2Efficacy and safety of low-dose interleukin-2 in combination with methotrexate in patients with active rheumatoid arthritis:a randomized,double-blind,placebo-controlled phase 2 trial显示文摘Rheumatoid arthritis(RA)is an aggressive autoimmune arthritis,and current therapies remain unsatisfactory due to low remission rate and substantially adverse effects.Low-dose interleukin-2(Ld-IL2)is potentially a therapeutic approach to further improve the disease.This randomized,double-blind,placebo-controlled trial was undertaken to evaluate the efficacy and safety of Ld-IL2 in patients with active RA.Patients were randomly assigned(1:1)to receive Ld-IL2,defined as a dose of 1 million IU,or placebo in a 12-week trial with a 12-week follow-up.Three cycles of Ld-IL2 or placebo were administered subcutaneously every other day for 2 weeks(a total of 7 doses),followed by a 2-week break.All patients received a stable dose of methotrexate(MTX).The primary outcomes were the proportion of patients achieving the ACR20,DAS28-ESR<2.6,and the change from baseline in CDAI or SDAI at week 24.Secondary endpoints included other clinical responses and safety.The primary outcomes were achieved in the perprotocol population.The improvements from baseline in CDAI and SDAI were significantly greater across time points for the LdIL2+MTX group(n=17)than for the placebo+MTX group(n=23)(P=0.018 and P=0.015,respectively).More patients achieved ACR20 response in the Ld-IL2+MTX group than those in the placebo+MTX group at week 12(70.6%vs 43.5%)and at week 24(76.5%vs 56.5%)(P=0.014).In addition,low Treg and high IL-21 were associated with good responses to Ld-IL2.Ld-IL-2 treatment was well-tolerated in this study.These results suggested that Ld-IL2 was effective and safe in RA.ClinicalTrials.gov number:NCT 02467504.Xiaoying Zhang Miao Miao Ruijun Zhang Xu Liu Xiaozhen Zhao Miao Shao Tian Liu Yuebo Jin Jiali Chen Huixin Liu Xia Zhang Yun Li Yunshan Zhou Yue Yang Ru Li Haihong Yao Yanying Liu Chun Li Yuhui Li Limin Ren Yin Su Xiaolin Sun Jing He Zhanguo Li 2022Signal Transduction and Targeted Therapy2022,7,4:7

4Role of chemerin/CMKLR1 in the maintenance of earlypregnancy显示文摘Chemerin is a cytokine that attracts much attention in the reproductive process. This study aimed toexplore the effects of chemerin and its receptor chemokine-like receptor 1 (CMKLR1) on the maintenance of earlypregnancy. The expression levels of chemerin and CMKLR1 in the decidua tissues of 20 early normal pregnantwomen and 20 early spontaneous abortion women were examined by Western blot and real-time polymerase chainreaction analyses. CMKLR1 receptor antagonist (a-NETA) was then intrauterinely injected into normal pregnantmice model to assess its effect on the outcome of pregnancy and the phosphorylation rate of ERK1/2 in deciduatissues. We found that the expression level of chemerin in women who had experienced early spontaneous abortionwas lower than in those who had experienced normal early pregnancy (P 〈 0.01); conversely, CMKLR1expression was higher in the former than in the latter (P 〈 0.01). In a pregnant-mouse model, the embryoresorption rate of a-NETA group was higher than that in the negative control group (61.5% vs. 10.8%) (P 〈 0.001).Compared with the control group, ERK1/2 phosphorylation in decidua tissues decreased in the a-NETA-treatedgroup (P 〈 0.01). 5Crucial function of histone lysine methylation in plant reproduction显示文摘Unlike animals, plants do not set aside germ cells early in development. In angiosperm species, reproduction occurs in the adult plant upon flowering. The multicellular male and female gametophytes differentiate from meiotic products within reproductive floral organs. Double fertilization is another remarkable feature of most angiosperm species. The zygote derived from fertilization of the egg cell by one of the sperm cells and the endosperm from fertilization of the central cell by the second sperm cell develop in a coordinated manner together and enclosed in the sporophytic maternal integuments, forming the seed. Understanding plant reproduction is biologically pertinent and agronomically and ecologically important. Here, we describe the known functions of histone lysine methylations in various steps of reproduction in the reference plant Arabidopsis thaliana. 9Whirligig‑Inspired Hybrid Nanogenerator for Multi‑strategy Energy Harvesting显示文摘Portable energy solutions are highly desired in the era of the Internet of Things for powering various distributed micro-electronic devices.At the same time,the energy crisis and catastrophic global warming are becoming serious problems in the world,emphasizing the urgent need for clean and renewable energy.Here,we report a low-cost,high-performance,and portable hand-driven whirligig structured triboelectric–electromagnetic hybrid nanogenerator(whirligig-HNG)for multi-strategy energy harvesting.The whirligig-HNG comprises a dynamic supercoiling TENG via the pulling-strings and inner-distributed EMGs(variable number)in the rotator.The whirligig structure can readily convert linear displacement in low frequency into rotary motion in extremely high frequency.Based on this ingenious design,the whirligig-HNG is capable to harvest the triboelectric energy from the supercoiling/uncoiling process from the pulling strings and simultaneously utilize the high-frequency rotation energy via electromagnetic induction.We have systematically investigated the working mecha-nism of the whirligig-HNG for coupled energy harvesting and compared the individual characteristics of TENG and EMG.The whirligig-HNG is successfully demonstrated to light up more than 100 commercial light-emitting diodes(LEDs)and drive portable electronics.This research presents the enormous potential of whirligig-HNG as a manual and portable power supply for powering various portable electronics.Xiaozhen Dan Ran Cao Xiaole Cao Yifei Wang Yao Xiong Jing Han Lan Luo Jiahong Yang Nuo Xu Jia Sun Qijun Sun Zhong Lin Wang 2023Advanced Fiber Materials2023,5,1:0
10Utilizing dual-pathway energy transfer in upconversion nanoconjugates for reinforced photodynamic therapy显示文摘Enhancing the therapeutic effect of existing treatments or developing new non-invasive treatments are important measures to achieve high-efficiency treatment of malignant tumors.Photodynamic therapy(PDT)is an emerging treatment modality,and the key for achieving high-efficiency PDT is to select light with strong tissue penetration depth and enhance the generation of reactive oxygen species(ROS).Although the upconversion nanoparticles(UCNPs)modified with the photosensitizers could achieve PDT with strong penetration depth under near-infrared light irradiation,the ROS generated by traditional single-pathway PDT is still insufficient.Herein,we developed a novel nanoconjugate(UCNP-Ce6/AIEgen)for dual-pathway reinforced PDT,in which the UCNPs were co-modified with chlorin e6(Ce6)and luminogen with aggregation-induced emission(AIEgen).Due to the presence of AIEgen,UCNP-Ce6/AIEgen could avoid aggregation-caused luminescence quenching in biological water environments and convert upconversion luminescence(UCL)of UCNPs to Ce6-activatable fluorescence.Therefore,under the irradiation of 808 nm laser,UCNP-Ce6/AIEgen can not only undergo direct lanthanide-triplet energy transfer to activate Ce6,but also convert the UCL of UCNPs to the light that can activate Ce6 through Fӧrster resonance energy transfer to generate more ROS,thus promoting tumor cell apoptosis.This work broadens the applications of nanoconjugates of lanthanide-based inorganic materials and organic dyes,and provides a conception for reinforced PDT of tumors.Ruohao Zhang Yu Lu Yifei Zhou Kehong Lv Xinyu Fu Jitong Gong Shuang Yao Xiaozhen Wang Jing Feng Hongjie Zhang 2024Nano Research2024,17,4:0

12Construction of coxsackievirus B5 viruses with luciferase reporters and their applications in vitro and in vivo显示文摘Coxsackievirus belongs to the Picornaviridae family and is one of the major pathogens that cause hand,foot and mouth disease(HFMD)in infants and children with potential serious complications and even deaths.The pathogenesis of this virus is not fully elucidated and no vaccine or antiviral drug has been approved.In this study,a full-length infectious cDNA clone of coxsackievirus B5 virus was assembled and the recombinant virus displayed similar growth kinetics and ability to cause cytopathic effects as the parental virus.Luciferase reporter was then incorporated to generate both full-length and subgenomic replicon(SGR)reporter viruses.The full-length reporter virus is suitable for high-throughput antiviral screening,while the SGR is a useful tool to study viral-host interactions.More importantly,the full-length reporter virus has also been shown to infect the suckling mouse model and the reporter gene could be detected using an in vivo imaging system,thus providing a powerful tool to track viruses in vivo.In summary,we have generated coxsackievirus B5 reporter viruses and provided unique tools for studying virus-host interactions in vitro and in vivo as well as for high-throughput screenings(HTS)to identify novel antivirals.Shangrui Guo Meng Xun Tingting Fan Xinyu Li Haoyan Yao Xiaozhen Li Bo Wu Hang Yang Chaofeng Ma Hongliang Wang 2023Virologica Sinica2023,38,4:0
13Gastric adenocarcinoma with hepatoid adenocarcinoma:A case report and literature review显示文摘Objective Hepatoid adenocarcinoma of the stomach is exceedingly rare. Only a few cases have been reported worldwide. In this report, we describe our pathological findings along with a review of the literature to improve our understanding of the disease and prevent misdiagnosis, as well as to provide evidence for its treatment and prognosis.Methods A 68-year-old male patient was admitted to our hospital(Dalian Municipal Central Hospital, Dalian, China) complaining of upper abdominal pain. Physical examination and regular laboratory blood tests showed no obvious abnormalities. A contrast-enhanced computed tomography scan displayed a 3.0 cm × 3.0 cm mass in the gastric antrum, but no metastasis was found in the adjacent organs.Results A radical gastrectomy was performed and postoperative histopathology showed a moderately differentiated adenocarcinoma with partial hepatoid adenocarcinoma.Conclusion Hepatoid adenocarcinoma of the stomach is a rare malignant neoplasm. The diagnostic criteria for hepatoid adenocarcinoma of the stomach depends mainly on the clinical, radiographic, and histopathological findings.
