 2 | Air route network optimization in fragmented airspace based on cellular automata显示文摘Air route network optimization,one of the essential parts of the airspace planning,is an effective way to optimize airspace resources,increase airspace capacity,and alleviate air traffic congestion.However,little has been done on the optimization of air route network in the fragmented airspace caused by prohibited,restricted,and dangerous areas(PRDs).In this paper,an air route network optimization model is developed with the total operational cost as the objective function while airspace restriction,air route network capacity,and non-straight-line factors(NSLF) are taken as major constraints.A square grid cellular space,Moore neighbors,a fixed boundary,together with a set of rules for solving the route network optimization model are designed based on cellular automata.The empirical traffic of airports with the largest traffic volume in each of the 9 flight information regions in China's Mainland is collected as the origin-destination(OD) airport pair demands.Based on traffic patterns,the model generates 35 air routes which successfully avoids 144 PRDs.Compared with the current air route network structure,the number of nodes decreases by 41.67%,while the total length of flight segments and air routes drop by 32.03% and 5.82% respectively.The NSLF decreases by 5.82% with changes in the total length of the air route network.More importantly,the total operational cost of the whole network decreases by 6.22%.The computational results show the potential benefits of the model and the advantage of the algorithm.Optimization of air route network can significantly reduce operational cost while ensuring operation safety. | Shijin WANG Xi CAO Haiyun LI Qingyun LI Xu HANG Yanjun WANG | 2017 | Chinese Journal of Aeronautics2017,30,3: | 16 |
| 3 | PrP mutants with different numbers of octarepeat sequences are more susceptible to the oxidative stress显示文摘One of the physiological functions of cellular prion protein(PrP C )is believed to work as a cellular resistance to oxidative stress,in which the octarepeats region within PrP plays an important role.However,the detailed mechanism is less clear.In this study,the expressing plasmids of wild-type PrP (PrP-PG5)and various PrP mutants containing 0(PrP-PG0),9(PrP-PG9)and 12(PrP-PG12)octarepeats were generated and PrP proteins were expressed both in E.coli and in mammalian cells.Protein aggregation and formation of carbonyl groups were clearly seen in the recombinant PrPs expressed from E.coli after treatment of H2O2.MTT and trypan blue staining assays revealed that the cells expressing the mutated PrPs within octarepeats are less viable than the cells expressing wild-type PrP.Statistically significant high levels of intracellular free radicals and low levels of glutathione peroxidase were observed in the cells transfected with plasmids containing deleted or inserted octarepeats.Remarkably more productions of carbonyl groups were detected in the cells expressing PrPs with deleted and inserted octarepeats after exposing to H2O2.Furthermore,cells expressing wild-type PrP showed stronger resistant activity to the challenge of H2O2 at certain extent than the mutated PrPs and mock. | 4 | Smartphone-powered iontophoresis-microneedle array patch for controlled transdermal delivery显示文摘The incidence rate of diabetes has been increasing every year in nearly all nations and regions.The traditional control of diabetes using transdermal insulin delivery by metal needles is generally associated with pain and potential infections.While microneedle arrays(MAs)have emerged as painless delivery techniques,the integration of MA systems with electronic devices to precisely control drug delivery has rarely been realized.In this study,we developed an iontophoresis-microneedle array patch(IMAP)powered by a portable smartphone for the active and controllable transdermal delivery of insulin.The IMAP in situ integrates iontophoresis and charged nanovesicles into one patch,achieving a one-step drug administration strategy of“penetration,diffusion and iontophoresis”.The MA of the IMAP is first pressed on the skin to create microholes and then is retracted,followed by the iontophoresis delivery of insulin-loaded nanovesicles through these microholes in an electrically controlled manner.This method has synergistically and remarkably enhanced controlled insulin delivery.The amount of insulin can be effectively regulated by the IMAP by applying different current intensities.This in vivo study has demonstrated that the IMAP effectively delivers insulin and produces robust hypoglycemic effects in a type-1 diabetic rat model,with more advanced controllability and efficiency than delivery by a pristine microneedle or iontophoresis.The IMAP system shows high potential for diabetes therapy and the capacity to provide active as well as long-term glycemic regulation without medical staff care. | Jingbo Yang Yanjun Li Rui Ye Ying Zheng Xiangling Li Yuzhen Chen Xi Xie Lelun Jiang | 2020 | Microsystems & Nanoengineering2020,6,1: | 3 |
| 5 | The impact of lipids on the cancer—immunity cycle and strategies for modulating lipid metabolism to improve cancer immunotherapy显示文摘Lipids have been found to modulate tumor biology,including proliferation,survival,and metastasis.With the new understanding of tumor immune escape that has developed in recent years,the influence of lipids on the cancer—immunity cycle has also been gradually discovered.First,regarding antigen presentation,cholesterol prevents tumor antigens from being identified by antigen presenting cells.Fatty acids reduce the expression of major histocompatibility complex class I and costimulatory factors in dendritic cells,impairing antigen presentation to T cells.Prostaglandin E2(PGE2)reduce the accumulation of tumor-infiltrating dendritic cells.Regarding T-cell priming and activation,cholesterol destroys the structure of the T-cell receptor and reduces immunodetection.In contrast,cholesterol also promotes T-cell receptor clustering and relative signal transduction.PGE2 represses T-cell proliferation.Finally,regarding T-cell killing of cancer cells,PGE2 and cholesterol weaken granule-dependent cytotoxicity.Moreover,fatty acids,cholesterol,and PGE2 can improve the activity of immunosuppressive cells,increase the expression of immune checkpoints and promote the secretion of immunosuppressive cytokines.Given the regulatory role of lipids in the cancer—immunity cycle,drugs that modulate fatty acids,cholesterol and PGE2 have been envisioned as effective way in restoring antitumor immunity and synergizing with immunotherapy.These strategies have been studied in both preclinical and clinical studies. | Mingming Zheng Wenxin Zhang Xi Chen Hongjie Guo Honghai Wu Yanjun Xu Qiaojun He Ling Ding Bo Yang | 2023 | Acta Pharmaceutica Sinica B2023,13,4: | 1 |

| 9 | The regulatory roles of DDIT4 in TDCIPP-induced autophagy and apoptosis in PC12 cells显示文摘Tris(1,3-dichloro-2-propyl) phosphate(TDCIPP) is a commonly used organophosphatebased flame retardant and can bio-accumulate in human tissues and organs. As its structure is similar to that of neurotoxic organophosphate pesticides, the neurotoxicity of TDCIPP has raised widespread concerns. TDCIPP can increase neuronal apoptosis and induce autophagy.However, its regulatory mechanism remains unclear. In this study, we found that the expression upregulation of the DNA Damage-Inducible Transcript 4(DDIT4) protein, which might play essential roles in TDCIPP-induced neuronal autophagy and apoptosis, was observed in TDCIPP-treated differentiated rat PC12 cells. Furthermore, we determined the protective effect of the DDIT4 suppression on the autophagy and apoptosis induced by TDCIPP using Western blot(WB) and Flow cytometry(FACS) analysis. We observed that TDCIPP treatment increased the DDIT4, the autophagy marker Beclin-1, and the microtubule-associated protein light chain 3-II(LC_(3)II) expressions and decreased the mTOR phosphorylation levels. Conversely, the suppression of DDIT4 expression increased the p-mTOR expression and decreased cell autophagy and apoptosis. Collectively, our results revealed the function of DDIT4 in cell death mechanisms triggered by TDCIPP through the m TOR signaling axis in differentiated PC12 cells. | 12 | Mevalonate improves anti-PD-1/PD-L1 efficacy by stabilizing CD274 mRNA显示文摘Mevalonate metabolism plays an important role in regulating tumor growth and progression;however,its role in immune evasion and immune checkpoint modulation remains unclear.Here,we found that non-small cell lung cancer(NSCLC)patients with higher plasma mevalonate response better to antiPD-(L)1 therapy,as indicated by prolonged progression-free survival and overall survival.Plasma mevalonate levels were positively correlated with programmed death ligand-1(PD-L1)expression in tumor tissues.In NSCLC cell lines and patient-derived cells,supplementation of mevalonate significantly upregulated the expression of PD-L1,whereas deprivation of mevalonate reduced PD-L1 expression.Mevalonate increased CD274 mRNA level but did not affect CD274 transcription.Further,we confirmed that mevalonate improved CD274 mRNA stability.Mevalonate promoted the affinity of the AU-rich elementbinding protein HuR to the 3'-UTR regions of CD274 mRNA and thereby stabilized CD274 mRNA.By in vivo study,we further confirmed that mevalonate addition enhanced the anti-tumor effect of anti-PD-L1,increased the infiltration of CD8^(+)T cells,and improved cytotoxic function of T cells.Collectively,our findings discovered plasma mevalonate levels positively correlated with the therapeutic efficacy of anti-PD-(L)1 antibody,and provided the evidence that mevalonate supplementation could be an immunosensitizer in NSCLC. | Wenxin Zhang Xiaohui Pan Yanjun Xu Hongjie Guo Mingming Zheng Xi Chen Honghai Wu Fengming Luan Qiaojun He Ling Ding Bo Yang | 2023 | Acta Pharmaceutica Sinica B2023,13,6: | 0 |
