Wnt/β-catenin signalling:function, biological mechanisms, and therapeutic opportunities显示文摘The Wnt/β-catenin pathway comprises a family of proteins that play critical roles in embryonic development and adult tissue homeostasis. The deregulation of Wnt/β-catenin signalling often leads to various serious diseases, including cancer and non-cancer diseases.Although many articles have reviewed Wnt/β-catenin from various aspects,a systematic review encompassing the origin, composition, function, and clinical trials of the Wnt/β-catenin signalling pathway in tumour and diseases is lacking. In this article,we comprehensively review the Wnt/β-catenin pathway from the above five aspects in combination with the latest research. Finally,we propose challenges and opportunities for the development of small-molecular compounds targeting the Wnt signalling pathway in disease treatment.Jiaqi Liu Qing Xiao Jiani Xiao Chenxi Niu Yuanyuan Li Xiaojun Zhang Zhengwei Zhou Guang Shu Gang Yin 2022Signal Transduction and Targeted Therapy2022,7,2:28
Phase Transformation Behavior and Microstructural Control of High-Cr Martensitic/Ferritic Heat-resistant Steels for Power and Nuclear Plants: A Review显示文摘The martensitic/ferritic steels have been used as boiler and turbine materials in power plants, and also been selected as potential materials for structural materials in nuclear reactors. In this paper, the kinetic analysis of the martensite formation and microstructural control of high-Cr martensitic/ferritic steels are reviewed. A modular approach, incorporating Fisher partitioning nucleation and anisotropic growth for impingement, was proposed to describe the martensite formation kinetics under different cooling rates. The kinetic analysis suggested a thermal-activated growth feature occurring during the martensitic transformation of martensitic steels. The microstructure can be tuned by composition optimization and various combinations of heat treatment parameters(temperature, time, severe and minor deformation). For the application in power plant, the potential of boundary-design, refinement of original austenite grain size and the final martensitic lath, pinning effect of stable carbides, in improving the performances of martensitic/ferritic steels at elevated temperatures should be investigated more thoroughly.Furthermore, efforts should be made to explore the effects of retained austenite on the improvement of high-temperature creep strength. For the application of nuclear plants, attempts should also be made to produce Fe powders with uniformly distributed oxide particles by chemical reactions.Xiaosheng Zhou Chenxi Liu Liming Yu Yongchang Liu Huijun Li 2015Journal of Materials Science & Technology2015,31,3:26
Stem cell-based bone and dental regeneration: a view of microenvironmental modulation显示文摘In modern medicine, bone and dental loss and defects are common and widespread morbidities, for which regenerative therapy has shown great promise.Mesenchymal stem cells, obtained from various sources and playing an essential role in organ development and postnatal repair, have exhibited enormous potential for regenerating bone and dental tissue.Currently, mesenchymal stem cells (MSCs)-based bone and dental regeneration mainly includes two strategies: the rescue or mobilization of endogenous MSCs and the application of exogenous MSCs in cytotherapy or tissue engineering.Nevertheless, the efficacy of MSCbased regeneration is not always fulfilled, especially in diseased microenvironments.Specifically, the diseased microenvironment not only impairs the regenerative potential of resident MSCs but also controls the therapeutic efficacy of exogenous MSCs,both as donors and recipients.Accordingly, approaches targeting a diseased microenvironment have been established, including improving the diseased niche to restore endogenous MSCs,enhancing MSC resistance to a diseased microenvironment and renormalizing the microenvironment to guarantee MSC-mediated therapies. Moreover, the application of extracellular vesicles (EVs) as cell-free therapy has emerged as a promising therapeutic strategy. In this review,we summarize current knowledge regarding the tactics of MSC-based bone and dental regeneration and the decisive role of the microenvironment, emphasizing the therapeutic potential of microenvironment-targeting strategies in bone and dental regenerative medicine.Chenxi Zheng Ji Chen Shiyu Liu Yan Jin 2019International Journal of Oral Science2019,11,3:18
SHP2 inhibition triggers anti-tumor immunity and synergizes with PD-1 blockade显示文摘Tyrosine phosphatase SHP2 is a promising drug target in cancer immunotherapy due to its bidirectional role in both tumor growth promotion and T-cell inactivation. Its allosteric inhibitor SHP099 is known to inhibit cancer cell growth both in vitro and in vivo. However, whether SHP099-mediated SHP2 inhibition retards tumor growth in vivo via anti-tumor immunity remains elusive. To address this, a CT-26 colon cancer xenograft model was established in mice since this cell line is insensitive to SHP099.Consequently, SHP099 minimally affected CT-26 tumor growth in immuno-deficient nude mice, but significantly decreased the tumor burden in CT-26 tumor-bearing mice with intact immune system.SHP099 augmented anti-tumor immunity, as shown by the elevated proportion of CD8tIFN-γtT cells and the upregulation of cytotoxic T-cell related genes including Granzyme B andPerforin, which decreased the tumor load. In addition, tumor growth in mice with SHP2-deficient T-cells was markedly slowed down because of enhanced anti-tumor responses. Finally, the combination of SHP099 and antiPD-1 antibody showed a higher therapeutic efficacy than either monotherapy in controlling tumor growthin two colon cancer xenograft models, indicating that these agents complement each other. Our study suggests that SHP2 inhibitor SHP099 is a promising candidate drug for cancer immunotherapy.Mingxia Zhao Wenjie Guo Yuanyuan Wu Chenxi Yang Liang Zhong Guoliang Deng Yuyu Zhu Wen Liu Yanhong Gu Yin Lu Lingdong Kong Xiangbao Meng Qiang Xu Yang Sun 2019Acta Pharmaceutica Sinica B2019,9,2:18
Formation mechanism and control methods of acicular ferrite in HSLA steels:A review显示文摘High strength low alloy(HSLA) steels have been widely used in pipelines, power plant components, civil structures and so on,due to their outstanding mechanical properties as high strength and toughness, and excellent weldability.Multi-phase microstructures containing acicular ferrite or acicular ferrite dominated phase have been proved to possess good comprehensive properties in HSLA steels. This paper mainly focuses on the formation mechanisms and control methods of acicular ferrite in HSLA steels.Effect of austenitizing conditions, continuous cooling rate, and isothermal quenching time and temperature on acicular ferrite transformation was reviewed.Furthermore, the modified process to control the formation of multi-phase microstructures containing acicular ferrite,as intercritical heat treatments, step quenching treatments and thermo-mechanical controlled processing, was summarized. The favorable combination of mechanical properties can be achieved by these modified treatments.Yi Shao Chenxi Liu Zesheng Yan Huijun Li Yongchang Liu 2018Journal of Materials Science & Technology2018,34,5:13
Precipitation and hot deformation behavior of austenitic heat-resistant steels: A review显示文摘The austenitic heat resistant-steels have been considered as important candidate materials for advanced supercritical boilers, nuclear reactors, super heaters and chemical reactors, due to their favorable combination of high strength, corrosion resistance, perfect mechanical properties, workability and low cost.Since the precipitation behavior of the steels during long-term service at elevated temperature would lead to the deterioration of mechanical properties, it is essential to clarify the evolution of secondary phases in the microstructure of the steels. Here, a summary of recent progress in the precipitation behavior and the coarsening mechanism of various precipitates during aging in austenitic steels is made. Various secondary phases are formed under service conditions, like MX carbonitrides, M_(23)C_6 carbides, Z phase, sigma phase and Laves phase. It is found that the coarsening rate of M_(23)C_6 carbides is much higher than that of MX carbonitrides. In order to understand the thermal deformation mechanism, a constitutive equation can be established, and thus obtained processing maps are beneficial to optimizing thermal processing parameters, leading to improved thermal processing properties of steels.Yinghui Zhou Yongchang Liu Xiaosheng Zhou Chenxi Liu Jianxin Yu Yuan Huang Huijun Li Wenya Li 2017Journal of Materials Science & Technology2017,33,12:12
Global Dynamic Molecular Profiling of Stomatal Lineage Cell Development by Single-Cell RNA Sequencing显示文摘The regulation of stomatal lineage cell development has been extensively investigated. However,a comprehensive characterization of this biological process based on single-cell transcriptome analysis has not yet been reported. In this study,we performed RNA sequencing on 12844 individual cells from the cotyledons of 5-day-old Arabidopsis seedlings. We identified 11 cell clusters corresponding mostly to cells at specific stomatal developmental stages using a series of marker genes.Comparative analysis of genes with the highest variable expression among these cell clusters revealed transcriptional networks that regulate development from meristemoid mother cells to guard mother cells.Examination of the developmental dynamics of marker genes via pseudo-time analysis revealed potential interactions between these genes.Collectively, our study opens the door for understanding how the identified novel marker genes participate in the regulation of stomatal lineage cell development.Zhixin Liu Yaping Zhou Jinggong Guo Jiaoai Li Zixia Tian Zhinan Zhu Jiajing Wang Rui Wu Bo Zhang Yongjian Hu Yijing Sun Yan Shangguan Weiqiang Li Tao Li Yunhe Hu Chenxi Guo Jean-David Rochaix Yuchen Miao Xuwu Sun 2020Molecular Plant2020,13,8:11
Cotton bollworm resistance to Bt transgenic cotton: A case analysis显示文摘Cotton bollworm (Helicoverpa armigera) is one of the most serious insect pests of cotton. Transgenic cotton expressing Cry toxins derived from a soil bacterium, Bacillus thuringiensis (Bt), has been produced to target this pest. Bt cotton has been widely planted around the world, and this has resulted in efficient control of bollworm populations with reduced use of synthetic insecticides. However, evolution of resistance by this pest threatens the continued success of Bt cotton. To date, no field populations of bollworm have evolved significant levels of resistance; however, several laboratory-selected Cry-resistant strains of H. armigera have been obtained, which suggests that bollworm has the capacity to evolve resistance to Bt. The development of resistance to Bt is of great concern, and there is a vast body of research in this area aimed at ensuring the continued success of Bt cotton. Here, we review studies on the evolution of Bt resistance in H. armigera, focusing on the biochemical and molecular basis of Bt resistance. We also discuss resistance management strategies, and monitoring programs implemented in China, Australia, and India.LIU ChenXi, LI YunHe, GAO YuLin, NING ChangMing & WU KongMing State Key Laboratory of Plant Disease and Insect Pests, Institute of Plant Protection, Chinese Academy of Agricultural Science, Beijing 100193, China 2010Science China(Life Sciences)2010,53,8:10
Cystathionine-γ-lyase ameliorates the histone demethylase JMJD3-mediated autoimmune response in rheumatoid arthritis显示文摘Cystathionine-γ-lyase(CSE),an enzyme associated with hydrogen sulfide(H2S)production,is an important endogenous regulator of inflammation.Jumonji domain-containing protein 3(JMJD3)is implicated in the immune response and inflammation. Here,we investigated the potential contribution of JMJD3 to endogenous CSE-mediated inflammation in rheumatoid arthritis(RA).Upregulated CSE and JMJD3 were identified in synovial fibroblasts(SFs)from RA patients as well as in the joints of arthritic mice.Knocking down CSE augmented inflammation in IL-1β-induced SFs by increasing JMJD3 expression. In addition,CSE−/−mice with collagen-induced arthritis(CIA)developed severe joint inflammation and bone erosion.Conversely, overexpressing CSE inhibited JMJD3 expression by the transcription factor Sp-1 and was accompanied by reduced inflammation in IL-1β-treated SFs.Furthermore,JMJD3 silencing or the administration of the JMJD3 inhibitor GSK-J4 significantly decreased the inflammatory response in IL-1β-treated SFs,mainly by controlling the methylation status of H3K27me3 at the promoter of its target genes.GSK-J4 markedly attenuated the severity of arthritis in CIA mice.In conclusion, suppressing JMJD3 expression by the transcription factor Sp-1 is likely responsible for the ability of CSE to negatively modulate the inflammatory response and reduce the progression of RA.Weijun Wu Ming Qin Wanwan Jia Zheng Huang Zhongzheng Li Di Yang Mengwei Huang Chenxi Xiao Fen Long Jianchun Mao Philip K.Moore Xinhua Liu Yi Zhun Zhu 2019Cellular & Molecular Immunology2019,16,8:9
Microstructures and mechanical properties of friction stir welds on 9% Cr reduced activation ferritic/martensitic steel显示文摘In this study, the microstructures and mechanical properties of 9%Cr reduced activation ferritic/martensitic(RAFM) steel friction stir welded joints were investigated. When a W-Re tool is used, the recommended welding parameters are 300 rpm rotational speed,60 mm/min welding speed and10 kn axial force. In stir zone(SZ),austenite dynamic recrystallization induced by plastic deformation and the high cooling rates lead to an obvious refinement of prior austenite grains and martensite laths. The microstructure in SZ contains lath martensite with high dislocation density,a lot of nano-sized MX and M_3C phase particles, but almost no M_(23)C_6 precipitates. In thermal mechanically affect zone(TMAZ)and heat affect zone(HAZ),refinement of prior austenite and martensitic laths and partial dissolution of M_(23)C_6 precipitates are obtained at relatively low rotational speed. However, with the increase of heat input, coarsening of martensitic laths, prior austenite grains, and complete dissolution of M_(23)C_6 precipitates are achieved.Impact toughness of SZ at-20?C is slightly lower than that of base material(BM),and exhibits a decreasing trend with the increase of rotational speed.Chao Zhang Lei Cui Yongchang Liu Chenxi Liu Huijun Li 2018Journal of Materials Science & Technology2018,34,5:8
Effects of total soy saponins on free radicals in the quadriceps femoris, serum testosterone,LDH,and BUN of exhausted rats显示文摘Purpose:The aim of this study was to investigate the impact of total soy saponins(TS) on the free radical metabolism from the quadriceps femoris muscle, serum testosterone, lactate dehydrogenase(LDH),and blood urea nitrogen(BUN) in rats exercised to exhaustion. Methods:A one-time exhausted treadmill exercise session was used.Sprague-Dawley rats were divided into 4 groups:a control group—animals receiving no TS and no exercise(NTSNE),animals receiving TS but no exercise group(TSNE),animals receiving no TS but exercised to exhaustion group(NTSE),and animals receiving TS and exercised to exhaustion group(TSE).The TSNE and TSE groups were fed TS at a dosage of 20 mg/kg body weight once per day for 2 weeks. The NTSE group was given a placebo, and the NTSNE group was not given any treatment. The NTSE and TSE groups were exercised at speed of 30 m/min on treadmill until exhausted. The exercise time and exercise distance were recorded when the rats became exhausted and the rats were then decapitated and anatomized immediately.A 10% homogenate of the quadriceps femoris tissue was prepared. The levels of superoxide dismutase(SOD),catalase(CAT),malondialdehyde(MDA),glutathione peroxidase(GSH-Px),glutathione reductase(GR),reduced glutathione(GSH),total antioxidant capacity(T-AOC),LDH,BUN,and serum testosterone were tested. Results:TS significantly increased the exercise time to exhaustion by 20.62%(p < 0.05).The MDA levels were decreased significantly in the TSNE group than in NTSNE group(p < 0.05);the T-AOC levels increased significantly in the TSNE group than in the other 3 groups(p < 0.01,p < 0.05,p < 0.05).The LDH activity significantly increased in the NTSE group than in TSNE group(p < 0.05).The BUN levels significantly increased in the NTSE group than in the other 3 groups(p < 0.01,p < 0.01,p < 0.05),and significantly increased in the TSE group than in NTSNE and TSNE groups(both p < 0.01).The serum testosterone levels increased significantly in the TSNE group than in the other 3 groups(all p < 0.01).SOD,CAT,GSH-Px,GR,and GSH were not statistically different among the groups. Conclusion:TS can significantly improve the exercised rats' serum testosterone level and antioxidant activity in their quadriceps femoris to varying degrees, decrease MDA and serum LDH and BUN levels, increase the exercise time, and delay the occurrence of the fatigue.Zhigang Liu Ruixin Nie Yun Liu Zhouhong Li Chenxi Yang Zhengying Xiong 2017Journal of Sport and Health Science2017,6,3:8
Variation of activation energy determined by a modified Arrhenius approach:Roles of dynamic recrystallization on the hot deformation of Ni-based superalloy显示文摘The hot deformation behaviors of Ni18 Cr9 Co9 Fe5 Nb3 Mo superalloy were explored in the formation temperature range free ofγ’phase with various strain rates applied. The hot deformation behaviors are initially modeled with Arrhenius equation which gives an average activation energy of 581.1 kJ mol^(-1).A modified Arrhenius approach, including the updated Zener-Hollomon parameter is proposed to consider the change of activation ene rgy under different deformation conditions which turns out a relatively accurate computation for activation energy of hot deformation,i.e.,the standard variance for modified model calculated in the covered deformation condition is just 35.4%of that for Arrhenius equation. The modified model also proposes a map for activation ene rgy which ranges from 571.5-589.0 kJ mol^(-1)for various deformation conditions.Microstructural features of the representative superalloy specimens were characterized by electron backscattered diffraction(EBSD)techniques in order to clarify the influence of activation energy on the microstructural formation. It is found that the Ni-based superalloy samples with higher activation energy are promoted by the degree of dynamic recrystallization which suggests that the rise in activation energy gives either a better recrystallization rate or finer grains.Peiru Yang Chenxi Liu Qianying Guo Yongchang Liu 2021Journal of Materials Science & Technology2021,,13:8
Controlled synthesis of high-quality W-Y2O3 composite powder precursor by ascertaining the synthesis mechanism behind the wet chemical method显示文摘As an emerging preparation technology, wet chemical method has been employed widely to produce lots of alloy materials such as W and Mo based alloys, owing to its unique technical advantages.Ascertaining the synthesis mechanism behind wet chemical method is indispensable for controlled synthesis of highquality W-Y2 O3 composite powder precursor. The co-deposition mechanism of yttrium and tungsten component behind the wet chemical method of preparing yttrium-doped tungsten composite nanopowder was investigated systematically in this work.A series of co-deposited composite powders fabricated under different acidity conditions were used as research targets for investigating the effect of surface composition and structure on co-deposition efficiency. It was found that white tungstic acid has more W-OH bonds and much higher co-deposition efficiency with Y^3+ions than yellow tungstic acid.It is illustrated that the coordination reaction between W-OH bonds on tungstic acid particles and Y^3+ions brings the co-deposition of yttrium and tungsten component into being.Through displacing H^+ions in W-OH bonds,Y^3+ions can be adsorbed on the surface of or incorporated into tungstic acid particles in form of ligand.Consequently,to control and regulate Y2 O3 content in powder precursor accurately,H^+ion concentration in wet chemical reaction should be in range of 0.55-2.82 mol L^-1 to obtain white tungstic acid.Besides,H^+ion concentration also has prominent effect on the grain size and morphology of reduced powder precursor. The optimal value should be around 1.58 mol L^-1,which can lead to minimum W grain size(about 17 nm) without bimodal structure. The chemical mechanism proposed in this work could produce great sense to preparation of high-quality precursor for sintering high-performance Y2 O3 dispersion strengthened W based alloys. Our work may also shed light on the approach to exploit analogous synthesis mechanism in other alloy systems.Zhi Dong Nan Liu Weiqiang Hu Zongqing Ma Chong Li Chenxi Liu Qianying Guo Yongchang Liu 2020Journal of Materials Science & Technology2020,36,1:7
Tailoring the secondary phases and mechanical properties of ODS steel by heat treatment显示文摘The oxide dispersion strengthened(ODS) steel with the nominal composition of Fe–14 Cr–2 W–0.3 Ti–0.2 V–0.07 Ta–0.3 Y2O3(wt%) was fabricated by mechanical alloying and hot isostatic pressing(HIP). In order to optimize the relative volume fraction of secondary phases, the as-HIPed ODS steel was annealed at 800°C, 1000°C, 1200°C for 5 h, respectively. The microstructures and different secondary phases of the as-HIPed and annealed ODS samples were identified by scanning electron microscopy(SEM), transmission electron microscopy(TEM), and X-ray diffraction(XRD). The tensile properties of all the ODS steels at room temperature were also investigated. The results indicate that annealing is an effective way to control the microstructure and the integral secondary phases. The annealing process promotes the dissolution of M23C6 particles, thus promoting the precipitation of TiC.No obvious coarsening of Y2Ti2O7 nanoparticles can be observed during annealing. The tensile results indicate that the annealed ODS sample with the optimized secondary phases and high density possesses the best mechanical properties.Qian Zhao Zongqing Ma Liming Yu Huijun Li Chenxi Liu Chong Li Yongchang Liu 2019Journal of Materials Science & Technology2019,35,6:6

Danggui Buxue Tang ameliorates bleomycin-induced pulmonary fibrosis in rats through inhibiting transforming growth factor-β1/Smad3/plasminogen activator inhibitor-1 signaling pathway显示文摘OBJECTIVE: To investigate the effect of Danggui Buxue Tang(DBT), a decoction from Traditional Chinese Medicine, on bleomycin-induced pulmonary fibrosis in rats, and to propose the possible underlying mechanism.METHODS: Forty male Sprague-Dawley rats were randomly divided into sham group, model group, prednisone group and DBT group. Pulmonary fibrosis rat model was established by intratracheal injection with bleomycin. Body weight and lung index were monitored. Histopathologic examination and collagen deposition were determined using Hematoxylin and eosin(HE) and Masson's trichrome staining. Immunohistochemistry staining was applied to observe the expression of alpha-smooth muscle actin(α-SMA). m RNA expression of α-SMA,collagen Ⅰ and collagen Ⅲ were measured by realtime fluorescence quantitative PCR(RT-q PCR). Inflammatory cytokines, including tumor necrosis factor alpha(TNF-α), interleukin-6(IL-6) and IL-1β in serum were detected by Enzyme-linked immunosorbent assay. Alkali hydrolysis method was conducted to investigate the content of hydroxyproline(HYP). Transforming growth factor-β1(TGF-β1),Smad3 and plasminogen activator inhibitor-1(PAI-1) protein level were examined by Western blot assay.RESULTS: DBT significantly reduced the severity of bleomycin-induced pulmonary fibrosis and inflammation as indicated by minimizing the lost of weight, and by lowering the levels of lung index, inflammation score, Ashcroft score, collagen volume fraction(%), HYP, α-SMA, collagen Ⅰ, collagen Ⅲ,TNF-α, IL-6, IL-1β, TGF-β1, Smad3 and PAI-1, consistent with the effect of prednisone.CONCLUSION: Our findings suggest that DBT is able to ameliorate the pulmonary fibrosis, the possible mechanism may involve inhibition of pulmonary inflammation and collagen deposition, possibly via suppressing TGF-β1/Smad3/PAI-1 signaling pathway.Wang Jiepeng Fang Chaoyi Wang Shaoxian Fang Fang Chu Xinqiao Liu Na Lu Chenxi Wang Shuo Li Wei 2020Journal of Traditional Chinese Medicine2020,40,2:5
Dimethyloxallyl glycine/nanosilicates-loaded osteogenic/angiogenic difunctional fibrous structure for functional periodontal tissue regeneration显示文摘The coupled process of osteogenesis-angiogenesis plays a crucial role in periodontal tissue regeneration.Although various cytokines or chemokines have been widely applied in periodontal in situ tissue engineering, most of them are macromolecular proteins with the drawbacks of short effective half-life,poor stability and high cost, which constrain their clinical translation. Our study aimed to develop a difunctional structure for periodontal tissue regeneration by incorporating an angiogenic small molecule, dimethyloxalylglycine(DMOG),and an osteoinductive inorganic nanomaterial, nanosilicate(nSi)into poly(lactic-co-glycolic acid)(PLGA)fibers by electrospinning. The physiochemical properties of DMOG/nSi-PLGA fibrous membranes were characterized.Thereafter, the effect of DMOG/nSi-PLGA membranes on periodontal tissue regeneration was evaluated by detecting osteogenic and angiogenic differentiation potential of periodontal ligament stem cells(PDLSCs)in vitro.Additionally, the fibrous membranes were transplanted into rat periodontal defects, and tissue regeneration was assessed with histological evaluation,micro-computed tomography(micro-CT),and immunohistochemical analysis.DMOG/nSi-PLGA membranes possessed preferable mechanical property and biocompatibility.PDLSCs seeded on the DMOG/nSi-PLGA membranes showed up-regulated expression of osteogenic and angiogenic markers, higher alkaline phosphatase(ALP)activity, and more tube formation in comparison with single application.Further,in vivo study showed that the DMOG/nSi-PLGA membranes promoted recruitment of CD90+/CD34stromal cells, induced angiogenesis and osteogenesis, and regenerated cementum-ligament-bone complex in periodontal defects.Consequently, the combination of DMOG and nSi exerted admirable effects on periodontal tissue regeneration.DMOG/nSi-PLGA fibrous membranes could enhance and orchestrate osteogenesis-angiogenesis,and may have the potential to be translated as an effective scaffold in periodontal tissue engineering.Lingling Shang Ziqi Liu Baojin Ma Jinlong Shao Bing Wang Chenxi Ma Shaohua Ge 2021Bioactive Materials2021,6,4:5
Microstructure evolution and mechanical properties of linear friction welded S31042 heat-resistant steel显示文摘S31042 heat-resistant steel was joined by linear friction welding(LFW) in this study. The microstructure and the mechanical properties of the LFWed joint were investigated by optical microscopy, scanning electronic microscopy, transmission electron microscopy, hardness test and tensile test. A defect-free joint was achieved by using LFW under reasonable welding parameters. The dynamic recrystallization of austenitic grains and the dispersed precipitation of NbCrN particles resulting from the high stress and high temperature in welding, would lead to a improvement of mechanical property of the welded joint. With increasing the distance from the weld zone to the parent metal, the austenitic grain size gradually increases from ~1μm to ~150μm, and the microhardness decreases from 301 HV to 225 HV. The tensile strength(about 731 MPa) of the welded joint is comparable to that of the S31042 in the solution-treated state.Yanmo Li Yongchang Liu Chenxi Liu Chong Li Zongqing Ma Yuan Huang Zumin Wang Wenya Li 2018Journal of Materials Science & Technology2018,34,4:5
Near-Real-Time Carbon Emission Accounting Technology Toward Carbon Neutrality显示文摘Climate change is the greatest environmental threat to humans and the planet in the 21st century.Global anthropogenic greenhouse gas emissions are one of the main causes of the increasing number of extreme climate events.Cumulative carbon dioxide(CO_(2))emissions showed a linear relationship with cumulative temperature rise since the pre-industrial stage, and this accounts for approximately 80%of the total anthropogenic greenhouse gases. Therefore, accurate and reliable carbon emission data are the foundation and scientific basis for most emission reduction policymaking and target setting.Currently,China has made clear the ambitious goal of achieving the peak of carbon emissions by 2030 and achieving carbon neutrality by 2060.The development of a finer-grained spatiotemporal carbon emission database is urgently needed to achieve more accurate carbon emission monitoring for continuous implementation and the iterative improvement of emission reduction policies.Near-real-time carbon emission monitoring is not only a major national demand but also a scientific question at the frontier of this discipline. This article reviews existing annual-based carbon accounting methods, with a focus on the newly developed real-time carbon emission technology and its current application trends. We also present a framework for the latest near-real-time carbon emission accounting technology that can be widely used. The development of relevant data and methods will provide strong database support to the policymaking for China’s“carbon neutrality”strategy. Finally, this article provides an outlook on the future of real-time carbon emission monitoring technology.Zhu Liu Taochun Sun Ying Yu Piyu Ke Zhu Deng Chenxi Lu Da Huo Xiang Ding 2022Engineering2022,8,7:5
Improvement in affinity and thermostability of a fully human antibody against interleukin-17A by yeast-display technology and CDR grafting显示文摘Monoclonal antibodies(m Abs) are widely used in many fields due to their high specificity and ability to recognize a broad range of antigens. IL-17 A can induce a rapid inflammatory response both alone and synergistically with other proinflammatory cytokines. Accumulating evidence suggests that therapeutic intervention of IL-17 A signaling offers an attractive treatment option for autoimmune diseases and cancer. Here, we present a combinatorial approach for optimizing the affinity and thermostability of a novel anti-h IL-17 A antibody. From a large na?ve phage-displayed library, we isolated the anti-IL-17 A m Ab 7 H9 that can neutralize the effects of recombinant human IL-17 A. However, the modest neutralization potency and poor thermostability limit its therapeutic applications. In vitro affinity optimization was then used to generate 8 D3 by using yeast-displayed random mutagenesis libraries. This resulted in four key amino acid changes and provided an approximately 15-fold potency increase in a cell-based neutralization assay. Complementarity-determining regions(CDRs) of 8 D3 were further grafted onto the stable framework of the hu Fv 4 D5 to improve thermostability. The resulting hybrid antibody9 NT/S has superior stabilization and affinities beyond its original antibody. Human fibrosarcoma cellbased assays and in vivo analyses in mice indicated that the anti-IL-17 A antibody 9 NT/S efficiently inhibited the secretion of IL-17 A-induced proinflammatory cytokines. Therefore, this lead anti-IL-17 A m Ab might be used as a potential best-in-class candidate for treating IL-17 A related diseases.Wei Sun Zhaona Yang Heng Lin Ming Liu Chenxi Zhao Xueying Hou Zhuowei Hu Bing Cuin 2019Acta Pharmaceutica Sinica B2019,9,5:4
