118beta-glycyrrhetinic acid induces ROS-mediated apoptosis to ameliorate hepatic fibrosis by targeting PRDX1/2 in activated HSCs显示文摘Hepatic stellate cells(HSCs)are essential drivers of fibrogenesis.Inducing activated-HSC apoptosis is a promising strategy for treating hepatic fibrosis.18beta-glycyrrhetinic acid(18b-GA)is a natural compound that exists widely in herbal medicines,such as Glycyrrhiza uralensis Fisch,which is used for treating multiple liver diseases,especially in Asia.In the present study,we demonstrated that 18b-GA decreased hepatic fibrosis by inducing the apoptosis in activated HSCs.18b-GA inhibited the expression of a-smooth muscle actin and collagen type Ⅰ alpha-1.Using a chemoproteomic approach derived from activity-based protein profiling,together with cellular thermal shift assay and surface plasmon resonance,we found that 18b-GA covalently targeted peroxiredoxin 1(PRDX1)and peroxiredoxin 2(PRDX2)proteins via binding to active cysteine residues and thereby inhibited their enzymatic activities.18b-GA induced the elevation of reactive oxygen species(ROS),resulting in the apoptosis of activated HSCs.PRDX1 knockdown also led to ROS-mediated apoptosis in activated HSCs.Collectively,our findings revealed the target proteins and molecular mechanisms of 18b-GA in ameliorating hepatic fibrosis,highlighting the future development of 18b-GA as a novel therapeutic drug for hepatic fibrosis.Qian Zhang Piao Luo Liuhai Zheng Jiayun Chen Junzhe Zhang Huan Tang Dandan Liu Xueling He Qiaoli Shi Liwei Gu Jiahao Li Qiuyan Guo Chuanbin Yang Yin Kwan Wong Fei Xia Jigang Wang 2022Journal of Pharmaceutical Analysis2022,12,4:3
2Design and analysis of a class of redundant collaborative manipulators with 2D large rotational angles显示文摘The parallel spindle heads with high rotational capability are demanded in the area of multi-axis machine tools and 3D printers.This paper focuses on designing a class of 2R1T(R:Rotation;T:Translation)parallel spindle heads and the corresponding collaborative 5-axis manipulators with 2-dimension(2D)large rotational angles.In order to construct 2D rotational degrees of freedom(DOFs),a platform with 2D revolute joints is proposed first.Based on the constraint screw theory,the feasible limbs that can be connected in the platform are synthesized.In order to provide constant rotational axis for the platform,a class of redundant limbs are designed.A class of redundant 2R1T parallel spindle heads is obtained by connecting the redundant limbs with the platform and the redundant characteristics are verified by the modified Grübler-Kutzbach criterion.The corresponding 5-axis collaborative manipulators are presented by constructing a 2-DOF series translational bottom moving platform.The inverse kinematics and the orientation workspace as well as the decoupling characteristics of this type of 2R1T parallel spindle heads are analyzed.The results show that these manipulators have large 2D rotational angles than the traditional A3/Z3 heads and can be potentially used in the application of multi-axis machine tools and the 3D printers.Xiaodong JIN Yuefa FANG Dan ZHANG Xueling LUO 2020Frontiers of Mechanical Engineering2020,15,1:2

4Single-cell transcriptome analysis reveals the regulatory effects of artesunate on splenic immune cells in polymicrobial sepsis显示文摘Sepsis is characterized by a severe and life-threatening host immune response to polymicrobial infection accompanied by organ dysfunction.Studies on the therapeutic effect and mechanism of immunomodulatory drugs on the sepsis-induced hyperinflammatory or immunosuppression states of various immune cells remain limited.This study aimed to investigate the protective effects and underlying mechanism of artesunate(ART)on the splenic microenvironment of cecal ligation and puncture-induced sepsis model mice using single-cell RNA sequencing(scRNA-seq)and experimental validations.The scRNA-seq analysis revealed that ART inhibited the activation of pro-inflammatory macrophages recruited during sepsis.ART could restore neutrophils’chemotaxis and immune function in the septic spleen.It inhibited the activation of T regulatory cells but promoted the cytotoxic function of natural killer cells during sepsis.ART also promoted the differentiation and activity of splenic B cells in mice with sepsis.These results indicated that ART could alleviate the inflammatory and/or immunosuppressive states of various immune cells involved in sepsis to balance the immune homeostasis within the host.Overall,this study provided a comprehensive investigation of the regulatory effect of ART on the splenic microenvironment in sepsis,thus contributing to the application of ART as adjunctive therapy for the clinical treatment of sepsis.Jiayun Chen Xueling He Yunmeng Bai Jing Liu Yin Kwan Wong Lulin Xie Qian Zhang Piao Luo Peng Gao Liwei Gu Qiuyan Guo Guangqing Cheng Chen Wang Jigang Wang 2023Journal of Pharmaceutical Analysis2023,13,7:1

8kdm4aa is required for reproduction and development of zebrafish显示文摘Lysine-specific demethylase 4A(KDM4A)catalyzes demethylation of histone lysine residues,which regulates chromatin state and transcription.In drosophila and mice,KDM4A plays an important role in multiple biological processes including development,aging,metabolism,and immunity,however the functions of KDM4A in fish are still unclear.There are two copies of the kdm4a gene in zebrafish,namely kdm4aa and kdm4ab,kdm4aa was edited using CRISPR/Cas9 technology in the present study,then homozygous kdm4aa mutants(kdm4aa^(-/-)��)were obtained,and loss of kdm4aa was confirmed by sequencing and increased H3K9me3.Whole-mount in situ hybridization showed that kdm4aa is widely expressed during the embryonic development of zebrafish.Compared with WT zebrafish,kdm4aa^(-/-)��zebrafish showed no significant difference in gamete formation and fertilization,but the survival rate of kdm4aa^(-/-)��embryos dramatically reduced to 21%at 26 hpf.Further observation showed that about 80%of survived kdm4aa^(-/-)��zebrafish experienced disruption in stripe formation,and 10%of survived kdm4aa^(-/-)��zebrafish underwent vertebral malformation.Alizarin red S staining demonstrated the abnormal spinal development in kdm4aa^(-/-)��zebrafish.These results indicated that kdm4aa is required for normal embryonic development of zebrafish,loss of kdm4aa function leads to decreased survival during the early stages of zebrafish development and morphological variation in adult zebrafish.Lijie Han Juntao Luo Songya Qu Xueling Shi Junfang Zhang Bingshe Han 2023Aquaculture and Fisheries2023,8,6:0
9Characteristics and Natural Gas Origin of Middle-Late Triassic Marine Source Rocks of the Western Sichuan Depression, SW China显示文摘A scientific exploration well(CK1) was drilled to expand the oil/gas production in the western Sichuan depression, SW, China. Seventy-three core samples and four natural gas samples from the Middle–Late Triassic strata were analyzed to determine the paleo-depositional setting and the abundance of organic matter(OM) and to evaluate the hydrocarbon-generation process and potential. This information was then used to identify the origin of the natural gas. The OM is characterized by medium n-alkanes(n C15–n C19), low pristane/phytane and terrigenous aquatic ratios(TAR), a carbon preference index(CPI) of ~1, regular steranes with C29 > C27 > C28, gammacerane/C30 hopane ratios of 0.15–0.32, and δDorg of-132‰ to-58‰, suggesting a marine algal/phytoplankton source with terrestrial input deposited in a reducing–transitional saline/marine sedimentary environment. Based on the TOC, HI index, and chloroform bitumen 'A' the algalrich dolomites of the Leikoupo Formation are fair–good source rocks;the grey limestones of the Maantang Formation are fair source rocks;and the shales of the Xiaotangzi Formation are moderately good source rocks. In addition, maceral and carbon isotopes indicate that the kerogen of the Leikoupo and Maantang formations is type Ⅱ and that of the Xiaotangzi Formation is type Ⅱ–Ⅲ. The maturity parameters and the hopane and sterane isomerization suggest that the OM was advanced mature and produced wet–dry gases. One-dimensional modeling of the thermal-burial history suggests that hydrocarbon-generation occurred at 220–60 Ma. The gas components and C–H–He–Ar–Ne isotopes indicate that the oilassociated gases were generated in the Leikoupo and Maantang formations, and then, they mixed with gases from the Xiaotangzi Formation, which were probably contributed by the underlying Permian marine source rocks. 12Aligned 3D porous polyurethane scaffolds for biological anisotropic tissue regeneration显示文摘A green fabrication process(organic solvent-free)of artificial scaffolds is required in tissue engineering field.In this work,a series of aligned three-dimensional(3D)scaffolds are made from biodegradable waterborne polyurethane(PU)emulsion via directional freeze–drying method to ensure no organic byproducts.After optimizing the concentration of polymer in the emulsion and investigating different freezing temperatures,an aligned PUs scaffold(PU14)generated from 14wt%polymer content and processed at196C was selected based on the desired oriented porous structure(pore size of 32.569.3 lm,porosity of 92%)and balanced mechanical properties both in the horizontal direction(strength of 41.3 kPa,modulus of 72.3 kPa)and in the vertical direction(strength of 45.5 kPa,modulus of 139.3 kPa).The response of L929 cells and the regeneration of muscle tissue demonstrated that such pure material-based aligned 3D scaffold can facilitate the development of orientated cells and anisotropic tissue regeneration both in vitro and in vivo.Thus,these pure material-based scaffolds with ordered architecture have great potentials in tissue engineering for biological anisotropic tissue regeneration,such as muscle,nerve,spinal cord and so on.Weiwei Lin Wanling Lan Yingke Wu Daiguo Zhao Yanchao Wang Xueling He Jiehua Li Zhen Li Feng Luo Hong Tan Qiang Fu 2020Regenerative Biomaterials2020,7,1:0
13Enhanced cyanogen chloride formation after UV/PS and UV/H_(2)O_(2) pre-oxidation and chlorination in natural river water显示文摘Ultraviolet/persulfate (UV/PS) and Ultraviolet/hydrogen peroxide (UV/H_(2)O_(2)) have attracted much attention in recent years as advanced oxidation processes for water treatment.However,it is not all clear how these two methods affect the formation of cyanogen chloride(CNCl) in the subsequent water chlorination process.In this study,it was found that both UV/H_(2)O_(2)and UV/PS pre-oxidation promoted the formation of CNCl in six actual water samples collected from urban rivers.Glycine,uric acid,arginine and histidine were investigated as the model compounds to explore the effects of different methods on the production of CNCl.The results showed that compared with chlorination alone,pre-oxidation by UV/H_(2)O_(2)and UV/PS can reduce the production of CNCl for glycine and uric acid by up to 95%during post-chlorination process.However,they can greatly promote the formation of CNCl for arginine and histidine by up to 120-fold.In a more detailed investigation,pre-oxidation of histidine formed highly reactive intermediates to chlorine,leading to increased CNCl formation and chlorine consumption.The results showed that the precursors of CNCl was altered after pre-oxidation,and need to be re-evaluated.Jie Xu Yang Guo Qian Yang Xueling Bai Runhua Lu Menghui Liu Zichen Kuang Luo Zhang Jing Li 2023Journal of Environmental Sciences2023,,4:0
