| 2 | Induced Th2 dominant immune response in APPswe, PSEN1dE9 transgenic mice after nasal immunization with an adenoviral vector encoding 10 tandem repeats of beta-amyloid 3-10显示文摘Immunotherapy for Alzheimer's disease (AD) is effective in improving cognitive function in transgenic mouse models of AD. Because the AN1792 [beta-amyloid (Aβ) 1-42] vaccine was halted because of T cell mediated meningoencephalitis, many scientists are searching for a novel vaccine to avoid the T cell mediated immune response caused by the Aβ1-42. Importantly, the time when the immunization is begun can influence the immune effect. In this study, an adenovirus vaccine was constructed containing 10 × Aβ3-10 repeats and gene adjuvant CpG DNA. Transgenic AD mice were immunized intranasally for 3 months. After 10 × Aβ3-10 vaccine immunization, high titers of anti-Aβ42 IgG1 predominant antibodies were induced. In spatial learning ability and probe tests, the 10 × Aβ3-10 immunized mice showed significantly improved memories compared to control mice. The 10 × Aβ3-10 vaccine resulted in a robust Th2 dominant humoral immune response and reduced learning deficits in AD mice. In addition, the 10 × Aβ3-10 vaccine might be more efficient if administered before Aβ aggregation at an early stage in the AD mouse brain. | 3 | Comparison of lateral episiotomy,traditional midwifery without episiotomy,and hands-off techniques delivery on pelvic floor function显示文摘Background Prolonged vaginal delivery times and episiotomy are associated with damage to the muscles of the pelvic floor,and with complications that include postpartum sexual dysfunction and urinary stress incontinence.This study was to compare the pelvic floor function among postpartum women who underwent three midwifery practices during labor:lateral episiotomy,traditional midwifery without episiotomy,and hands-off techniques delivery.Methods We included 320 primipara who were full-term pregnancy and underwent vaginal delivery in Department of Obstetrics,Peking University People’s Hospital,China.117 women had episiotomy during labor and were classified into the lateral episiotomy group;103 had traditional midwifery without episiotomy and were classified into the traditional midwifery practice group;and 100 experienced hands-off techniques during labor and were classified into the hands-off techniques delivery group.The duration of the second stage of labor,and postpartum pelvic floor muscle strength were compared between the three study groups.Results There were no significant differences between the three groups for the duration of the second stage of labor and postpartum pelvic floor muscle strength(P>0.05).However,the incidence of postpartum urinary stress incontinence and sexual dysfunction in the hands-off techniques delivery group was significantly lower than in the lateral episiotomy groups and the traditional midwifery practice group(P<0.01).Conclusions Hands-off techniques delivery could reduce damage to the pelvic floor and improve the quality of life for women after delivery. | Jingjing Gong Lili Xing Xiaodan Li Xiaona Wang Huijuan Chen | 2021 | Gynecology and Obstetrics Clinical Medicine2021,1,1: | 2 |
| 4 | Roadmap of environmental health research on emerging contaminants: Inspiration from the studies on engineered nanomaterials显示文摘Research on the environmental health of emerging contaminants is critical to understand their risks before causing severe harm.However,the low environmental concentrations,complex behaviors,and toxicology of emerging contaminants present enormous challenges for researchers.Here,we reviewed the research on the environmental health of engineered nanomaterials(ENMs),one of the typical emerging contaminants,to enlighten pathways for future research on emerging contaminants at their initial exploratory stage.To date,some developed pretreatment methods and detection technologies have been established for the determination of ENMs in natural environments.The mechanisms underlying the transfer and transformation of ENMs have been systematically explored in laboratory studies.The mechanisms of ENMs-induced toxicity have also been preliminarily clarified at genetic,cellular,individual,and short food chain levels,providing not only a theoretical basis for revealing the risk change and environmental health effects of ENMs in natural environments but also a methodological guidance for studying environmental health of other emerging contaminants.Nonetheless,due to the interaction of multiple environmental factors and the high diversity of organisms in natural environments,health effects observed in laboratory studies likely differ from those in natural environments.We propose a holistic approach and mesocosmic model ecosystems to systematically carry out environmental health research on emerging contaminants,obtaining data that determine the objectivity and accuracy of risk assessment. | Xiaona Li Feng He Zhenyu Wang Baoshan Xing | 2022 | Eco-Environment & Health2022,1,3: | 1 |

| 7 | Exploration of the potential mechanism of Danggui Shaoyao powder in the treatment of endometriosis based on bioinformatics显示文摘Background:Endometriosis is a hormone-and organ-dependent disease with unclear pathogenesis.Danggui Shaoyao powder(DSP)has been used for the treatment of endometriosis for many years and has been proven to be effective,but its underlying molecular mechanisms remain unclear.In this study,we explored the molecular targets and pathways of DSP in the treatment of endometriosis mainly from the perspective of network pharmacology,and provided some novel ideas for managing this disease.Methods:In the present study,we focused on the underlying mechanisms between DPS and endometriosis,via oral bioavailability screening,drug similarity assessment,target identification,network analysis,drug relocation,and molecular docking using network pharmacology.Results:By bioinformatics,54 active components of DSP were predicted from the 534 components provided by the TCMSP database.We then found 85 validated targets and 6059 predicted targets for herbal components,and 2241 targets for endometriosis.After mapping,there were 255 targets in common.After that,network analysis was performed to screen key networks and core targets.Ninetynine indirect targets were collected from 14 direct ones.GO and KEGG analyses identified 63 enriched functions and 10 pathways(P<.01).According to the disease target,dipyridamole could undergo drug repositioning for endometriosis(score?1).The mechanism of action of dipyridamole on endometriosis was compared with that of DSP.Finally,eight targets,namely,AR,ESR1,HMGCR,NOS2,NR3C1,PPARG,PTGES,and PTGS2,were validated by molecular docking,suggesting that the probable molecular mechanisms by which DSP treats endometriosis are through seven validated pathways:hsa05200,hsa04915,hsa01100,hsa00900,hsa04920,hsa01130,and hsa00590.Conclusion:In this study,we uncovered the mechanism by which DSP can treat endometriosis using a drugecomponentetarget interaction network.This approach can provide some novel ideas for the development of Chinese medicine and the clinical management of endometriosis. | Xiuping Zhang Lulu Zhang Qi Wang Xinhui Sun Yang Dong Yu Xing Xiaona Ma | 2019 | Journal of Traditional Chinese Medical Sciences2019,6,4: | 1 |
| 8 | A new DNA vaccine fused with the C3d-p28 induces a Th2 immune response against amyloid-beta显示文摘To enhance anti-amyloid-beta(Aβ) antibody generation and induce a Th2 immune response,we constructed a new DNA vaccine p(Aβ3-10)10-C3d-p28.3 encoding ten repeats of Aβ3-10 and three copies of C3d-p28 as a molecular adjuvant.In this study,we administered this adjuvant intramuscularly to female C57BL/6J mice at 8-10 weeks of age.Enzyme linked immunosorbent assay was used to detect the titer of serum anti-Aβ antibody,isotypes,and cytokines in splenic T cells.A 3(4,5-cimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay was used to detect the proliferation rate of splenic T cells.Brain sections from a 12-month-old APP/PS1 transgenic mouse were used for detecting the binding capacities of anti-Aβ antibodies to Aβ plaques.The p(Aβ3-10)10-C3d-p28.3 vaccine induced high titers of anti-amyloid-β antibodies,which bound to Aβ plaques in APP/PS1 transgenic mouse brain tissue,demonstrating that the vaccine is effective against plaques in a mouse model of Alzheimer's disease.Moreover,the vaccine elicited a predominantly IgG1 humoral response and low levels of interferon-γ in ex vivo cultured splenocytes,indicating that the vaccine could shift the cellular immune response towards a Th2 phenotype.This indicated that the vaccine did not elicit a detrimental immune response and had a favorable safety profile.Our results indicate that the p(Aβ3-10)10-C3d-p28.3 vaccine is a promising immunotherapeutic option for Aβ vaccination in Alzheimer's disease. | Wanshu Guo Sha Sha Tongzi Jiang Xiaona Xing Yunpeng Cao | 2013 | Neural Regeneration Research2013,8,27: | 0 |

ELISA analysis of serum showed Aβ42 antibody titers were significantly increased in mice immunized with Aβ1–42 and AdCpG-(Aβ3–10)10. Concanavalin A and AdCpG-(Aβ3–10)10 stimulation significantly increased the number of proliferating spleen cells cultured from AdCpG(Aβ3–10)10 and Aβ42 groups compared with the control group. In the AdCpG(Aβ3–10)10 group, levels of interleukin(IL)-4 and IL-10 were increased, while those of IL-2 and interferon-γ were decreased. In the Aβ42 group, levels of IL-4, IL-10, IL-2 and interferon-γ were all increased. Experimental findings indicate that AdCpG-(Aβ3–10)10 vaccine can produce strong T helper 2(Th2) humoral immune responses in addition to the production of Aβ42 antibody.
