| 2 | AFF3-DNA methylation interplay in maintaining the mono-allelic expression pattern of XIST in terminally differentiated cells显示文摘X chromosome inactivation and genomic imprinting are two classic epigenetic regulatory processes that cause mono-allelic gene expression.In female mammals,mono-allelic expression of the long non-coding RNA gene X-inactive specific transcript(XIST)is essential for initiation of X chromosome inactivation upon differentiation.We have previously demonstrated that the central factor of super elongation complex-like 3(SEC-L3),AFF3,is enriched at gamete differentially methylated regions(DMRs)of the imprinted loci and regulates the imprinted gene expression.Here,we found that AFF3 can also bind to the DMR downstream of the XIST promoter.Knockdown of AFF3 leads to de-repression of the inactive allele of X IST in terminally differentiated cells.In addition,the binding of AFF3 to the XIST DMR relies on DNA methylation and also regulates DNA methylation level at DMR region.However,the KAP1-H3K9 methylation machineries,which regulate the imprinted loci,might not play major roles in maintaining the mono-aUelic expression pattern of XIST in these cells.Thus,our results suggest that the differential mechanisms involved in the X IST DMR and gDMR regulation,which both require AFF3 and DNA methylation. | Yue Zhang Chao Wang Xiaoxu Liu Qian Yang Hongliang Ji Mengjun Yang Manman Xu Yunyan Zhou Wei Xie Zhuojuan Luo Chengqi Lin | 2019 | Journal of Molecular Cell Biology2019,11,9: | 3 |
| 3 | Can Culture Lead to Export?Empirical Data on the Belt and Road Initiative显示文摘The B&R Initiative is a modernized and specific manifestation of China's'going global'development strategy. It undertakes the historic mission of promoting communication and exchange between cultures and nations, but also has a positive role in the development of international economy and trade relations. As a messenger of the essence of Chinese culture, Confucius Institutes have a positive impact on the development of Asian and world civilizations. Using Difference-in-Difference estimation methods to study the statistical data from the countries along the B&R routes, this paper examines the relationship between cultural and commodity outputs and comes to the following conclusions: Cultural output is an important factor in increasing commodity exports; Confucius Institutes help increase China's exports to the countries along the B&R routes; and the growth in cultural exports shows regional differences, hysteresis, and fluctuations. As China's cultural outputs are still at an immature stage, their promotional effects on exports are unstable. | 4 | Fluorobenzene diluted low-density electrolyte for high-energy density and high-performance lithium-sulfur batteries显示文摘The mass fraction of electrolytes is the crucial factor affecting the energy density of lithium-sulfur(Li-S)batteries. Due to the high porosity within the C/S cathode, high concentration of polysulfides, and side reaction in lithiun metal anode under lean electrolyte, it is extremely challenging to improve performance while reducing the electrolyte volume. Here, we report a novel electrolyte with relatively low density(1.16 g cm^(-2)), low viscosity(1.84 m Pa s), and high ionic conductivity, which significantly promotes energy density and cyclability of Li-S batteries under practical conditions. Moreover, such electrolyte enables a hybrid cathode electrolyte interphase(CEI) and solid electrolyte interface(SEI) layer with plentiful Li F, which leads to fast kinetics of ions transport and stable cyclability even under low temperatures.Compared to Li-S batteries in electrolyte employing 1,1,2,2-tetrafluoroethyl 2,2,3,3-tetrafluoropropyl ether(TTE) diluent, the ultra-thick cathode(20 mg cm^(-2)) shows a high capacity of 9.48 m Ah cm^(-2)and excellent capacity retention of 80.3% over 191 cycles at a low electrolyte-to-sulfur ratio(E/S = 2) and negative-to-positive capacity ratio(N/P = 2.5), realizing a 19.2% improvement in energy density in coin cells(from 370 to 441 Wh kg^(-1)) and a high energy density up to 467 Wh kg^(-1) in pouch cells. | 6 | Proscillaridin A inhibits hepatocellular carcinoma progression through inducing mitochondrial damage and autophagy显示文摘Hepatocellular carcinoma(HCC)is one of the leading causes of cancer-related deaths globally.At present,drug options for systemic treatment of HCC are very limited.There is an urgent need to develop additional effective drugs for HCC treatment.In the present study,we found that proscillaridin A(ProA),a cardiac glycoside,exerted a strong anticancer effect on multiple HCC cell lines.ProA significantly inhibited the cell proliferation,migration,and invasion of HCC cells.ProA also had a marked inhibitory effect on the progression of HCC in the MHCC97H xenograft nude mouse model.ProA-mediated suppression of HCC was closely related to cell apoptosis.ProA-treated HCC cells displayed significant mitochondrial damage and elevated reactive oxygen species production,resulting in profound cell apoptosis.Meanwhile,ProA also played a role in autophagy induction in HCC cells.Defects in autophagy partially relieved ProA’s anticancer effect in HCC cells.Our findings demonstrate that ProA can effectively inhibit HCC progression and may serve as a potential therapeutic agent for HCC treatment. | Mengjun Luo Yanfeng Liu Nannan Liu Weiqing Shao Lijun Ming Jing Liu Youhua Xie | 2021 | Acta Biochimica et Biophysica Sinica2021,53,1: | 1 |
| 7 | Establishment of an in vitro reporter system for screening HBx-targeting molecules显示文摘Chronic hepatitis B virus (HBV) infection remains a global public health problem. HBV-encoded X protein (HBx) is a multifunctional regulator that is required to initiate and maintain productive HBV infection, and is involved in HBV-related hepatocellular carci noma (HCC). Inhibitors that in terfere with the functions of HBx could be useful not only for the inhibition of HBV replication but also for the prevention or treatment of HBV-related HCC. To screen molecules that target HBx on a large scale remains a tech nical challenge due to a lack of sensitive and high-throughput system .In this work, we established an in vitro bioluminescent reporter system for screening HBx-targeting molecules. The system is based on a secretory fusion protein that combines HBx and NanoLuc (HBx-Nluc). The measured activity of NanoLuc in the culture supernatant of HBx-Nluc-expressing cells directly reflects the level of secreted HBx-Nluc. HBx protein-targeting intracellular anti-HBx single-chain variable fragment and RNA-targeting shRNA significantly reduced the secreted NanoLuc activity in HBx-Nluc-expressing cells. | 8 | Tuning Solid Interfaces via Varying Electrolyte Distributions Enables High-Performance Solid-State Batteries显示文摘Solid/solid interface is the major challenge for high-performance solid-state batteries.Solid electrolytes(SEs)play a crucial role in the fabrication of effective interfaces in solid-state batteries.Herein,the electrolyte distribution with varied particle sizes is tuned to construct solid-state batteries with excellent performance at different operating temperatures.Solid-state batteries with the configuration S/L(small-sized SE in composite cathode and large-sized SE in electrolyte layer)show the best performance at room temperature(168 mA h g^(−1) at 0.2 C,retention of 99%,100 cycles)and−20°C(89 mA h g^(−1) at 0.05 C),while the configuration S/S displays better performance at elevated temperature.The superior performance of S/L battery is associated with faster lithium-ion dynamics due to the better solid/solid interface between active materials and electrolytes.Moreover,the inferior performance at 60℃is caused by the formation of voids and cracks in the electrolyte layer during cycling.In contrast,the S/S battery delivers superior performance at elevated operating temperature because of the integrated structure.This work confirms that tailoring electrolyte size has significant effect on fabricating all-climate solid-state batteries. | Linfeng Peng Chuang Yu Ziqi Zhang Ruonan Xu Mengjun Sun Long Zhang Shijie Cheng Jia Xie | 2023 | Energy & Environmental Materials2023,6,2: | 0 |
