2Relationship between genetic polymorphisms of metabolizing enzymes CYP2E1, GSTM1 and Kazakh's esophageal squamous cell cancer in Xinjiang, China显示文摘AIM: To analyze the relationship between genetic polymorphisms of metabolizing enzymes CYP2E1, GSTM1 andKazakh's esophageal squamous cell cancer in China.METHODS: The genotypes of cytochromes P450 (CYP) 2E1 and glutathione S-transferase (GST) M1 were investigated by polymerase chain reaction (PCR) and restriction fragment length polymorphism (RFLP) following PCR in 104 Kazakh's patients with esophageal cancer (EC) and 104 non-cancer controls.RESULTS: The frequency of CYP2E1 c1/c1 genotype was significantly higher in patients with cancer (77.9%) thanin control subjects (24.0%) (P<0.05; OR, 11.13; 95%CI,5.84-21.22). The difference of GSTM1 null was significantly more frequent in the cancer (34.6%) vsthe control group (3.8%) (P<0.05; OR, 13.24; 95%CI, 4.50-38.89). On the other hand, the combination of GSTM1 presence and CYP2E1 c1/c1 genotypes increased the risk for cancer (P<0.05;OR, 13.42; 95%CI, 6.29-28.3).CONCLUSION: The CYP2E1 c1/c1, GSTM1 deletion genotypes are genetically susceptible biomarkers for ESCC in Kazakh population. Individuals with allele c1 of RsaI polymorphic locus for CYP2E1 may increase the risk of ESCC. 3p53 polymorphism in human papillomavirus-associated Kazakh's esophageal cancer in Xinjiang,China显示文摘AIM: To investigate the relationship between p53 codon 72 polymorphism and human papillomavirus (HPV) type 16 infection in Kazakh's esophageal cancer (EC) in Xinjiang, China.METHODS: Encoding regions of p53codon 72 and HPV-16 E6 were amplified by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and polymerase chain reaction (PCR) methods using pairs of primary esophageal squamous cell carcinoma (SCC) tissue and corresponding normal mucosa, which were collected from 104 patients of Kazakh in Xinjiang, China. RESULTS: Only arginine allele was detected in 70.1% (39/55) of HPV-16-E6-positive cases but only in 40.8% (20/49) of HPV-16-E6-negative cases (P<0.05; OR, 3.53; 95% CI, 1.57-7.98). In contrast, such a significant correlation between p53 polymorphism and HPV infection was not evident in corresponding normal mucosae. The allele frequency of Arg allele in cancer cases (0.68) was higher than that in normal mucosa samples (0.54) (P<0.05; OR, 1.80; 95% CI, 1.21-2.69).CONCLUSION: p53 codon 72 Arg homozygous genotype is one of the high-risk genetic factors for HPV-associated SCC of Kazakh. 4Comparative proteome analysis of untreated and Helicobacter pylori-treated HepG2显示文摘AIM: To investigate the pathological effect of Helicobacter pylori(H pylori) on human hepatic ceils, proteomic methods were used to find and to identify proteins that were overexpressed in HepG2 cells treated by H pylori.METHODS: H pylori was co-cultured with HepG2 for 6 h. Two-dimensional gel electrophoresis was used to gainthe protein expression pattern of untreated and H pyloritreated HepG2. After staining and image analysis,spots of interest were isolated and subjected to massspectrometry.RESULTS: Seven proteins, which were up-regulated inH p ylori-treated HepG2 cells, were identified. 5Effects of sodium β-aescin on expression of adhesion molecules and migration of neutrophils after middle cerebral artery occlusion in rats显示文摘AIM: To investigate the effects of sodium β-aescin on neutrophil migration and expression of adhesion molecules (ICAM-1 and E-selectin) after middle cerebral artery occlusion (MCAO) in rats. METHODS: Rats were pretreated with sodium β-aescin for 7 d and then subjected to cerebral ischemia/reperfusion (I/R) injury induced by an MCAO. After a 2-h ischemia and a 24-h reperfusion, the infarct volume and neurological deficit were determined by the method of TTC staining and the Longa’s score. The effect of sodium β-aescin on the migration of neutro- phils was evaluated by measuring the activity of myeloperoxidase (MPO) enzyme. The expressions of adhesion molecules were determined by immunohistochemistry and Western blot. RESULTS: Sodium β-aescin significantly reduced the cerebral infarct volume and ameliorated the neurological deficit (P<0.05 or P<0.01). The MPO activity and the expressions of ICAM-1 and E-selectin in the vehicle-treated rats were increased significantly (P<0.01) after cerebral I/R. After treatment with sodium β-aescin, the enzymatic activity of MPO and the expressions of these adhesion molecules were significantly reduced compared with the vehicle-treated group (P<0.05 or P<0.01). 6Advances in direct production of value-added chemicals via syngas conversion显示文摘Syngas conversion to fuels and chemicals is one of the most challenging subjects in the field of C1 chemistry. It is considered as an attractive alternative non-petroleum-based production route. The direct synthesis of olefins and alcohols as high value-added chemicals from syngas has drawn particular attention due to its process simplicity, low energy consumption and clean utilization of carbon resource, which conforms to the principles of green carbon science. This review describes the recent advances for the direct production of lower olefins and higher alcohols via syngas conversion. Recent progress in the development of new catalyst systems for enhanced catalytic performance is highlighted. 7Cloning and expression of ornithine decarboxylase gene from human colorectal carcinoma显示文摘AIM: To construct and express ODC recombinant gene for further exploring its potential use in early diagnosis of colorectal carcinoma.METHODS: Total RNA was extracted from colon cancer tissues and amplified by reverse-transcription PCR with two primers, which span the whole coding region of ODC. The synthesized ODC cDNA was cloned into vector pQE-30 at restriction sites BamH I and Sal I which constituted recombinant expression plasmid pQE30-ODC. The sequence of inserted fragment was confirmed by DNA sequencing,the fusion protein including 6His-tag was facilitated for purification by Ni-NTA chromatographic column.RESULTS: ODC expression vector was constructed and confirmed with restriction enzyme digestion and subsequent DNA sequencing. The DNA sequence matching on NCBI Blast showed 99 % affinity. The vector was transformed into E.coli M15 and expressed. 8Effects of β-aescin on apoptosis induced by transient focal cerebral ischemia in rats显示文摘AIM: To investigate the effects of β-aescin on apoptosis induced by transient focal brain ischemia in rats. METHODS: Rats were pretreated with β-aescin for 7 d and then subjected to brain ischemia/reperfusion (I/R) injury induced by a middle cerebral artery occlusion, After 2 h ischemia and 24 h reperfusion, Hematoxylin-Eosin (HE) staining, in situ end-labeling of nuclear DNA fragmentation (TUNEL) were employed to determine the level of apoptosis, The expressions of caspase-3 and Bcl-2 in the cortex were determined by immunohistochemistry and Western blot. The release of cytochrome c was analyzed by Western blot. RESULTS: The increased numbers of HE- and TUNEL-positive staining cells were significantly observed at 24 h after reperfusion, The immunoreactivity was inhibited by β-aescin (30, 60 mg/kg) (P<0.01 or P<0.05 vs vehicle-treated), After cerebral I/R, cytochrome c was released into the cytosol and caspase-3 was activated, whereas Bcl-2 expression was inhibited. β-Aescin (30, 60 mg/kg) markedly inhibited the expression of caspase-3 and the release of cytochrome c, and up-regulated the expression of Bcl-2 (P<0.05, P<0.01 vs vehicle-treated). CONCLUSION: β-Aescin could potently inhibit caspase-3 activation and the release of cytochrome c, increasing the expression of Bcl-2 after cerebral I/R in rats, These findings on the inhibitory effects of β-aescin on brain ischemic injury-induced apoptosis might have important theoretical basis for the treatment on ischemic cerebrovascular diseases.Xia-minHU YanZHANG Fan-dianZENG 2004Acta Pharmacologica Sinica2004,25,10:6
9Nonlinear system PID-type multi-step predictive control显示文摘A compound neural network was constructed during the process of identification and multi-step prediction. Under the PlD-type long-range predictive cost function, the control signal was calculated based on gradient algorithm. The nonlinear controller' s structure was similar to the conventional PID controller. The parameters of this controller were tuned by using a local recurrent neural network on-line. 13Human endostatin gene transfer,either naked or with liposome,has the same inhibitory effect on growth of mouse liver tumor cells in vivo显示文摘AIM: To explore a safe and efficient strategy of tumortherapy using anti-angiogenetic agents.METHODS: Endostatin gene with a signal sequence ofhuman IgG y chain was amplified by PCR and cloned intopVAXl plasmid which was the only vector authorized byFDA in clinical trial to construct a recombinant plasmidnamed as pVAX-sEN. The recombinant plasmid wasdetected with EcoRI/KpnI and DNA sequencing. BALB/c micebearing hepatocarcinoma cell line H22 were treated withnaked pVAX-sEN or liposome-DNA complex in which thedose of DNA and the ratio of DNA and liposome weredifferent from each other. To compare the efficiency ofgene transfection, expression of endostatin at the treatedtumor site was assayed with ELISA. To investigate the effectof pVAXI-sEN on hepatocellular carcinoma, pVAX-sENeither naked or in liposome-DNA complex was injectedinto BALB/c mice bearing H22, then the diameter of tumorswas measured, microvessel density was detected byimmunohistochemistry, endostatin expression in vivo wasassayed at different time points.RESULTS: DNA sequencing showed the endostatin genewith the signal peptide was correctly cloned. In situ geneexpression assay indicated that both the ratio of DNAand liposome and the dose of DNA could affect the genetransfection efficiency. Interestingly, naked pVAX-sEN hada similar in situ endostatin expression to pVAX-sEN withliposome. 14An overview on nonlinear porous flow in low permeability porous media显示文摘This paper gives an overview on nonlinear porous flow in low permeability porous media, reveals the microscopic mechanisms of flows, and clarifies properties of porous flow fluids. It shows that, deviating from Darcy's linear law, the porous flow characteristics obey a nonlinear law in a low-permeability porous medium, and the viscosity of the porous flow fluid and the permeability values of water and oil are not constants. Based on these characters, a new porous flow model, which can better describe low permeability reservoir, is established. This model can describe various patterns of porous flow, as Darcy's linear law does. 16A novel method for preparation of tissue microarray显示文摘AIM:To improve the technique of tissue microarray (tissue chip).METHODS: A new tissue microarraying method was invented with a common microscope installed with a special holing needle, a sampling needle, and a special box fixing paraffin blocks on the microscope slide carrier. 17Risk prediction models for lung cancer:Perspectives and dissemination显示文摘Objective: The objective was to systematically assess lung cancer risk prediction models by critical evaluation of methodology, transparency and validation in order to provide a direction for future model development.Methods: Electronic searches(including PubMed, EMbase, the Cochrane Library, Web of Science, the China National Knowledge Infrastructure, Wanfang, the Chinese BioMedical Literature Database, and other official cancer websites) were completed with English and Chinese databases until April 30 th, 2018. Main reported sources were input data, assumptions and sensitivity analysis. Model validation was based on statements in the publications regarding internal validation, external validation and/or cross-validation.Results: Twenty-two studies(containing 11 multiple-use and 11 single-use models) were included. Original models were developed between 2003 and 2016. Most of these were from the United States. Multivariate logistic regression was widely used to identify a model. The minimum area under the curve for each model was 0.57 and the largest was 0.87. The smallest C statistic was 0.59 and the largest 0.85. Six studies were validated by external validation and three were cross-validated. In total, 2 models had a high risk of bias, 6 models reported the most used variables were age and smoking duration, and 5 models included family history of lung cancer.Conclusions: The prediction accuracy of the models was high overall, indicating that it is feasible to use models for high-risk population prediction. However, the process of model development and reporting is not optimal with a high risk of bias. This risk affects prediction accuracy, influencing the promotion and further development of the model. 18Improvement of tribological behavior of a Ti-Al-V alloy by nitrogen ion implantation显示文摘The tribological properties especially wear and hardness of a Ti-Al-V alloy with nitrogen implantation (energy 60 keV) were investigated. The implantation was carried out at fluences range from 1×1016 to 4×1017 ions/cm2. Glancing angle X-ray diffraction (GAXRD) and X-ray photoelectron spectroscopy (XPS) analyses were performed to obtain surface characterization of the implanted sample. The unimplanted and implanted samples were also annealed at 600 ℃ in order to understand the influence of annealing on the tribological properties of Ti-Al-V. The hardness shows significant improvement at the higher fluence. After annealing at 600 ℃, the friction coefficient exhibits a relative decrease for the nitrogen-implanted samples. In addition, the wear rates of the implanted samples exhibits a great decrease after annealing at 600 ℃.
