Targeted Mutagenesis of NAC Transcription Factor Gene,OsN AC041,Leading to Salt Sensitivity in Rice显示文摘Salinity is a major abiotic stress factor that seriously affects plant growth. Many genes are involved in the response to salt stress with various metabolism pathways. A number of plant transcription factor family genes have been found to be involved in the salt stress response, and NAM, ATAF and CUC(NAC) transcription factors are thought to act as active regulators during abiotic stress, especially salt stress. In this study, we detected a rice NAC transcription factor coding gene, OsNAC041, and confirmed that it influenced the germination of seeds under salt stress and salt tolerance of plants. OsNAC041 was primarily expressed in the leaves and located in the nucleus. Furthermore, the CRISPR/Cas9 method was used to obtain a targeted osnac041 mutant, of which the plant height was higher than that of the wild-type, showing increased salt sensitivity. Moreover, RNA-seq analysis revealed a number of differentially expressed genes(DEGs) involved in several important signaling pathways in the osnac041 mutant. Subsequently, Kyoto Encyclopedia of Genes and Genomes annotation also revealed differential expression of DEGs associated with mitogen-activated protein kinase signaling, peroxisome, eukaryotictype ABC transporters, photosynthesis and plant hormones, which are involved in stress-related signaling pathways. Overall, our study suggested that OsNAC041 was involved in the salt stress response in rice. These findings not only provide empirical evidence of OsNAC041 function, but also provide new insight into its potential application in rice resistance breeding. | WANG Bo ZHONG Zhaohui ZHANG Huanhuan WANG Xia LIU Binglin YANG Lijia HAN Xiangyan YU Deshui ZHENG Xuelian WANG Chunguo SONG Wenqin CHEN Chengbin ZHANG Yong | 2019 | Rice science2019,26,2: | 8
The relationship between viscosity and glass forming ability of Al-(Ni)-Yb alloy systems显示文摘The dynamic viscosity of Al-Yb and Al-Ni-Yb superheated melts was measured using a torsional oscillation viscometer. The results show that the temperature dependence of viscosity fits the Arrhenius law well and the fitting factors are calculated. The amorphous ribbons of these alloys were produced by the melt spinning technique and the thermal properties were characterized by using a differential scanning calorimetry (DSC). E (the activation energy for viscous flow), which reflects the change rate of viscosity, has a good negative relation with the GFA in both Al-Yb and Al-Ni-Yb systems. However, there is no direct relation between liquidus viscosity (ηL) and GFA. The superheated fragility M can predict GFA in Al-Yb or Al-Ni-Yb alloy system. | JIA Ran, BIAN XiuFang, Lü XiaoQian, SONG KaiKai & LI XueLian Key Laboratory for Liquid-Solid Structural Evolution and Processing of Materials (Ministry of Education), Shandong University, Jinan 250061, China | 2010 | Science China(Physics,Mechanics & Astronomy)2010,53,3: | 6
Kruppel-like factor 10 protects against acute viral myocarditis by negatively regulating cardiac MCP-1 expression显示文摘Viral myocarditis(VMC)is a cardiac disease associated with myocardial inflammation and injury induced by virus infection.Cardiomyocytes have recently been regarded as key players in eliciting and modulating inflammation within the myocardium.Kruppel-like factor 10(KLF10)is a crucial regulator of various pathological processes and plays different roles in a variety of diseases. However, its role in VMC induced by coxsackievirus B3(CVB3)infection remains unknown. In this study,we report that cardiac KLF10 confers enhanced protection against viral myocarditis. We found that KLF10 expression was downregulated upon CVB3 infection.KLF10 deficiency enhanced cardiac viral replication and aggravated VMC progress.Bone marrow chimera experiments indicated that KLF10 expression in nonhematopoietic cells was involved in the pathogenesis of VMC.We further identified MCP-1 as a novel target of KLF10 in cardiomyocytes, and KLF10 cooperated with histone deacetylase 1(HDAC1)to negatively regulate MCP-1 expression by binding its promoter, leading to activation of MCP-1 transcription and recruitment of Ly6C^(high) monocytes/macrophages into the myocardium. This novel mechanism of MCP-1 regulation by KLF10 might provide new insights into the pathogenesis of VMC and a potential therapeutic target for VMC. | Jie Yang Hongkai Zhang Xuelian Wang Jing Guo Lin Wei Yahui Song Yuan Luo YinXia Zhao Malayannan Subramaniam Thomas C.Spelsberg Lie Wang Wei Xu Min Li | 2021 | Cellular & Molecular Immunology2021,18,9: | 3
A durable 4-1BB-based CD19 CAR-T cell for treatment of relapsed or refractory non-Hodgkin lymphoma显示文摘Objective:Previous studies reported that 4-1BB-based CD19 chimeric antigen receptor(CAR)-T cells were more beneficial for the clinical outcomes than CD28-based CAR-T cells, especially the lower incidence rate of severe adverse events. However, the median progression-free survival(mPFS)of 4-1BB-based product Kymriah was shorter than that of CD28-based Yescarta(2.9 months vs.5.9 months),suggesting that Kymriah was limited in the long-term efficacy. Thus,a safe and durable 4-1BB-based CD19 CAR-T needs to be developed. Methods:We designed a CD19-targeted CAR-T(named as IM19)which consisted of an FMC63 scFv,4-1BB and CD3ζintracellular domain and was manufactured into a memory T-enriched formulation.A phase I/II clinical trial was launched to evaluate the clinical outcomes of IM19 in relapsed or refractory(r/r)B cell non-Hodgkin lymphoma(B-NHL).Dose-escalation investigation(at a dose of 5×10^(5)/kg,1×10^(6)/kg and 3×106/kg)was performed in 22 r/r B-NHL patients. All patients received a single infusion of IM19 after 3-day conditional regimen. Results:At month 3,the overall response rate(ORR)was 59.1%,the complete response rate(CRR)was 50.0%.The mPFS was 6 months and the 1-year overall survival rate was 77.8%.Cytokine release syndrome(CRS)occurred in 13 patients(59.1%),with 54.5%of grade 1−2 CRS.Only one patient(4.5%)experienced grade 3 CRS and grade 3 neurotoxicity. Conclusions:These results demonstrated the safety and durable efficacy of a 4-1BB-based CD19 CAR-T,IM19,which is promising for further development and clinical investigation. | Zhitao Ying Ting He Shanzhao Jin Xiaopei Wang Wen Zheng Ningjing Lin Meifeng Tu Yan Xie Lingyan Ping Weiping Liu Lijuan Deng Yanping Ding Xuelian Hu Bing Bu Xin’an Lu Yuqin Song Jun Zhu | 2022 | Chinese Journal of Cancer Research2022,34,1: | 2

A palaeoearthquake event and its age revealed by the travertine layer along the Litang fault in the southeastern margin of the Qinghai-Tibetan plateau显示文摘The Litang fault(LTF),located in the southeast of the Qinghai-Tibetan Plateau,is known for its high level of present-day seismicity, whereas its Pleistocene activity has been scarcely documented. This study focused on a tract of banded travertine deposits precipitated from thermal waters along the NW–SE-trending LTF trace. The role of travertine deposits in recording neotectonic activity has been studied by identifying their internal structure.Typical soft-sediment deformation structures observed within the banded travertines include micro folds, liquefied breccia, and liquefied diapirs. These deformed structures, which are restricted to a single unit separated unconformably by undeformed layers, can be traced for tens of meters, indicating that they were formed by seismic shaking triggered by LTF activity. The deformation of the banded travertine layers is attributed to the combined effects of seismic shaking, liquefaction, and fluidization, and it can be related to a paleo earthquake event with a magnitude of MS>5.The U-series ages obtained from the banded travertine deposits perturbed by the earthquakes are in the range of 130.59–112.94 ka,indicating an important fault-assisted neotectonic activity that occurred during the Middle–Late Pleistocene.Analysis of such structures,in combination with the use of U-series dating methods, can yield a reliable timing of neotectonic activity and provide new evidence for under-standing the seismotectonic setting of the Litang area. | Yao Yang Mingjian Liang Chao Ma Jun Li Hualiang Shen Fang Du Song Luo Shao Liu Xuelian Rui | 2023 | Earthquake Research Advances2023,3,3: | 0
Implementing the free HPV vaccination for adolescent girls aged below 14 in Shenzhen,Guangdong Province of China:experience, challenges, and lessons显示文摘Cervical cancer is a major public health concern in China, accounting for almost one-fifth of the global incidence and mortality. The recently prequalified domestic bivalent human papillomavirus(HPV)vaccine offers a practical and feasible preventive measure. In response to the global call for action, the National Health Commission issued an Action Plan to eliminate cervical cancer by 2030,with promotion of the HPV vaccination for school-aged girls as a critical step.Despite this, implementation of the vaccination has been patchy, with very low coverage among eligible girls. To address this, from December 2021 to December 2022,a demonstration project was launched in Shenzhen,Guangdong Province,to promote the inclusion of HPV vaccine in local immunisation programme and to address existing barriers to implementation.Using multiple sources of data, this article presents a case study of the demonstration project, analysing its impact on rolling out HPV vaccination among eligible girls and identifying any challenges encountered during implementation. The demonstration project has shown promising results in increasing the HPV vaccination rate, promoting public awareness and acceptance of the domestic HPV vaccine, and establishing a model for quickly scaling up the vaccination at the municipal level. The success of the project can be attributed to several factors, including strong governmental commitment, sufficient funding,multi-sectoral collaboration, ensured vaccine accessibility and affordability, improved vaccination services, and effective health education and communication strategies.Lessons learned from Shenzhen can provide valuable insights for future advocacy and implementation of the vaccination in other areas of China, but challenges must be addressed to achieve universal coverage. These include addressing vaccine hesitancy, expanding the programme to cover a broader age range, and ensuring consistent quality of vaccination services in primary care facilities.Overcoming these challenges will require innovative strategies,public-private partnerships, and sustained funding and resources.Future research should focus on evaluating the long-term effectiveness of the vaccination programme and identifying contextual factors that may impact its implementation in different settings.Overall, the effective control of cervical cancer in China will rely on the“political will”to ensure the incorporation of preventive interventions into policies and universal programme coverage. | Dadong Wu Peiyi Liu Danhong Song He Wang Siqi Chen Wanyi Tang Xuelian Zhao Fanghui Zhao Yueyun Wang | 2023 | Infectious Diseases of Poverty2023,12,5: | 0

Expert consensus on prevention and cardiopulmonary resuscitation for cardiac arrest in COVID-19显示文摘Background:Cardiopulmonary resuscitation(CPR)strategies in COVID-19 patients differ from those in patients suffering from cardiogenic cardiac arrest.During CPR,both healthcare and non-healthcare workers who provide resuscitation are at risk of infection. The Working Group for Expert Consensus on Prevention and Cardiopulmonary Resuscitation for Cardiac Arrest in COVID-19 has developed this Chinese Expert Consensus to guide clinical practice of CPR in COVID-19 patients.Main recommendations:1)A medical team should be assigned to evaluate severe and critical COVID-19 for early monitoring of cardiac-arrest warning signs.2)Psychological counseling and treatment are highly recommended, since sympathetic and vagal abnormalities induced by psychological stress from the COVID-19 pandemic can induce cardiac arrest.3)Healthcare workers should wear personal protective equipment(PPE).4)Mouth-to-mouth ventilation should be avoided on patients suspected of having or diagnosed with COVID-19.5)Hands-only chest compression and mechanical chest compression are recommended.6)Tracheal-intubation procedures should be optimized and tracheal-intubation strategies should be implemented early.7)CPR should be provided for 20-30 min.8)Various factors should be taken into consideration such as the interests of patients and family members, ethics, transmission risks, and laws and regulations governing infectious disease control.Changes in management:The following changes or modifications to CPR strategy in COVID-19 patients are proposed:1)Healthcare workers should wear PPE.2)Hands-only chest compression and mechanical chest compression can be implemented to reduce or avoid the spread of viruses by aerosols.3)Both the benefits to patients and the risk of infection should be considered.4)Hhealthcare workers should be fully aware of and trained in CPR strategies and procedures specifically for patients with COVID-19. | Wei Song Jie Wei Xiangdong Jian Deren Wang Yanhong Ouyang Yuanshui Liu Xianjin Du Ying Chen Yingqi Zhang Heping Xu Shuming Xianyu Qiong Ning Xiang Li Xiaotong Han Feng Zhan Tao Yu Wenteng Chen Jun Zhang Wenwei Cai Sheng’ang Zhou Shengyang Yi Yu Cao Xiaobei Chen Shunjiang Xu Zong’an Liang Duohu Wu Fen Ai Zhong Wang Qingyi Meng Yuhong Mi Sisen Zhang Rongjia Yang Shouchun Yan Wenbin Han Yong Lin Chuanyun Qian Wenwu Zhang Yan Xiong Jun Lv Baochi Liu Yan Cao Xiaojun He Xuelian Sun Yufang Cao Tian’en Zhou | 2021 | Asian Pacific Journal of Tropical Medicine2021,14,6: | 0
Effects of grade, academic performance, and sex on spatial working memory and attention in primary school children:a cross-sectional observational study显示文摘Objective:Only a few studies have investigated the development trends in spatial working memory and attention among a large sample of primary school students over a wide range of ages, while the efficiency of learning and memory processes is fundamental to academic performance, particularly for children, who are in a key developmental stage when their life opportunities can be altered. We aimed to explore how the spatial working memory and attention of primary school children are affected by grade, academic performance, and sex. Methods:In this cross-sectional observational study,students(144 boys,139 girls,6–13 years old)were recruited from Experimental Primary School Affiliated to Shanghai Jiao Tong University in Shanghai,China in 2018.The study protocol was reviewed and approved by the Bio-Ethics Board of the Bio-X Institutes,Shanghai Jiao Tong University.Based on a simple Spatial Working Memory and Attention Test on Paired Symbols test,we evaluate 283 participants’working memory and attention ability. Results:Attention and working memory performance were enhanced as a positive function of grade in primary school children, and students who showed better academic achievement also performed better on the working memory task. However, attention and working memory performance were not affected by sex. Conclusion:Attention and working memory performance of primary school students develop with grade and corresponds to Better academic performance.Attention and working memory ability do not differ significantly between boys and girls. | Nengpeng Zhan Xuelian Fan Fengtao Shen Lulu Song Chenhuan Zhou Jiayi Xiao Xun Wu Leonardo Jiahao Li Jiayao Xi Sophia Jiayi Li Suhua Zeng Can Li Lihui Wang Weidong Li | 2022 | Journal of Bio-X Research2022,5,2: | 0
Thermal History of the Naruo Porphyry Deposit in the Duolong Ore District,Western Tibet:Evidence from U-Pb,^(40)Ar/^(39)Ar and(U-Th)/He Thermochronology显示文摘The Naruo porphyry copper deposit containing more than 2 Mt of copper is located in the Duolong ore district in the west of the Bangongco–Nujiang belt in central Tibet.New zircon U-Pb,biotite^(40)Ar/^(39)Ar,zircon(U-Th)/He ages, published age data together with thermal modeling were presented in this paper to investigate the thermal history of Naruo deposit.Thermal modeling reveals a prolonged magmatic-hydrothermal evolution firstly cooling from~700℃to~350℃at 120 Ma,then cooling to 230℃at 106 Ma and maintaining at 200℃from 106 to 90 Ma which is attributed to multiple magmatic events and thermal effect of strike-slip fault.Affected by thrust nappe structure, the sample was consistent with 120℃from 70 to 63 Ma.The Naruo deposit started to experience exhumation at a rate of~0.07 km/Myr since 60 Ma which is related to India-Asia collision. The prolonged magmatic-hydrothermal evolution process might have important influence on the Naruo deposit. The ore-related intrusions preserved in the foot walls of strike-slip fault and thrust nappe structure are the objects of future exploration in the Duolong ore district. | YANG Huanhuan FANG Xiang SONG Yang LIN Bin LI Yanbo WANG Qin LIU Zhibo HE Long ZHANG Qi FU Xuelian | 2022 | Acta Geologica Sinica(English Edition)2022,96,6: | 0
The Systemic Immune Inflammatory Index Predicts No-Reflow Phenomenon after Primary Percutaneous Coronary Intervention in Older Patients with STEMI显示文摘Purpose:Coronary no-reflow phenomenon(NRP),a common adverse complication in patients with ST-segment eleva-tion myocardial infarction(STEMI)treated by percutaneous coronary intervention(PCI),is associated with poor patient prognosis. In this study, the correlation between the systemic immune-inflammation index(SII)and NRP in older patients with STEMI was studied,to provide a basis for early identification of high-risk patients and improve their prognosis.Materials and methods:Between January 2017 and June 2020,578 older patients with acute STEMI admitted to the Department of Cardiology of Hebei General Hospital for direct PCI treatment were selected for this retrospective study.Patients were divided into an NRP group and normal-flow group according to whether NRP occurred during the operation.Clinical data and the examination indexes of the two groups were collected.Logistic regression was used to analyze the independent predictors of NRP,and the receiver operating characteristic curve was used to further analyze the ability of SII to predict NRP in older patients with STEMI.Results:Multivariate logistic analysis indicated that hypertension(OR=2.048,95%CI:1.252–3.352,P=0.004),lymphocyte count(OR=0.571,95%CI:0.368–0.885,P=0.012),platelet count(OR=1.009,95%CI:1.005–1.013,P<0.001),hemoglobin(OR=1.015,95%CI:1.003–1.028,P=0.018),multivessel disease(OR=2.237,95%CI:1.407–3.558,P=0.001),and SII≥1814(OR=3.799,95%CI:2.190–6.593,P<0.001)were independent predictors of NRP after primary PCI in older patients with STEMI.Receiver operating characteristic curve analysis demonstrated that SII had a high predictive value for NRP(AUC=0.738;95%CI:0.686–0.790),with the best cut-off value of 1814,a sensitivity of 52.85%and a specificity of 85.71%.Conclusion:For older patients with STEMI undergoing primary PCI,SII is a valid predictor of NRP. | Jiaqi Wang Feifei Zhang Man Gao Yudan Wang Xuelian Song Yingxiao Li Yi Dang Xiaoyong Qi | 2023 | Cardiovascular Innovations and Applications2023,7,1: | 0
