| 4 | Ectopic expression of VRT-A2 underlies the origin of Triticum polonicum and Triticum petropavlovskyi with long outer glumes and grains显示文摘Polish wheat (Triticum polonicum) is a unique tetraploid wheat species characterized by an elongated outer glume. The genetic control of the long-glume trait by a single semi-dominant locus, P1 (from Polish wheat), was established more than 100 years ago, but the underlying causal gene and molecular nature remain elusive. Here, we report the isolation of VRT-A2, encoding an SVP-clade MADS-box transcription factor, as the P1 candidate gene. Genetic evidence suggests that in T. polonicum, a naturally occurring sequence rearrangement in the intron-1 region of VRT-A2 leads to ectopic expression of VRT-A2 in floral organs where the long-glume phenotype appears. Interestingly, we found that the intron-1 region is a key ON/OFF molecular switch for VRT-A2 expression, not only because it recruits transcriptional repressors, but also because it confers intron-mediated transcriptional enhancement. Genotypic analyses using wheat accessions indicated that the P1 locus is likely derived from a single natural mutation in tetraploid wheat, which was subsequently inherited by hexaploid T. petropavlovskyi. | 5 | Clinical Predictors of Functional Cure in Children 1–6 Years-old with Chronic Hepatitis B显示文摘Background and Aims:Hepatitis B surface antigen(HBsAg)clearance is significantly more common in children with chronic hepatitis B(CHB)than in adults;however,the possible influencing factors related to HBsAg loss have yet to be found.This study aimed to explore the efficacy of long-term interferon(IFN)αtherapy in treating children with CHB and analyzed the factors influencing functional cure after treatment.Methods:A total of 236 children aged 1–6 years and diagnosed with CHB via liver biopsy were included in the study,all receiving IFNαtreatment(IFNα-2b monotherapy,IFNα-2b followed by lamivudine[LAM]or IFNα-2b combined with LAM)and followed up for 144 weeks.A comprehensive analysis was conducted on clinical data,including biochemical items,serum markers of hepatitis B virus(HBV)and immunological indexes,and logistic regression analysis was used to screen the influencing factors related to HBsAg loss.Results:The cumulative loss rates of HBsAg were 79.5%,62.1%and 42.1%at 144 weeks after the start of treatment in the 1–3 years-old group,3–5 years-old group and 5–7 years-old group,respectively(p<0.05).IFNα-2b combined with LAM treatment displayed the highest HBsAg loss rates compared with monotherapy and sequential treatment(p=0.011).Younger baseline age and lower HBsAg levels were independent factors for the prediction of HBsAg loss(p<0.05).The baseline PreS1 and hepatitis B core antibody levels in the HBsAg loss group were lower than those in the HBsAg non-loss group.In addition,the PreS1 level was positively corelated with the level of HBsAg,HBV DNA and liver inflammation.Conclusions:Long-term treatment with IFNαwas effective in achieving HBsAg loss in CHB children aged 1–6 years-old.Age less than 3 years-old and lower HBsAg levels are independent predictors of functional cure in children with CHB. | Jing Pan Haiyan Wang Tiantian Yao Xuejiao Liao Hao Cheng Suthat Liangpunsakul Yan Wang Min Zhang Zheng Zhang | 2022 | Journal of Clinical and Translational Hepatology2022,10,3: | 3 |
| 6 | BYSL contributes to tumor growth by cooperating with the mTORC2 complex in gliomas显示文摘Objective:BYSL,which encodes the Bystin protein in humans,is upregulated in reactive astrocytes following brain damage and/or inflammation.We aimed to determine the role and mechanism of BYSL in glioma cell growth and survival.Methods:BYSL expression in glioma tissues was measured by quantitative real-time PCR,Western blot,and immunohistochemistry.In vitro assays were performed to assess the role of BYSL in cell proliferation and apoptosis.Protein interactions and co-localization were determined by co-immunoprecipitation and double immunofluorescence.The expression and activity of the AKT/m TOR signaling molecules were determined by Western blot analysis,and the role of BYSL in glioma growth was confirmed in an orthotopic xenograft model.Results:The BYSL m RNA and protein levels were elevated in glioma tissues.Silencing BYSL inhibited glioma cell proliferation,impeded cell cycle progression,and induced apoptosis,whereas overexpressing BYSL protein led to the opposite effects.We identified a complex consisting of BYSL,RIOK2,and m TOR,and observed co-localization and positive correlations between BYSL and RIOK2 in glioma cells and tissues.Overexpressing BYSL or RIOK2 increased the expression and activity of AKT/m TOR signaling molecules,whereas downregulation of BYSL or RIOK2 decreased the activity of AKT/m TOR signaling molecules.Silencing BYSL or RIOK2 decreased the growth of the tumors and prolonged the lifespan of the animals in an orthotopic xenograft model.Conclusions:High expression of BYSL in gliomas promoted tumor cell growth and survival both in vitro and in vivo.These effects could be attributed to the association of BYSL with RIOK2 and m TOR,and the subsequent activation of AKT signaling. | Shangfeng Gao Zhuang Sha Junbo Zhou Yihao Wu Yunnong Song Cheng Li Xuejiao Liu Tong Zhang Rutong Yu | 2021 | Cancer Biology & Medicine2021,18,1: | 3 |

| 13 | Role of Ras-related Nuclear Protein/Polypyrimidine Tract Binding Protein in Facilitating the Replication of Hepatitis C Virus显示文摘Background and Aims:Ras-related nuclear(RAN)protein is a small GTP-binding protein that is indispensable for the translocation of RNA and proteins through the nuclear pore complex.Recent studies have indicated that RAN plays an important role in virus infection.However,the role of RAN in hepatitis C virus(HCV)infection is unclear.The objective of this study was to investigate the role and underlying mechanisms of RAN in HCV infection.Methods:Huh7.5.1 cells were infected with the JC1-Luc virus for 24 h and then were incubated with complete medium for an additional 48 h.HCV infection and RAN expression were determined using luciferase assay,quantitative reverse transcription-PCR and western blotting.Small interfering RNA was used to silence RAN.Western blotting and immunofluorescence were used to evaluate the cytoplasmic translocation of polypyrimidine tract-binding(PTB),and coimmunoprecipitation was used to examine the interaction between RAN and PTB.Results:HCV infection significantly induced RAN expression and cytoplasmic redistribution of PTB.Knockdown of RAN dramatically inhibited HCV infection and the cytoplasmic accumulation of PTB.Colocalization of RAN and PTB was determined by immunofluorescence,and a direct interaction of RAN with PTB was demonstrated by coimmunoprecipitation.Conclusions:PTB in the host cytoplasm is directly associated with HCV replication.These findings demonstrate that the involvement of RAN in HCV infection is mediated by influencing the cytoplasmic translocation of PTB. | Jihua Xue Jun Cheng Xuejiao Ma Yixian Shi Huafa Yin Yufeng Gao Jiabin Li | 2021 | Journal of Clinical and Translational Hepatology2021,9,4: | 0 |

| 16 | Effect of Dandelion(Taraxacum mongolicum Hand.-Mazz.)Intercropping with Different Plant Spacing on Blight and Growth of Pepper(Capsicum annuum L.)显示文摘Intercropping of crops that can secrete bacteriostatic active substances can not only inhibit the occurrence of disease but also have an important effect on plant growth.However,the effects of dandelion intercropping on pepper blight control and pepper growth remain unclear.In this study,the control effect of dandelion on pepper blight was studied by inoculating the pepper leaves with Phytophthora infestans,and it also discusses the correlation of the occurrence of pepper epidemic disease with the pepper canopy environment,soil environment,pepper photo-synthesis,and yield index.The results showed that best plant distance for dandelion intercropping was 20 cm(P20),and the control effect reached 43.31%.As compared to the CK,SOD enzyme,POD enzyme,and PAL enzyme were significantly up-regulated during the growth of pepper;chlorophyll content in pepper leaves was significantly increased;photosynthetic characteristics were significantly increased;stem diameter and yield of crop pepper were effectively improved;and the quality of the pepper product was better,but intercropping dandelion resulted in a significant decrease of nutrients in the soil environment of pepper,so a reasonable intercropping distance was needed.The correlation analysis shows that the incidence of pepper blight(A)was significantly positively correlated with soil temperature(Q),intercellular carbon dioxide(L),and canopy air temperature(O).The incidence of capsicum blight(A)was significantly negatively correlated with chlorophyll content(F),net photo-synthetic rate(K),stomatal conductance(M),ww rate(N),soil sucrase activity(W),vitamin C(AB),and leaf PAL enzyme(J).Finally,it was deduced that intercropping dandelion could effectively control the occurrence of pepper blight while also demonstrating a complex interaction with the pepper growing environment. | Peixin Li Hanbing Liu Yingtong Chen Xin Zhang Ning Cao Ying Sun Meimei Jia Mengran Wu Xuejiao Tong Xinmei Jiang Xihong Yu Yao Cheng | 2023 | Phyton-International Journal of Experimental Botany2023,92,8: | 0 |
