1Efficient inversions and duplications of mammalian regulatory DNA elements and gene clusters by CRISPR/Cas9显示文摘The human genome contains millions of DNA regulatory elements and a large number of gene clusters,most of which have not been tested experimentally.The clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated nuclease 9(Cas9)programed with a synthetic single-guide RNA(sgRNA)emerges as a method for genome editing in virtually any organisms.Here we report that targeted DNA fragment inversions and duplications could easily be achieved in human and mouse genomes by CRISPR with two sgRNAs.Specifically,we found that,in cultured human cells and mice,efficient precise inversions of DNA fragments ranging in size froma few tens of bp to hundreds of kb could be generated.In addition,DNA fragment duplications and deletions could also be generated by CRISPR through trans-allelic recombination between the Cas9-induced double-strand breaks(DSBs)on two homologous chromosomes(chromatids).Moreover,junctions of combinatorial inversions and duplications of the protocadherin(Pcdh)gene clusters induced by Cas9 with four sgRNAs could be detected.In mice,we obtained founders with alleles of precise inversions,duplications,and deletions of DNA fragments of variable sizes by CRISPR.Interestingly,we found that very efficient inversions were mediated by microhomology-mediated end joining(MMEJ)through short inverted repeats.We showed for the first time that DNA fragment inversions could be transmitted through germlines in mice.Finally,we applied this CRISPR method to a regulatory element of the Pcdha cluster and found a new role in the regulation of members of the Pcdhg cluster.This simple and efficient method should be useful in manipulating mammalian genomes to study millions of regulatory DNA elements as well as vast numbers of gene clusters.Jinhuan Li Jia Shou YaGuo Yuanxiao Tang YonghuWu Zhilian Jia Yanan Zhai Zhifeng Chen Quan Xu Qiang Wu 2015Journal of Molecular Cell Biology2015,7,4:4

5Deflection estimation of industrial robots with flexible joints显示文摘The application of industrial robots in manufacturing industries has received considerable concerns due to the high flexibility,multifunctionality,and cost-efficiency.It is well known that the robot positioning accuracy is susceptible to the load and motion of robots owing to the insufficient stiffness of robots.Therefore,the machining accuracy improvement has been a research focus in the robotic manufacturing industries in the last decade.To overcome the measurement difficulty of the joint torque and position as well as the complex dynamic coupling between rotors and links,two forward dynamics algorithms for the robot deflection estimation are proposed in this paper.The robot kinematics and dynamics algorithms considering the dynamic coupling between rotors and links are developed based on Lie theory.The forward dynamics equations of robots are solved via the proposed algorithms:the implicit numerical integration algorithm and numerical iterative estimation algorithm.When only the motor position is available,the implicit numerical integration algorithm is employed to solve the forward dynamics equations to estimate the joint torque and position.At the same time,when both the motor position and torque are available,the forward dynamics equations can be reorganized as algebraic equations and solved by the numerical iterative estimation algorithm.Simulations of a 6-DOF serial robot are performed to verify the accuracy of the proposed algorithms.Huan Liu Yaguo Lei Xiao Yang Wenlei Song Junyi Cao 2022Fundamental Research2022,2,3:1

8Remaining Useful Life Prediction With Partial Sensor Malfunctions Using Deep Adversarial Networks显示文摘In recent years,intelligent data-driven prognostic methods have been successfully developed,and good machinery health assessment performance has been achieved through explorations of data from multiple sensors.However,existing datafusion prognostic approaches generally rely on the data availability of all sensors,and are vulnerable to potential sensor malfunctions,which are likely to occur in real industries especially for machines in harsh operating environments.In this paper,a deep learning-based remaining useful life(RUL)prediction method is proposed to address the sensor malfunction problem.A global feature extraction scheme is adopted to fully exploit information of different sensors.Adversarial learning is further introduced to extract generalized sensor-invariant features.Through explorations of both global and shared features,promising and robust RUL prediction performance can be achieved by the proposed method in the testing scenarios with sensor malfunctions.The experimental results suggest the proposed approach is well suited for real industrial applications.Xiang Li Yixiao Xu Naipeng Li Bin Yang Yaguo Lei 2023IEEE/CAA Journal of Automatica Sinica2023,10,1:1
