1Preventive effect of chrysin on experimental autoimmune uveitis triggered by injection of human IRBP peptide 1-20 in mice显示文摘Uveitis is a common cause of blindness worldwide.Experimental autoimmune uveitis(EAU)is an animal model of noninfectious uveitis.Chrysin(5,7-dihydroxyflavone)is a member of the flavonoid family and has anti-inflammatory effects.We immunized C57BL/6J mice with human interphotoreceptor retinoid-binding protein peptide 1–20 to induce EAU.Chrysin was administered intragastrically at 25 mg/kg daily to the chrysin-treated mice from 3 days before immunization to 21 days after immunization.Vehicle was administered to the mice in the control group according to the same protocol.Lower clinical and histopathological scores,increased integrity of the blood–retinal barrier(BRB)and higher expression of tight junction proteins were observed in the chrysin-treated mice.Chrysin significantly decreased the proportions of Th1,Th17 and CD4^(+)CD3^(+)CD62L^(+)Th0 cells,and increased the proportion of Treg cells.Both macrophage infiltration and the expression of inducible nitric oxide synthase in the retina were efficiently inhibited by chrysin treatment.In chrysin-treated mice,the expression of interferon-γ,interleukin(IL)-17A,IL-6,IL-1βand tumor necrosis factor-αwas reduced in the retina,whereas higher levels of transforming growth factor-βwere detected.Furthermore,NF-κBp65 was downregulated after chrysin treatment.In conclusion,as an anti-inflammatory molecule,chrysin exerts a preventive effect on EAU by modulating the balance among helper T-cell subsets and suppressing ocular inflammation,thereby maintaining the integrity of the BRB.Xiangda Meng Sijie Fang Zhuhong Zhang Yang Wang Caiyun You Jingkai Zhang Hua Yan 2017Cellular & Molecular Immunology2017,14,8:3
2Effect of Image Noise on the Classification of Skin Lesions Using Deep Convolutional Neural Networks显示文摘Skin lesions are in a category of disease that is both common in humans and a major cause of death.The classification accuracy of skin lesions is a crucial determinant of the success rate of curing lethal diseases.Deep Convolutional Neural Networks(CNNs)are now the most prevalent computer algorithms for the purpose of disease classification.As with all algorithms,CNNs are sensitive to noise from imaging devices,which often contaminates the quality of the images that are fed into them.In this paper,a deep CNN(Inception-v3)is used to study the effect of image noise on the classification of skin lesions.Gaussian noise,impulse noise,and noise made up of a compound of the two are added to an image dataset,namely the Dermofit Image Library from the University of Edinburgh.Evaluations,based on t-distributed Stochastic Neighbor Embedding(t-SNE)visualization,Receiver Operating Characteristic(ROC)analysis,and saliency maps,demonstrate the reliability of the Inception-v3 deep CNN in classifying noisy skin lesion images.Xiaoyu Fan Muzhi Dai Chenxi Liu Fan Wu Xiangda Yan Ye Feng Yongqiang Feng Baiquan Su 2020Tsinghua Science and Technology2020,25,3:2
3Cholesterol modulating the orientation of His17 in hepatitis C virus p7 (5a) viroporin--A molecular dynamic simulation study显示文摘Protein p7 of HCV is a 63 amino acid channel forming membrane protein essential for the progression of viral infection and the sensitivity of this channel to small-molecule inhibitors renders p7 a potential target for novel therapies against HCV infection. Previous biochemical experiments suggested that the His17 of p7 is a pore-lining residue and solvated-exposed to participate in channel gating. However, a recent NMR structural identi fication of the p7 hexamer in dodecylphosphocholine(DPC) micelles indicated that the His17 is embedded into the protein matrix. In this work, we performed molecular dynamic simulations to bridge the controversial observations. Our results illustrated that by incorporating the cholesterol into DOPC membranes to mimic an actual membrane-like composition,the orientation of His17 in the hexameric bundles spontaneously access to the central pore region,indicating a versatile property of the p7 viroporin conformation that could be voluntarily in fluenced by its surrounding environments.Yuebin Zhang Xiangda Peng Hong Ren Huiying Chu Yan Li Guohui Li 2018Chinese Chemical Letters2018,29,5:0
