1Changes in heavy metal contents in animal feeds and manures in an intensive animal production region of China显示文摘The 360 feed and manure samples were collected from 150 animal farms in Jiangsu Province, China and analyzed for heavy metals. Concentrations of Zn and Cu in animal feeds were 15.9–2041.8 and undetected–392.1 mg/kg respectively, while Hg, As, Pb, Cd, and Cr in all feeds were below 10 mg/kg. Concentrations of Cu, Zn, and Cr in animal manures were 8.4–1726, 39.5–11379, and 1.0–1602 mg/kg respectively, while As, Cd, Hg, and Pb were < 10 mg/kg. The concentration of Cu, Zn, As and Cr in animal feed and manure were positively correlated(p < 0.001), but the Cd, Hg, and Pb were not statistically correlated between the feed and the manure. Concentrations of Cu and Zn were highest in pig feed and manure, followed by poultry and dairy feeds and manures. During 1990– 2008, Cu, Zn, As, Cr, Cd contents increased by 771%, 410%, 420%, 220%, and 63% in pig manure, 212%, 95%, 200%, 791%, and –63% in dairy manure, and 181%, 197%, 1500%, 261, and 196% in poultry manure. Most of the increases occurred from 2002 to 2008, which reflects the extensive use of feed additives after 2002. In contrast, Pb and Hg in manures continuously decreased from 1990 to 2008. 2Pretreatment with Danhong injection protects the brain against ischemia-reperfusion injury显示文摘Danhong injection(DHI),a Chinese Materia Medica standardized product extracted from Radix Salviae miltiorrhizae and Flos Carthami tinctorii,is widely used in China for treating acute ischemic stroke.In the present study,we explored the neuroprotective efficacy of DHI in a rat model of temporary middle cerebral artery occlusion,and evaluated the potential mechanisms underlying its effects.Pretreatment with DHI(0.9 and 1.8 mL/kg)resulted in a significantly smaller infarct volume and better neurological scores than pretreatment with saline.Furthermore,DHI significantly reduced the permeability of the blood-brain barrier,increased occludin protein expression and decreased neutrophil infiltration,as well as profoundly suppressing the upregulation of matrix metallopeptidase-9 expression seen in rats that had received vehicle.Matrix metallopeptidase-2 expression was not affected by ischemia or DHI.Moreover,DHI(1.8 mL/kg)administered 3 hours after the onset of ischemia also improved neurological scores and reduced infarct size.Our results indicate that the neuroprotective efficacy of DHI in a rat model of cerebral ischemia-reperfusion injury is mediated by a protective effect on the blood-brain barrier and the reversal of neutrophil infiltration.Shaoxia Wang Hong Guo Xumei Wang Lijuan Chai Limin Hu Tao Zhao Buchang Zhao Xiaoxu Tan Feifei Jia 2014Neural Regeneration Research2014,9,15:12
3The influence of volcanism on paleoclimate in the northeast of China:Insights from Jinchuan peat,Jilin Province,China显示文摘Just like contemporary sediments, peat itself is a good repository of information about climate change, the effects of volcanic activity on climate change have been truly recorded in peat, since it is a major archive of volcanic eruption incidents. A section of sand was identified as tephra from the Jinchuan peat, Jilin Province, China, for the grains look like slag with surface bubbles and pits, characterized by high porosity, and loose structure with irregular edges and corners. According to the peat characteristics of uniform deposition, the tephra was dated at 2002-1976 a B.P. by way of linear interpolation, so the time of volcanic eruption was 15 B.C.-26 A.D. (the calibrated age). While the geochemical characteristics of tephra in this study are quite the same as those of tephra from the Jinlongdingzi volcano at Longgang and from alkaline basaltic magma, with the contents of SiO2<55%, and the similar contents to Al2O3 and Fe, but the contents of Na2O>K2O. We speculated that the tephra in this study came from the Longgang volcano group. Compared with 11 recorded volcanic eruption events as shown on the carbon and oxygen isotope curves of the Jinchuan peat cellulose, it is obviously seen that adjacent or large-scale volcanic eruptions are precisely corresponding to the minimum temperature and humidity. It seems that these volcanic eruptions indeed affected the local climate, leading to the drop of regional temperature and humidity. As a result, there was prevailing a cold and dry climate there, and all these changes can be well recorded in peat. 4Effects of folic acid on in vitro astrocytic differentiation of neural stem cells from neonatal rats显示文摘BACKGROUND:Folic acid is essential for normal functioning of the nervous system.Previous studies have focused on the effects of folic acid on astrocyte proliferation. OBJECTIVE:To explore the effects of folic acid on astrocyte differentiation of neural stem cells (NSCs) and the related mechanisms in vitro. DESIGN,TIME AND SETTING:A randomized,controlled,grouping experiment was performed in Tianjin Medical University between August 2007 and October 2008. MATERIALS:Folic acid and 5-bromo-2-deoxyuridine(BrdU) were obtained from Sigma,MO,USA. Primary antibodies[rabbit anti-rat nestin,β-tubulin-Ⅲ,glial fibrillary acidic protein,and neurogenin1 (Ngn1);mouse anti-rat BrdU andβ-actin monoclonal antibodies]were purchased from Santa Cruz Biotechnology,USA. METHODS:At 6 days of NSC proliferation from 24-hour-old neonatal rats,BrdU incorporation assay was performed.Seven days after primary culture,NSCs were induced to differentiate with medium containing 5%fetal bovine serum.Cultured NSCs were assigned to three groups:control,low-dose (liquid media with 8 mg/L folic acid),and high-dose folic acid(liquid media with 44 mg/L folic acid). MAIN OUTCOME MEASURES:At day 7 after primary culture,the cells were identified as NSCs by immunocytochemical methods.Double-label immunofluorescence technique for glial fibrillary acidic protein/BrdU detected differentiated cells 7 days after induction.Western blot was used to analyze expression of Ngn1 protein in NSCs. RESULTS:In serum-free suspension medium,neurospheres comprised a large number of Nestin-, glial fibrillary acidic protein-,β-tubulin-Ⅲ-,and BrdU-positive cells.Compared with the control group, high-dose folic acid supplementation led to an marked increase in the percentage of glial fibrillary acidic protein/BrdU-positive cells(P<0.05),and significantly decreased Ngn1 protein expression (P<0.05). 5Geochemistry of rare-earth elements in shallow groundwater, northeastern Guangdong Province, China显示文摘Shallow groundwater and hot springs were collected from northeastern Guangdong Province, Southeast China, to determine the concentrations and fractionation patterns of rare-earth elements(REE). The results show that the La, Ce and Nd of REEs are abundant in groundwater and rock samples, and the ∑REE contents in groundwater and rock samples range from 126.5 to 2875.3 ng/L, and 79.44 to 385.85 mg/L, respectively. The shallow groundwater has slightly HREE-enriched PAAS-normalized patterns. However, the granitic rocks PAAS-normalized patterns, with remarkable negative Eu anomalies, are different from that of shallow groundwater. The enrichment of HREE is considered to be controlled by REE complexation and readsorption for most groundwater has Ce and Eu positive anomalies. The Ce and Eu anomalies in groundwater are controlled by redox conditions. Moreover, the Fe-contain sediments dissolution and/or the reduction of Fe oxyhydroxides are another factor contributing to Ce anomalies. 6Recommendation for Cardiac Magnetic Resonance Imaging-Based Phenotypic Study:Imaging Part显示文摘Cardiac magnetic resonance(CMR)imaging provides important biomarkers for the early diagnosis of many cardiovascular diseases and has been reported to reveal phenome-wide associations of cardiac/aortic structure and functionality in popula-tion studies.Nevertheless,due to the complexity of operation and variations among manufactural vendors,magnetic field strengths,coils,sequences,scan parameters,and image analysis approaches,CMR is rarely used in large cohort studies.Existing guidelines mainly focused on the diagnosis of cardiovascular diseases,which did not aim to basic research.The purpose of this study was to propose a recommendation for CMR based phenotype measurements for cohort study.We classify the imaging sequences of CMR into three categories according to the importance and universality of correspond-ing measurable phenotypes.The acquisition time and repeatability of the phenotypic measurement were also taken into consideration during the categorization.Unlike other guidelines,this recommendation focused on quantitative measurement of large amount of phenotypes from CMR.Chengyan Wang Yan Li Jun Lv Jianhua Jin Xumei Hu Xutong Kuang Weibo Chen He Wang 2021Phenomics2021,1,4:1
7Overexpression of MdATG8i improves water use efficiency in transgenic apple by modulating photosynthesis, osmotic balance, and autophagic activity under moderate water deficit显示文摘Water deficit is one of the major limiting factors for apple(Malus domestica)production on the Loess Plateau,a major apple cultivation area in China.The identification of genes related to the regulation of water use efficiency(WUE)is a crucial aspect of crop breeding programs.As a conserved degradation and recycling mechanism in eukaryotes,autophagy has been reported to participate in various stress responses.However,the relationship between autophagy and WUE regulation has not been explored.We have shown that a crucial autophagy protein in apple,MdATG8i,plays a role in improving salt tolerance.Here,we explored its biological function in response to long-term moderate drought stress.The results showed that MdATG8i-overexpressing(MdATG8i-OE)apple plants exhibited higher WUE than wild-type(WT)plants under long-term moderate drought conditions.Plant WUE can be increased by improving photosynthetic efficiency.Osmoregulation plays a critical role in plant stress resistance and adaptation.Under long-term drought conditions,the photosynthetic capacity and accumulation of sugar and amino acids were higher in MdATG8i-OE plants than in WT plants.The increased photosynthetic capacity in the OE plants could be attributed to their ability to maintain optimal stomatal aperture,organized chloroplasts,and strong antioxidant activity.MdATG8i overexpression also promoted autophagic activity,which was likely related to the changes described above.In summary,our results demonstrate that MdATG8i-OE apple lines exhibited higher WUE than WT under long-term moderate drought conditions because they maintained robust photosynthesis,effective osmotic adjustment processes,and strong autophagic activity.Xin Jia Ke Mao Ping Wang Yu Wang Xumei Jia Liuqing Huo Xun Sun Runmin Che Xiaoqing Gong Fengwang Ma 2021Horticulture Research2021,8,1:1

10Noninvasive Assessment of Liver Fibrosis and Inflammation in Chronic Hepatitis B:A Dual-task Convolutional Neural Network(DtCNN)Model Based on Ultrasound Shear Wave Elastography显示文摘Background and Aims:Liver stiffness(LS)measured by shear wave elastography(SWE)is often influenced by hepat-ic inflammation.The aim was to develop a dual-task convo-lutional neural network(DtCNN)model for the simultaneous staging of liver fibrosis and inflammation activity using 2D-SWE.Methods:A total of 532 patients with chronic hepatitis B(CHB)were included to develop and validate the DtCNN model.An additional 180 consecutive patients between De-cember 2019 and April 2021 were prospectively included for further validation.All patients underwent 2D-SWE examina-tion and serum biomarker assessment.A DtCNN model con-taining two pathways for the staging of fibrosis and inflam-mation was used to improve the classification of significant fibrosis(≥F2),advanced fibrosis(≥F3)as well as cirrhosis(F4).Results:Both fibrosis and inflammation affected LS measurements by 2D-SWE.The proposed DtCNN performed the best among all the classification models for fibrosis stage[significant fibrosis AUC=0.89(95%CI:0.87-0.92),ad-vanced fibrosis AUC=0.87(95%CI:0.84-0.90),liver cirrho-sis AUC=0.85(95%CI:0.81-0.89)].The DtCNN-based pre-diction of inflammation activity achieved AUCs of 0.82(95%CI:0.78-0.86)for grade≥A1,0.88(95%CI:0.85-0.90)grade≥A2 and 0.78(95%CI:0.75-0.81)for grade≥A3,which were significantly higher than the AUCs of the single-task groups.Similar findings were observed in the prospec-tive study.Conclusions:The proposed DtCNN improved di-agnostic performance compared with existing fibrosis staging models by including inflammation in the model,which sup-ports its potential clinical application.Chengyan Wang Lili Zheng Yan Li Shujun Xia Jun Lv Xumei Hu Weiwei Zhan Fuhua Yan Ruokun Li Xinping Ren 2022Journal of Clinical and Translational Hepatology2022,10,6:0
11WWP2 ameliorates oxidative stress and inflammation in atherosclerotic mice through regulation of PDCD4/HO-1 pathway显示文摘WWP2 is a HECT-type E3 ubiquitin ligase that regulates various physiological and pathological activities by binding to different substrates,but its role in atherosclerosis(AS)remains largely unknown.The objective of the present study is to investigate the role and underlying molecular mechanisms of WWP2 in endothelial injury.We found that WWP2 expression is significantly decreased in Apolipoprotein E(ApoE)-/-mice.Overexpression of WWP2 attenuates oxidative stress and inflammation in AS mice,while knockdown of WWP2 has opposite effects.WWP2 overexpression alleviates oxidized low-density lipoprotein(ox-LDL)-induced human umbilical vein endothelial cell(HUVEC)injury,evidenced by the decreased oxidative stress levels and the secretion of inflammatory cytokines.Programmed cell death 4(PDCD4)is identified as a potential substrate of WWP2.Co-immunoprecipitation(Co-IP)further demonstrates that WWP2 interacts with PDCD4,which is enhanced by ox-LDL treatment.Furthermore,the level of PDCD4 ubiquitination is significantly increased by WWP2 overexpression under the condition of MG132 treatment,while WWP2 knockdown shows opposite results.Subsequently,rescue experiments demonstrate that WWP2 knockdown further aggravates oxidative stress and inflammation in ox-LDL-treated HUVECs,while knockdown of PDCD4 alleviates this effect.Moreover,the use of sn-protoporphyrin(SnPP),an inhibitor of HO-1 pathway,confirms that PDCD4 enhances endothelial injury induced by ox-LDL through inhibiting HO-1 pathway.In conclusion,our results suggest that WWP2 protects against atherosclerosis progression via the PDCD4/HO-1 pathway,which may provide a novel treatment strategy for atherosclerosis.Xingye Wang Lu Ma Songlin Zhang Qiang Song Xumei He Jun Wang 2022Acta Biochimica et Biophysica Sinica2022,54,8:0

14NLRP3 inflammasome mediates angiotensinⅡ-induced isletβcell apoptosis显示文摘Elevation of angiotensinⅡ(AngⅡ)in the serum of patients with diabetes is known to promote apoptosis of isletβcells,but the underlying mechanism remains unclear.The aim of the present study was to explore the role of Nod-like receptor protein 3(NLRP3)inflammasome in AngⅡ-induced apoptosis of pancreatic isletβcells and investigate the possible underlying mechanism.The effect of AngⅡon INS-1 cell(a rat insulinoma cell line)viability was detected by CCK-8 method.The cell apoptosis was detected by flow cytometry and western blot analysis.The effect of AngⅡon the expressions of thioredoxin-interacting protein(TXNIP)and NLRP3 protein was detected by western blot analysis.The expression of TXNIP mRNA was detected by real-time polymerase chain reaction.The results showed that AngⅡwas able to reduce INS-1 cell viability and promote apoptosis and at the same time up-regulate the expressions of TXNIP and NLRP3 components.AngⅡ-induced apoptosis was inhibited after administration of the NLRP3 inhibitor MCC950,and TXNIP silencing could reduce the NLRP3 expression and apoptosis,while both effects of AngⅡon TXNIP-NLRP3 and its apoptosis-inducing effect were inhibited by angiotensinⅡtypeⅠreceptor(AT1R)blocker Telmisartan.Our results demonstrated that the TXNIP-NLRP3 inflammasome pathway mediated AngⅡ-induced INS-1 cell apoptosis and might hopefully become a novel target for the treatment of diabetes mellitus.Jin Wang Yanjin Feng Haiyan Huo Xumei Zhang Jiping Yue Wenting Zhang Zi Yan Xiangying Jiao 2019Acta Biochimica et Biophysica Sinica2019,51,5:0
