| 1 | Segmentation of Hematoxylin-Eosin stained breast cancer histopathological images based on pixel-wise SVM classifier显示文摘Hematoxylin-Eosin(HE) staining is the routine diagnostic method for breast cancer(BC), and large amounts of HE stained histopathological images are available for analysis. It is emergent to develop computational methods to efficiently and objectively analyze these images, with the aim of providing potentially better diagnostic and prognostic information for BC. This work focuses on analyzing our in-house HE stained histopathological images of breast cancer tissues. Since tumor nests(TNs) and stroma morphological characteristics can reflect the biological behaviors of breast invasive ductal carcinoma(IDC), accurate segmentation of TNs and the stroma is the first step towards the subsequent quantitative analysis. We first propose a method based on the pixel-wise support vector machine(SVM) classifier for segmenting TNs and the stroma, then extract four morphological characters related to the TNs from the images and investigate their relationships with the patients' 8-year disease free survival(8-DFS). The evaluation result shows that the classification based segmentation method is able to distinguish between TNs and stroma with 87.1% accuracy and 80.2% precision,suggesting that the proposed method is promising in segmenting HE stained IDC histopathological images. | 2 | Bacterial Diversity of Gut Content in Sea Cucumber(Apostichopus japonicus) and Its Habitat Surface Sediment显示文摘This study investigated the bacterial diversity of gut content of sea cucumber(Apostichopus japonicus) and its habitat surface sediment in a bottom enhancement area using PCR-based denaturing gradient gel electrophoresis(DGGE) technique. Bacterial diversity evaluation showed that the value of the Shannon-Wiener index of gut content in different intestinal segments of A. japonicus varied between 2.88 and 3.00, lower than that of the surrounding sediment(3.23). Phylogenetic analysis showed that bacterial phylotypes in gut content and the surrounding sediment of A. japonicus were closely related to Proteobacteria includingγ-, α-, δ-and ε-proteobacteria, Bacteroidetes, Firmicute, and Actinobacteria, of which γ-proteobacteria were predominant. These results suggested that the sea cucumber A. japonicus was capable of feeding selectively, and PCR-DGGE was applicable for characterizing the bacterial community composition in gut content and the surrounding sediment of sea cucumber. | 10 | Targeting Kindlin-2 in adipocytes increases bone mass through inhibiting FAS/PPARγ/FABP4 signaling in mice显示文摘Osteoporosis(OP)is a systemic skeletal disease that primarily affects the elderly population,which greatly increases the risk of fractures.Here we report that Kindlin-2 expression in adipose tissue increases during aging and high-fat diet fed and is accompanied by decreased bone mass.Kindlin-2 specific deletion(K2KO)controlled by Adipoq-Cre mice or adipose tissue-targeting AAV(AAV-Rec2-CasRx-sgK2)significantly increases bone mass.Mechanistically,Kindlin-2 promotes peroxisome proliferator-activated receptor gamma(PPARγ)activation and downstream fatty acid binding protein 4(FABP4)expression through stabilizing fatty acid synthase(FAS),and increased FABP4 inhibits insulin expression and decreases bone mass.Kindlin-2 inhibition results in accelerated FAS degradation,decreased PPARγactivation and FABP4 expression,and therefore increased insulin expression and bone mass.Interestingly,we find that FABP4 is increased while insulin is decreased in serum of OP patients.Increased FABP4 expression through PPARγactivation by rosiglitazone reverses the high bone mass phenotype of K2KO mice.Inhibition of FAS by C75 phenocopies the high bone mass phenotype of K2KO mice.Collectively,our study establishes a novel Kindlin-2/FAS/PPARγ/FABP4/insulin axis in adipose tissue modulating bone mass and strongly indicates that FAS and Kindlin-2 are new potential targets and C75 or AAV-Rec2-CasRx-sgK2 treatment are potential strategies for OP treatment. | Wanze Tang Zhen Ding Huanqing Gao Qinnan Yan Jingping Liu Yingying Han Xiaoting Hou Zhengwei Liu Litong Chen Dazhi Yang Guixing Ma Huiling Cao | 2023 | Acta Pharmaceutica Sinica B2023,13,11: | 0 |
| 11 | A Preliminary Survey of Arbuscular Mycorrhizal Fungi Associated with Marsh Plants in Lhalu Wetland,Suburban Lhasa,South Tibet显示文摘A survey was made of the spore community of arbuscular mycorrhizal fungi (AMF) and root colonization by AMF in 16 plant species in Lhalu wetland on the outskirts of Lhasa city in Tibet. It was found that 13 of the 16 plant species investigated (81. 5% ) formed arbuscular mycorrhizal structures and dark septate endophytic fungi colonized the roots of most species. Total AMF colonization ranged from 0 to 82. 6% in dicots and 0 to 54. 5% in monocots. Both total AMF and arbuscular colonization were greater in dicots than that in monocots. A total of 48 taxa representing 7 genera of AMF were isolated and identified. Of these,9 species belonged to Acaulospora,2 to Appendicispora,34 to Glomus,and 1 each to Pacispora,Paraglomus and Scutellospora. Spores of Glomus aggregatum,G. deserticola and G. etunicatum were most common and abundant in the spore survey. Spores of 8 to 26 AMF species were isolated from the rhizosphere soil of individual plant species. Spore densities in soil associated with the 16 plant species ranged from 20 to 475 per 20 g soil,with an average of (92. 3 ± 14. 6). Species richness of AMF ranged from 6 to 12. 7. There were no significant differences between dicots and monocots in AMF spore density or species richness. | 12 | Mechanical Stress Induces Neuroendocrine and Immune Responses of Sea Cucumber(Apostichopus japonicus)显示文摘Grading procedure in routine sea cucumber hatchery production is thought to affect juvenile sea cucumber immunological response. The present study investigated the impact of a 3-min mechanical perturbation mimicking the grading procedure on neuroendocrine and immune parameters of the sea cucumber Apostichopus japonicus. During the application of stress, concentrations of noradrenaline and dopamine in coelomic fluid increased significantly, indicating that the mechanical perturbation resulted in a transient state of stress in sea cucumbers. Coelomocytes concentration in coelomic fluid increased transiently after the beginning of stressing, and reached the maximum in 1 h. Whereas, coelomocytes phagocytosis at 3 min, superoxide anion production from 3 min to 0.5 h, acid phosphatase activity at 0.5 h, and phenoloxidase activity from 3 min to 0.5 h were all significantly down-regulated. All of the immune parameters recovered to baseline levels after the experiment was conducted for 8 h, and an immunostimulation occurred after the stress considering the phagocytosis and acid phosphatase activity. The results suggested that, as in other marine invertebrates, neuroendocrine/immune connections exist in sea cucumber A. japonicus. Mechanical stress can elicit a profound influence on sea cucumber neuroendocrine system. Neuroendocrine messengers act in turn to modulate the immunity functions.
