| 2 | SlMYB75,an MYB-type transcription factor,promotes anthocyanin accumulation and enhances volatile aroma production in tomato fruits显示文摘Genetic manipulation of genes to upregulate specific branches of metabolic pathways is a method that is commonly used to improve fruit quality.However,the use of a single gene to impact several metabolic pathways is difficult.Here,we show that overexpression of the single gene SlMYB75(SlMYB75-OE)is effective at improving multiple fruit quality traits.In these engineered fruits,the anthocyanin content reached 1.86mg g−1 fresh weight at the red-ripe stage,and these SlMYB75-OE tomatoes displayed a series of physiological changes,including delayed ripening and increased ethylene production.In addition to anthocyanin,the total contents of phenolics,flavonoids and soluble solids in SlMYB75-OE fruits were enhanced by 2.6,4,and 1.2 times,respectively,compared to those of wild-type(WT)fruits.Interestingly,a number of aroma volatiles,such as aldehyde,phenylpropanoid-derived and terpene volatiles,were significantly increased in SlMYB75-OE fruits,with some terpene volatiles showing more than 10 times higher levels than those in WT fruits.Consistent with the metabolic assessment,transcriptomic profiling indicated that the genes involved in the ethylene signaling,phenylpropanoid and isoprenoid pathways were greatly upregulated in SlMYB75-OE fruits.Yeast one-hybrid and transactivation assays revealed that SlMYB75 is able to directly bind to the MYBPLANT and MYBPZM cis-regulatory elements and to activate the promoters of the LOXC,AADC2 and TPS genes.The identification of SlMYB75 as a key regulator of fruit quality attributes through the transcriptional regulation of downstream genes involved in several metabolic pathways opens new avenues towards engineering fruits with a higher sensory and nutritional quality. | Wei Jian Haohao Cao Shu Yuan Yudong Liu Juanfang Lu Wang Lu Ning Li Jianhui Wang Jian Zou Ning Tang Chan Xu Yulin Cheng Yanqiang Gao Wanpeng Xi Mondher Bouzayen Zhengguo Li | 2019 | Horticulture Research2019,6,1: | 39 |
| 3 | Manipulation of SlMXl for enhanced carotenoids accumulation and drought resistance in tomato显示文摘Plants are the ultimate source of nutrients in the human diet. To ensure adequate availability of high quality food for an increasing world population, traits including improved tolerance of stresses and nutrient levels need to be selected in crops, both individually and in combination. Here we report the identification of Sl MX1 encoding a MIXTA-like MYB transcription factor in tomato that simultaneously modulates drought resistance and metabolic processes through regulating key structural and regulatory genes of the corresponding pathways. Over-expression of Sl MX1 results in substantially increased drought tolerance and improved fruit quality, while knocking down Sl MX1 resulted in the opposite phenotypes. | 4 | N_2O Emission from Paddy Field under Different Rice Planting Modes显示文摘Measurements of N2O emissions from conventional rice cultivation (CRC), CRC with straw mulching, system of rice intensification (SRI) and SRI with plastic film mulching were conducted through static chamber/gas-chromatography techniques. The results show that daily fluctuation of N2O emissions in jointing stage are much higher than in others. A type peak of N2O seasonal emission presented between jointing and bearing stages companying with high daily average temperature and low precipitation. Biomass and leaf stomatal conductance were observed. Total quantities of N2O emission were budgeted. The results showed that after jointing stage the average N2O emission flux of SRI with plastic film mulching increased significantly than CRC with straw mulching and SRI, the leaf stomatal conductance of those showed the same trend (p< 0.05 ). Yield and total quantity of N2O emission in CRC with straw mulching enhanced 13.7% and 10.7% compared with those of CRC, respectively. | 5 | Intrastriatal glial cell line-derived neurotrophic factors for protecting dopaminergic neurons in the substantia nigra of mice with Parkinson disease显示文摘BACKGROUND:Substantia nigra is deep in position and limited in range,the glial cell line-derived neurotrophic factor(GDNF)injection directly into substantia nigra has relatively greater damages with higher difficulty.GDNF injection into striatum,the target area of dopaminergic neuron,may protect the dopaminergic neurons in the compact part of substantia nigra through retrograde transport.OBJECTIVE:To investigate the protective effect of intrastriatal GDNF on dopaminergic neurons in the substantia nigra of mice with Parkinson disease(PD),and analyze the action pathway.DESIGN:A controlled observation.SETTING:Neurobiological Laboratory of Xuzhou Medical College.MATERIALS:Twenty-four male Kunming mice of 7-8 weeks old were used.GDNF,1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)were purchased from Sigma Company(USA);LEICAQWin image processing and analytical system.METHODS:The experiments were carried out in the Neurobiological Laboratory of Xuzhou Medical College from September 2005 to October 2006.The PD models were established in adult KunMing mice by intraperitoneal injection of MPTP.The model mice were were randomly divided into four groups with 6 mice in each group:GDNF 4-day group,phosphate buffer solution(PSB)4-day group,GDNF 6-day group and PSB 6-day group.Mice in the GDNF 4 and 6-day groups were administrated with 1 μL GDNF solution(20 μg/L,dispensed with 0.01 mol/L PBS)injected into right striatum at 4 and 6 days after model establishment.Mice in the PSB 4 and 6-day groups were administrated with 0.01 mol/L PBS of the same volume to the same injection at corresponding time points.② On the 12th day after model establishment,the midbrain tissue section of each mice was divided into 3 areas from rostral to caudal sides.The positive neurons of tyroxine hydroxylase(TH)and calcium binding protein(CB)with obvious nucleolus and clear outline were randomly selected for the measurement,and the number of positive neurons in unit area was counted.MAIN OUTCOME MEASURES:Number of positive neurons of TH and CB in midbrain substantia nigra of mice in each group.RESULTS:All the 24 mice were involved in the analysis of results.The numbers of TH+ and CB+ neurons in the GDNF 4-day group(54.33±6.92,46.33±5.54)were obviously more than those in the PBS 4-day group(27.67±5.01,21.50±5.96,P < 0.01).The numbers of TH+ and CB+ neurons in the GDNF 6-day group(75.67±5.39,69.67±8.69)were obviously more than those in the PBS 6-day group(27.17±4.50,21.33±5.72,P < 0.01)and those in the GDNF 4-day group(P < 0.01).CONCLUSION:Intrastriatal GDNF can protect dopaminergic neurons in substantia nigra of PD mice,and it may be related to the increase of CB expression. | Chenghua Xiao Yanqiang Wang Hongmei Liu Hongjun Wang Junping Cao Dianshuai Gao | 2007 | Neural Regeneration Research2007,2,4: | 4 |

| 7 | Expression of calbindin D28K in substantia nigra of model rats with Parkinson disease显示文摘BACKGROUND: Previous researches suggested that expression level of calbindin D28K mRNA decreased in substantia nigra (SN) of model rats with Parkinson disease (PD), and this might be related to the decrease of anti-degeneration potentials of dopaminergic neurons. OBJECTIVE: To observe expression changes of calbindin D28K in SN dopaminergic neurons during their degeneration and death in midbrain of PD model rats. DESIGN: Completely randomized grouping design. SETTING: Department of Neurobiology, Xuzhou Medical College. MATERIALS: A total of 92 healthy male SD rats, with the age of 3 months, weighing 200-250 g, were selected from Experimental Animal Center of Xuzhou Medical College [certification: SCXK (su) 2003-0003]. Calbindin D28K(CB), tyroxine hydroxylase (TH), ABC kit, 6-hydroxydopamine (6-OHDA) and Nissl dyes were provided by Sigma Company, and sheep serum was provided by Beijing Zhongshan Company. METHODS: The experiment was carried out in the Neurobiological Center of Xuzhou Medical College from October 2003 to October 2004. ① With lot method, rats were divided into blank control group (n=28), experimental control group (n=28) and experimental group (n=36). Rats in experimental group were injected with 6-OHDA at right corpus striatum for PD modeling; rats in experimental control group were injected with saline at the same site; rats in blank control group did not give any injections. ② On the 7th, 14th, 21st and 28th days, SN segments on right midbrain from every 5 rats in experimental group were fixed, embedded with paraffin and cut into successively coronary pieces. Rats in other two groups were treated with the same methods and then stained with Nissl to show neuronal form. Meanwhile, CB and TH antibodies staining with immunohistochemistry were used to show CB containing dopaminergic neurons and dopaminergic neurons, and cells were calculated and observed under optic microscope. ③ On the 14th and 28th days, every 4 rats in experimental group and every 4 rats in control group were selected to obtain their brains and separate SN on the injured side. Western blot was used to detect expression of calbindin D28K, protein band was scanned with imaging equipment, and data were analyzed with LabWorks software. MAIN OUTCOME MEASURES: ① On the 7th, 14th, 21st and 28th days, Nissl staining results of SN neurons and immunohistochemical staining results of CB and TH antibodies; ② On the 14th and 28th days, Western blot results of calbindin D28K in SN neurons. RESULTS: Among 92 rats, 2 rats in experimental group died after 1 day due to 6-OHDA injection and other 90 rats were involved in the final analysis. ① Nissl staining results: On the 7th day of 6-OHDA injection, most neuronal somas on right SN pars compacta were shown as deep pycnosis or lysis breakage; on the 14th and 21st days, amount of neurons was decreased remarkably; on the 28th day, most neurons in SN pars compacta disappeared. ② Results of immunohistochemical staining: Amount of positive neurons of calbindin D28K in right SN pars compacta was not changed on the 7th day after 6-OHDA injection; on the 14th day, the amount was higher in experimental group than that in both control groups (P < 0.01) and was decreased till the 21st day, but it was still higher than that in the two control groups (P < 0.05); on the 28th day, positive neurons of calbindin D28K nearly disappeared, and the amount was lower than that in the two control groups (P < 0.01). In addition, on the 7th day after 6-OHDA injection into corpus striatum, positive TH neurons decreased 24% in right SN, and there was significant difference from that in control groups; on the 14th, 21st and 28th days, positive TH neurons decreased 37%, 46% and 64%, respectively. ③ Western blot results: On the 14th day after 6-OHDA injection into corpus striatum, expression of calbindin D28K in right SN was higher in experimental group than that in both control groups (P < 0.05); however, on the 28th day, the expression was lower than that in the two control groups (P < 0.01). | 8 | Hydrogen peroxide and applications in green hydrocarbon nitridation and oxidation显示文摘Basic organic chemicals and high value–added products are mainly produced by hydrocarbon nitridation and oxidation.However,several drawbacks limit the application of the traditional oxidation and nitridation technologies in the future,such as complex processes,poor intrinsic safety,low atom utilization,and serious environmental pollution.The green nitridation and oxidation technologies are urgently needed.Hydrogen peroxide,a well–known green oxidant,is widely used in green hydrocarbon oxidation and nitridation.But its industrial production in China adopts fixed–bed technology,which is fall behind slurry–bed technology adopted by advanced foreign chemical companies,limiting the development of hydrogen peroxide industry and green hydrocarbon nitridation or oxidation industry.This article reviews the industrial production technologies of hydrogen peroxide and basic organic chemicals such as caprolactam,aniline,propene oxide,epichlorohydrin,phenol,and benzenediol,especially introduces the green production technologies of basic organic chemicals related with H_(2)O_(2).The article also emphasis on the efforts of Chinese researchers in developing its own slurry–bed technology of hydrogen peroxide production,and corresponding green hydrocarbon nitridation or oxidation technologies with hydrogen peroxide.Compared with traditional nitridation or oxidation technologies,green production technologies of caprolactam,propene oxide,epichlorohydrin,and benzenediol with hydrogen peroxide promote the nitrogen atom utilization from 60%to near 100%and the carbon atom utilization from 80%to near 100%.The waste emissions and environmental investments are reduced dramatically.Technological blockade against the green chemical industry of China are partially broken down,and technological upgrade in the chemical industry of China is guaranteed. | Yanqiang Shi Yuetong Xia Guangtong Xu Langyou Wen Guohua Gao Baoning Zong | 2022 | Chinese Journal of Chemical Engineering2022,35,1: | 2 |

| 11 | Multi-site anchoring of single-molecule for efficient and stable perovskite solar cells with lead shielding显示文摘The emergence of perovskite solar cells(PSCs) has greatly promoted the progress of photovoltaic technologies.The rapid development of PSCs has been driven by the advances in optimizing perovskite films and their adjacent interfaces.However,the polycrystalline perovskite layers in most highly efficient PSCs still contain various defects that greatly limit photovoltaic performance and stability of the devices.Herein,we introduce a multifunctional additive ethylene diamine tetra methylene phosphonic sodium(EDTMPS) with multiple anchor points into the precursor of perovskite to improve the efficiency and stability of PSCs and provide in-situ protection of lead leakage.The addition of EDTMPS acts as a crystal growth controller and passivation agent for perovskite films,thereby slowing down the crystallization rate of the film and obtaining high-quality perovskite films.Our study also provides an insight into how the modifier modulate the interfacial energy level arrangement as well as affect transfer of charge carriers and their recombination under photoinduced excitation.As a result,the power conversion efficiency(PCE) of single subcell with a working area of 0.255 cm^(2) increases significantly from 20.03% to 23.37%.Moreover,we obtained a PCE of 19.16% for the 25 cm^(2) module.Importantly,the unencapsulated EDTMP-modified PSCs exhibit better operational and thermal stability,as well as in-situ absorption of leaked lead ions. | Zhengyan He Shufang Zhang Yushuang Gao Quanming Geng Xiangrui Jia Shuo Yang Zhiqiang Zhang Yanpu Zheng Yanqiang Hu Changlin Yao Qi Zhang | 2023 | Journal of Energy Chemistry2023,,12: | 0 |
| 12 | Skincare plants of the Naxi of NW Yunnan, China显示文摘The Naxi of Northwest Yunnan,China use medicinal plants to treat skin conditions related to traditional lifestyles in extreme environments.However,modernization endangers both the medicinal plants used to treat skin conditions and traditional knowledge.Therefore,investigation and documentation of the medicinal plants used and associated traditional knowledge is necessary.In this study,we conducted an ethnobotanical survey in 12 Naxi communities in Northwest Yunnan.For this purpose,we used semistructured surveys to interview 840 informants from Naxi communities.We used informant consensus factor and use frequency as quantitative indices to evaluate the importance of medicinal plant species.A total of 161 medicinal plant species belonging to 69 families were documented.The highest informant consensus factor(ICF)values were recorded for skin nourishing(ICF?0.849),frostbite and chapped skin(ICF?0.833).These skin treatments are highly related to the environment and lifestyle of Naxi communities.The main active compounds of plants used to treat skin conditions in Naxi communities are known to have skin-treating properties.This study reveals that the skin conditions treated by the Naxi are associated with traditional medicine culture and social economic development.In addition,this study uses ethnobotanical indices to explain how skin condition treatments are linked to the natural environment of Naxi communities. | Yanqiang Zhao Zexing Yang Bayi Lang Manfred Shao Wu Meng Dayuan Xue Lu Gao Lixin Yang | 2020 | Plant Diversity2020,42,6: | 0 |
