1ZBP1(DAI/DLM-1) promotes osteogenic differentiation while inhibiting adipogenic differentiation in mesenchymal stem cells through a positive feedback loop of Wnt/β-catenin signaling显示文摘The lineage specification of mesenchymal stem/stromal cells(MSCs) is tightly regulated by a wide range of factors. Recently, the versatile functions of ZBP1(also known as DAI or DLM-1) have been reported in the blood circulation and immune systems.However, the biological function of ZBP1 during the lineage specification of MSCs is still unknown. In the present study, we found that ZBP1 was upregulated during osteogenesis but downregulated during adipogenesis in mouse bone marrow-derived MSCs(m BMSCs). ZBP1 was highly expressed in osteoblasts but expressed at a relatively low level in marrow adipocytes. Knockdown of ZBP1 inhibited alkaline phosphataseactivity, extracellular matrix mineralization, and osteogenesis-related gene expression in vitro and reduced ectopic bone formation in vivo. Knockdown of ZBP1 also promoted adipogenesis in MSCs in vitro. Conversely, the overexpression of ZBP1 increased the osteogenesis but suppressed the adipogenesis of MSCs. When the expression of ZBP1 was rescued, the osteogenic capacity of ZBP1-depleted m BMSCs was restored at both the molecular and phenotypic levels.Furthermore, we demonstrated that ZBP1, a newly identified target of Wnt/β-catenin signaling, was required for β-catenin translocation into nuclei. 2A Simple Approach to Bioconjugation at Diverse Levels: Metal-Free Click Reactions of Activated Alkynes with Native Groups of Biotargets without Prefunctionalization显示文摘Te efcient bioconjugation of functional groups/molecules to targeted matrix and bio-related species drives the great development of material science and biomedicine,while the dilemma of metal catalysis,uneasy premodifcation,and limited reaction efciency in traditional bioconjugation has restricted the booming development to some extent.Here,we provide a strategy for metal-free click bioconjugation at diverse levels based on activated alkynes.As a proof-of-concept,the abundant native groups including amine,thiol,and hydroxyl groups can directly react with activated alkynes without any modifcation in the absence of metal catalysis.Trough this strategy,high-efcient modifcation and potential functionalization can be achieved for natural polysaccharide,biocompatible polyethylene glycol(PEG),synthetic polymers,cell penetrating peptide,protein,fast whole-cell mapping,and even quick diferentiation and staining of Gram-positive bacteria,etc.Terefore,current metal-free click bioconjugation strategy based on activated alkynes is promising for the development of quick fuorescence labeling and functional modifcation of many targets and can be widely applied towards the fabrication of complex biomaterials and future in vivo labeling and detection.Xianglong Hu Xueqian Zhao Benzhao He Zheng Zhao Zheng Zheng Pengfei Zhang Xiujuan Shi Ryan T.K.Kwok Jacky W.Y.Lam Anjun Qin Ben Zhong Tang 2018Research2018,,1:7

4Impairment of cognition and sleep after acute ischaemic stroke or transient ischaemic attack in Chinese patients: design, rationale and baseline patient characteristics of a nationwide multicentre prospective registry显示文摘background and aim Cognitive impairment and sleep disorder are both common poststroke conditions and are closely related to the prognosis of patients who had a stroke.The Impairment of CognitiON and Sleep after acute ischemic stroke or transient ischemic attack in Chinese patients(ICONS)study is a nationwide multicentre prospective registry to investigate the occurrence and associated factors of cognitive impairment and sleep disorder after acute ischaemic stroke(AIS)or transient ischaemic attack(TIA).Methods Consecutive AIS or TIA in-hospital patients within 7 days after onset were enrolled from 40 participating sites in China.Comprehensive baseline clinical and imaging data were collected prospectively.Blood and urine samples were also collected on admission and follow-up visits.Patients were interviewed face to face for cognition and sleep related outcomes at 2 weeks,3,6 and 12 months after AIS/TIA and followed up for clinical outcomes by telephone annually over 5 years.results Between August 2015 and January 2018,a total of 2625 patients were enrolled.92.65% patients had AIS and 7.35% patients had TIA.Overall,the average age was 61.04 years,and 72.38% patients were male.Median National Institutes of Health Stroke Scale score was 3 in AIS patients.Conclusions The ICONS study is a large-scale nationwide prospective registry to investigate occurrence and the longitudinal changes of cognitive impairment and sleep disorder after AIS or TIA.Data from this registry may also provide opportunity to evaluate associated factors of cognitive impairment or sleep disorder after AIS or TIA and their impact on clinical outcome.Yongjun Wang Xiaoling Liao Chunxue Wang Ning Zhang Lijun Zuo Yang Yang Yuesong Pan Xianglong Xiang Jing Jing Xia Meng Xingquan Zhao Yilong Wang Jiong Shi Hao Li 2021Stroke & Vascular Neurology2021,6,1:4
5The spatial multiscale variability of heavy metals based on factorial kriging analysis: A case study in the northeastern Beibu Gulf显示文摘Factorial kriging analysis is applied to the research on the spatial multiscale variability of heavy metals in submarine. It is used to analyze the multiscale spatial structures of seven heavy metals, Ni, Cu, Zn, Pb, Cr, As and Cd in the surface sediment from the northeastern of Beibu Gulf, identify and separate spatial variations at different scales of heavy metals, and discuss the provenance of heavy metals and the influencing factors. The results show that the existence of three-scale spatial variations those consist of nugget effect, a spherical structure with range of 30 km(short-range scale) and a spherical structure with range of 140 km(long-range scale) in the linear model of coregionalization fitted. The spatial distribution features of seven heavy metals at short-range scale reflect 'spot-like' or 'stripe-like' local-scale spatial variations; the spatial distribution features of the seven heavy metals at long-range scale represent 'slice-like' regional-scale spatial variations. At local scale, Zn, Cr, Ni,Cu, Pb and Cd are derived primarily from parent materials of Hainan Island, Leizhou Peninsula and Guangxi land, whose spatial distribution characteristics are controlled by granularity of sediments, while As is influenced dominantly by human pollution components from Hainan Island and Leizhou Peninsula. At regional scale, Zn,Cr, Ni and Cu originate primarily from parent rock materials of Leizhou Peninsula and Hainan Island, secondly from Guangxi land; As originated primarily from parent rock materials from Hainan Island, secondly from Leizhou Peninsula and Guangxi land. These metals are transported and migrated with sediments dominated by the anticlockwise circulation of Beibu Gulf year-round, deposited in 'convergence center', forming the whole sedimentary pattern in direction of NWW-NNW at regional scale. 6Do migrant and resident species differ in the timing of increases in reproductive and thyroid hormone secretion and body mass? A case study in the comparison of pre-breeding physiological rhythms in the Eurasian Skylark and Asian Short-toed Lark显示文摘Background:Physiological preparation for reproduction in small passerines involves the increased secretion of reproductive hormones, elevation of the metabolic rate and energy storage, all of which are essential for reproduction. However, it is unclear whether the timing of the physiological processes involved is the same in resident and migrant species that breed in the same area. To answer this question, we compared temporal variation in the plasma concentration of luteinizing hormone(LH), testosterone(T), estradiol(E_2), triiothyronine(T_3) and body mass, between a migrant species, the Eurasian Skylark(Alauda arvensis) and a resident species, the Asian Short-toed Lark(Calandrella cheleensis), both of which breed in northeastern Inner Mongolia, China, during the 2014 and 2015 breeding seasons.Methods:Twenty adult Eurasian Skylarks and twenty Asian Short-toed Larks were captured on March 15, 2014 and 2015 and housed in out-door aviaries. Plasma LH, T(males), E_2(females), T_3 and the body mass of each bird were measured every six days from March 25 to May 6.Results:With the exception of T, which peaked earlier in the Asian Short-toed Lark in 2014, plasma concentrations of LH, T, E_2 and T_3 of both species peaked at almost the same time. However, Asian Short-toed Larks attained peak body mass earlier than Eurasian Skylarks. Plasma T_3 concentrations peaked 12 days earlier than plasma LH in both species. Generally, plasma LH, T, E_2, T_3 and body mass, peaked earlier in both species in 2014 than 2015.Conclusions:The timing of pre-reproductive changes in the endocrine system and energy metabolism can be the same in migrant and resident species; however, residents may accumulate energy reserves faster than migrants. Although migration does not affect the timing of pre-breeding reproductive and metabolic changes, migrant species may need more time to increase their body mass. 8Microstructure evolution and superelasticity behavior of Ti-Ni-Hf shape memory alloy composite with multi-scale and heterogeneous reinforcements显示文摘In the present study,the in-situ TiB whisker was introduced into the Ti-Ni-Hf shape memory alloy composite by the in-situ reaction of the Ti-Ni-Hf alloy powder and TiB2 powders.The(Ti,Hf)2 Ni phase also precipitated,accompanied with the formation of TiB phase.Moreover,the residual TiB2 particles can be observed,as the TiB2 content was higher than 0.7 wt%.Thereinto,the larger scale reinforcements constituted the quasi-continuous network structure.The smaller scale reinforcements distributed in the interior of the network structure.The two-scale reinforcements showed the uniform distribution at macroscopic level and inhomogeneous distribution at microscopic level.The single stage B19?B2 martensitic transformation occurred in the Ti-Ni-Hf composites.In addition,the martensitic transformation temperatures continuously decreased with the increased TiB2 content owing to the compositional and mechanical effect.The moderate TiB2 addition not noly enhanced the matrix strength,but also significantly improved the superelasticity.The excellent superelaticity with the completely recoverable strain of 4%can be obtained in the Ti-Ni-Hf composite containing 0.7 wt%TiB2.Xiaoyang Yi Bin Sun Weihong Gao Xianglong Meng Zhiyong Gao Wei Cai Liancheng Zhao 2020Journal of Materials Science & Technology2020,42,7:2

15Genome sequence of Gossypium anomalum facilitates interspecific introgression breeding显示文摘Crop wild relatives are an important reservoir of natural biodiversity. However, incorporating wild geneticdiversity into breeding programs is often hampered by reproductive barriers and a lack of accurate genomicinformation. We assembled a high-quality, accurately centromere-anchored genome of Gossypium anomalum, a stress-tolerant wild cotton species. We provided a strategy to discover and transfer agronomicallyvaluable genes from wild diploid species to tetraploid cotton cultivars. With a (Gossypium hirsutum 3 G.anomalum)2 hexaploid as a bridge parent, we developed a set of 74 diploid chromosome segment substitution lines (CSSLs) of the wild cotton species G. anomalum in the G. hirsutum background. This set of CSSLsincluded 70 homozygous substitutions and four heterozygous substitutions, and it collectively containedabout 72.22% of the G. anomalum genome. Twenty-four quantitative trait loci associated with plant height,yield, and fiber qualities were detected on 15 substitution segments. Integrating the reference genome withagronomic trait evaluation of the CSSLs enabled location and cloning of two G. anomalum genes thatencode peroxiredoxin and putative callose synthase 8, respectively, conferring drought tolerance andimproving fiber strength. 18An on-line free handwritten Chinese character recognition method based on component cascaded HMMs显示文摘This paper presents a cascaded Hidden Markov Model (HMM), which allows state's transition, skip and duration. The cascaded HMM extends the way of HMM pattern description of Handwritten Chinese Character (HCC) and depicts the behavior of handwritten curve more reliably in terms of the statistic probability. Hence character segmentation and labeling are unnecessary. Viterbi algorithm is integrated in the cascaded HMM after the whole sample sequence of a HCC is input. More than 26,000 component samples are used for training 407 handwritten component HMMs. At the improved training stage 94 models of 94 Chinese characters are gained by 32,000 samples. 19Feature extraction for classification of different weather conditions显示文摘Classification of different weather conditions provides a first step support for outdoor scene modeling,which is a core component in many different applications of outdoor video analysis and computer vision.Features derived from intrinsic properties of the visual effects of different weather conditions contribute to successful classification.In this paper,features representing both the autocorrelation of pixel-wise intensities over time and the max directional length of rain streaks or snowflakes are proposed.Based on the autocorrelation of each pixel’s intensities over time,two temporal features are used for coarse classification of weather conditions according to their visual effects.On the other hand,features are extracted for fine classification of video clips with rain and snow.The classification results on 249 video clips associated with different weather conditions indicate the effectiveness of the extracted features,by using C-SVM as the classifier.Xudong ZHAO Peng LIU Jiafeng LIU Xianglong TANG 2011Frontiers of Electrical and Electronic Engineering in China2011,6,2:1
