5Carbon monoxide-induced adventitious rooting of hypo-cotyl cuttings from mung bean seedling显示文摘In this paper, the regulatory role of ex-ogenous carbon monoxide (CO) during the genera-tion and development of adventitious roots in mung bean (Phaseolus radiatus L. cv. Mingguang) hypo-cotyl cuttings was surveyed. The results indicated that, like nitric oxide (NO), CO donor Hematin in-duced adventitious rooting in dose- and time-de- pendent manners. These responses were also prov- en by the addition of gaseous CO aqueous solution with different concentrations. Additionally, the above CO-mediated effect was differently reversed when CO scavenger hemoglobin (Hb), CO specific syn-thetic inhibitor ZnPPIX, the auxin transport inhibitor NPA and nitric oxide synthase (NOS) inhibitor L-NAME were added with Hematin, respectively. 6Involvement of carbon monoxide produced by heme oxygenase in ABA-induced stomatal closure in Vicia faba and its proposed signal transduction pathway显示文摘Carbon monoxide (CO) has recently proven to be an important bioactive or signaling molecule in mammalian cells. Its effects are mainly mediated by nitric oxide (NO) and cyclic GMP (cGMP). In Vicia faba leaves, CO production and heme oxygenase (HO) activity, an important CO synthetic enzyme, are first reported to increase in response to ABA treatment, which could result in stomatal closure. Inter- estingly, ABA-induced stomatal closure in V. faba guard cells is partially blocked when the synthetic CO inhibitor ZnPP, or the CO/NO scavenger Hb is added. Furthermore, we show that, exogenously applied CO donor, hematin, and CO aqueous solution not only result in the enhancement of CO release, but also time-dependently induce stomatal closure, and the latter is mimicked by the application of an NO donor SNP. The above-mentioned stomatal closure effects are differentially reversed by the addition of tungstate, a potent inhibitor of NO synthetic enzyme nitrate reductase (NR), the specific NO scavenger cPTIO, ZnPP, or Hb. During treatment for 4 h, SNP, 0.01% CO aqueous solution or hematin significantly triggers NO synthesis, whereas cPTIO, or tungstate approximately fully inhibits NO fluorescence. Ad- ditionally, application of the GC inhibitor ODQ blocks CO-induced stomatal closure. This inhibition could be reversed when 8-Br-cGMP is added. 7Endogenous Hydrogen Peroxide Plays a Positive Role in the Upregulation of Heme Oxygenase and Acclimation to Oxidative Stress in Wheat Seedling Leaves显示文摘Pretreatment of lower H2O2 doses (0.05, 0.5 and 5 mM) for 24 h was able to dose-dependently attenuate lipid peroxidation in wheat seedling leaves mediated by further oxidative damage elicited by higher dose of H2O2 (150 mM) for 6 h, with 0.5 mM H2O2 being the most effective concentrations. Further results illustrated that 0.5 mM H2O2 pretreatment triggered the biphasic production of H2O2 during a 24 h period. We also noticed that only peak I (0.25 h) rather than peak Ⅱ (4 h) was approximately consistent with the enhancement of heme oxygenase (HO) activity, HO-1 gene expression. Meanwhile, enhanced superoxide dismutase (SOD) activity, Mn-SOD and Cu,Zn-SOD transcripts might be a potential source of peak I of endogenous H2O2. Further results confirmed that 0.5 mM H2O2 treatment for 0.5 h was able to upregulate HO gene expression, which was detected by enzyme activity determination, semi-quantitative reverse transcription-polymerase chain reaction and western blotting. Meanwhile, the application of N,N'-dimethylthiourea, a trap for endogenous H2O2, not only blocked the upregulation of HO, but also reversed the corresponding oxidation attenuation. 12Ultrathin Al2O3-coated reduced graphene oxide membrane for stable lithium metal anode显示文摘Lithium(Li) metal has been considered as the most attractive anode materials for Li-ion batteries(LIBs)due to its high theoretic specific capacity. The formation of unstable solid electrolyte interphase(SEI) and dendritic Li on the metal anode, however, hindered its practical application. Herein, to address the issues, a Li-free electrode with ultrathin Al_2O_3 coated on reduced graphene oxide(rGO) membrane that covers a Cu foil current collector was developed. The composite electrode exhibits excellent interfacial protection of lithium metal deposited between Cu foil and rGO electrochemically. Firstly, it affords good Li^+ permeability from the electrolyte. Secondly, the ultrathin Al_2O_3 has sufficient mechanical strength to inhibit the penetration of Li dendrite. 13Whole-Genome Resequencing of a Worldwide Collection of Rapeseed Accessions Reveals the Genetic Basis of Ecotype Divergence显示文摘Rapeseed (Brassica napus),an important oilseed crop,has adapted to diverse climate zones and latitudes by forming three main ecotype groups,namely winter,semiwinter,and spring types. However,genetic variations underlying the divergence of these ecotypes are largely unknown. Here,we report the global pattern of genetic polymorphisms in rapeseed determined by resequencing a worldwide collection of 991 germplasm accessions.A total of 5.56 and 5.53 million singlenucleotide polymorphisms (SNPs)as Well as 1.86 and 1.92 million InDels were identified by mapping reads to the reference genomes of 'Darmor-bzh'and 'Tapidor,'respectively.We generated a map of allelic drift paths that shows splits and mixtures of the main populations,and revealed an asymmetric evolution of the two subgenomes of B.napus by calculating the genetic diversity and linkage disequilibrium parameters.Selective-sweep analysis revealed genetic changes in genes orthologous to those regulating various aspects of plant development and response to stresses.A genome-wide association study identified SNPs in the promoter regions of FLOWERING LOCUS T and FLOWERING LOCUS C orthologs that corresponded to the different rapeseed ecotype groups. 14Risk factors for hepatic decompensation in patients with primary biliary cirrhosis显示文摘AIM:To examine the clinical features and analyze prognostic factors in a prospective study of primary biliary cirrhosis(PBC) patients.METHODS:From 1995 to 2010,PBC patients without hepatic decompensation seen at the Peking Union Medical College Hospital were enrolled.Clinical signs and manifestations(pruritus,persistent fatigue,jaundice and pain in the right hypochondrium),laboratory parameters(auto-antibodies for autoimmune hepatic disease,biliary and hepatic enzymes,immunoglobulin,bilirubin,and albumin) and imaging findings were recorded at entry and at specific time points during follow-up.Cox regression and Kaplan-Meier analyses,respectively,assessed the risk factors for hepatic decompensation and survival.RESULTS:Two hundred and sixty-two PBC patients were enrolled with a median follow-up of 75.2 mo(range,21-201 mo).The 240 patients were aged 51.5 ± 10.2 years at diagnosis and 91.6% were female.Two hundred and forty-five(93.5%) were seropositive for anti-mitochondrial antibodies.At presentation,170 patients(64.9%) were symptomatic,while 96 patients(36.6%) had extra-hepatic autoimmune disease.During the follow-up period,62(23.7%) patients developed hepatic decompensation of whom four underwent liver transplantation and 17 died.The cumulative survival rate and median survival time were 83.9% and 181.7 mo,respectively.Cox regression analysis revealed that an incomplete ursodeoxycholic acid(UDCA) response or inconsistent treatment [P < 0.001;hazard risk(HR) 95%CI = 2.423-7.541],anti-centromere antibodies(ACA) positivity(P < 0.001;HR 95%CI = 2.516-7.137),alanine aminotransferase ratio(AAR) elevations(P < 0.001;HR 95%CI = 1.357-2.678),and histological advanced liver disease(P = 0.006;HR 95%CI = 1.481-10.847) were predictors of hepatic decompensation.The clinical features and survival of PBC in China are consistent with those described in Western countries.CONCLUSION:Incomplete UDCA response or inconsistent treatment,ACA positivity,AAR elevations,and advanced histological stage are predictors of decompensation.Tian-Yan Shi Li-Na Zhang Hua Chen Li Wang Min Shen Xuan Zhang Feng-Chun Zhang 2013World Journal of Gastroenterology2013,19,7:9
15Is urban development an urban river killer? A case study of Yongding Diversion Channel in Beijing, China显示文摘The high population and concrete environment alter urban areas by changing temperature, rainfall runoff, and water resource utilization activities. This study was conducted to investigate the water quality features of the Yongding Diversion Channel in Beijing, China, and its relationship with rainfall and urban development. Monthly water quality data were obtained from April to October of 2004 at monitoring sites of Sanjiadian, Gaojing, Luodaozhuang, and Yuyuangtan. The monthly water quality grades from 2007 to 2011 were also investigated and compared with those of other rivers. Dissolved oxygen and pH showed greater decreases after one or two moderate rainfall events than several light rainfall events. The potassium permanganate index(CODMn), ammonia nitrogen(NH3-N) and total phosphorus(TP) increased more after several light rainfall events than after one or two moderate or heavy rainfall events. Pollutant concentrations(CODMn, NH3-N, TP) in downstream regions showed greater changes than those in upstream areas after heavy rainfall events. Intense human activities around the channel greatly influenced the water quality of the channel in rainy season because of runoff pollution; however, heavy rainfall had a strong dilution effect on the pollutant concentrations in rivers. Overall, urban development has obviously deteriorated the water quality of the Yongding Diversion Channel as indicated by an increase in the water quality index from 3.22 in 2008 to 4.55 in2010. 17针栓式喷注器单元液/液撞击的一次破碎形态的仿真研究显示文摘目的:针对液/液针栓式喷注器单元,研究其在不同径向射流喷注速度下的一次破碎形态,并阐明在一次破碎下喷雾半锥角的形成、压力场分布、喷雾粒径分布与速度场分布。创新点:通过流体体积函数转换离散相(VOF-to-DPM)模型,结合网格自适应(AMR)技术还原了针栓式喷注器单元液/液撞击的一次破碎形态。方法:1.通过VOF-to-DPM模型完成一次破碎过程中对液相的捕捉;2.采用计算流体动力学后处理(CFD-post)模块进行后处理,得到一次破碎下喷雾半锥角的形成以及压力场、喷雾粒径与速度场的分布云图;3.在仿真计算过程中使用AMR技术减少计算量,节约时间成本与计算资源。结论:1.速度大的径向射流在穿透轴向液膜后会形成一个蘑菇状的头部;扰动在蘑菇状顶端下方形成涡,有助于蘑菇状顶端边缘破碎的发生。2.气动力和表面张力对轴向液膜破碎过程中产生的直液丝和环状液丝的破碎起到重要作用;由于液滴的聚合现象,轴向液膜的直径在破碎过程中并不是单调递减的。3.喷雾半锥角的大小和径向射流速度的大小成正比;一次破碎首先发生在轴向液膜前沿、径向射流头部以及撞击点附近。4.在表面张力的作用下,轴向液膜内边缘的速度较小,直径较大;当径向射流的速度增大时,轴向液膜内边缘的速度值减小得更加明显。Rui ZHOU Chi-bing SHEN Xuan JIN 2020Journal of Zhejiang University-Science A(Applied Physics & Engineering)2020,21,8:8
