| 1 | Anti-tumor effect of pEgr-interferon-γ-endostatin gene-radiotherapy in mice bearing Lewis lung carcinoma and its mechanism显示文摘Background Gene-radiotherapy, the combination of gene therapy and radiation therapy, is a new paradigm for cancer treatment. To enhance anti-tumor effect of gene-radiotherapy, in this study we construct a radiation-inducible dual-gene co-expression vector pEgr-interferon(IFN)-γ- endostatin and studied the anti-tumor effect of pEgr-IFN-γ-endostatin gene-radiotherapy in mice bearing Lewis lung carcinoma and its mechanism.Methods Gene recombinant technique was used to construct dual-gene co-expression plasmid pEgr-IFN-γ-endostatin, and single-gene expression plasmid pEgr-IFN-γ and pEgr-endostatin. The plasmids packed by liposome were injected locally into the tumors of the mice, and the tumors were irradiated with 5 Gy X-ray 36 hours later. The tumor growth rate at different time and mean survival period of the mice were observed. Cytotoxic activity of splenic cytotoxic T-lymphocyte (CTL), natural killer (NK) cell and tumor necrosis factor (TNF)-α secretion activity of peritoneal macrophages of the mice in various groups were evaluated 15 days after irradiation. The intratumor micro-vessel density was evaluated by immunohistochemical staining 10 days after irradiation.Results The tumor growth rate of the mice in dual-gene-radiotherapy group was significantly lower than those in control group, 5 Gy group and single-gene-radiotherapy group at different time after gene-radiotherapy, and the mean survival period of which was longer. Cytotoxic activity of splenic CTL, NK and TNF-α secretion activity of peritoneal macrophages of the mice in dual-gene-radiotherapy group were significantly higher than those in control group, 5 Gy X-ray irradiation group and pEgr-endostatin gene-radiotherapy group 15 days after irradiation. The intratumor micro-vessel density of the mice in dual-gene-radiotherapy group was significantly lower than those in control group, 5 Gy X-ray irradiation group and pEgr-IFN-γgene-radiotherapy group. | 4 | Numerical study of corner separation in a linear compressor cascade using various turbulence models显示文摘Three-dimensional corner separation is a common phenomenon that significantly affects compressor performance.Turbulence model is still a weakness for RANS method on predicting corner separation flow accurately.In the present study,numerical study of corner separation in a linear highly loaded prescribed velocity distribution(PVD) compressor cascade has been investigated using seven frequently used turbulence models.The seven turbulence models include Spalart–Allmaras model,standard k–e model,realizable k–e model,standard k–x model,shear stress transport k–x model,v^2–f model and Reynolds stress model.The results of these turbulence models have been compared and analyzed in detail with available experimental data.It is found the standard k–e model,realizable k–e model,v^2–f model and Reynolds stress model can provide reasonable results for predicting three dimensional corner separation in the compressor cascade.The Spalart–Allmaras model,standard k–x model and shear stress transport k–x model overestimate corner separation region at incidence of 0°.The turbulence characteristics are discussed and turbulence anisotropy is observed to be stronger in the corner separating region. | Liu Yangwei Yan Hao Liu Yingjie Lu Lipeng Li Qiushi | 2016 | Chinese Journal of Aeronautics2016,29,3: | 11 |
| 5 | ^(7)Be and^(210)Pb radioactivity and implications on sources of surface ozone at Mt.Waliguan显示文摘Beryllium-7 (7Be) and lead-210 (210Pb) radioac- tivity in aerosols collected, from October 2002 to January 2004 at Mt. Waliguan, by the Global Atmospheric Watch (GAW) Station, Qinghai Province is presented. The data were analyzed together with simultaneously measured surface ozone concentrations. We found that short time variations of 7Be and 210Pb were linked to alternations of synoptic weather around the Mt. Waliguan region. 210Pb showed the lowest concentration in summer while Be showed no obvious sea- 7 sonal changes. Relatively high Be and 7 210 Pb radioactivity was observed at Mt. Waliguan when compared with the ob- servations at other mountain sites in other parts of the world. Surface ozone and 7Be showed a consistent seasonalvariation. Surface ozone correlated fairly well with 7Be/210Pb ratio. This suggested that vertical transport from higher altitudes of the atmosphere has predominant effects on the budget of surface ozone at Mt. | 6 | Distribution Characteristics and Metallogenic Regularity of Graphite Deposits in Qinling Orogen,China显示文摘Qinling orogen is one of the five main repository distribution provinces of large scale graphite resources. Graphite occurrence strata are multitudinous including Neo Archaean group to Neopaleozoic. Mineral deposit types are complete consisting of crystal flaky graphite deposit and aphanitic graphite deposit, ore types of the former are main graphite gneiss, graphite schist and graphitized marble, and ore types of the latter are main graphite layer metamorphosised from coal bed and graphitic carbargilite. At present, most graphite deposits with low research degree only implement preliminary investigations of the graphite resource, which has a good prospect. Based on the basic material of dozens of graphite deposits, spots and plays discovered in the Qinling orogen, this paper applied geologic multidisciplinary analysis method to disclose the graphite deposit types of the Qinling orogen, mainly through outdoors geologic survey and specimen examination with analysis of some typical deposits: regional metamorphism crystal graphite deposits and thermo-contact metamorphic aphanitic graphite deposits, with the control of latitudinally trending regional tectonic, graphite deposits of the Qinling orogen distribute sublatitudinally trending three large ore belts: I the Neoarchean–Proterozoic basement crystal graphite enrichment deposits zone along the south margin of North China plate; II the Carboniferous intermountain basin group aphanitic graphite enrichment deposits zone near the Shangdan suture of the West Qinling; III Paleozoic crystal graphite enrichment deposits zone in the Qinling paleomicroplate of the west part of the East Qinling.Conclusions are reached from multiple inquiries: Qinling multi-cycle complex continental collision orogen zone has experienced multiple tectonic framework transformation and polyphase tectonic thermal event from Neo Archean Erathem adjointing multiple graphite tectonic mineralization cycle. In the light of chief control of mineralization and ore types, each of the three large graphite deposits belts of the Qinling orogen has its own genesis: I graphite deposits enriched metallogenetic zone is that regional tectogenetic movement from Neo Archaean caused polyphase metapepsis, which superimposed up and reconstructed Neo Archean Erathem to Proterozoic basement, forming regional metamorphic big flake crystal graphite deposits; II graphite deposits enriched metallogenetic zone is that polyphase regional tectogenetic movement associated with multiple heating caused by multiphase igneous intrusion, which resulted in original coal layers emerging multiple thermo-contact metamorphisms and formed thermo-contact metamorphic aphanitic graphite deposits. III graphite deposits enriched metallogenetic zone is that Palaeozoic cap–rock experienced polyphase metapepsis, forming regional metamorphic finely flake crystal graphite deposits. | 7 | Modified k-ω model using kinematic vorticity for corner separation in compressor cascades显示文摘A new method of modifying the conventional k-ω turbulence model for corner separation is proposed in this paper. The production term in the ω equation is modified using kinematic vorticity considering fluid rotation and deformation in complex geometric boundary conditions. The corner separation flow in linear compressor cascades is calculated using the original k-ω model, the modified k-ω model and the Reynolds stress model(RSM). The numerical results of the modified model are compared with the available experimental data, as well as the corresponding results of the original k-ω model and RSM. In terms of accuracy, the modified model, which significantly improves the performance of the original k-ω model for predicting corner separation, is quite competitive with the RSM. | 8 | A combined study of SHRIMP U-Pb dating, trace element and mineral inclusions on high-pressure metamorphic overgrowth zircon in eclogite from Qinglongshan in the Sulu terrane显示文摘Methods recently advanced for discrimination on the genesis of metamorphic zircon, such as analysis of mineral inclusions and trace elements, provide us powerful means to distinguish zircon overgrowth during high-pressure metamorphism. Zircons in ultrahigh-pressure eclogite from Qinglongshan in the Sulu terrane were studied by the SHRIMP U-Pb method in combining with trace element and mineral inclusion analyses. No inherited core was identified in the analyzed zircons by means of cathodoluminescence images. The occurrence of high-pressure metamorphic mineral inclusions in zircon, such as garnet, omphacite, rutile, and the flat HREE pattern in zircon indicate that the zircon formed at high-pressure metamorphic conditions. | 11 | Transplantation of human limbal cells cultivated on amniotic membrane for reconstruction of rat corneal epithelium after alkaline burn显示文摘Background The transplantation of limbal epithelial cells cultivated on amniotic membrane is a newly developed treatment for limbal stem cell deficiency. The purpose of our study was to investigate the biological characteristics of limbal epithelial cells and evaluate the effect of transplantation of cultivated human limbal epithelial cells on ocular surface reconstruction in limbal stem cell deficiency rat model. Methods Human limbal cells were isolated and cultivated in vitro. Cytokertins 3, 12, and 19 (K3, K12 and K19) and p63 were detected by immunofluorescent staining or RT-PCR. BrdU labelling test was used to identify the slow cycling cells in the cultures. Limbal stem cell deficiency was established in rat cornea by alkali burn. Two weeks after injury, the rats received transplants of human limbal stem cells cultivated on amniotic membrane carrier. The therapeutic effect was evaluated by slit lamp observation, Hemotoxin and Eosin (HE) staining and immunofluorescent staining.Results On day 7 in primary culture, p63 and K19 were strongly expressed by most cells but only a few cells expressed K3. On days 14 and 21, p63 and K19 were still expressed by a majority of cells, but the expressive intensity of p63 decreased in a number of cells, while the proportion of K3 positive cells increased slightly and some cells coexpressed p63 and K3. RT-PCR showed that gene expression of both p63 and K12 were positive in cultivated limbal cells, but in mature superficial epithelial cells, only K12 was detected. BrdU labelling test showed that most cells were labelled with BrdU after 7 days’ labelling and BrdU label retaining cells were observed after chasing for 21 days with BrdU free medium. For in vivo test, slit lamp observation, HE staining and immunofluorescent staining showed that the rats receiving transplant of human limbal stem cells cultivated on amniotic membrane grew reconstructed corneas with intact epithelium, improved transparency and slight or no neovascularization. A majority of epithelial cells of the reconstructed cornea were positive to antihuman nuclear antibody and cells expressing K3 were found mainly in superfacial epithelium.Conclusions Limbal stem cells can be cultivated in vitro: the cells are characterized by high proliferation and slow cycling and identified as p63/K19 positive and K3/K12 negative. During culture, some stem cells can proliferate and differentiate into mature cornea epithelial cells. Amniotic membrane is a suitable carrier for limbal stem cells. | 15 | Phenotypic and genetic parameter estimation of juvenile growth and bottom color traits in half-smooth tongue sole,Cynoglossus semilaevis显示文摘Half-smooth tongue sole(Cynoglossus semilaevis) is a commercially important fish species. There were no reports about accurate genetic parameters of growth traits in C. semilaevis, despite efforts aimed at the culture of this species. Therefore, the main goal of this study was to estimate the parameters for the early growth traits(total length, body depth and body weight) of C. semilaevis based on the data obtained from 80 families. The trait of fish with pure white bottom or not(named as the trait of bottom color) was also analyzed. All the heritabilities estimated for the traits were medium, which varied from 0.210 to 0.362. Genetic correlations among growth traits were highly positive, varying from 0.913 to 0.959, indicating that selection of one of the three traits would result in correlated increase in the others. Also, genetic correlations between bottom color and growth traits were positive and varying from 0.241 to 0.353, suggesting that selection for the bottom color can be utilized to enhance the selection of growth traits. In addition, fish from the top performing 16 full-sib families were selected as broodstock, based on the above analysis and the comparison of family breeding value. | 16 | Postharvest Biological Control of Apple Anthracnose by Bacillus subtilis BS80-6,A Mutant Strain through Ion Implantation显示文摘The original Bacillus subtilis was isolated from the soil of an apple orchard,and B. subtilis strain BS80-6,selected from Bacillus subtilis by ion implantation,was used as an antagonist for postharvest biological control against apple anthracnose( colletotrichum gloeosporioides). The mechanisms of action and efficacy of Bacillus subtilis strain BS80-6 against apple anthracnose caused by colletotrichum gloeosporioides were studied in vitro and on apples in controlled and semi-commercial conditions. An application of cell suspension( 108 cells per mL) of the antagonist in artificial wounds of apples reduced growth of C. gloeosporioides after storage at different temperatures. The inhibitory actions of cell culture to hyphal growth and to spores germination were 86. 6% and 98. 65%,respectively,and the control efficacy of cell culture against the disease was 60. 34% at room temperature. The results showed that BS80-6 was more effective against apple anthracnose.The cell culture of BS80-6 received better control efficacy against apple anthracnose than culture filtrates and autoclaved cell. All treatments significantly inhibited the disease both in vitro and in vivo compared with control. Fruits treated with cell culture and stored at 10 ℃ had lower infection rate,more delayed formation of acervwlus and smaller lesion diameter than those stored at 20 ℃. There was better control efficacy in fruits inoculated with spores after application of BS80-6. The mechanisms of BS80-6 activity appeared to involve production of an antifungal substance,deformation of hyphal cell and disintegration of cell wall.
