2Wheat breeding history reveals synergistic selection of pleiotropic genomic sites for plant architecture and grain yield显示文摘Diversity surveys of crop germplasm are important for gaining insights into the genomic basis for plant architecture and grain yield improvement,which is still poorly understood in wheat.In this study,we exome sequenced 287 wheat accessions that were collected in the past 100 years.Population genetics analysis identified that 6.7%of the wheat genome falls within the selective sweeps between landraces and cultivars,which harbors the genes known for yield improvement.These regions were asymmetrically distributed on the A and B subgenomes with regulatory genes being favorably selected.Genome-wide association study(GWAS)identified genomic loci associated with traits for yield potential,and two underlying genes,TaARF12 encoding an auxin response factor and TaDEP1 encoding the G-proteinγ-subunit,were located and characterized to pleiotropically regulate both plant height and grain weight.Elite single-nucleotide haplotypes with increased allele frequency in cultivars relative to the landraces were identified and found to have accumulated over the course of breeding.Interestingly,we found that TaARF12 and TaDEP1 function in epistasis with the classical plant height Rht-1 locus,leading to propose a“Green Revolution”-based working model for historical wheat breeding.Collectively,our study identifies selection signatures that fine-tune the gibberellin pathway during modern wheat breeding and provides a wealth of genomic diversity resources for the wheat research community.Aili Li Chenyang Hao Zhenyu Wang Shuaifeng Geng Meiling Jia Fang Wang Xiang Han Xingchen Kong Lingjie Yin Shu Tao Zhongyin Deng Ruyi Liao Guoliang Sun Ke Wang Xingguo Ye Chengzhi Jiao Hongfeng Lu Yun Zhou Dengcai Liu Xiangdong Fu Xueyong Zhang Long Mao 2022Molecular Plant2022,15,3:4
3Large-size niobium disulfide nanoflakes down to bilayers grown by sulfurization显示文摘Atomically thin layers of group VB transition metal dichalcogenides (TMDs)provide a unique platform for studying two-dimensional (2D) superconductivityand charge density waves. Thus far, the bottom-up synthesis of these 2D TMDshas often involved precursors that are corrosive or toxic, and their lateral sizesare typically only a few micrometers. In this paper, we report the growth ofNbS2 nanoflakes with a thickness down to bilayers and a lateral dimension upto tens of micrometers without using harsh chemical species. NbS2 nanoflakeseither standing or lying with respect to the sapphire substrate were obtained bysulfurization of niobium oxide films that were prepared via pulsed laser deposition.Standing nanoflakes are considered to grow epitaxiaUy on the sapphire substrateaccording to their ordered orientation, while lying nanoflakes with randomorientations were grown directly on top of the niobium oxide films. The Ramanspectra of the 3R-phase exhibit strong dependence on the layer thickness, wherethe A1 mode softens as the layer number decreases. In contrast to the stable bulkNbS2, the ultra-thin nanoflakes were oxidized on their top surfaces after prolongedexposure to air, as revealed by X-ray photoelectron spectroscopy. 5Trajectory planning and motion control of full-row seedling pick-up arm显示文摘Frequent start-stops of automatic seedling transplanters would lengthen the seedling picking time due to alternating movement of the motor,and cause positioning errors and frequent motor vibrations when starting the machine.To solve these problems and ensure the pick-up continuity of automatic transplanters,in this study the smooth circular arc interpolation algorithm and the least square method for multinomial curve fitting were used to conduct the trajectory planning of automatic transplanter based on the movement of the pick-up arm of the transplanter.Velocity and time curves were fitted in segments to acquire motion control parameters and further tracking control was conducted.The mathematical model of the control system was established using Simulink,and simulation analysis and system debugging test were performed.Experiments show that the trajectory curves obtained by MATLAB’s data processing can realize the continual and smooth motion trajectory of the pick-up arm;stepper motor velocity control can effectively track the planning curve and improve the seedling pick-up efficiency.The pick-up and pushing time of each motion of the planned plug seedling transplanter reduced by 1.0678 s and the start-stop times reduced from 6 to 1,which solved the frequent motor vibrations when starting the machine and improved the stability of the system.In addition,overstep and out-of-step of the motor at the start-stop moment were avoided and displacement error was reduced.Ling Ren Ning Wang Weibin Cao Jiangquan Li Xingchen Ye 2020International Journal of Agricultural and Biological Engineering2020,13,3:1

8The soft glumes of common wheat are sterile-lemmas as determined by the domestication gene Q显示文摘The Q gene in common wheat encodes an APETALA2(AP2) transcription factor that causes the free threshing attribute. Wheat spikelets bearing several florets are subtended by a pair of soft glumes that allow free liberation of seeds. In wild species, the glumes are tough and rigid,making threshing difficult. However, the nature of these 'soft glumes', caused by the domestication allele Q is not clear. Here, we found that over expression of Q in common wheat leads to homeotic florets at glume positions. We provide phenotypic, microscopy, and marker genes evidence to demonstrate that the soft glumes of common wheat are in fact lemma-like organs, or so-called sterile-lemmas. By comparing the structures subtending spikelets in wheat and other crops such as rice and maize, we found that AP2 genes may play conserved functions in grasses by manipulating vestigial structures, such as floret-derived soft glumes in wheat and empty glumes in rice. 11minimal genome design to maximally guarantee fertile inter-subspecific hybrid rice显示文摘Hybrid rice has made considerable contributions to achieve the ambitious goal of food security for the world's population.Hybrid rice from indica/xian and japonica/geng subspecies shows much higher heter-osis and is thereby an important innovation in promoting rice production in the next decade.However,such inter-subspecific hybrid rice has long suffered from serious hybrid sterility,which is a major challenge that needs to be addressed.In this study,we performed a genome design strategy to produce fertile inter-subspecific hybrid by creation of wide compatibility varieties that are able to overcome hybrid sterility.Based on combined genetic analyses in two indica-japonica crosses,we determined that four hybrid ste-rility loci,S5,f5,pf12 and Sc,are the major QTLs controlling inter-subspecific hybrid sterility and thus the minimal targets that can be manipulated for breeding sub-specific hybrid rice.We then cloned the pf12 lo-cus,one of the most effective loci for hybrid male sterility,by map-based cloning,and showed that artificial disruption of pf12A gene at this locus could successfully rescue hybrid fertility.We further dissected the genetic basis of wide compatibility using three pairwise crosses from a wide-compatibility variety Dular and representative indica and japonica varieties.On this basis,we constructed and assembled different combinations of naturally compatible alleles of four loci,S5,Sc,pf12,and f5,and found that the improved lines could fully recover pollen and embryo sac fertility in test-crossed F,s,thereby completely fulfilling the demands of inter-subspecific hybrid spikelet fertility in agricultural production.This breeding scheme would facilitate redesign of future inter-subspecific hybrid rice with a higher yield potential.Penghui Zhou Zhengji Wang Xingchen Zhu Yao Tang Liang Ye Huihui Yu Yating Li Ningke Zhang Ting Liu Tian Wang Yuying Wu Dengyun Cao Yuan Chen Xu Li Qinglu Zhang Jinghua Xiao Sibin Yu Qifa Zhang Jiaming Mi Yidan Ouyang 2023Molecular Plant2023,16,4:0
12Artemisinins inhibit oral candidiasis caused by Candida albicans through the repression on its hyphal development显示文摘Candida albicans is the most abundant fungal species in oral cavity. As a smart opportunistic pathogen, it increases the virulence byswitching its forms from yeasts to hyphae and becomes the major pathogenic agent for oral candidiasis. However, the overuse ofcurrent clinical antifungals and lack of new types of drugs highlight the challenges in the antifungal treatments because of the drugresistance and side effects. Anti-virulence strategy is proved as a practical way to develop new types of anti-infective drugs. Here,seven artemisinins, including artemisinin, dihydroartemisinin, artemisinic acid, dihydroartemisinic acid, artesunate, artemether andarteether, were employed to target at the hyphal development, the most important virulence factor of C. albicans. Artemisininsfailed to affect the growth, but significantly inhibited the hyphal development of C. albicans, including the clinical azole resistantisolates, and reduced their damage to oral epithelial cells, while arteether showed the strongest activities. The transcriptomesuggested that arteether could affect the energy metabolism of C. albicans. Seven artemisinins were then proved to significantlyinhibit the productions of ATP and cAMP, while reduced the hyphal inhibition on RAS1 overexpression strain indicating thatartemisinins regulated the Ras1-cAMP-Efg1 pathway to inhibit the hyphal development. Importantly, arteether significantlyinhibited the fungal burden and infections with no systemic toxicity in the murine oropharyngeal candidiasis models in vivo causedby both fluconazole sensitive and resistant strains.
