1Delineating spatiotemporal and hierarchical development of human fetal innate lymphoid cells显示文摘Whereas the critical roles of innate lymphoid cells(ILCs)in adult are increasingly appreciated,their developmental hierarchy in early human fetus remains largely elusive.In this study,we sorted human hematopoietic stem/progenitor cells,lymphoid progenitors,putative ILC progenitor/precursors and mature ILCs in the fetal hematopoietic,lymphoid and non-lymphoid tissues,from 8 to 12 post-conception weeks,for single-cell RNA-sequencing,followed by computational analysis and functional validation at bulk and single-cell levels.We delineated the early phase of ILC lineage commitment from hematopoietic stem/progenitor cells,which mainly occurred in fetal liver and intestine.We further unveiled interleukin-3 receptor as a surface marker for the lymphoid progenitors in fetal liver with T,B,ILC and myeloid potentials,while IL-3RA–lymphoid progenitors were predominantly B-lineage committed.Notably,we determined the heterogeneity and tissue distribution of each ILC subpopulation,revealing the proliferating characteristics shared by the precursors of each ILC subtype.Additionally,a novel unconventional ILC2 subpopulation(CRTH2–CCR9+ILC2)was identified in fetal thymus.Taken together,our study illuminates the precise cellular and molecular features underlying the stepwise formation of human fetal ILC hierarchy with remarkable spatiotemporal heterogeneity.Chen Liu Yandong Gong Han Zhang Hua Yang Yang Zeng Zhilei Bian Qian Xin Zhijie Bai Man Zhang Jian He Jing Yan Jie Zhou Zongcheng Li Yanli Ni Aiqing Wen Yu Lan Hongbo Hu Bing Liu 2021Cell Research2021,31,10:2

4Cancer-macrophage hybrid membrane-camouflaged photochlor for enhanced sonodynamic therapy against triple-negative breast cancer显示文摘Sonodynamic therapy(SDT)has aroused considerable momentum in cancer therapy due to its abilities of deep penetration,low toxicity,and noninvasion,while insufficient tumor accumulation of sonosensitizers is a major obstacle for SDT effect.Here,we developed a 4T1 cancer cell-macrophage hybrid membrane(HM)-camouflaged sonosensitizer nanoplatform by encapsulating photochlor(HPPH)-loaded albumin nanoparticles(PHNPs).The experimental results proved that the HM-coated biomimetic NPs(PHNPs@HM)could express the characteristic membrane proteins of both cancer cells and macrophages,remarkedly enhancing the effective targeting and endocytosis to 4T1 cells through homologous adhesion recognition and immune escaping.Meanwhile,as a novel sonosensitizer,HPPH could generate amount of reactive oxygen species(ROS)under ultrasound(US)irradiation and exhibit obvious SDT efficiency to inhibit 4T1 tumor growth through ROS-induced cell apoptosis.This study provides a novel and multifunctional biomimetic sonosensitizer system to enhance SDT efficiency.Lishan Zhang Ting Yin Baozhen Zhang Chong Yan Chengyu Lu Lanlan Liu Ze Chen Hui Ran Qingxia Shi Hong Pan Aiqing Ma Lintao Cai 2022Nano Research2022,15,5:0
5A study of TCM master Yan Zhenghua’s medication rule in prescriptions for digestive system diseases based on Apriori and complex system entropy cluster显示文摘Objective:To explore Yan Zhenghua’s drug selection rule for treating digestive system diseases using data mining.Methods:The 609 medical records of digestive system diseases treated by Yan Zhenghua were collected and the herbs in these recipes were examined using a data mining technique.The correlativity between herb pairs and association rules was studied using an Apriori algorithm and the correlativity among multi-herbs was studied using a complex system entropy cluster technique.Results:Yan Zhenghua’s treatment of digestive system diseases featured 15 herbs prescribed at least 159 times each,22 herb pairs prescribed at least 155 times each,and eight frequently used herb core combinations.A confidence greater than 0.91 and a support level greater than 20%were achieved using the modified mutual information method.Conclusion:The data mining results conformed to findings from clinical practice.The data mining method is a valuable technique with which to study the experience of famous,elderly traditional Chinese medicine physicians.Jiarui Wu Weixian Guo Yan Tang Aiqing Han Bing Yang Dan Zhang Bing Zhang 2015Journal of Traditional Chinese Medical Sciences2015,2,4:0
