2Targeting BMI-1-mediated epitheliale-mesenchymal transition to inhibit colorectal cancer liver metastasis显示文摘Liver is the most common metastatic site for colorectal cancer(CRC),there is no satisfied approach to treat CRC liver metastasis(CRCLM).Here,we investigated the role of a polycomb protein BMI-1 in CRCLM.Immunohistochemical analysis showed that BMI-1 expression in liver metastases was upregulated and associated with T4 stage,invasion depth and right-sided primary tumor.Knockdown BMI-1 in high metastatic HCT116 and LOVO cells repressed the migratory/invasive phenotype and reversed epithelialemesenchymal transition(EMT),while BMI-1 overexpression in low metastatic Ls174 T and DLD1 cells enhanced invasiveness and EMT.The effects of BMI-1 in CRC cells were related to upregulating snail via AKT/GSK-3βpathway.Furthermore,knockdown BMI-1 in HCT116 and LOVO cells reduced CRCLM using experimental liver metastasis mice model.Meanwhile,BMI-1 overexpression in Ls174 T and DLD1 significantly increased CRCLM.Moreover,sodium butyrate,a histone deacetylase and BMI-1 inhibitor,reduced HCT116 and LOVO liver metastasis in immunodeficient mice.Our results suggest that BMI-1 is a major regulator of CRCLM and provide a potent molecular target for CRCLM treatment.Zhiyao Xu Zhuha Zhou Jing Zhang Feichao Xuan Mengjing Fan Difan Zhou Zhenyu Liuyang Ximei Ma Yiyang Hong Yihong Wang Sherven Sharma Qinghua Dong Guanyu Wang 2021Acta Pharmaceutica Sinica B2021,11,5:8
3High salt diet accelerates the progression of murine lupus through dendritic cells via the p38 MAPK and STAT1 signaling pathways显示文摘The increased incidence of systemic lupus erythematosus(SLE)in recent decades might be related to changes in modern dietary habits.Since sodium chloride(NaCl)promotes pathogenic T cell responses,we hypothesize that excessive salt intake contributes to the increased incidence of autoimmune diseases,including SLE.Given the importance of dendritic cells(DCs)in the pathogenesis of SLE,we explored the influence of an excessive sodium chloride diet on DCs in a murine SLE model.We used an induced lupus model in which bone marrow-derived dendritic cells(BMDCs)were incubated with activated lymphocyte-derived DNA(ALD-DNA)and transferred into C57BL/6 recipient mice.We observed that a high-salt diet(HSD)markedly exacerbated lupus progression,which was accompanied by increased DC activation.NaCl treatment also stimulated the maturation,activation and antigenpresenting ability of DCs in vitro.Pretreatment of BMDCs with NaCl also exacerbated BMDC-ALD-DNA-induced lupus.These mice had increased production of autoantibodies and proinflammatory cytokines,more pronounced splenomegaly and lymphadenopathy,and enhanced pathological renal lesions.The p38 MAPK–STAT1 pathway played an important role in NaClinduced DC immune activities.Taken together,our results demonstrate that HSD intake promotes immune activation of DCs through the p38 MAPK–STAT1 signaling pathway and exacerbates the features of SLE.Thus,changes in diet may provide a novel strategy for the prevention or amelioration of lupus or other autoimmune diseases.Ze Xiu Xiao Xiaojiang Hu Ximei Zhang Zhigang Chen Julie Wang Ke Jin Feng Lin Cao Baoqing Sun Joseph A.Bellanti Nancy Olsen Song Guo Zheng 2020Signal Transduction and Targeted Therapy2020,5,1:6
4Profiling of circular RNA N^(6)-methyladenosine in moso bamboo(Phyllostachys edulis)using nanopore-based direct RNA sequencing显示文摘N6-methyladenosine(m^(6)A)is a prevalent modification in messenger RNAs and circular RNAs that play important roles in regulating various aspects of RNA metabolism.However,the occurrence of the m^(6)A modification in plant circular RNAs has not been reported.A widely used method to identify m^(6)A modifications relies on m^(6)A-specific antibodies followed by next-generation sequencing of precipitated RNAs(MeRIP-Seq).However,one limitation of MeRIP-Seq is that it does not provide the precise location of m^(6)A at single-nucleotide resolution.Although more recent sequencing techniques such as Nanopore-based direct RNA sequencing(DRS)can overcome such limitations,the technology does not allow sequencing of circular RNAs,as these molecules lack a poly(A)tail.Here,we developed a novel method to detect the precise location of m^(6)A modifications in circular RNAs using Nanopore DRS.We first enriched our samples for circular RNAs,which we then fragmented and sequenced on the Nanopore platform with a customized protocol.Using this method,we identified 470 unique circular RNAs from DRS reads based on the back-spliced junction region.Among exonic circular RNAs,about 10%contained m^(6)A sites,which mainly occurred around acceptor and donor splice sites.This study demonstrates the utility of our antibody-independent method in identifying total and methylated circular RNAs using Nanopore DRS.This method has the additional advantage of providing the exact location of m^(6)A sites at single-base resolution in circular RNAs or linear transcripts from non-coding RNA without poly(A)tails.Yongsheng Wang Huihui Wang Feihu Xi Huiyuan Wang Ximei Han Wentao Wei Hangxiao Zhang Qianyue Zhang Yushan Zheng Qiang Zhu Markus V.Kohnen Anireddy S.N.Reddy Lianfeng Gu 2020Journal of Integrative Plant Biology2020,62,12:3
5Parameter estimates of Heston stochastic volatility model with MLE and consistent EKF algorithm显示文摘Heston model is the most famous stochastic volatility model in finance. This paper considers the parameter estimation problem of Heston model with both known and unknown volatilities. First, parameters in equity process and volatility process of Heston model are estimated separately since there is no explicit solution for the likelihood function with all parameters. Second, the normal maximum likelihood estimation(NMLE) algorithm is proposed based on the It?o transformation of Heston model. The algorithm can reduce the estimate error compared with existing pseudo maximum likelihood estimation. Third, the NMLE algorithm and consistent extended Kalman filter(CEKF) algorithm are combined in the case of unknown volatilities. As an advantage, CEKF algorithm can apply an upper bound of the error covariance matrix to ensure the volatilities estimation errors to be well evaluated. Numerical simulations illustrate that the proposed NMLE algorithm works more efficiently than the existing pseudo MLE algorithm with known and unknown volatilities. Therefore, the upper bound of the error covariance is illustrated. 6Credit Scoring Based on the Set-Valued Identification Method显示文摘Credit scoring is one of the key problems in financial risk managements.This paper studies the credit scoring problem based on the set-valued identification method,which is used to explain the relation between the individual attribute vectors and classification for the credit worthy and credit worthless lenders.In particular,system parameters are estimated by the set-valued identification algorithm based on a given recognition criteria.In order to illustrate the efficiency of the proposed method,practical experiments are conducted for credit card applicants of Australia and credit card holders from Taiwan,respectively.The empirical results show that the set-valued model has a higher prediction accuracy on both small and large numbers of data set compared with logistic regression model.Furthermore,parameters estimated by the set-valued identification method are more stable,which provide a meaningful and logical explanation for extracting factors that influence the borrowers’credit scorings.WANG Ximei HU Min ZHAO Yanlong DJEHICHE Boualem 2020Journal of Systems Science & Complexity2020,33,5:2

8Transcriptome and QTL analyses reveal candidate genes for fiber quality in Upland cotton显示文摘With increasing demand for high-quality cotton,it is desirable to identify genes involved in fiber development for molecular improvement of cotton.In this study,780 differentially expressed genes(DEGs)were identified in developing fibers at 10 days post-anthesis(DPA)in Gossypium hirsutum acc.DH962 and G.hirsutum cv.Jimian 5 using RNA-seq.Of 15 stable QTL for fiber quality identified in the same two parents in previous studies,4,3,6,1,and 1 QTL were associated with fiber length(FL),fiber strength(FS),micronaire(MIC),fiber elongation(FE)and fiber length uniformity ratio(FU),respectively.Integration of DEGs and QTL allowed the identification of 31 genes in 9 QTL regions,of which 25 were highly expressed in fibers based on the transcriptome datasets and 9 were preferentially expressed in different stages of fiber development.Gh_A01G0453(GhDTX19),Gh_D07G1581 and Gh_D04G0942 were expressed specifically in 5 and 10 DPA fibers,with Gh_D04G0942 showing low expression in other tissues except pistil.Gh_D07G1799(GhGAUT9),Gh_D11G0326(GhVPS29),Gh_D11G0333(GhTCP14),and Gh_D11G0334(GhNRP2)were preferentially expressed in 5 or 10 DPA fibers;Gh_A01G0397(GhABCG10)and Gh_D07G1744 were expressed specifically in 20 and 25 DPA fibers.These results suggest candidate genes for molecular improvement of cotton fiber quality.Hantao Wang Ruiting Zhang Chao Shen Ximei Li De Zhu Zhongxu Lin 2020The Crop Journal2020,8,1:2

14A Super-robust Armoured Superhydrophobic Surface with Excellent Anti-icing Ability显示文摘It has been proved that the construction of interconnected armour on superhydrophobic surface could significantly enhance the mechanical robustness.Here,a new kind of armour with frame/protrusion hybrid structure was achieved by nanosecond laser technology.Then,this armoured superhydrophobic surface demonstrated excellent durability,which could withstand linear abrasion(~3 N press)800 cycles,water jet test(1.0 MPa pressure)40 times and 100℃treatment 18 days.Particularly,the armoured superhydrophobic sample shows outstanding anti-icing ability,which can speed up the supercooled water dropping(no adhesion within 2 h),increase the freezing delay time by~3 times and maintain low adhesion force(less than 35 kPa)after 30 icing/de-icing cycles.Further finite element analysis and theoretical modeling proved that the developed frame/protuberance hybrid structure could effectively enhance the durability.The relatively low surface accuracy in this study can significantly reduce processing cost,which provides a bright future for the practical application of armour superhydrophobic materials.Peng Wang Hui Zhao Boyuan Zheng Ximei Guan Bin Sun Yongli Liao Ying Yue Wei Duan Haimin Ding 2023Journal of Bionic Engineering2023,20,5:1

18Self-assembled all-inclusive organic-inorganic nanoparticles enable cascade reaction for the detection of glucose显示文摘Traditional colorimetric glucose biosensor generally involves complex assay procedures.Free labile enzymes and peroxidase substrates are used separately for triggering a chromogenic reaction.These limits result in inferior enzyme stability and defective enzymatic catalytic efficiency,making it hard to routinely utilize them for the direct and fast test of glucose.In this work,we provide an all-inclusive substrates/enzymes nanoparticle employed 3,3’5,5’-tetramethylbenzidine(TMB) as chromogenic substrates and glucose oxidase(GOx)/horseradish peroxidase(HRP) as signal amplifier enzymes(TMB-GH NPs) by the molecule self-assembly technique.The 'all-inclusive' nanoparticles can realize the tandem colorimetric reactions,and the oxidation product of TMB(ox-TMB) exhibits a strong NIR laserdrive n photothermal effect,thus allowing qua ntitative photothermal detection of glucose.Owing to the restriction of the molecular motion of GOx,HRP,and TMB,the distance of mass transfer between substrates was s hortened largely,leading to improved catalytic activity for glucose.Overall,our strategy will simplify the analysis procedure,furthermore,these integrated nanoparticles not only display higher stability and activity than that of the free GOx/HRP system and possesses an excellent performance for colorimetric and photothermal bioassay of glucose simultaneously.We believe that this unique technique will give good inspirations to develop simple and precise methods for bioassay.Ximei Sun Yan Li Qian Yang Yunwei Xiao Yuting Zeng Jindi Gong Ziyu Wang Xiaofeng Tan He Li 2021Chinese Chemical Letters2021,32,5:1
