1High-throughput screening for small molecule inhibitors of the type-I interferon signaling pathway显示文摘Interferons(IFNs) are cytokines with fundamental roles in resistance to infections, cancer and other diseases. Type-I IFNs, interferon α(IFN-α) and interferon β(IFN-β), act through a shared receptor complex(IFNAR) comprised of IFNAR1 and IFNAR2 subunits. Binding of type-I IFN to IFNAR1 will robustly activate Janus activated kinase-signal transducer and activator of transcription(JAK-STAT)signaling pathway. Aberrant activation of the type-I IFN response results in a spectrum of disorders called interferonopathies. The purpose of this research is to develop an assay for high-throughput screening(HTS) of small molecule inhibitors of the type-I IFN signaling pathway. Inhibition of type-I IFN signaling can be beneficial in terms of therapeutic use and understanding the underlying mechanism of action. We report here a HTS campaign with the secreted embryonic alkaline phosphatase(SEAP) reporter gene assay against 32,000 compounds which yielded 25 confirmed hits. 7Analytical model of non-uniform charge distribution within the gated region of GaN HEMTs显示文摘A physics-based analytical expression that predicts the charge,electrical field and potential distributions along the gated region of the GaN HEMT channel has been developed.Unlike the gradual channel approximation(GCA),the proposed model considers the non-uniform variation of the concentration under the gated region as a function of terminal applied volt-ages.In addition,the model can capture the influence of mobility and channel temperature on the charge distribution trend.The comparison with the hydrodynamic(HD)numerical simulation showed a high agreement of the proposed model with numerical data for different bias conditions considering the self-heating and quantization of the electron concentration.The ana-lytical nature of the model allows us to reduce the computational and time cost of the simulation.Also,it can be used as a core expression to develop a complete physics-based transistorⅣmodel without GCA limitation.Amgad A.Al-Saman Eugeny A.Ryndin Xinchuan Zhang Yi Pei Fujiang Lin 2023Journal of Semiconductors2023,44,8:0
8Fact-condition statements and super relation extraction for geothermic knowledge graphs construction显示文摘Researchers utilize information from the geoscience literature to deduce the regional or global geological evolution.Traditionally this process has relied on the labor of researchers.As the number of papers continues to increase,acquiring domain-specific knowledge becomes a heavy burden.Knowledge Graph(KG)is proposed as a new knowledge representation technology to change this situation.However,the super relation is not considered in the previous KG,which bridges the geological phenomenon(fact)and its precondition(condition).For instance,in the statement(“the late Archean was a crucial transition period in the history of global geodynamics”),the condition statement(“crucial transition for global geodynamics”)works as the complementary fact statement(“the late Archean was a crucial transition period”),which defines the scale of crucial transition accurately in the late Archean.In this study,fact-condition statement extraction is introduced to construct a geological knowledge graph.A rule-based multi-input multi-output model(R-MIMO)is proposed for information extraction.In the R-MIMO,fact-condition statements and their super relation are considered and extracted for the first time.To verify its performances,a GeothCF dataset with 1455 fact tuples and 789 condition tuples is constructed.In experiments,the R-MIMO model achieves the best performance by using BERT as encoder and LSTM-d as decoder,achieving F180.24%in tuple extraction and F170.03%in tag prediction task.Furthermore,the geothermic KG with super relation is automatically constructed for the first time by trained R-MIMO,which can provide structured data for further geothermic research.Qizhi Chen Hong Yao Shengwen Li Xinchuan Li Xiaojun Kang Wenwen Lai Jian Kuang 2023Geoscience Frontiers2023,14,5:0
