| 1 | Effects of Coal Rank and High Organic Sulfur on the Structure and Optical Properties of Coal-based Graphene Quantum Dots显示文摘Coal-based graphene quantum dots(GQDs) were successfully produced via a one-step chemical synthesis from six different coal ranks, from which two superhigh organic sulfur(SHOS) coals were selected as natural S-doped carbon sources for the preparation of S-doped GQDs. The effects of coal properties on coal-based GQDs were analyzed by means of high-resolution transmission electron microscopy(HRTEM), X-ray diffraction(XRD), Fourier transform infrared(FTIR) spectroscopy, X-ray photoelectron spectroscopy(XPS), ultraviolet-visible(UV-Vis) absorption spectroscopy, and fluorescence emission spectra. It was shown that all coal samples can be used to prepare GQDs, which emit bluegreen and blue fluorescence under ultraviolet light. Anthracite-based GQDs have a hexagonal crystal structure without defects, the largest size, and densely arranged carbon rings in their lamellae; the highrank bituminous coal-based GQDs are relatively reduced in size, with their hexagonal crystal structure being only faintly visible; the low-rank bituminous coal-based GQDs are the smallest, with sparse lattice fringes and visible internal defects. As the metamorphism of raw coals increases, the yield decreases and the fluorescence quantum yield(QY) initially increases and then decreases. Additionally, the surface of GQDs that were prepared using high-rank SHOS coal(high-rank bituminous coal) preserves rich sulfur content even after strong oxidation, which effectively adjusts the bandgap and improves the fluorescence QY. | 2 | Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. | 4 | Sequence framework of two different kinds of margins and their response to tectonic activity during the Middle-Late Triassic, Ordos Basin显示文摘Two kinds of margin respectively occur in the Ordos Basin during the Middle-Late Triassic (Yanchang Age), one is foreland margin developed under the background of flexural subsidence by thrusting intensively in the southwest margin, and the other is intracratonic basin margin by stable subsidence in northern and central parts of the basin. The Middle-Late Triassic Yanchang Formation can be divided into four regional third-order sequences, which are separated by gentle angular unconformity or regional erosion surface, made up of lowstand system tract (LST), expanding system tract (EST) and highstand system tract (HST) from lower to upper within a sequence. But there are distinct differences of the sequence framework between the southwest margin and northern and central parts of the basin. The southwest margin develops heavy conglomerate layer and unconformity as a result of orogeny by thrusting, and the intracratonic basin margin by stable subsidence in the northern and central parts grows aggradational sandstone, conglomerate in fluvio-delta system and parallel unconformity. The depositional framework of southwest margin reflects the tectonic evolution from flexural subsidence by thrusting to rebounded uplift. The formation of sequence boundary is related to the resilient uplift and erosion. The sequence stratigraphic framework and depositional system tract configuration in the foreland basin are controlled by structural activity of the fold and thrust belt, and the sequence succession reflects episodic thrusting of the Middle-Late Triassic toward the foreland basin. The sequence evolution in northern and central parts reflects the depositional succession of fluvio-delta system under intracratonic background, composed of coarse-grained sediment in braided channel deposit at the lower, meandering channel deposit in the middle and fine-grained sediment in the flood plain at the upper, dominated by lake level fluctuation. | 11 | Polyguluronate Sulfate and Its Oligosaccharides but not Heparin Promotes FGF19/FGFR1c Signaling显示文摘Fibroblast growth factor 19(FGF19) functions as a hormone by affecting glucose metabolism. FGF19 improves glucose tolerance when overexpressed in mice with impaired glucose tolerance or diabetes. A functional cellular FGF19 receptor consists of FGF receptor(FGFR) and glycosaminoglycan complexed with either α Klotho or β Klotho. Interestingly, in mice with diet-induced diabetes, a single injection of FGF1 is enough to restore blood sugar levels to a healthy range. FGF1 binds heparin with high affinity whereas FGF19 does not, indicating that polysaccharides other than heparin might enhance FGF19/FGFR signaling. Using a FGFs/FGFR1 c signaling-dependent Ba F3 cell proliferation assay, we discovered that polyguluronate sulfate(PGS) and its oligosaccharides, PGS12 and PGS25, but not polyguluronate(PG), a natural marine polysaccharide, enhanced FGF19/FGFR1 c signaling better than that of heparin based on ~3H-thymidine incorporation. Interestingly, PGS6, PGS8, PGS10, PGS12, PGS25, and PGS, but not PG, had comparable FGF1/FGFR1 c signal-stimulating activity compared to that of heparin. These results indicated that PGS and its oligosaccharides were excellent FGF1/FGFR1 c and FGF19/FGFR1 c signaling enhancers at cellular level. | 13 | Melatonin ameliorates bleomycin-induced pulmonary fibrosis via activating NRF2 and inhibiting galectin-3 expression显示文摘Pulmonary fibrosis(PF)is a chronic interstitial lung disease with no effective therapies.Galectin-3(Gal-3),a marker of oxidative stress,plays a key role in the pathogenesis of PF.Fibroblast-myofibroblast differentiation(FMD)is an important source of fibrotic cells in PF.Previous studies showed that melatonin(MT)exerted anti-fibrotic effect in many diseases including PF through its antioxidant activity.In the present study we investigated the relationships among Gal-3,NRF2,ROS in FMD and their regulation by MT.We established an in vitro model of FMD in TGF-β1-treated human fetal lung fibroblast1(HFL1)cells and a PF mouse model via bleomycin(BLM)intratracheal instillation.We found that Gal-3 expression was significantly increased both in vitro and in vivo.Knockdown of Gal-3 in HFL1 cells markedly attenuated TGF-β1-induced FMD process and ROS accumulation.In TGF-β1-treated HFL1 cells,pretreatment with NRF2-specific inhibitor ML385(5μM)significantly increased the levels of Gal-3,α-SMA and ROS,suggesting that the expression of Gal-3 was regulated by NRF2.Treatment with NRF2-activator MT(250μM)blockedα-SMA and ROS accumulation accompanied by reduced Gal-3 expression.In BLM-induced PF model,administration of MT(5 mg·kg^(−1)·d^(−1),ip for 14 or 28 days)significantly attenuated the progression of lung fibrosis through up-regulating NRF2 and down-regulating Gal-3 expression in lung tissues.These results suggest that Gal-3 regulates TGF-β1-induced pro-fibrogenic responses and ROS production in FMD,and MT activates NRF2 to block FMD process by down-regulating Gal-3 expression.This study provides a useful clue for a clinical strategy to prevent PF. | Yue-jiao Lan Ming-han Cheng Hui-min Ji Yu-qian Bi Yong-yue Han Chong-yang Yang Xuan Gu Jian Gao Hong-liang Dong | 2023 | Acta Pharmacologica Sinica2023,44,5: | 2 |

| 19 | All-trans retinoic acid regulates the expression of MMP-2 and TGF-β2 via RDH5 in retinal pigment epithelium cells显示文摘·AIM: To investigate the effect of all-trans retinoic acid(ATRA) on retinol dehydrogenase 5(RDH5), matrix metalloproteinase-2(MMP-2) and transforming growth factor-β2(TGF-β2) transcription levels, and the effect of RDH5 on MMP-2 and TGF-β2 in retinal pigment epithelium(RPE) cells.·METHODS: After adult RPE cell line-19(ARPE-19 cells) intervened with gradient concentrations of ATRA(0-20 μmol/L) for 24h, flow cytometry was used to detect the proliferation and apoptosis of cells in each group, and quantitative realtime polymerase chain reaction(q RT-PCR) was used to detect RDH5, MMP-2 and TGF-β2 m RNA expression. Then, after ARPE-19 cells transfected with three different si RNA targets for 48h, the RDH5 knockdown efficiency of each group and expression of MMP-2 and TGF-β2 m RNA within them was detected by q RT-PCR. ·RESULTS: Flow cytometry results showed that ATRA could inhibit the proliferation of RPE cells and promote the apoptosis of RPE cells, and the difference of apoptosis was statistically significant when the ATRA concentration exceeded 5 μmol/L and compared with the normal control group(P=0.027 and P=0.031, respectively). q RT-PCR results showed that ATRA could significantly inhibit the expression level of RDH5 m RNA(P<0.001) and promote the expression of MMP-2 and TGF-β2 m RNA(P=0.03 and P<0.001, respectively) in a dose-dependent manner, especially when treated with 5 μmol/L ATRA. The knockdown efficiency of RDH5 si RNA varies with different targets, among which RDH5 si RNA-435 had the highest knockdown efficiency, i.e., more than 50% lower than that of the negative control group(P=0.02). When RDH5 was knocked down for 48h, the results of q RT-PCR showed that the expressions of MMP-2 and TGF-β2 m RNA were significantly up-regulated(P<0.001).·CONCLUSION: ATRA inhibits the expression of RDH5 and promotes MMP-2 and TGF-β2, and further RDH5 knockdown significantly upregulates MMP-2 and TGF-β2.
