1SlMYB75,an MYB-type transcription factor,promotes anthocyanin accumulation and enhances volatile aroma production in tomato fruits显示文摘Genetic manipulation of genes to upregulate specific branches of metabolic pathways is a method that is commonly used to improve fruit quality.However,the use of a single gene to impact several metabolic pathways is difficult.Here,we show that overexpression of the single gene SlMYB75(SlMYB75-OE)is effective at improving multiple fruit quality traits.In these engineered fruits,the anthocyanin content reached 1.86mg g−1 fresh weight at the red-ripe stage,and these SlMYB75-OE tomatoes displayed a series of physiological changes,including delayed ripening and increased ethylene production.In addition to anthocyanin,the total contents of phenolics,flavonoids and soluble solids in SlMYB75-OE fruits were enhanced by 2.6,4,and 1.2 times,respectively,compared to those of wild-type(WT)fruits.Interestingly,a number of aroma volatiles,such as aldehyde,phenylpropanoid-derived and terpene volatiles,were significantly increased in SlMYB75-OE fruits,with some terpene volatiles showing more than 10 times higher levels than those in WT fruits.Consistent with the metabolic assessment,transcriptomic profiling indicated that the genes involved in the ethylene signaling,phenylpropanoid and isoprenoid pathways were greatly upregulated in SlMYB75-OE fruits.Yeast one-hybrid and transactivation assays revealed that SlMYB75 is able to directly bind to the MYBPLANT and MYBPZM cis-regulatory elements and to activate the promoters of the LOXC,AADC2 and TPS genes.The identification of SlMYB75 as a key regulator of fruit quality attributes through the transcriptional regulation of downstream genes involved in several metabolic pathways opens new avenues towards engineering fruits with a higher sensory and nutritional quality.Wei Jian Haohao Cao Shu Yuan Yudong Liu Juanfang Lu Wang Lu Ning Li Jianhui Wang Jian Zou Ning Tang Chan Xu Yulin Cheng Yanqiang Gao Wanpeng Xi Mondher Bouzayen Zhengguo Li 2019Horticulture Research2019,6,1:39
2Establishment of intestinal organoid cultures modeling injury-associated epithelial regeneration显示文摘The capacity of 3D organoids to mimic physiological tissue organization and functionality has provided an invaluable tool to model development and disease in vitro.However,conventional organoid cultures primarily represent the homeostasis of self-organizing stem cells and their derivatives.Here,we established a novel intestinal organoid culture system composed of 8 components,mainly including VPA,EPZ6438,LDN193189,and R-Spondin 1 conditioned medium,which mimics the gut epithelium regeneration that produces hyperplastic crypts following injury;therefore,these organoids were designated hyperplastic intestinal organoids(Hyper-organoids).Single-cell RNA sequencing identified different regenerative stem cell populations in our Hyper-organoids that shared molecular features with in vivo injury-responsive Lgr5+stem cells or Clu+revival stem cells.Further analysis revealed that VPA and EPZ6438 were indispensable for epigenome reprogramming and regeneration in Hyper-organoids,which functioned through epigenetically regulating YAP signaling.Furthermore,VPA and EPZ6438 synergistically promoted regenerative response in gut upon damage in vivo.In summary,our results demonstrated a new in vitro organoid model to study epithelial regeneration,highlighting the importance of epigenetic reprogramming that pioneers tissue repair.Molong Qu Liang Xiong Yulin Lyu Xiannian Zhang Jie Shen Jingyang Guan Peiyuan Chai Zhongqing Lin Boyao Nie Cheng Li Jun Xu Hongkui Deng 2021Cell Research2021,31,3:9
3A framework for equipment systems-of-systems effectiveness evaluation using parallel experiments approach显示文摘Equipment systems-of-systems(So S) effectiveness evaluation can provide important reference for construction and optimization of the equipment So S. After discussing the basic theory and methods of parallel experiments, we depict an So S effectiveness analysis and evaluation method using parallel experiments theory in detail. A case study is carried out which takes the missile defense system as an example. An artificial system of the missile defense system is constructed with the multi-agent modeling method. Then, single factor, multiple factors and defense position deployment computational experiments are carried out and evaluated with the statistical analysis method. Experiment results show that the altitude of the secondary interception missile is not the key factor which affects So S effectiveness and putting the defense position ahead will increase defense effectiveness. 4Zanthoxylum-specific whole genome duplication and recent activity of transposable elements in the highly repetitive paleotetraploid Z.bungeanum genome显示文摘Zanthoxylum bungeanum is an important spice and medicinal plant that is unique for its accumulation of abundant secondary metabolites,which create a characteristic aroma and tingling sensation in the mouth.Owing to the high proportion of repetitive sequences,high heterozygosity,and increased chromosome number of Z.bungeanum,the assembly of its chromosomal pseudomolecules is extremely challenging.Here,we present a genome sequence for Z.bungeanum,with a dramatically expanded size of 4.23 Gb,assembled into 68 chromosomes.This genome is approximately tenfold larger than that of its close relative Citrus sinensis.After the divergence of Zanthoxylum and Citrus,the lineage-specific whole-genome duplication event q-WGD approximately 26.8 million years ago(MYA)and the recent transposable element(TE)burst~6.41 MYA account for the substantial genome expansion in Z.bungeanum.The independent Zanthoxylum-specific WGD event was followed by numerous fusion/fission events that shaped the genomic architecture.Integrative genomic and transcriptomic analyses suggested that prominent speciesspecific gene family expansions and changes in gene expression have shaped the biosynthesis of sanshools,terpenoids,and anthocyanins,which contribute to the special flavor and appearance of Z.bungeanum.In summary,the reference genome provides a valuable model for studying the impact of WGDs with recent TE activity on gene gain and loss and genome reconstruction and provides resources to accelerate Zanthoxylum improvement.Shijing Feng Zhenshan Liu Jian Cheng Zihe Li Lu Tian Min Liu Tuxi Yang Yulin Liu Yonghong Liu He Dai Zujun Yang Qing Zhang Gang Wang Jisen Zhang Huifeng Jiang Anzhi Wei 2021Horticulture Research2021,8,1:5
5Multi-agent decision support system for missile defense based on improved PSO algorithm显示文摘Ballistic missile defense system(BMDS) is important for its special role in ensuring national security and maintaining strategic balance. Research on modeling and simulation of the BMDS beforehand is essential as developing a real one requires lots of manpower and resources. BMDS is a typical complex system for its nonlinear, adaptive and uncertainty characteristics.The agent-based modeling method is well suited for the complex system whose overall behaviors are determined by interactions among individual elements. A multi-agent decision support system(DSS), which includes missile agent, radar agent and command center agent, is established based on the studies of structure and function of BMDS. Considering the constraints brought by radar,intercept missile, offensive missile and commander, the objective function of DSS is established. In order to dynamically generate the optimal interception plan, the variable neighborhood negative selection particle swarm optimization(VNNSPSO) algorithm is proposed to support the decision making of DSS. The proposed algorithm is compared with the standard PSO, constriction factor PSO(CFPSO), inertia weight linear decrease PSO(LDPSO),variable neighborhood PSO(VNPSO) algorithm from the aspects of convergence rate, iteration number, average fitness value and standard deviation. The simulation results verify the efficiency of the proposed algorithm. 7Comprehensive metabolic profiling of Alismatis Rhizoma triterpenes in rats based on characteristic ions and a triterpene database显示文摘Alismatis Rhizoma(AR)is widely used in Chinese medicine,and its major bioactive components,triterpenes,reportedly possess various pharmacological activities.Therefore,it is very important to study the metabolism of triterpenes in vivo.However,the metabolism of AR triterpene extract has not been comprehensively elucidated due to its complex chemical components and metabolic pathways.In this study,an ultra-performance liquid chromatography quadrupole time-of-flight mass spectrometry method,which was based on the characteristic ions from an established database of known triterpenes,was used to analyze the major metabolites in rats following the oral administration of Alismatis Rhizoma extracts(ARE).As a result,a total of 233 constituents,with 85 prototype compounds and 148 metabolites,were identified for the first time.Hydrogenation,oxidation,sulfate and glucuronidation conjugation were the major metabolic pathways for triterpenes in AR.In addition,the mutual in vivo transformation of known ARE triterpenes was discovered and confirmed for the first time.Those results provide comprehensive insights into the metabolism of AR in vivo,which will be useful for future studies on its pharmacodynamics and pharmacokinetics.Moreover,this established strategy may be useful in metabolic studies of similar compounds.Lu Wang Sen Li Jiaxin Li Zhongzhe Cheng Yulin Feng Hui Ouyang Zhifeng Du Hongliang Jiang 2021Journal of Pharmaceutical Analysis2021,11,1:2

18New insights in the battle between wheat and Puccinia striiformis显示文摘Wheat stripe rust caused by Puccinia striiformis f.sp.tritici(Pst)poses a great threat to wheat production worldwide.The rapid change in virulence of Pst leads to a loss of resistance in currently resistant wheat cultivars,which results in frequent disease epidemics.Therefore,a major focus is currently placed on investigating the molecular mechanisms underlying this rapid variation of pathogenicity and coevolving wheat resistance.Limited by the lack of a system for stable transformation of Pst and the difficulties in wheat transformation,it is not easy to generate deeper insights into the wheat-Pst interaction using established genetic methods.Nevertheless,considerable effort has been made to unravel the wheat-Pst interaction and significant progress is being made.Histology and cytology have revealed basic details of infection strategies and defense responses during wheat-Pst interactions,identified cellular components involved in wheat-Pst interactions,and have helped to elucidate their role in the infection process or in plant defense responses.Transcriptome and genome sequencing has revealed the molecular features and dynamics of the wheat-Pst pathosystem.Extensive molecular analyses have led to the identification of major components in the wheat resistance response and in Pst virulence.Studies of wheat-Pst interactions have now entered a new phase in which cellular and molecular approaches are being used.This review focuses on the cellular biology of wheat-Pst interactions and integrates the emerging data from molecular analyses with the histocytological observations.Chunlei TANG Xiaojie WANG Yulin CHENG Minjie LIU Mengxin ZHAO Jinping WEI Zhensheng KANG 2015Frontiers of Agricultural Science and Engineering2015,2,2:1
