| 1 | The interaction of CpEBF1 with CpMADSs is involved in cell wall degradation during papaya fruit ripening显示文摘Ethylene plays a pivotal role in climacteric fruit ripening;whereas 1-MCP,a non-toxic antagonist of ethylene,prevents ethylene-dependent responses and fruit ripening.In this study,a short-term treatment(1 h)with 400 nL L^(−1)1-MCP delayed the ripening of harvested papaya.However,long-term application of 1-MCP(400 nL L^(−1),16 h)resulted in abnormal fruit ripening,with the fruits exhibiting normal yellowing without softening,significantly higher cellulose and lignin contents,and intact cell walls(CW).Furthermore,we found that long-term treatment with 1-MCP significantly inhibited the expression of CpEBF1,an EIN3-binding F-box-1 gene.A protein interaction analysis using yeast two-hybrid,BiFC and GST pull-down assays showed that CpEBF1 interacts with the CpMADS1/3 and CpEIL1 proteins.The interaction of CpEBF1 with CpMADS1/3 further activated the activities of CW-degradation gene promoters.Subcellular localization showed that these proteins were localized in the nucleus.Additionally,the expression levels of CpMADS1/3,CpEIL1,and several CW-degradation-related genes were significantly downregulated by long-term 1-MCP treatment.Therefore,we propose that the inhibited expression of CpEBF1 and CpMADS1/3 resulted in the repressed activation of CW-degradation-related genes via their interaction,thereby resulting in fruit softening disorders. | Xiaochun Ding Xiaoyang Zhu Lanlan Ye Shuangling Xiao Zhenxian Wu Weixin Chen Xueping Li | 2019 | Horticulture Research2019,6,1: | 7 |
| 2 | Ability to detect weak effective seismic signals by utilizing chaotic vibrator system显示文摘Taking the advantage of CVS (Chaotic Vibrator System) sensitivity of large-scale periodic phase-state response to quasi-periodic or periodic signals,a series of numerical experiments were made to understand the ability of CVS to detect weak effective seismic signals in the common-shot seismic record distorted by strong stochastic noise. The re-sults demonstrate that the large-scale periodic phase-states of CVS are correlated with the signal composition of the quasi-periodic wavelet sequence constructing from horizontal moveout of seismic events,noise strength and the noise distortion de-gree to signal. For the same kind of events,the higher the noise distortion degree is,the lower the detectable SNR can be reached by CVS. For seismic data with the same noise distortion degree,the closer the scanning seismic velocity (the trial moveout ve-locity) approaches to the accurate velocity,the higher the detectable SNR can be reached by CVS. | 3 | Rapid Detection of Wheat Blast Pathogen Magnaporthe oryzae Triticum Pathotype Using Genome-Specific Primers and Cas12a-mediated Technology显示文摘Wheat blast,caused by the fungus Magnaporthe oryzae Triticum(MoT)pathotype,is a devastating disease persistent in South America and Bangladesh.Since MoT generally fails to cause visual symptoms in wheat until the heading stage when the infection would have advanced,disease control by fungicide application solely based on the detection of visual symptoms is ineffective.To develop an accurate and sensitive method to detect MoT at the seedling and vegetative stages for disease control,we sequenced the genomes of two MoT isolates from Brazil and identified two DNA fragments,MoT-6098 and MoT-6099,that are present in the MoT genome but not in the genome of the rice-infecting Magnaporthe oryzae Oryzae(MoO)pathotype.Using polymerase chain reaction(PCR),we confirmed the specificity of the two markers in 53 MoT and MoO isolates from South America and Bangladesh.To test the efficiency of the two markers,we first established a loop-mediated isothermal amplification(LAMP)method to detect MoT at isothermal conditions,without the use of a PCR machine.Following this,we used the Cas12a protein and guide RNAs(gRNAs)to target the MoT-6098 and MoT-6099 sequences.The activated Cas12a showed indiscriminate single-stranded deoxyribonuclease(ssDNase)activity.We then combined targetdependent Cas12a ssDNase activation with recombinase polymerase amplification(RPA)and nucleic acid lateral flow immunoassay(NALFIA)to develop a method that accurately,sensitively,and cost-effectively detects MoT-specific DNA sequences in infected wheat plants.This novel technique can be easily adapted for the rapid detection of wheat blast and other important plant diseases in the field. | Houxiang Kang Ye Peng Kangyu Hua Yufei Deng Maria Bellizzi Dipali Rani Gupta Nur Uddin Mahmud Alfredo S.Urashima Sanjoy Kumar Paul Gary Peterson Yilin Zhou Xueping Zhou Md Tofazzal Islam Guo-Liang Wang | 2021 | Engineering2021,7,9: | 4 |

| 5 | Pathway-pathway network-based study of the therapeutic mechanisms by which salvianolic acid B regulates cardiovascular diseases显示文摘Investigation of the potential therapeutic mechanisms of drug candidates is an essential step in the process of new drug discovery.With the rapid development of systems biology,recent network analyses of proteins,drugs,and diseases have enabled great progress in delineating the molecule mechanisms of drug candidates.However,most analyses perform a direct association between gene/protein and disease levels without considering the intermediate biological pathways regulated by the drugs.Given that a protein performs its biological roles through pathways,we propose using a novel pathway-pathway network analysis to investigate the potential therapeutic functions of the drug candidates.Many studies have demonstrated that salvianolic acid B(SalB) of Salvia miltiorrhiza is an effective therapy for cardiovascular diseases(CVD).Using molecular docking methods to identify direct interacting targets of Sal B,we collected all Sal B-regulated proteins with supporting experimental evidence in PubMed abstracts.FDA-approved CVD drugs and their corresponding targets were also collected.From a traditional drug-protein network analysis,we found that Sal B could affect ACE and REN of the renin-angiotensin-aldosterone system to relax vessels and alleviate hypertension.Subsequent pathway-pathway network analysis was attempted to study the mechanisms of Sal B in treating CVD,and demonstrated that Sal B regulates immunity/inflammation,apoptosis,ion transport and basic metabolism processes in the treatment of CVD.Regulating the immune/inflammation process may be the major mechanism of Sal B.We believe that pathwaypathway network analysis is a novel method for studying the therapeutic mechanisms of herbal ingredients. | YE Li HE Yuan YE Hao LIU XuePing YANG LinLin CAO ZhiWei TANG KaiLin | 2012 | Chinese Science Bulletin2012,57,14: | 1 |

| 7 | Variation Laws and Influencing Factors of pH in Fishery Waters in Quzhou City显示文摘[Objectives] This study was conducted to strengthen the understanding of the variation laws of pH in fishery waters. [Methods]Based on the investigation data of pH, dissolved oxygen(Do), water temperature(T) and chlorophyll a(Chl. a) of the fishery waters in Quzhou City from 2017 to 2020, the seasonal variation characteristics and influencing factors of pH in the fishery waters were analyzed and discussed. [Results] The pH of the fishery waters in Quzhou City was in the range of 6.40-11.3, and was characterized by the highest seasonal variation in spring and the lowest in winter. The correlation analysis showed that water temperature and pH were significantly positively correlated in spring and summer(P<0.05), and the photosynthesis of phytoplankton was the main reason that affected the pH of water bodies in spring and summer. There was a negative correlation between water temperature and pH in winter, and the pH in the natural waters had a significant negative correlation with water temperature(P<0.05). The balance of carbonate system in the water bodies was the main reason that affected the pH of water in winter. The level of water eutrophication(Chl. a) affected the pH of fishery waters to a certain extent. When Chl. a<10 μg/L, there was a significant positive correlation between the pH of fishery waters and Chl. a(P<0.05). Various environmental factors had different effects on the pH of different types of fishery waters. The stepwise regression analysis showed that the pH of natural waters was mainly affected by Chl. a, and the pH of aquaculture waters was mainly affected by water temperature and dissolved oxygen. | 8 | Identification and Biological Characteristics of Edwardsiella tarda Isolated from Pelteobagrus fulvidraco显示文摘Over the past five years,an epidemic disease broke out in Pelteobagrus fulvidraco ponds of Zhejiang Province. The diseased fish mainly exhibits head splitting and surface bleeding; dissection results reveal hepatonephromegaly and abdominal bleeding. A bacterial strain HSY201301 was isolated from the liver tissue of P. fulvidraco with typical symptoms. Artificial infection experiment confirmed that the isolated strain had a strong virulence to healthy P. fulvidraco,leading to similar symptoms to naturally infected P. fulvidraco. The isolated strain was identified as an Edwardsiella tarda strain according to conventional morphological,physiological,biochemical characteristics and 16 S rRNA gene sequence. Results of drug susceptibility test indicated that the isolated strain was sensitive to ciprofloxacin,doxitard,penicillin,doxycycline,and rocephin. The results showed that the content of methylmercury in the crab meat was higher than that of the crab butter. According to China's national standard and industry standard,the content of methylmercury in the crab meat was lower than the limit and accorded with relevant regulations. | 12 | Ultrasound microbubbles combined with liposome-mediated pNogo-R shRNA delivery into neural stem cells显示文摘In the present study, ultrasound-mediated microbubble destruction (UMMD) alone and combined with liposome technology was used as a novel nonviral technique to transfect a Nogo receptor (Nogo-R) shRNA plasmid (pNogo-R shRNA) into neural stem cells (NSCs). Using green fluorescent protein as a reporter gene, transfection efficiency of NSCs was significantly higher in the group transfected with UMMD combined with liposomes compared with that of the group transfected with UMMD or liposomes alone, and did not affect cell vitality. In addition, Nogo-R mRNA and protein expression was dramatically decreased in the UMMD combined with liposome-mediated group compared with that of other groups after 24 hours of transfection.
