1PINK1 kinase dysfunction triggers neurodegeneration in the primate brain without impacting mitochondrial homeostasis显示文摘In vitro studies have established the prevalent theory that the mitochondrial kinase PINK1 protects neurodegeneration by removing damaged mitochondria in Parkinson's disease(PD).However,difficulty in detecting endogenous PINK1 protein in rodent brains and cell lines has prevented the rigorous investigation of the in vivo role of PINK1.Here we report that PINK1 kinase form is selectively expressed in the human and monkey brains.CRISPR/Cas9-mediated deficiency of PINK1 causes similar neurodegeneration in the brains of fetal and adult monkeys as well as cultured monkey neurons without affecting mitochondrial protein expression and morphology.Importantly,PINK1 mutations in the primate brain and human cells reduce protein phosphorylation that is important for neuronal function and survival.Our findings suggest that PINK1 kinase activity rather than its mitochondrial function is essential for the neuronal survival in the primate brains and that its kinase dysfunction could be involved in the pathogenesis of PD.Weili Yang Xiangyu Guo Zhuchi Tu Xiusheng Chen Rui Han Yanting Liu Sen Yan Qi Wang Zhifu Wang Xianxian Zhao Yunpeng Zhang Xin Xiong Huiming Yang Peng Yin Huida Wan Xingxing Chen Jifeng Guo Xiao-Xin Yan Lujian Liao Shihua Li Xiao-Jiang Li 2022Protein & Cell2022,13,1:3
2MXene based mechanically and electrically enhanced film for triboelectric nanogenerator显示文摘The development of triboelectric nanogenerator(TENG)technology which can directly convert ambient mechanical energy into electric energy may affect areas from green energy harvesting to emerging wearing electronics.And,the material of triboelectric layer is critical to the mechanical robustness and electrical output characteristics of the TENGs.Herein,a MXene enhanced electret polytetrafluoroethylene(PTFE)film with a high mechanical property and surface charge density is developed.The MXene/PTFE composite film was synthesized by spraying and annealing treatment.With the doping of MXene,the crystallinity of composite film could be tuned,leading to an enhancement in the tensile property of 450%and reducing the wear volume about 80%in the friction test.Furthermore,the as-fabricated TENG with this composite film outputs 397 V of open-circuit voltage,21µA of short-circuit current,and 232 nC of transfer charge quantity,which are 4,6,and 6 times higher than that of the TENG made by pure PTFE film,respectively.Therefore,this work provides a creative strategy to simultaneously improve the mechanical property and electrical performance of the TENGs,which have great potential in improving device stability under a complex mechanical environment.Yuyu Gao Guoxu Liu Tianzhao Bu Yaoyao Liu Youchao Qi Yanting Xie Shaohang Xu Weili Deng Weiqing Yang Chi Zhang 2021Nano Research2021,14,12:2
3Distribution of aromatic amines,phenols,chlorobenzenes,and naphthalenes in the surface sediment of the Dianchi Lake,China显示文摘Organic pollutants are widespread environmental pollutants with high toxicity,persistence,and bioaccumulation.Our aim was to investigate the distribution of aromatic amines,phenols,chlorobenzenes,and naphthalenes in the surface sediment of the Dianchi Lake,China.Nineteen surface sediment samples were collected from the Dianchi Lake,and 40 types of organic pollutants were analyzed via gas chromatography-mass spectrometry.The total organic pollutant concentrations in the surface sediment of the Dianchi Lake varied from 27.4 to 1.62×10^3 ng/g.The concentrations of phenols were much higher than those in other water bodies but still within a controllable range,whereas the concentrations of the other organic pollutant classes were similar or even lower.The detection ratio of 3-or 4-methylphenol was the highest(100.00%)among the pollutants.The average total organic pollutant concentrations decreased in the following order:Caohai(540 ng/g)>the middle of Waihai(488 ng/g)>the edge of Waihai(351 ng/g)>Haigeng Dam(90.4 ng/g).Pearson analysis showed a strong correlation among 1-methylnaphthalene,2-methylnaphthalene,1,3-dinitronaphthalene,and 1,4-dinitronaphthalene(P<0.01).Caohai,the north lakeshore of Waihai and the south of Waihai showed higher risk because of high concentration;meanwhile,1,4-dichlorobenzene,3-or 4-methylphenol and 1,2,4-trichlorobenzene were mor e likely to cause risks.Xinyi Hu Ting Yang Chen Liu Jun Jin Bingli Gao Xuejun Wang Min Qi Baokai Wei Yuyu Zhan Tan Chen Hongtao Wang Yanting Liu Dongrui Bai Zhu Rao Nan Zhan 2020Frontiers of Environmental Science & Engineering2020,14,4:1

6Type I photosensitizer based on AIE chromophore tricyano-methylene-pyridine for photodynamic therapy显示文摘Image guided photodynamic therapy(PDT)combines fluorescence tracing and phototherapy,which can achieve a more accurate and effective treatment effect.However,traditional photosensitizers are limited by the aggregation-caused fluorescence quenching(ACQ)effect and low reactive oxygen species(ROS)generation in a hypoxic environment,resulting in poor imaging and treatment effect.Herein,we report a tricyano-methylene-pyridine(TCM)-based Type I aggregation-induced emission(AIE)photosensitizer(TCM-MBP),the strong elec-tron acceptance(D-A)effect extends the wavelength to near-infrared(NIR)region to reduce the autofluorescence interference,and oxygen atoms provide lone pair electrons to enhance the inter system crossing(ISC)rate,thereby promoting the generation of more triplet states to produce ROS.The AIE photosensitizer TCM-MBP exhibited low oxygen dependence,NIR emission,and higher ROS production compared to commercially avail-able Ce_(6) and RB.After encapsulation with DSPE-PEG 2000,TCM-MBP nanoparticles(TCM-MBP NPs)could penetrate to visualize cells and efficiently kill cancer cells upon light irradiation.This study provides an oxygen-independent AIE photosensitizer,which has great potential to replace the commercial ACQ photosensitizers.Chao Pan Weijun Zhao Xiaolei Zhao Zhenxing Liu Xiangyu Li Yanting Lyu Xupeng Wu Zhirong Zhu Wei-Hong Zhu Qi Wang 2023Green Chemical Engineering2023,4,3:0

9Discovery of a subtype-selective, covalent inhibitor against palmitoylation pocket of TEAD3显示文摘The TEA domain(TEAD)family proteins(TEAD1-4)are essential transcription factors that control cell differentiation and organ size in the Hippo pathway.Although the sequences and structures of TEAD family proteins are highly conserved,each TEAD isoform has unique physiological and pathological functions.Therefore,the development and discovery of subtype selective inhibitors for TEAD protein will provide important chemical probes for the TEAD-related function studies in development and diseases.Here,we identified a novel TEAD 1/3 covalent inhibitor(DC-TEADin1072)with biochemical IC50 values of 0.61±0.02 and 0.58±0.12μmol/L against TEAD1 and TEAD3,respectively.Further chemical optimization based on DC-TEAD in 1072 yielded a selective TEAD3 inhibitor DCTEAD3 in03 with the IC_(50) value of 0.16±0.03μmol/L,which shows 100-fold selectivity over other TEAD isoforms in activity-based protein profiling(ABPP)assays.In cells,DC-TEAD3 in03 showed selective inhibitory effect on TEAD3 in GAL4-TEAD(1-4)reporter assays with the IC50 value of1.15μmol/L.When administered to zebrafish juveniles,experiments showed that DC-TEAD3 in03 reduced the growth rate of zebrafish caudal fins,indicating the importance of TEAD3 activity in controlling proportional growth of vertebrate appendages.Tian Lu Yong Li Wenchao Lu TWGM Spitters Xueyu Fang Jun Wang Simian Cai Jing Gao Yanting Zhou Zhe Duan Huan Xiong Liping Liu Qi Li Hualiang Jiang Kaixian Chen Hu Zhou Hua Lin Huijin Feng Bing Zhou Christopher L.Antos Cheng Luo 2021Acta Pharmaceutica Sinica B2021,11,10:0
