1Effect of electroacupuncture on expression of Ang/Tie-2 mRNA and protein in rats with acute cerebral infarction显示文摘OBJECTIVE: To identify the intervention mechanism of the effect of electroacupuncture on the expression of Ang/Tie-2 m RNA and protein in rats with acute cerebral infarction induced by middle cerebral artery occlusion(MCAO).METHODS: Altogether 120 Wistar rats were subjected to MCAO by inserting a nylon filament, andthen divided into 3 groups: control group, injured group and electroacupuncture group. The injured and electroacupuncture groups were further divided into the following 7 subgroups according to the time after MCAO: 3, 6, 12, 24 h, 3, 7 and 12 d, with 8 rats in each subgroup. The electroacupuncture group was given electroacupuncture treatment at Shuigou(GV 26) instantly after operation. The rats were killed at different time points according to their groups, and then the expression levels of Ang/Tie-2 m RNA and protein were detected using Real-Time polymerase chain reaction and immunohistochemical staining.RESULTS: The m RNA and protein expression levels of Ang/Tie-2 in the electroacupuncture group were significant higher than that in the injured group.CONCLUSION: The results suggested that electroacupuncture could significantly regulate the expression of Ang/Tie-2 m RNA and protein in the rats with acute cerebral infarction induced by MCAO, and enhance angiogenesis after ischemic penumbra.Li Jing Bai Zonglu Du Yuanhao Li Yongfeng Zhang Xuezhu Pang Bo Zhang Jingjing 2017Journal of Traditional Chinese Medicine2017,37,5:17
2An update: the role of Nephrin inside and outside the kidney显示文摘Nephrin is a key molecule in podocytes to maintain normal slit diaphragm structure. Nephin interacts with many other podocyte and slit diaphragm protein and also mediates important cell signaling pathways in podocytes. Loss of nephrin during the development leads to the congenital nephrotic syndrome in children. Reduction of nephrin expression is often observed in adult kidney diseases including diabetic nephropathy and HIV-associated nephropathy. The critical role of nephrin has been confirmed by different animal models with nephrin knockout and knockdown. Recent studies demonstrate that knockdown of nephrin expression in adult mice aggravates the progression of unilateral nephrectomy and Adriamycin-induced kidney disease. In addition to its critical role in maintaining normal glomerular filtration unit in the kidney, nephrin is also expressed in other organs. However, the exact role of nephrin in kidney and extra-renal organs has not been well characterized. 4Accurate authentication of Dendrobium officinale and its closely related species by comparative analysis of complete plastomes显示文摘Owing to its great medicinal and ornamental values, Dendrobium officinale is frequently adulterated with other Dendrobium species on the market. Unfortunately, the utilization of the common DNA markers ITS,ITS2, and mat Ktrbc L is unable to distinguish D. officinale from 5 closely related species of it(D. tosaense, D.shixingense, D. flexicaule, D. scoriarum and D. aduncum). Here, we compared 63 Dendrobium plastomes comprising 40 newly sequenced plastomes of the 6 species and 23 previously published plastomes. The plastomes of D. officinale and its closely related species were shown to have conserved genome structure and gene content. Comparative analyses revealed that small single copy region contained higher variation than large single copy and inverted repeat regions, which was mainly attributed to the loss/retention of ndh genes.Furthermore, the intraspecific sequence variability among different Dendrobium species was shown to be diversified, which necessitates a cautious evaluation of genetic markers specific for different Dendrobium species.By evaluating the maximum likelihood trees inferred from different datasets, we found that the complete plastome sequence dataset had the highest discriminatory power for D. officinale and its closely related species,indicating that complete plastome sequences can be used to accurately authenticate Dendrobium species.& 2018 Chinese Pharmaceutical Association and Institute of Materia Medica, Chinese Academy of Medical Sciences. 5Remodeling of motor cortex function in acute cerebral infarction patients following human urinary kallidinogenase A functional magnetic resonance imaging evaluation after 6 months显示文摘A total of 29 patients were treated within 48 hours after acute subcortical cerebral infarction with Xuesaitong or Xuesaitong plus human urinary kallidinogenase for 14 days.Neurological deficits,activity of daily living,and evaluations of distal upper limb motor functions at the 6-month follow-up showed that patients treated with Xuesaitong plus human urinary kallidinogenase recovered better than with Xuesaitong alone.In addition,functional MRI revealed that activation sites were primarily at the ipsilesional side of injury in all patients.Human urinary kallidinogenase induced hyperactivation of the ipsilesional primary sensorimotor cortex,premotor cortex,supplementary motor area,and contralesional posterior parietal cortex.Results showed that human urinary kallidinogenase improved symptoms of neurological deficiency by enhancing remodeling of long-term cortical motor function in patients with acute cerebral infarction.Xuezhu Song Lixin Han Yan Liu 2012Neural Regeneration Research2012,7,11:10
6Coupled BEM-FEM Analysis of the Large-Diameter Cylinder Structure System显示文摘A method of coupled BEM-FEM analysis for the elastic spatial structure system is presented. It can be applied to the calculation of the stress and deformation of the large-diamater cylinder structure system and it is suitable for symmetric or non-symmetric structures under the distributed or concentrated load. 7Effect of electro-scalp acupuncture on acute ischemic stroke: a randomized, single blind, trial显示文摘OBJECTIVE: To determine the clinical efficacy and safety of electro-scalp acupuncture in the treatment of patients with acute ischemic stroke.METHODS: Totally 74 patients with acute ischemic stroke were enrolled and divided into either body acupuncture(Control) or electro-scalp acupuncture(ESA) groups according to randomized controlled principle. The patients in the control group weregiven body acupuncture treatment once daily for28 d, whereas except for the body acupuncture,electro-scalp acupuncture was additional treatment given to the ESA group. Neurological deficits,everyday motor function and muscle strength were evaluated at baseline and the 28 th d by NIH Stroke Scale(NIHSS), Fugl-Meyer Assessment(FMA) and Modified Barthel Index Score(MBI), respectively.RESULTS: There were not obvious between-group differences in the baseline efficacy parameters(NIHSS, FMA and MBI)(all P > 0.05), whereas significant between-group differences were found in post-treatment NIHSS, FMA-UE and MBI scores(all P < 0.05). 9PKM2-dependent glycolysis promotes the proliferation and migration of vascular smooth muscle cells during atherosclerosis显示文摘Increased glycolysis is involved in the proliferation and migration of vascular smooth muscle cells(VSMCs).Pyruvate kinase isoform M2(PKM2),a key rate-limiting enzyme in glycolysis,accelerates the proliferation and migration of tumor cells.Although the intracellular mecha nisms associated with oxidized low-density lipoprotein(oxLDL)-stimulated VSMC proliferation and migration have been extensively explored,it is still unclear whether oxLDL promotes the proliferation and migration of VSMCs by enhancing PKM2-dependent glycolysis.In the present study,we detected PKM2 expression and pyruvate kinase activity in oxLDL-treated VSMCs and explored the regulation of PKM2 in oxLDL-treated VSMCs and apoE^-/-mice.The results showed that PKM2 expression in VSMCs was higher in the intima than in the media in plaques from atherosclerotic rabbits.Moreover,PKM2 level in VSMCs was increased during atherosclerosis progression in apoE^-/-mice.Both PKM2 expression and pyruvate kinase activity were found to be upregulated by oxLDL stimulation in VSMCs.Shikonin(SKN),a specific inhibitor of PKM2,was found to inhibit the oxLDL-induced proliferation and migration in VSMCs,in addition to delaying the atherosclerosis progression in apoE^-/-mice.More importantly,oxLDL increased glucose uptake,ATP and lactate production,and the extracellular acidification rate in VSMCs,which could be reversed by SKN.Meanwhile,oxygen consumption rate was unchanged after oxLDL stimulation,suggesting that glycolysis is the main contributor to the energy supply in oxLDL-treated VSMCs.Our results suggest that oxLDL induces VSMC proliferation and migration by upregulating PKM2-dependent glycolysis,thereby contributing to the atherosclerosis progression.Thus,targeting PKM2-dependent glycolysis might provide a novel therapeutic approach for the treatment of atherosclerosis.Xuezhu Zhao Fancheng Tan Xiaoru Cao Zhengyu Cao Bicheng Li Zhaoqian Shen Ye Tian 2020Acta Biochimica et Biophysica Sinica2020,52,1:3

12Synthesis of Amphiphilic Starch Derivatives Using One-pot Synthesis Procedure显示文摘Amphiphilic starch derivatives with high content of functional groups were prepared from potato starch using a one-pot synthesis method with a single reaction medium for the entire procedure. Potato starch was benzylated, followed by the introduction of hydroxypropyltrimethylammonium(HPMA) moieties without the purification of intermediates. The synthesis was performed under heterogeneous conditions, leading to the formation of benzyl 2-hydroxypropyltri methylammonium starch chloride(BnHPMAS) with a total degree of substitution(DS) of up to 1.4. This process improved the efficiency of the preparation of amphiphilic starch derivatives and reduced the time and resources consumed by avoiding a separation process and purification of the intermediate compounds.The DS of BnHPMAS was in the range of 0.36 to 1.4, which could be tuned by varying the molar ratio of the reagents to repeating unit or by changing the reaction temperature, time, and medium. The structure of the amphiphilic starches was characterized using elemental analysis, size exclusion chromatography,fourier transform infrared spectroscopy(FT-IR), and nuclear magnetic resonance(NMR) spectroscopy. 13Study of the mechanism of cationic drug release increase coated with Sureleaseafter curing显示文摘This paper describes the investigation of the release mechanism of the cationic drug coated with Sureleaseafter curing compared with nonionic and anionic drug.The release rate of cured propafenone hydrochloride pellets coated with Sureleasewas increased significantly compared with uncured ones.The changes in release rate after seal-coating proved the migration of drug into the polymer film.Based on a comparative study of ammonia permeation tests and the impact of ammonia on drug with different properties,it was shown that the change in release of the cationic drug after curing was related to the combination of cationic drug and ammonia in Surelease.Also,the scanning electron microscopy results showed that curing and dissolution can both prevent polymer film coalescence.During coating,the evaporated ammonia combined with the cationic drug and the polymer film coalesced.However,the combined ammonia could be decomposed during curing and interrupt the coalescence of the polymer-coat,leading to increased release.In an optimized process,the ammonia in the Sureleasecan be evaporated quickly and the ionization of the drug will be reduced.All these factors can contribute to the formation of the polymer film.Yanting Hou Huiying Wang Xuezhu Zhang Meijuan Zou Gang Cheng 2013Asian Journal of Pharmaceutical Sciences2013,8,5:1
