1Comparative cytotoxicity of fourteen trivalent and pentavalent arsenic species determined using real-time cell sensing显示文摘The occurrence of a large number of diverse arsenic species in the environment and in biological systems makes it important to compare their relative toxicity. The toxicity of arsenic species has been examined in various cell lines using different assays, making comparison difficult. We report real-time cell sensing of two human cell lines to examine the cytotoxicity of fourteen arsenic species: arsenite(As~Ⅲ), monomethylarsonous acid(MMA~Ⅲ) originating from the oxide and iodide forms, dimethylarsinous acid(DMA~Ⅲ), dimethylarsinic glutathione(DMAG~Ⅲ), phenylarsine oxide(PAO~Ⅲ), arsenate(AsV), monomethylarsonic acid(MMA~Ⅴ), dimethylarsinic acid(DMA~Ⅴ),monomethyltrithioarsonate(MMTTA~Ⅴ), dimethylmonothioarsinate(DMMTA~Ⅴ),dimethyldithioarsinate(DMDTA~Ⅴ), 3-nitro-4-hydroxyphenylarsonic acid(Roxarsone, Rox),and 4-aminobenzenearsenic acid(p-arsanilic acid, p-ASA). Cellular responses were measured in real time for 72 hr in human lung(A549) and bladder(T24) cells. IC50 values for the arsenicals were determined continuously over the exposure time, giving rise to IC50 histograms and unique cell response profiles. Arsenic accumulation and speciation were analyzed using inductively coupled plasma-mass spectrometry(ICP-MS). On the basis of the 24-hr IC50 values, the relative cytotoxicity of the tested arsenicals was in the following decreasing order: PAO~Ⅲ? MMA~Ⅲ≥ DMA~Ⅲ≥ DMAG~Ⅲ≈ DMMTA~Ⅴ≥ As~Ⅲ? MMTTA~Ⅴ> AsV> DMDTA~Ⅴ>DMA~Ⅴ> MMA~Ⅴ≥ Rox ≥ p-ASA. Stepwise shapes of cell response profiles for DMA~Ⅲ, DMAG~Ⅲ,and DMMTA~Ⅴcoincided with the conversion of these arsenicals to the less toxic pentavalent DMA~Ⅴ. 2Methylated and thiolated arsenic species for environmental and health research——A review on synthesis and characterization显示文摘Hundreds of millions of people around the world are exposed to elevated concentrations of inorganic and organic arsenic compounds, increasing the risk of a wide range of health effects. Studies of the environmental fate and human health effects of arsenic require authentic arsenic compounds. We summarize here the synthesis and characterization of more than a dozen methylated and thiolated arsenic compounds that are not commercially available. We discuss the methods of synthesis for the following14 trivalent(Ⅲ) and pentavalent() arsenic compounds: monomethylarsonous acid(MMA~Ⅲ), dicysteinylmethyldithioarsenite(MMA~Ⅲ(Cys)_2), monomethylarsonic acid(MMA~Ⅴ),monomethylmonothioarsonic acid(MMMTAⅤ) or monothio-MMA~Ⅴ, monomethyldithioarsonic acid(MMDTA~Ⅴ) or dithio-MMA~Ⅴ, monomethyltrithioarsonate(MMTTA~Ⅴ) or trithio-MMA~Ⅴ,dimethylarsinous acid(DMA~Ⅲ), dimethylarsino-glutathione(DMA~Ⅲ(SG)), dimethylarsinic acid(DMA~Ⅴ), dimethylmonothioarsinic acid(DMMTA~Ⅴ) or monothio-DMAⅤ, dimethyldithioarsinic acid(DMDTA~Ⅴ) or dithio-DMA~Ⅴ, trimethylarsine oxide(TMAO~Ⅴ), arsenobetaine(AsB), and an arsenicin-A model compound. We have reviewed and compared the available methods,synthesized the arsenic compounds in our laboratories, and provided characterization information. 3HPLC-DRC-ICP-MS检测四种尿砷化合物显示文摘拟建立检测尿液中As(Ⅲ)、DMA(Ⅴ)、MMA(Ⅴ)和As(Ⅴ)4种砷化合物形态的HPLC—DRC-ICP—MS的方法。采用DRCTM(反应气为O2)与HPLC联用作为检测器,Phenomenex ODS-3作为分离柱,5 mmol/L四丁基羟胺,3 mmol/L丙二酸,5%(体积分数)甲醇为流动相(pH=5.0~6.0),建立了同时分析尿液中As(Ⅲ)、DMA(Ⅴ)、MMA(Ⅴ)和As(Ⅴ)的方法。实验结果显示:该分析方法对上述4种砷化合物的检测限均在0.1μg/L以下,在1~20μg/L范围内线性关系良好,准确度和精密度良好,回收率为89%~106%,样品中的Cl^-1对测定没有影响。所建立的HPLC-DRC-ICP—MS分析方法稳定、可靠,适用于快速、批量测定尿砷形态。邓国栋 郑宝山 王学 李社红 王明国 李波 胡军 Xiufen LU X Chris Le 2008地球与环境2008,36,3:2

12Study of vascular smooth muscle cell calcification induced by hyperphosphate and intervented by phosphonoformic acid显示文摘Objective:To evaluate the effects of different concentrations of phosphate on calcium deposition and osteocalcin level in cultured bovine aortic smooth muscle cell,investigate the mechanism of hyperphosphatemia to evoke calcification of vascular smooth muscle cell and observe the effects of phosphonoformic acid(PFA) in different concentrations on vascular calcification.Methods:The bovine aortic smooth muscle cells(BASMC) were cultured.Calcium deposition and the expression of osteocalcin of BASMC in different concentrations of phosphate(1.5 mmol/L and 2.0 mmol/L) and PFA were determined by o-cresolphthalein complexone and radioimmunity methods,respectively.Osteocalcin mRNA expressions were determined by RT-PCR.Results:After six or nine days of BASMC cultured,the calcium deposition in Pi 2.0 mmol/L group was more than that in Pi 1.5 mmol/L group[(77.187±11.692)μg/(mg·protein) vs(25.768±1.750)μg/(mg·protein),P < 0.01 and(125.399±16.677)μg/(mg·protein) vs(29.046±2.635)μg/(mg·protein),P < 0.01 respectively].The calcium deposition was dependent on time and dosage of phosphate treatment.After 72 h culture the osteocalcin in Pi 2.0 mmol/L group was more than that in Pi 1.5 mmol/L group[in supernatant,(1.503±10-2±2.601×10-3)ng/(μg· protein) vs(2.981×10-3±8.382×10-4)ng/(μg·protein),P < 0.001],the same was found in osteocalcin mRNA expression[OC/GAPDH,(1.906±0.132) vs(0.748±0.037),P < 0.001].Compared to Pi 1.5mmol/L group,bovine smooth muscle cells(BSMC) cultured in media containing Pi 2.0 mmol/L phosphate levels increased calcium deposition[On day 6,(77.187±11.692)μg/(mg·protein) vs(25.768±1.750)μg/(mg·protein),P < 0.001].Elevated phosphate treatment of BSMCs also enhanced the expression of the osteoblastic differentiation marker osteocalcin[On day 3,Pi 2.0 mmol/L group vs Pi 1.5mmol/L group,(1.503×10-2±2.601×10-3)ng/(μg·protein) vs(2.981×10-3±8.382×10-4)ng/(μg·protein),P < 0.001].PFA decreased ciacium deposition and osteocalcin expres-sion statistically[Pi 2.0 mmol/L+PFA1.0 mmol/L group vs Pi 2.0mmol/L group ,ciacium deposition,(37.729±5.899)μg/(mg· protein) vs(77.187±11.692)μg/(mg·protein),P < 0.001];Osteocalcin in supernatant,(4.529±10-3±1.250×10-3)ng/(μg·protein) vs(1.503×10-2±2.601×10-3) ng/(μg·protein),P < 0.001;osteocalcin mRNA expression,OC/GAPDH,(0.642±0.092) vs(1.89±0.165),P < 0.01].Conclusion:Hyperphosphate may directly promote calcium deposition and the osteocalcin expression of BASMCs.It may be a new explanation for the phenomenon of vascular calcification in hyperphosphatemic conditions.Hyperphosphatemia is an independent factor to stimulate vascular calcification.PFA can inhibit calcium deposition and osteocalcin expression induced by elevated phosphate.PFA may be a new medicine to treat vascular calcification induced by elevated phosphate.Weiping Lu Xiaoyun Wang Xiufen Zhao 2007Journal of Nanjing Medical University2007,21,6:1

14Effects of Dietary Intake of Arsenosugars and Other Organic Arsenic Species on Studies of Arsenic Methylation Efficiency in Humans显示文摘Extensive research has used dimethylarsinic acid(DMA)in urine as a marker of arsenic methylation.The premise is that humans methylate inorganic arsenicals to monomethylarsonic acid(MMA)and DMA and excrete these arsenic species into the urine.However,DMA in urine not only comes from the methylation of inorganic arsenic but also could be a result of metabolism of other arsenic species,such as arsenosugars and arsenolipids.Most environmental health and epidemiological studies of arsenic methylation might have overlooked confounding factors that contribute to DMA in urine.Here we critically evaluate reported studies that used methylation indexes,concentration ratios of methylated arsenicals,or the percentage of DMA in urine as markers of arsenic methylation efficiency.Dietary intake of arsenosugars potentially confounds the calculation and interpretation of the arsenic methylation efficiencies.Many studies have not considered incidental dietary intake of arsenosugars,arsenolipids,and other organic arsenic species.Future studies should consider the dietary intake of diverse arsenic species and their potential effect on the urinary concentrations of DMA.Tetiana Davydiuk Jeffrey Tao Xiufen Lu X.Chris Le 2023Environment & Health2023,1,4:0
15Measurement of reflected second harmonics and nonlinearity parameter using a transducer with complex structure显示文摘Measurement of nonlinearity parameter using the second-harmonic reflective model is studied. A new kind of compound transducer is designed and fabricated for this purpose. With this transducer and the finite amplitude insert-substitution method, an experimental system to measure the nonlinearity parameter using reflective model is developed. 16The influence of molecular structure of protein on the acoustic nonlinearity parameter B/A显示文摘In this paper, the influence of molecular structure of proteins on the acoustic non-linearity parameter B/A has been studied. By using biochemical technique, three proteins such as bovine serum albumin, bovine hemoglobin and pepsin were employed and their quarternary,tertiary and secondary protein structures were perturbed by the denaturing agent sodium do-decyl sulfate (SDS) solution, while their primary structures of protein were not changed. This means that only the structural features were altered and the chemical composition maintained unchanged. B/A has been measured by using improved thermodynamic method. The experi-mental results show that when the concentration of SDS solution increases, the alterations of the protein structure increase, and the contribution of protein to the B/A value decreases. 17Study of acoustic nonlinearity parameter B/A for seawater显示文摘The B/ A values of NaCl solution and arhficial seawater (prepared by LymanFleming formula) with different salinities and temperatures are measured and compared with the calculated B / A values in literature. 18Synthesis,Style Editing,and Animation of 3D Cartoon Face显示文摘As a popular kind of stylized face,cartoon faces have rich application scenarios.It is challenging to create personalized 3D cartoon faces directly from 2D real photos.Besides,in order to adapt to more application scenarios,automatic style editing,and animation of cartoon faces is also a crucial problem that is urgently needed to be solved in the industry,but has not yet had a perfect solution.To solve this problem,we first propose“3D face cartoonizer”,which can generate high-quality 3D cartoon faces with texture when fed into 2D facial images.We contribute the first 3D cartoon face hybrid dataset and a new training strategy which first pretrains our network with low-quality triplets in a reconstruction-then-generation manner,and then finetunes it with high-quality triplets in an adversarial manner to fully leverage the hybrid dataset.Besides,we implement style editing for 3D cartoon faces based on k-means,which can be easily achieved without retrain the neural network.In addition,we propose a new cartoon faces'blendshape generation method,and based on this,realize the expression animation of 3D cartoon faces,enabling more practical applications.Our dataset and code will be released for future research.Ming Guo Feng Xu Shunfei Wang Zhibo Wang Ming Lu Xiufen Cui Xiao Ling 2024Tsinghua Science and Technology2024,29,2:0
19Arsenic speciation in freshwater fish:challenges and research needs显示文摘Food and water are the main sources of human exposure to arsenic.It is important to determine arsenic species in food because the toxicities of arsenic vary greatly with its chemical speciation.Extensive research has focused on high concentrations of arsenic species in marine organisms.The concentrations of arsenic species in freshwater fish are much lower,and their determination presents analytical challenges.In this review,we summarize the current state of knowledge on arsenic speciation in freshwater fish and discuss challenges and research needs.Fish samples are typically homogenized,and arsenic species are extracted using water/methanol with the assistance of sonication and enzyme treatment.Arsenic species in the extracts are commonly separated using high-performance liquid chromatography(HPLC)and detected using inductively coupled plasma mass spectrometry(ICPMS).Electrospray ionization tandem mass spectrometry,used in combination with HPLC and ICPMS,provides complementary information for the identification and characterization of arsenic species.The methods and perspectives discussed in this review,covering sample preparation,chromatography separation,and mass spectrometry detection,are directed to arsenic speciation in freshwater fish and applicable to studies of other food items.Despite progress made in arsenic speciation analysis,a large fraction of the total arsenic in freshwater fish remains unidentified.It is challenging to identify and quantify arsenic species present in complex sample matrices at very low concentrations.Further research is needed to improve the extraction efficiency,chromatographic resolution,detection sensitivity,and characterization capability.Karen S.Hoy Tetiana Davydiuk Xiaojian Chen Chester Lau Jordan R.M.Schofield Xiufen Lu Jennifer A.Graydon Ruth Mitchell Megan Reichert X.Chris Le 2023Food Quality and Safety2023,7,3:0
